Abstract
본 발명에 따른 VSIG4 단백질 (VSIG4.Ig)을 주사하면 간염의 발생을 예방할 뿐 아니라, 이미 형성된 간염의 경우에도 치료 효과가 있기 때문에 VSIG4를 간염 치료의 표적으로 사용하여 간염 특히, 자가면역 간염의 치료 및 예방에 세포 치료제 또는 단백질 치료제의 형태로 유용하게 사용할 수 있다.
Description
도 2는 Balb/c WT T-cells (105) 에 대한 세포 주기 분석 결과이다(R2, Sub-G0 phase; R3, G0/G1 phase; R4, S-phase; R5, G2/M phase).
도 3은 ConA 처리 2시간 전 B6 WT 마우스에 대한 가용 VSIG4.Ig 의 in vivo 처리 결과이다. 간 MNC는 세포내 IFN-γ, TNF-α 및 IL-17A 측면에서 조사되었다.
도 4는 는 VSIG4.Ig (400 μg/mouse)의 in vivo 주입으로 치사량(30 mg/kg)의 ConA의 처리로 인해 유발된 급성 간염에 대한 마우스의 생존률 분석 결과이다.
도 5는 30 mg/kg의 ConA 처리 시 VSIG4.Ig 전처리에 따른 헤마톡실린-에오신 염색된 마우스 간조직의 현미경 관찰 결과이다.
도 6은 치사량의 ConA 처리 3시간 후 VSIG4.Ig (400 μg/mouse)를 주입하여 검토한 ConA-유발 간 손상(면역 매개 간 손상)에서 VSIG4.Ig의 치료효과 검토 결과이다.
Claims (9)
- VSIG4 단백질 또는 이의 분절, 이의 활성화제 및 이의 발현조절제로 이루어진 군에서 선택된 어느 하나 또는 둘 이상의 단백질과 약제학적으로 허용가능한 담체를 포함하는 간염 예방 또는 치료용 약학조성물.
- 제1항에 있어서,
상기 VSIG4 단백질 분절은 VSIG4 단백질의 세포외 도메인 중 41 내지 113번의 아미노산 서열을 포함하는 것을 특징으로 하는 간염 예방 또는 치료용 약학조성물. - 제2항에 있어서,
상기 VSIG4 단백질 분절은 인간 항체의 Fc 부위와 융합시킨 재조합단백질 형태로 사용하는 것을 특징으로 하는 간염 예방 또는 치료용 약학조성물. - 제1항 내지 제3항 중 어느 한 항에 있어서,
상기 간염은 자가면역 간염인 것을 특징으로 하는 간염 예방 또는 치료용 약학조성물. - VSIG4-인코딩 유전자와 약제학적으로 허용가능한 담체를 포함하는 간염 예방 또는 치료용 약학조성물.
- 제5항에 있어서,
상기 VSIG4-인코딩 유전자는 바이러스성 또는 비-바이러스성 벡터를 사용한 것을 특징으로 하는 간염 예방 또는 치료용 약학조성물. - 제6항에 있어서,
상기 바이러스성 벡터는 아데노바이러스, 아데노-부속 바이러스, 헬퍼-의존형 아데노바이러스 및 레트로바이러스 벡터로 이루어진 군에서 선택된 것을 특징으로 하는 간염 예방 또는 치료용 약학조성물. - 간염을 예방하거나 치료할 필요가 있는 개체로부터 얻어진 생물학적 샘플에 임의의 화합물을 처리하는 단계; 및 VSIG4 단백질의 발현 수준을 확인하는 단계를 포함하는 간염 치료제의 스크리닝 방법.
- 간염을 예방하거나 치료할 필요가 있는 개체로부터 얻어진 생물학적 샘플에서 VSIG4의 발현 프로파일을 검출하는 단계; 및 상기 발현 프로파일을 정상대조군에서 VSIG4의 발현 프로파일과 비교하여 간염 여부를 판정하는 단계를 포함하는 간염의 진단을 위한 정보를 제공하는 방법.
