Abstract

PURPOSE: An anti-acne composition containing a germinated soybean extract and lactic acid bacteria which suppresses Propionibacterium acnes is provided to specifically suppress P. acnes which cause acne, to ensure antibacterial action against P. acne and harmful bacteria, and to prevent and treat acne diseases. CONSTITUTION: An anti-acne composition contains a germinated soybean extract and lactic acid bacteria which suppress Propionibacterium acnes. The extract is prepared using water, C1-C5 alcohols, propylene glycol, 1,3-butylene glycol, or a mixture thereof. 정상적인 섭식 습관을 갖는 20-30세의 건강한 성인 15명으로부터 분변을 수집하였다. 이후, BBL의 혐기성 샘플 컬렉션 & 트랜스포트 시스템(BBL's anerobic sample collection & transport system)을 이용하여 편성 혐기성(obligate anaerobic) 유산균을 분리하였다.Fecals were collected from 15 healthy adults aged 20-30 with normal eating habits. 수집된 인분의 적당량을 생리식염수 100 ㎖에 현탁한 후, 적당량을 취하여 5% 양 혈청이 함유된 BL 아가 배지(Blood-Liver, Nissue)에 도말하였다. 박트론 혐기 챔버(Bactron Anaerobic chamber; Sheldon Manufacturing Inc.)를 이용하여 37?에서 48시간 동안 배양하였다. 생존한 균주 중에서 0.5-2.0 mm 의 적갈색 중심 부분을 갖는 밝은 갈색, 유백색 또는 황갈색 형태의 비피도박테리움 속 미생물을 선별하였다. 선별된 유산균은 총 3 종이며, 이들을 각각 SPM 0308, SPM1005, SPM1207로 명명하였다.

배양된 P. acnes KCTC 3320을 서스펜션 형식으로 준비하고, 여기에 상기 분리된 유산균 SPM 0308, SPM1005, SPM1207을 1:1 비율로 혼합하고 8ml RC 배지에 첨가한 후 10초간 보텍스 믹스하였다. 그 후 무산소 환경(90% N2, 5% H2, 5% CO2) 하 120분간 37 ℃에서 배양시켰다. 대조군으로 P. acnes 서스펜션이 같은 양의 PBS와 혼합되었다. 배양 후, 각 서스펜션에 있는 P. acnes CFU가 평판계수법(plate count technique)으로 측정되었다. 그리고, P. acnes 생존률은 아래와 같은 식을 사용하여 측정하였다.

결과는 평균± 표준편차로 표현되었다. 자료의 통계적 평가를 위해, SPSS 15.0를 사용하여 one-way ANOVA가 적용되었다. 그 후 터키 검정법을 이용해 사후 검정 비교하였으며, 그 결과를 도 1에 나타내었다. 도 1에서 보는 바와 같이 SPM0308 에 의한 항균 효과와 대조군 및 다른 유산균 SPM1005, SPM1207 에 의한 항균 효과의 차이에 대한 통계처리 결과 p<0.05으로 SPM0308 에 의한 항균 효과와 대조군 및 다른 유산균에 의한 항균 효과의 차이가 매우 크다는 것을 알 수 있다.
