Abstract
본 발명에 따른 혐기성 배양 배지 제조용 산소제거 및 가스혼합 분배장치를 사용하면 기존의 혐기성 배양배지 제조장치의 산소제거컬럼의 막힘현상을 개선하고, 가스압을 일정하게 유지할 수 있으며, 나동선의 색깔변화로 쉽게 컬럼의 재활성화 여부를 파악할 수 있다. 또한 혐기챔버와 산소제거컬럼을 연결하여 저가, 저순도의 가스를 산소가 완벽하게 제거된 고순도의 가스로 전환하여 혐기챔버에 공급함으로써, 절대 혐기성 미생물의 고체배지 배양을 가능하게 하고 knock-out mutant의 제조를 가능하게 한다.
Description
도 2는 산소가 오염되어 산소제거컬럼이 어두운 색깔의 산화구리를 형성하고 있는 모습과 수소가스를 이용한 산소제거로 밝은 구리 색깔의 활성화된 산소제거컬럼의 모습을 나타낸 것이다.
도 3은 절대혐기배지제조 장치를 이용한 배지를 제조하는 모습을 나타낸 것이다.
도 4는 국내에서 배양이 되지 못했던 Methanobrevibacter 속의 메탄생성 고세균 배양을 확인한 결과를 나타낸 것이다.
도 5는 산소제거컬럼이 혐기챔버에 연결되어 저순도, 저가의 가스를 이용하여 산소가 완전히 제거된 고순도의 가스를 혐기챔버 내에 공급하고 이를 통해 완벽하게 혐기상태를 유지하는 혐기챔버를 나타낸 것이다.

Sludge fluid (슬러지 용액) |
0.5% yeast extract를 폐수처리 혐기조에서 구한 슬러지에 첨가하고 질소가스를 10분간 주입하여 37℃에서 24시간 배양한다. 슬러지를 13,000g에서 원심분리하여 상등액만을 취한뒤 살균한뒤 혐기상태의 병에 질소가스를 충진하여 보존. 슬러지용액은 상온에서 어두운 장소에 보관함. |

Claims (5)
- 나동선으로 구성된 산소제거 칼럼과 육안으로 나동선의 색변화를 통해 칼럼의 재생여부 확인이 가능하고 칼럼을 재생하여 사용할 수 있는 것을 특징으로 하는 혐기성 세균 배양용 산소제거 및 가스혼합 분배장치.
- 제1항에 있어서, 혐기성 세균은 난배양성 절대혐기 고세균인 것을 특징으로하는 산소제거 및 가스혼합 분배장치.
- 제2항에 있어서, 난배양성 절대혐기 고세균은 매탄생성 고세균인 Methanobrevibacter 속 Methanobacterium 속, Geobacter 속 및 Clostridium 속으로 구성된 군에서 선택되는 것을 특징으로 하는 산소제거 및 가스혼합 분배장치.
- 제1항의 혐기성 세균 배양용 산소제거 및 가스혼합 분배장치가 연결되어 구동되는 것을 특징으로 하는 혐기챔버.
- 제1항의 산소제거 및 가스혼합 분배장치를 이용하는 것을 특징으로 하는 혐기배지의 제조방법.
