<실험예 1> 괭생이모자반 추출물의 콜라겐 전사인자 (COL1A1 promotor)의 증가효과Experimental Example 1 Increase of Collagen Transcription Factor (COL1A1 Promotor)
상기의 방법으로 제조한 실시예 1의 괭생이모자반 추출물의 콜라겐 전사인자의 증가 효과를 확인하기 위해 luciferase가 결합되어 있는 plasmid 를 사용한 reporter gene assay를 수행하였다. 간단히 설명하면, NIH-3T3 세포주를 24웰 플레이트에 접종한 후 10% 우태아혈청과 1% 페니실린/스트렙토마이신을 함유한 DMEM 배지에서 24시간 동안 배양한다. 이후 전사인자가 활성화되면 이것을 luciferase로 보고할 수 있도록 제작된 plasmid인 COLlA1 promoter-luciferase reporter plasmid DNA (pGL3-Basic, Promega)를 Lipofectamin transfection 시약(Invitrogen사 제품)으로 transfection 하였다. Transfection 24시간 후에 괭생이모자반 추출물을 50㎍/ml 농도로 처리한 후 36시간 후에 세포를 lysis 시키고 luciferase활성을 luminometer로 측정하여 COL1A1 promoter 전사인자활성을 측정하였다. NIH-3T3를 105개의 농도로 6웰 플레이트에 배양하고, 24시간 후 해조류 추출물을 36시간 처리하였다. 해조류추출물 처리 36시간 후 세포를 모아서 역전사PCR(RT-PCR)을 수행하여, 전사된 COL1A1의 mRNA를 측정하여 정량하였다. NIH-3T3세포를 3x 104개의 농도로 24 웰 플레이트에 배양하고, 24시간 후, 추출물을 처리하여 36시간 배양하였다. 추출물 처리 36시간 후 세포를 PBS로 세척하고 계면활성제와 프로테아제 저해제를 처리하여 세포균등액을 얻었다. 세포균등액을 전기영동하여 nitrocellulose membrane으로 trasfer하고 collgen에 대한 1차 항체와 peroxidase가 결합된 2차항체로 immunbloting을 시행하여 콜라겐단백질 발현량을 측정하였다.NIH-3T3 cells were cultured in 24 well plates at a concentration of 3 × 10 4 , and after 24 hours, the extracts were treated and cultured for 36 hours. After 36 hours of extract treatment, cells were washed with PBS and treated with a surfactant and a protease inhibitor to obtain a cell homogenate. The cell homogenate was electrophoresed and trasfered to nitrocellulose membrane, and the expression level of collagen protein was measured by immunbloting with secondary antibody combined with primary antibody and peroxidase against collgen.
상기에서 살펴본 바와 같이 물, 알코올 또는 이들의 혼합물로 추출되는 괭생이모자반 추출물은, 콜라겐 전사인자 (COL1A1 promotor)의 전사인자의 활성을 증가시키고, 콜라겐 유전자 COL1A1의 mRNA를 증가시키며, 피부세포에서 콜라겐 생합성을 증가시켜 피부의 주 구성 성분인 콜라겐을 증가시킴으로써 피부의 염증억제, 주름 개선, 탄력 개선의 효과가 있으므로 화장품 등의 피부외용제로 사용될 수 있다.As described above, the extract of boksan motzaban, which is extracted with water, alcohol or a mixture thereof, increases the activity of the transcription factor of the collagen transcription factor (COL1A1 promotor), increases the mRNA of the collagen gene COL1A1, and collagen in skin cells.
