Abstract
본 발명은 금속성분의 동위원소의 양을 측정하는 방식에 의한 암의 존재 유무를 판단하는 방법에 의한 암의 진단에 관한 것으로, 암세포 조직 및 혈액에서 동위원소의 양을 측정하는 방법에 의하여 암조직의 존재 여부 및 암종의 종류와 관련된 진단을 가능하게 하는 것을 그 특징으로 하는 동위원소의 측정에 의한 인체 중의 암진단법에 관한 것이다.

Description
본 발명은 암의 진단에 관한 것으로 혈액 또는 조직 중의 동위원소의 양을 측정하여 비교 분석함으로써 암의 종류 및 유무를 판단하는 방법을 제공하는 것으로 극미량의 분석에 의하여 미세한 종양의 유무를 판단할 수 있게 하며, 단순하게 혈액 또는 조직의 분석을 이용하여 정확한 진단을 하게 함을 특징으로 한다.
동위원소는 아이소토프(isotope) 또는 동위체(同位體)라고도 한다. 화학원소는 서로 같아 화학적으로 거의 구별할 수가 없으나, 그것을 구성하고 있는 원자의 질량이 서로 다른 것을 동위원소라고 한다. 영어의 isotope는 그리스어인 isos(같은)와 topos(장소)의 합성어인데, 질량은 서로 달라도 원소의 주기율표에서 같은 장소에 배열되는 데서 1901년 영국의 화학자 F.소디가 그 개념을 확립시킴과 동시에 이 명칭을 붙였다고 한다.
일반적으로 어떤 원소의 화학적 성질은 그 원소를 구성하고 있는 원자의 원 자핵 내에 있는 양성자의 수, 즉 원자번호에 의해 결정되며 한편 원자의 질량은 양성자와 중성자의 수의 합, 즉 질량수에 거의 비례하므로, 동위원소란 같은 수의 양성자를 가지고 중성자의 수만이 다른 원자핵으로 이루어지는 원소들이라고 할 수 있다.
예를 들면, 자연계에 존재하는 산소는 대부분이 8개의 양성자와 8개의 중성자를 가지는 질량수 16인 원자핵으로 이루어져 있으나, 이 밖에 얼마 안되지만 9개의 중성자를 가지는 질량수 17인 것과 10개의 중성자를 가지는 질량수 18인 것이 혼재하고 있다. 마찬가지로 질소에는 11과 15의 질량수를 갖는 2종의 동위원소가 있고, 우라늄도 234, 235, 238의 질량수를 갖는 3종의 동위원소의 혼합물이 일체가 된 것으로서 자연계에 존재한다.
원자핵의 바깥궤도를 도는 전자(電子)의 수는 원자번호와 같으므로 동위원소는 모두 같은 수의 궤도전자를 가진다. 천연으로 존재하는 화학원소의 종류는 약 90종이며, 이에 대하여 천연의 동위원소는 약 300종이나 된다. 따라서 이것을 평균해 보면, 한 원소당 3종의 동위원소를 갖는 것이 되지만, 실제로는 주석(10개), 카드뮴(8개)과 같이 많은 동위원소를 갖는 것도 있고, 또 베릴륨 ·플루오르 ·나트륨 ·비스무트와 같이 천연으로는 동위원소가 없고 단 1종의 원자로 이루어져 있는 것도 있다.
일반적으로 어떤 천연원소가 얼마나 많은 동위원소를 가지고 있는가에 대해서는 뚜렷한 법칙성은 없지만, 원자번호가 홀수인 원소는 대부분 2종 이상의 동위원소를 갖지 않으며, 원자번호가 짝수인 원소는 비교적 많은 동위원소를 갖는 것이 많다는 사실이 인정된다. 자연계에 존재하는 원소는 이와 같은 동위원소의 혼합물인데, 그 혼합비는 지구상의 어느 곳에서 채취한 시료에 대해서도 거의 일정하다.
보통 원소의 원자량은 이들 혼합물을 일체로 간주했을 때의 평균적인 질량을 나타내는 것이며, 본래는 정수에 아주 가까운 값을 가져야 할 원자량이 정수가 아닌 중간적인 값을 갖는 일이 많은 것은 원소가 몇 개의 동위원소의 집합체이기 때문이다. 보통 이들 동위원소를 각각 다른 종류의 것(이것을 核種이라고 한다)으로서 나타낼 때는 산소 16 16O, 질소 14 14N, 우라늄 235 235U 등, 그것이 속해 있는 원소명에 질량수를 첨가하는 방법이 사용되고 있는데 그 중에는 수소와 같이 특별한 명칭(경수소 ·중수소 ·삼중수소)을 사용하는 것도 있다.
최근의 연구에 의하면 산소의 동위원소인 18O이 생물체에 독성을 보인다고 보고되고 있다. 수소의 동위원소인 중수소, 2H는 D2O의 형태로 미생물에 대하여 92%의 억제효과를 보였으며 생쥐에 있어 5일내 99.5%가 사망에 이르게 하는 결과를 보인다고 보고되기도 하였다. 따라서 본 발명자는 장기간 암환자의 치료 및 진단과정을 살펴본 결과 혈액 중의 동위원소의 함량 변화에 따라 암이 발생할 수 있으며 그 농도를 분석하는 방법에 의하여 암진단 및 암의 종류를 판단할 수 있다는 것을 알게 되었으며 이로써 본 발명을 완성하게 되었다. 방사능 오염지역에서는 과다한 암환자의 발생이 보고되고 있으며 이는 과다한 방사능에 조사한 인체에서 체내 동위원소의 함량이 증가되며 이로서 암의 발생이 연유된다는 것을 추론할 수 있으며 본 발명가는 이를 암환자의 혈액 및 조직을 분석하는 방법에 의하여 입증하여 본 발명을 완성하게 되었다.
본 발명은 혈액 또는 조직 중의 동위원소의 함량을 분석하는 방법에 의한 암진단에 관한 것으로, 현재 과학적인 한계로 인하여 1mm 이하의 암종에 대한 정확한 분석이 불가능한 현실에서, 혈액의 분석을 통한 암종 및 암발생유무를 보다 정확하게 판단하므로써 조기에 암을 진단하고, 이를 토대로 정밀검사하여 암을 치료할 수 있는 기준 및 근거를 제시할 수 있게 한다. 따라서 본 발명은 혈액 또는 생체조직 중의 동위원소의 함량을 분석하여 이를 정상인과 비교하는 방법에 의하여 암의 발생 유무 및 암종을 알 수 있게 하는 것을 그 목적으로 한다.
본 발명의 가장 근본적인 작용근거는 동위원소의 분석이다. 동위원소의 분석은 질량 스펙트럼 분석법(Mass Spectrometric Method)을 이용한다. 환자 또는 정상인의 혈액 또는 조직 중의 체액을 대상으로 질량스펙트럼분석에 의하여 동위원소의 농도를 분석하고, 이를 정상인과 비교하는 방법에 의하여 암종 및 암의 발생 유무를 진단할 수 있다.
본 발명의 효과는 극소량의 암조직 또는 현재 과학으로 진단 불가능한 크기의 암조직에 대해서도 동위원소의 양을 측정하여 비교하는 방식에 의하여 이를 정 확히 진단하게 함으로써 암의 치료율을 높이고, 건강을 회복할 수 있는 기회를 제공하여 건강한 삶을 영위하게 하는 데 매우 큰 도움을 줄 수 있다. 암은 조기진단시 그 치료 성공율이 매우 높은 것은 주지의 사실이다. 그러나 아직 이를 뒷밧침할 명확한 진단방법이 없는 것 또한 사실이다. 본 발명은 이러한 한계를 해결하여 암의 조기진단을 가능하게 하여 암환자의 적극적인 치료 및 그 성공율을 높이는데 크게 기여할 수 있다.
본 발명자는 정상인 증류수 및 암환자로 부터 혈액, 조직, 미네랄 워터, 전기분해수를 대상으로 동위원소의 양을 비교하기 위하여 시험에 사용하였다. 동위원소의 조성을 분석하기 위하여 EMAL-2(Energy Mass Analyzer)를사용하였다. 이는 이중 촛점형식 질량분광광도계(double-focus type mass spectrophotometer)이다. 원자의 이온화와 증기화를 위하여 레이저원(Lazer source)으로 개별 이온을 사용하였다. 표준시료를 이용하여 분석결과치를 보정하였으며 전체 10종의 안정한 동위원소를 사용하였으며, 이를 이용하여 조성비를 조사하였다. 사용한 원소로는 마그네슘, 실리콘, 황, 염소, 칼륨, 칼슘, 크롬, 철, 구리 등이다. 이들 원자의 동위원소 그룹은 마그네슘의 경우 분자량 24, 25, 26을 사용하였고, 실리콘의 경우 분자량 28, 29, 30을 사용하였으며, 황의 경우 분자량 32, 33, 34, 36을 사용하였고, 염소의 경우 분자량 35, 37을, 칼륨의 경우 분자량 39, 40, 41을, 칼슘의 경우 분자량 40, 42, 43, 44, 46, 48을, 크롬의 경우 분자량 50, 52, 53, 54를, 철의 경우 분자량 54, 56, 57, 58을, 구리의 경우 분자량 63, 65를 사용하였다. 또한 물을 제외한 다른 시료의 경우 안정한 동위원소의 분석을 위하여 진공오븐에서 섭씨 360도하에서 1시간 동안 건조하였다.
산소 또는 수소의 동위원소 조성의 미량변화에 있어 표준물질은 SMOW(Standard middle-ocean water)을 사용하였으며 조성의 변화는 1/1000을 표시하는 퍼밀(permill)을 사용하였다. 분석된 물은 4~5ml을 10-5~10-6mmHg의 진공도 하에서 800℃에서 우라늄과 반응시켜 수소의 형태로 발생되게 한 후 측정하였다.
수소의 동위원소 조성 분석을 위하여 독일 Varion사의 GD-150을 사용하였으며, 물 중의 산소의 분석 및 기체 시료의 분석을 위하여 우크라이나의 Sumi사의 Electron을 사용하였다. 이들 기기의 특성은 개별 원소의 동위원소의 동위원소 조성의 미세한 변화도 감지할 수 있게 하는 것으로서 시료와 표준물질을 동시에 분석 비교하게 하는 특성을 가지고 있다. 질량분석기는 2~3개의 이온 수집기를 가지고 있는 것 외에 동시에 2~3개의 이온성 전류를 측정하여 그들의 관계를 측정하게 한다. 혈액 시료 중의 동위원소의 분석은 질량분석기 EMAL-2를 사용하였다.
이하 본 발명을 하기의 실시예로서 상술하고자 한다. 다만, 본 발명의 범위가 하기의 실시예에 의하여 한정되지는 않는다.
실시예 1. 혈액 중의 중수소함량
중수소의 함량을 수소의 함량 대비로 ppm(part per million)의 형태로 나타 내었다. 즉, 수소 1H 1,000,000개 당 2D의 갯수이다.

암환자의 경우 정상인에 비하여 heavy isotope의 함량이 높았다. 그러나 36S의 경우 특이적으로 검출되지 않아 이는 상대적으로 결핍된다는 것을 알 수 있었다.

칼륨의 동위원소 중 40K의 함량이 특이적으로 암환자에게서는 0로 나타나 검출되지 않았다. 이는 정상인과 차별화되는 가장 특이한 부분 중의 하나이다. Heavy isotope의 경우 암환자군에서 높게 나타났다.

칼슘의 동위원소 중 heavy isotope의 경우 암환자 혈액에서 높은 함량을 보였다.
이상의 결과에서 알 수 있는 바와 같이, 혈액 또는 조직 중의 동위원소의 양을 측정하는 방법에 의하여 암의 존재 유무를 파악 할 수 있으며, 40K, 36S의 결핍이 특징적으로 암환자에게서 나타났다. 이는 암의 존재 유무를 판단할 수 있는 중요한 기준으로 이용될 수 있다.
본 발명은 혈액 또는 조직 중의 동위원소의 양을 측정하는 방법에 의하여 암의 존 재 유무 및 그 암종을 판단 할 수 있는 방법에 관한 것으로서 이 방법을 이용할 경우 암진단의 정확성을 더할 수 있어 암여부에 대한 오진을 최소화 할 수 있으며, 미세한 암종의 존재에 대해서도 적용가능하여 암의 조기진단을 통한 적극적 치료에 이용될 수 있어 국민의 건강증진에 기여 할 수 있으며, 기업의 이윤 창출에 도움을 줄 수 있다.
Claims (6)
- 혈액 또는 조직세포 중에 함유된 원소의 동위원소의 양을 측정하는 방법에 의하여 암의 존재 유무를 파악하는 방법.
- 상기 청구항 1에서 함유된 원소로서 수소, 산소, 마그네슘, 칼슘, 칼륨, 황, 염소, 규소, 철, 구리 중 하나로 이루어지는 것을 특징으로 하는 암의 진단방법.
- 상기 청구항 1에서 수소의 경우 중수소(2H)의 함량이 10% 이상 증가되는 것을 근거로 하는 암의 진단방법.
- 상기 청구항 1에서 산소의 경우 18O의 함량이 10% 이상 증가되는 것을 특징으로 하는 암의 진단방법.
- 상기 청구항 2에서 Heavy isotope의 함량이 10% 이상 증가되는 것을 근거로 하는 암의 진단방법.
- 상기 청구항 1에서 동위원소로서 40K, 36S 중 어느 한가지 이상의 결핍을 근 거로 하는 암의 진단방법.
