본 발명은 인체에 해를 끼치는 각종 세균 및 바이러스를 예방 및 박멸하기 위하여 인체에 무해한 물질들로 새로운 혼합물을 생성하고 이를 활용하여 각종 세균과 바이러스를 퇴치하고자 하는 기술에 관한 것이다.
세균이란 원핵세포(原核細胞)를 갖는 단세포의 미생물군을 의미하며, 박테리아라고도 한다. 넓은 뜻으로는 원핵균류(原核菌類)를 가리키나, 좁은 뜻으로는 원핵균류 중 미코플라스마류·점액세균류·스피로헤타류·방선균류는 제외된다. 보통 세균이라고 할 때는 좁은 뜻의 세균류에 포함되는 것을 가리킬 경우가 많다. 세균류는 세포벽이 있는 미소한 단세포체이며, 운동을 하지 않는 것도 있고 편모(鞭毛)로 운동하는 것도 있다. 영양법은 일반적으로 종속영양생활을 하지만, 일부에는 다른 생물군에서는 볼 수 없는 특이한 독립영양생활을 하는 것도 있다.
세균류는 부생(腐生)·기생·공생을 하여 종속영양생활을 하거나, 독립하여 필요로 하는 유기물을 자력으로 무기물에서 만들어낸다. 세균류는 지구상의 모든 곳에 살고 있는데 특히 비옥한 토양이나 물 속에 많으며, 1g 속에 30억 이상의 세균이 들어 있다. 보통 20∼50℃에서 생육하지만 이보다 낮거나 높은 온도에서 생육 하는 것도 있다. 세균류는 각종 영양법에 의해서 스스로 살아가는 한편, 생태계에서 주요 원소(탄소·질소·황)의 순환에 중요한 역할을 하고 있다. 또 그 작용으로 생긴 여러 가지 생산물은 인간에게 이용되고 있다. 예를 들면, 젖산·아세트산·글루콘산 등의 유기산, 글루탐산 및 그 밖의 아미노산·아밀라아제·프로테아제 등의 효소제, 아세톤, 부탄올, 비타민 C, 대용혈청이 되는 덱스트란, 항균성항생물질, 나아가서는 세균성백신, 면역혈청, 요구르트, 치즈 등이 있다.
(1). 영양법의 기본형:세균류가 유기물을 만들기 위해서는 탄소원과 수소공여체 및 에너지원이 필요하다. 이 3가지에 유의하면 영양법에는 다음과 같은 기본형이 있음을 알 수 있다. 우선 에너지원, 즉 ATP(아데노신 3인산)를 생성하는 인산화에는 산화환원반응에 의한 것과 광화학반응에 의한 것이 있다. 산화환원반응은 세균류에서 일반적으로 볼 수 있는 것으로, 해당계(解糖系)·시트르산회로계·전자전달계에서 일어난다. 이러한 기본형 모두를 볼 수 있는 것은 슈도모나스강(綱)에 한정되고, 그중 광화학반응에 의한 것은 홍색세균류에서 볼 수 있다.
(2). 산소와의 관계 : 세균류 영양법의 기본형은 분자상태의 산소와의 관계에 따라 다양화되어 있다. 혐기성(嫌氣性)인 것도 있으나 오히려 호기성인 것이 많다. 종속영양생활을 하는 세균의 영양법을 살펴보면 혐기성 종속영양에는 무기발효(부티르산균 등)와 무기호흡(바실루스 등에서 볼 수 있는 아세트산호흡 등)이 있다. 호기성 종속영양에는 유기발효(아세트산균 등)와 일반적인 산소호흡이 있다. 임의종속영양은 환경조건에 따라 무기발효를 하거나 산소호흡 또는 질산호흡을 한다(대장균). 산소호흡은 특히 토양 속에서 활발하게 일어나며, 이로 인해 유기물이 환원되어 이산화탄소가 생성된다. 체중 1kg당의 산소 소비량을 사람과 비교하면 세균류의 소비량은 사람의 수백배에서 수만배나 되고 아조토박터는 6만 배가 된다.
그 체제를 형태와 세표표층, 세포내 구조로 나누어 실펴보면 다음과 같다.
(1). 형태:세균류의 형태와 크기는 조건에 따라 달라지기도 하지만, 일반적으로 구균은 지름 0.5∼1㎛, 간균(桿菌)은 거의 구균과 같은 길이이며, 폭은 길이의 1/2 또는 1/7이다. 콤마균은 1회 만곡된 형태이고, 나선균은 4∼20회 말린 형태이다. 특징적인 세포집단을 형성하는 것에는 8개의 세포가 입방체모양으로 배열하는 8연구균, 세포가 연쇄되어 있는 연쇄구균·연쇄간균, 세포가 불규칙하게 덩어리져 있는 포도상구균 등이 있다. 이들과는 다른 특수한 세포집단으로 모상체(毛狀體)가 있다. 모상체는 다세포체가 아니지만 1줄로 늘어선 세포 사이가 밀집해 있고, 또 세포 사이에 분화가 일어나 지지세포(부착기)가 되는 것과 생식세포가 되는 것도 있다. 세균류의 편모는 진핵생물(眞核生物)의 편모보다 구조가 간단하고 미세구조나 운동형식도 다르다. 또 세균류의 편모는 탈락하기 쉽다. 형태적으로 보면, 티푸스균은 몸주위에 8∼12개의 편모를 갖는 주생다모균(周生多毛菌)이고, 콜레라균은 한쪽 끝에 1개의 편모가 있는 극생일모균(極生一毛菌)이다. 이 밖에 1극생·양극생 다모균도 있다.
(2). 세포표층:세균류의 세포막은 지질분자(脂質分子)로 이루어지는 2개의 층 안팎에 단백질이 산재해 있는 얇은 막이며 호흡에 관계하는 효소외에 소수의 세균류의 세포막에는 광합성에 관계하는 전자전달계효소가 함유되어 있다. 세포막의 바깥쪽에 있는 세포벽에는 단층인 것(그람 염색반응양성균)과 2층인 것(그람 염색 반응음성균)이 있다. 단층인 것은 펩티드글리칸과 테이코산이 주성분이고, 2층인 것은 소량의 펩티드글리칸과 리포다당·리포단백질로 이루어졌다. 그러나 대부분의 세균세포는 적어도 어느 시기에는 점액층으로 둘러싸이게 된다. 점액층은 폴리펩티드인 경우와 다당류(多糖類)인 경우가 있으며 세포를 유해한 것으로부터 보호하는 역할을 하고 있다. 세균에 따라서는 협막이 이 역할을 하는 것도 있다. 협막은 한천모양의 두꺼운 층으로 그 주성분은 협막다당류이다. 예를 들면 연쇄구균에서는 히알루론산이다.
(3) 세포내 구조:세균류의 원핵(핵산 DNA)은 김자염색 등에 선택적으로 염색되며 염색된 입상부(粒狀部)는 핵양체(核樣體)라고 불린다. DNA는 세포막 또는 리소좀과 연락하고 있다. 리소좀은 세포막의 일부가 함입하여 말려들어 생긴 막구조물이다. 세균류의 리보솜(RNA-단백질복합체)은 진핵세포의 것보다 작고, 세포 안에 거의 균등하게 함유되어 있다. 또 세포 안에는 다당체·지질(脂質) 등의 과립, 황·탄산칼슘의 결정 등도 함유되어 있다. 폴리메타인산이 주성분인 볼루틴이라는 저장물질은 이염과립(異染顆粒)이라고도 하며, 디프테리아균에는 반드시 들어 있다. 또 세균류의 세포에는 색소체와 액포가 없고, 체포질에는 유동성이 없다.
세포의 생식을 살펴보면 다음과 같다.
세균류는 보통 2분법으로 증식하는데, 1회 분열에 15∼20분이 걸리고, 조건이 좋으면 1개가 24시간 동안에 1021개, 총중량으로 약 4t이 된다. 대장균 1마리를 필요한 무기염류 수㎎을 함유한 배양액 1㎖ 속에서 배양하면 36시간 동안에 수십억 개나 된다. 질산균 등은 출아법(出芽法)으로 증식한다. 또 모상체의 세포 안에는 고니디아라는 내생포자, 또는 극모(極毛)가 있는 유주자(遊走子)가 생긴다. 간균은 세포 안에 1개의 휴면포자를 형성한다. 대장균은 개체에 따라 편모 외에 다수의 짧은 섬모(纖毛)를 갖는 것이 있는데, 이것으로 섬모가 없는 개체와 결합하여 섬모를 통해 DNA의 일부를 교환한다. 이를 위접합(僞接合)이라 한다.
세균류는 리케차강(綱)·진정세균강·슈도모나스강의 셋으로 분류된다. 리케차강은 다른 어떤 세균보다도 작고 동물세포 안에 기생하며 그람 음성이다. 진정세균강은 종속영양균으로서, 편모가 없거나 주생편모(일부의 것은 극생편모)가 있으며 그람 양성 또는 음성이다. 슈도모나스강은 종족영양균이 기본이지만 독립영양균과 관련종속영양균도 포함되며, 편모가 없거나 극생편모(일부는 주생편모)를 가진다. 그람 음성인지의 여부는 분명하지 않다.
(1). 리케차강 ① 클라미디아목(目):앵무병클라미디아는 조류에 기생하여 고양이·사람에게 공기전염되고, 트라코마클라미디아는 사람·원숭이 등에 기생한다. ② 리케차목:쯔쯔가무시병 리케차는 털진드기의 일종인 붉은 쯔쯔가무시를 매개로 하여 쥐에서 사람에게 전염된다.
(2). 진정세균강 ① 진정세균목:널리 분포하고 있는 세균으로 그람 양성균에는 포도상구균·부티르산균·젖산균·디프테리아균 등이 있고, 그람음성균에는 뿌리혹박테리아(근류균)·아조토박터·대장균·콜레라균 등이 있다. ② 카리오파논목:길이 수십㎛의 모상체가 물 속이나 유기물에 부생하여 주생편모로 활발하게 운동하는 세균도 있고, 절지동물이나 소·양 등의 소화기관 안에 기생하는 것도 있 다.
(3). 슈도모나스강 ① 슈도모나스목:종속영양 또는 화학합성을 하는 세균으로 대부분 토양이나 물 속에서 산다. 아세트산균·녹농균·메틸산화균·질화세균·황산환원균·호염균·메탄세균 등이 있다. ② 홍색세균목:유황천·호소 빛 바닷물에도 사는 세균으로 수소공여체로는 황화수소·수소분자·지방산·케토산 등을 사용하는데, 산소를 발생하지 않는 세균형광합성을 한다. 무황홍색균·황홍색균·황록색균 등이 있다. ③ 황세균목:황화물이 있는 토양이나 물 속에 사는 세균으로 황화수소 기타의 화합물이나 황을 유기 또는 무기적으로 산화하여 화학합성 또는 종속영양생활을 한다. 여기에 속하는 세균에는 모상체도 있으며, 고체에 닿으면 활주운동이나 흔들운동을 한다.
바이러스는 세포성생물과는 분명하게 다른 분자적 구성과, 독특한 증식상(增殖相)을 가진 비세포성 구조체의 일군(一群)이다. 이 용어는 라틴어로 〈독(毒)〉을 의미하는 말로서, 비루스 또는 여과성병원체라고도 한다. 바이러스는 그 자체를 증식시킬 수 있는 세포 내에 들어가 숙주(宿主;바이러스의 기생 대상이 되는 생물)에 여러 가지 영향을 미친다. 예를 들면 담배모자이크바이러스(TMV)가 담뱃잎에 모자이크 증상을 일으키며, 구제역(口蹄疫)바이러스가 소 입의 점막 등에 수포(水疱)를 만든다. 이러한 여러 증상을 초래하는 것은 세균보다 작아서 광학현미경으로 관찰할 수 없으며, 세균여과기를 물과 함께 통과해버리는 여과성 입자인데, 이 입자가 병원성이라는 것을 19세기 말에 발견했다. 그 후 여러 가지 바이러스가 알려지고, 전자현미경에 의해 형태나 성상(性狀)에 대한 연구가 비약적으로 진행되었다.
바이러스는 증식을 위한 유전정보를 담당한 핵산(DNA 또는 RNA)을 가진 핵단백질의 분자로서, 크기는 극소세균인 리케차의 반 정도(20∼300nm)이다. 바이러스는 구성하는 핵산의 종류에 따라 DNA바이러스와 RNA바이러스로 크게 나뉘기도 하고, 숙주에 따라 식물바이러스·동물바이러스·세균바이러스·곤충바이러스로 구분되는데, 식물과 곤충, 또는 항온동물과 곤충 같은 식으로 2가지 숙주에서 증식하는 바이러스도 있다. 또 1종류의 숙주에만 기생하는 것이 있는가 하면, 오이모자이크바이러스처럼 숙주가 100종 이상인 것도 있다. 식물바이러스는 RNA바이러스이며, 단백질 이외에 폴리아민이나 금속을 함유한 바이러스도 있다. 또 지방·탄수화물을 함유한 바이러스도 발견되는데 이것들은 동물바이러스에 많다. 동물바이러스와 박테리오파지(세균바이러스 또는 파지라고도 한다)에는 DNA바이러스와 RNA바이러스가 있다. 박테리오파지 중에는 일반 세균이나 방선균(放線菌)에 붙는 것이 있다. 또 효모균에 붙는 바이러스나, 페니실리움·아스페르길로스 등과 같은 곰팡이 또는 버섯에 붙는 바이러스도 있는데 이들은 마이코파지라 한다.
바이러스를 구성하는 핵산은 한가닥사슬 또는 두가닥사슬인데, 가장 많이 발견되는 것은 이중나선 DNA와 한가닥사슬 RNA이다. 이러한 핵산이 중심이 되고 이 중심을 일정 구조의 단백각(蛋白殼;capsid)이 감싸고 있다. 이 중심과 단백각이 합쳐진 것이 뉴클레오캡시드라는 바이러스의 기본 구조이다. 어떤 것은 뉴클레오캡시드 바깥쪽에 엔벨로프(被膜)가 있다. 엔벨로프는 리포단백질막으로 되어 있으며, 표면에 돌기(스파이크)가 있는 경우도 있다. 캡소미어(구조단위)의 배열로 보면 캡시드의 구성 양식(대칭성)에는 등방대칭(대부분은 정이십면체)·나선대칭(TMV는 16 단위로써 一周)·복합대칭(T2파지나 폭스바이러스) 등 3가지가 있다. 오르토믹소바이러스는 나선체로서 원통형의 길게 늘인 끈모양이며, 엔벨로프에 싸여 비리온이 된다. 비리온이란 세포 밖에서 감염성을 갖는 바이러스를 말한다. 라브도바이러스는 짧은 막대모양이며, 한쪽 끝에서부터 엔벨로프에 싸여 있다. 아데노바이러스나 헤르페스바이러스 등은 캡소미어가 정삼각형으로 늘어서서 정이십면체를 구성한다.
바이러스의 증식이란 바이러스의 핵산이 숙주세포 안으로 들어가서 바이러스 자체의 합성을 하는 것이다. 바이러스는 숙주의 세포벽에 어떤 흡착부위(리포단백질이나 리포다당류)에 결합하고(吸着), 세포벽을 파괴한 핵산이 엔벨로프·캡시드로부터 나와서(脫殼) 세포 안으로 침입한다. 바이러스의 침입을 받은 세포는 본래의 대사메커니즘이 제어되고, 염색체는 분해되어서 바이러스핵산과 바이러스단백질이 생산되어 축적된다(暗黑期). 그 후 바이러스핵산과 바이러스단백질은 바이러스를 합성하여 많은 바이러스가 생긴다(潛伏期). 결국 바이러스는 숙주세포를 파괴하고 나온다(放出期). DNA바이러스의 증식에서는 DNA바이러스핵산으로부터 유전정보를 전사(轉寫)한 각종 전령 RNA(mRNA;messenger RNA)가 만들어진다. 이것에 의해 먼저 바이러스성분 합성을 위한 효소단백의 생합성이 진행되고, 다음에 바이러스의 성분이 되는 바이러스 DNA와 바이러스단백질이 만들어진다. 바이러스성분의 조립은 대부분 숙주핵 안에서 행해지는데, 이 사이에 숙주세포의 세포 내 활동은 제어된다. RNA바이러스의 증식에서는 RNA가 그대로 생기거나 또는 RNA로부터 유전정보를 전사한 mRNA가 생겨, 바이러스단백질과 바이러스RNA합성이 행해진다.
바이러스의 분류에 관해서는 A. 로프가 1962년 분류한 것이 가장 널리 이용된다. 먼저 바이러스를 물리화학적 성상에 따라 크게 나누고, 바이러스종의 세분(細分)은 혈청학(血淸學)적으로 행한다. 분류의 지표가 되는 주요형질을 들면 다음과 같다. ① 핵산의 형태 ② 캡시드의 구축(構築) ③ 엔벨로프의 유무 ④ 에테르감수성(感受性) 등이다.
[DNA바이러스]
(1). 파르보바이 러스(Parvovirus과):한가닥사슬 DNA, 지름 20nm, 캡소미어 32개, 엔벨로프는 없고, 에테르내성이다.
(2). 파포바바이러스(Papovavirus과):고리모양 이중나선 DNA, 지름 45∼50nm, 캡소미어 42∼72개. 정이십면체이며 엔벨로프는 없고 에테르내성이다. 종양을 만드는 성질이 있다.
(3). 아데노바이러스(Adenovirus과):이중나선 DNA, 지름 70∼90nm, 캡소미어 252개. 정이십면체로 엔벨로프는 없고, 에테르내성이다.
(4). 헤르페스바이러스(Herpesvirus과):이중나선 DNA, 지름 100nm, 캡소미어 162개. 정이십면체의 뉴클레오캡시드를 형성하며, 엔벨로프가 있고 에테르감수성이다.
(5). 폭스바이러스(Poxvirus과):약 200×300nm의 벽돌모양 바이러스. 이중나선 DNA이며 이중의 엔벨로프에 싸여 있고 대형이다. 숙주원형질 내에서 증식하는 유일한 바이러스이며 두창·백신 등의 바이러스가 대표적이다.
[RNA바이러스]
(1). 피코르나바이러스(Picornavirus과):한가닥사슬 RNA, 지름 20∼30nm, 캡소미어 32개. 엔벨로프는 없으며 에테르내성이다.
(2). 레오바이러스(Reovirus과):이중나선 RNA, 지름 60∼80nm. 엔벨로프는 없고 에테르내성이다. 캡소미어는 이중인데 그 수는 확실하게 밝혀져 있지 않다. 유유아설사증바이러스가 대표적이다.
(3). 토가바이러스(Togavirus과):한가닥사슬 RNA, 지름 40∼70nm, 캡소미어 32개. 엔벨로프가 있으며 에테르감수성이다. 절지동물 매개성인 것이 많으며 황열병(黃熱病)·풍진바이러스가 대표적이다.
(4). 아레나바이러스(Arenavirus과):한가닥사슬 RNA, 지름 50∼300nm. 엔벨로프가 있으며 에테르감수성이다. 전자불투과성 입자를 함유한다.
(5). 코로나바이러스(Coronavirus과):한가닥사슬 RNA, 지름 7∼9nm의 막대모양 캡시드에 싸여 있으며, 지름은 80∼130nm이다. 엔벨로프는 꽃잎모양의 돌기가 있어서 태양의 코로나와 같은 모양이다. 사람의 상기도(上氣道)로부터 분리되었다.
(6). 레트로바이러스(Retrovirus과):한가닥사슬 RNA, 지름 약 109nm. 엔벨로프가 있으며, 에테르감수성이다. 종창바이러스를 이 과에 포함시킨다.
RNA 외에 미량의 DNA와 효소를 함유하는 것이 특징이다.
(7). 분야바이러스(Bunyavirus과):한가닥사슬 RNA, 지름 90∼100nm. 엔벨로프가 있으며, 에테르나 열에 대한 감수성이 높다. 절지동물 매개성 바이러스.
(8). 오르토믹소바이러스(Orthomyxovirus과):한가닥사슬 RNA, 원통형 뉴클레오캡시드의 지름은 6∼9nm. 엔벨로프가 있으며 비리온은 지름 80∼120nm이며 구모 양 내지 실모양이다.
(9). 파라믹소바이러스(Paramyxovirus과):한가닥사슬 RNA이며 오르토믹소바이러스보다 약간 대형이다. 지름 150∼300nm. 뉴클레오캡시드는 18nm의 원통형이며 홍역바이러스·RS바이러스가 대표적이다.
(10). 라브도바이러스(Rhabdovirus과):한가닥사슬RNA, 지름 약 70nm의 탄환모양. 엔벨로프는 길이 약 175nm이며 광견병바이러스, 소의 수포성구내염바이러스가 대표적이다.

바이러스는 가장 미소한 감염성 인자이며, 종류도 많아 그 감염에 의해 일어나는 병도 많다. 의학의 경우, 바이러스는 일반적으로 감염하는 숙주에 따라 크게 동물바이러스·식물바이러스·세균바이러스로 나눈다. 동물바이러스는 사람 이외의 동물에서는 종양을 형성하는 경우도 많이 알려져 있으나, 사람에 대해서는 많은 감염증을 일으키는 것이 확인되어 있을 뿐 종양에 관해서는 확인되지 않았다. 사람과의 관계로 본 경우, 바이러스는 숙주의 개체·장기·조직에 대해서 선택적으로 병을 일으킬 수 있다. 예를 들면 일본뇌염바이러스에 감염된 경우 대부분의 사람은 발병하지 않거나(不顯性感染), 발병해도 경증으로 지나는데, 극히 소수의 사람이 중증이 된다(顯性感染). 이것은 개체에 대한 바이러스의 선택의 두드러진 예인데, 반대로 인플루엔자처럼 감염되면 거의 100% 현성감염을 일으키는 바이러스도 있다. 다음에 장기·조직에 대한 바이러스의 기호에 대해 설명한다. ① 호흡기계의 장기에 잘 침입하는 바이러스:라이노바이러스·아데노바이러스·코로나바이러스· 콕사키바이러스 등으로 비염·인두염·기관지염·폐렴 등을 일으킨다. ② 중추신경계에 잘 침입하는 바이러스:폴리오바이러스(유행성소아마비바이러스)·아데노바이러스·홍역바이러스·일본뇌염바이러스·광견병바이러스 등이며, 급성회백수염(急性灰白髓炎)·수막염·뇌염·광견병 등을 일으킨다. ③ 눈에 잘 침입하는 바이러스:아데노바이러스의 8형이나 엔테로바이러스의 70형으로, 유행성각막결막염이나 급성출혈성결막염을 일으킨다. ④ 소화기계의 장기에 잘 침입하는 바이러스:콕사키A 및 B군 바이러스, 또는 단순헤르페스바이러스가 구내염을 일으키고, 로타바이러스·카리시바이러스 등은 유아에 설사증을 일으킨다. ⑤ 피부에 유명한 병변을 일으키는 바이러스:병명으로서 홍역·풍진·수두 등이 있으며 모두 병명에 붙은 이름의 바이러스가 병원(病原)이다. 또 이와 달리 발병·경과의 관점에서 바이러스에 의한 병을 분류하면, 크게 급성감염증·지속성감염증·지발성감염증으로 나눌 수 있다. ① 급성감염증:홍역·수두·폴리오(유행성소아마비)·인플루엔자 등이 포함되는데, 이들 병은 임상적으로도 각각 특이한 증상을 나타내며 병원바이러스도 각각이어서 병원체와 병이 일대일로 대응한다. 그러나 급성감염증 중에서도 코감기라고 하는 임상증상을 일으키는 바이러스에는 라이노바이러스나 코로나바이러스 등이 있고, 유유아의 설사증을 일으키는 바이러스에는 로타바이러스·카리시바이러스·아스트로바이러스 등이 있는 등 여러 개의 병원체가 하나의 병에 대응하는 것도 있다. ② 지속성감염증:급성감염증에서는 증상이 회복되면 병원바이러스는 체내에서 소멸하는 것이 보통이다. 그런데 이것이 체내의 일부에 잔존하여 때로 재발하는 경우가 있다. 예를 들면 유유아기에 단순헤르페스가 감염하여도 증상이 나타나지 않 은 채 경과하고, 숙주의 저항력이 저하했을 때 구순헤르페스로서 수포가 생기는 경우가 있다. 이것은 삼차신경절(三叉神經節) 등에 바이러스가 잠복해 있었기 때문이며 수포액 속에 바이러스를 증명하는 것이 생긴다. 이것을 지속성감염증이라고 한다. ③ 지발성감염증:수개월에서 수년이라는 긴 잠복기 후에 발병하고, 천연성(遷延性;지연성) 경과를 취하고 예후가 나쁜 감염증을 말한다. 아급성경화성전뇌염이나 진행성다소성백질뇌증이 그 예이다. 한편 바이러스감염증의 전염경로에는 세균이나 리케차 등에 의한 감염증과 같이 비말감염·접촉감염·곤충매개감염 등이 있는 외에, 모친에서 자식에 전해지는 자궁내감염·경태반감염(B형간염바이러스)·산도감염(사이트메가로바이러스)이 있다. 바이러스감염증을 치료와 예방면에서 보면, 바이러스에는 세균이나 리케차에 대해 항생물질처럼 효과가 있는 항바이러스제는 없고, 면역글로불린제제를 잠복기 초기에 이용하면 유효한 것이 있는 정도이다. 그러나 최근에는 인터페론의 항바이러스작용이 주목되고 있다. 따라서 바이러스감염증에는 백신에 의한 예방(유행성소아마비·홍역·풍진·유행성이하선염·일본뇌염·인플루엔자 등)이 최고의 대책이다. 발병 후 치료로서는 대증요법(對症療法)과 합병증 예방의 치료가 행해지고 있을 뿐이다.
바이러스의 유전학상 성상을 대충 살펴보면 다음과 같다.
(1).파지의 용균성(溶菌性)과 용원성(溶原性)
파지(박테리오파지 또는 세균성바이러스)에 의해 감염된 대장균은 파지를 방출하는데 그것은 새로 합성된 효소가 세포벽을 부분적으로 분해해서 숙주를 용균시킴으로써 일어난다. 용균은 효소의 작용에 의해 세포가 녹는 현상이다. 실험적으로 는 한천 배지 위의 세균 집락(集落;colony)에 용균이 일어나면, 군데군데 둥글고 투명한 반점(溶菌斑)으로 나타난다. 이 반점의 모양이나 크기에는 특징이 있는데 파지가 갖는 유전형질의 표현형이라 여겨진다. 그런데 숙주에 침입한 파지 DNA가 이러한 용균과정을 거치지 않고 숙주 DNA의 일정 부위에 정착해서 그대로 복제되어 숙주의 세포분열과 보조를 같이 하는 경우가 있다. 이때 원래의 파지 DNA를 프로파지라 하며, 숙주를 용원균이라 한다. 이 용원균은 잠재적으로 비리온(감염성이 있는 바이러스 입자)의 생산능력을 가지고 있어서 그것을 자손에 전한다. 이 잠재적 능력의 유발에 의해 프로파지가 세포로부터 유리되어 활성화하면 숙주는 용균된다. 이렇게 해서 방출된 비리온은, 프로파지가 없는 동종(同種) 세균을 감염시켜 그 세균을 용균 또는 용원균으로 바꾼다(溶原化). 이렇게 파지의 용군·용원화·유발의 조합은 유전학적으로 설명될 수 있다.
(2).돌연변이
용원반이나 숙주의 범위(宿主域) 또는 온도감수성 등에 관계있는 바이러스의 유전자에 돌연변이가 생겨 변이주(變異株)가 발생하는 경우가 있다(원래 바이러스의 계통은 野生株라 한다). 예를 들면 대장균을 숙주로 하는 T계파지의 하나인 T4에 의한 용균반에는, 야생주에 의한 야생형용균반과는 크기가 다른 r형이 출현한다. 이것은 T4파지에 함유된 다른 3개의 유전자의 돌연변이에 의해 발생하는 것이며, r형의 출현은 대장균의 주(株)에 따라 달라진다. 또 야생주인 T2파지는 대장균의 어떤 주를 용균하지는 않으나, 변이주인 T2h는 용균반을 만든다. 또한 다른 변이주 T2hr의 용균반 모양은 T2h와 다르다. 이 T2hr와 T2 (변이주와의 비교표기에서는 T2h-r-)가 이중으로 감염(重感染)하면 새로 형성된 파지에는 모체 파지와 같은 것 외에 T2h와 T2r가 일정한 비율로 생긴다. 이것은 모체 DNA의 단편결합(재조합;recombination)에 의한 것이라 짐작된다. 이 T계파지의 연구에 의해 돌연변이의 메커니즘이나 유전암호, 또는 유전자와 단백질구조와의 관계라는 생물학의 기초적 문제를 이해할 수 있게 되었다.
동물바이러스의 유전지표에 관하여 알아보면 다음과 같다.
동물바이러스가 숙주에 감염을 일으킨 때에 나타나는 중요한 것으로서 간섭인자(干涉因子;인터페론 IF)·항체 등이 있다. 인터페론은 당을 함유한 단백질로서, 중감염의 경우에는 한쪽 바이러스의 증식을 저지하는 기능을 가지고 있다. 항체는 바이러스단백질이 항원이 되어 생성된 것인데, 다음 바이러스의 흡착 내지는 증식을 막는 작용을 한다. 이와 같이 항원성·인터페론감수성·증식능·병원성·숙주역 등은 동물바이러스의 유전지표로서 중요한 것이다. 인플루엔자바이러스는 S항원(핵단백질)에 의해 A·B·C형으로 분류되는데, A형은 표면의 V항원(당단백질)에 따라 A·A1 ·A2 ·A3 등이 있고, 변이하기 쉬운 것도 바이러스가 가지고 있는 유전적 성상(性狀)이 변하기 때문이다.
기존의 항균성 물질들은 인체에 유해한 성분들이 들어 있어 상용화가 어렵거나, 인체에 무해한 물질들로 구성되더라도 항균능력이 떨어져 실제로 적용하기가 어려운 경우가 많았다. 설사 다소의 효과가 있다고 하더라도 부분적으로 일부 세균 에만 그 효과를 발휘하여 범용성이 떨어지는 경우가 많았다.
따라서 본 발명이 이루고자 하는 기술적 과제는 상술한 바와 같은 다양한 문제점들을 극복하여 저렴한 가격으로 양산이 가능하며, 인체에 유해성이 없으며 세균 및 바이러스를 효과적으로 박멸시킬 수 있는 혼합물을 제공하는데 있다.
환경친화적인 화합물 및 혼합물을 활용하여 세균과 바이러스를 퇴치하는 방법으로 인체에 유해성이 전혀 없으며, 구제효율도 좋을 뿐만 아니라, 또한 구제시간이 단축되는 장점을 지니고 있다.
본 발명에 따른 새로운 항균성 기능물질은 "산화아연,지르코니아,유황,각섬석"으로 구성된다.
산화아연(ZnO)은 산소와 아연의 화합물이다. 화학식량은 81.4이다. 녹는점은 가압하에서 1975℃, 비결정형의 비중이 5.47, 결정형의 비중은 5.78이고, 용해도는 0.42㎎/100㎖(물 18℃)이다. 공업약품·의약품·안료 등에서는 아연화(亞鉛華)·아연백(亞鉛白) 등으로 부르며, 천연에서 홍아연석으로 산출된다. 옥살산아연 ZnC2O4를 400℃에서 열분해하거나 염기성탄산아연 ZnCO3·3Zn(OH)2를 열분해하여 만든다. 공업적으로는 금속아연을 연소시켜서 만든다. 순수한 것은 상온에서 무색의 결정(홍아연석은 육방정계) 또는 분말이다. 250℃ 이상으로 가열하면 노란색이 되지만 냉각과 동시에 원상태로 되돌아간다. 강열(强熱)하면 1atm에서는 약 1300℃에서 승 화되기 시작한다. 가시광선은 잘 반사하지만 자외선은 잘 흡수한다. 태양빛에 의해 인광(燐光)을 내며 음극선·양극선 등에 의해 초록·보라 등으로 발광하고, 가열하면 흰색의 열발광을 한다. 가열해도 분해되지 않는다. 양쪽성 산화물로서 산에는 녹아 아연염을 만들고, 수산화알칼리 수용액에는 녹아서 아연산염을 만든다. 또한 암모니아수·탄산암모늄수용액에서도 착염을 만들며 녹는다. 산화코발트 CoO와 강열하면 코발트그린이라는 안료를 얻을 수 있다. 백색 안료로서 페인트·그림물감 등으로 사용된다. 그 밖에 시멘트·에나멜·유탁(乳濁)유리 등의 용도가 있으며 고무충전제 등에도 사용된다. 입자가 고운 상질(上質)의 것은 의약품·화장품·치과충전제로서 이용되고 있으며, 메탄올(메틸알코올) 합성용 촉매이기도 하다.
지르코니아의 화학식은 ZrO2이다. 산화지르코늄(Ⅳ)이라고도 한다. 지르콘광상에서 산출되는 은백색의 단단한 희유금속 원소이다. 분자량 123.22. 녹는점 2700℃. 저온형(단사결정계)과 고온형(정방결정계)로 나뉜다. 굴절률이 크고 녹는점이 높으므로 내식성이 크다. 물에 녹지 않고, 황산·플루오르화수소산에 녹는다. 온도의 급변에 견디므로 내열재료나 도가니 등에도 널리 사용되고, 소결체(燒結體)는 관상용(觀賞用)으로 사용되며 안료(顔料)로도 사용된다.
유황에 대하여 살펴보자. 오래 전부터 그 존재가 알려져 온 황은 고대에는 황을 태워 그 연기에 쐬어서 소독하는 방법으로 사용되었다고 한다. 그 후 의약 또는 화약으로 널리 이용되어 왔다. 영어의 sulfur 또는 sulphur는 산스크리트의 '불의 근원'을 뜻하는 Sulvere로부터 유래된 라틴어 Sulphurium이 그 어원이다. 지구 상에는 자연황으로서 유리상태로 산출되지만 화합물로서도 광범위하게 다량 존재한다. 자연황은 화산지방에 많고 미국 ·일본 ·이탈리아 등이 주요 산지이다. 화합물로서는 황철석 ·황동석 ·방연석(方鉛石) ·섬아연석 ·진사(辰砂) 등의 황화물, 석고 ·중정석(童晶石) 등의 황산염으로 존재하며 그 외에 화산가스 ·온천 ·광천 등에도 황화수소 ·아황산가스(이산화황) ·황산 등의 형태로 존재한다. 또한 생물계에서도 중요한 원소이며, 단백질 속에 많이 함유되어 있다.
각섬석(角閃石, hornblende)은 각섬석족 중에서 가장 흔히 다량으로 나타나는 종류이며, 좁은 뜻으로 보통각섬석(common hornblende)이라고도 한다. 칼슘 ·나트륨 ·칼륨 ·마그네슘 ·철(Ⅱ) ·철(Ⅲ) ·알루미늄 ·수산기 ·플루오르 등을 함유하는 규산염이다. 화학조성이 아주 복잡하고 다양하여 한 화학식으로 나타내기가 어렵다. 약간 긴 마름모기둥 또는 6각기둥이나 때로는 짧은 주상결정을 보이며 쌍정(雙晶)일 때도 많다. 주면(柱面)에 평행한 두 방향으로 완전한 쪼개짐이 있는데 이들 둔 쪼개짐은 약 120°, 60°에서 서로 교차하므로, 이것에 의해서 90℃로 교차하는 휘석과 구별할 수 있다. 주로 안산암 ·현무암 등 화성암의 반정(斑晶)을 이루며, 또 심성암 ·반심성암 ·결정편암 등과 함께 산출되기도 한다.
상술한 재료들과 그 성분비를 표로 정리하면 다음과 같다.

상기 구성요소와 구성비는 반복실험을 통하여 최고의 황금비율을 찾아낸 결과이다. 이렇게 제조하여 만든 신 항균물질을 사용하여 각종 응용상품을 제조하여 시용하면 세균과 바이러스의 발생을 예방 및 구제할 수 있다.
그 작용 매커니즘을 살펴보면 상술한 항생물질들이 세균과 바이러스의 외피를 파괴하여 형체를 붕괴하고 또한 유전자에 손상을 주어 멸균기능을 수행한다.
세균의 경우를 살펴보면, 세균류는 안쪽에서부터 핵물질 ·세포질 ·세포막 ·세포벽(세균의 종류에 따라 세포벽이 없는 것도 있다) ·점액층 또는 캡슐[萊膜]의 순으로 되어 있다. 세포질에는 전자현미경으로 관찰되는 리보솜 ·메소솜 ·색소체, 그리고 무기영양분의 과립체 또는 함유물이 들어 있다. 본 발명에 따른 항균 신물질은 세균의 세포벽을 파괴하고 침투하여 유전자에 해당하는 DNA나 RNA에 치명적 손상을 초래하여 세균을 멸균한다.
바이러스의 경우를 살펴보기 전에 바이러스에 의해 감염된 세포와 정상세포 간의 차이점을 먼저 살펴볼 필요성이 있다. 가장 중요한 차이점은 다음과 같다. ① 생체 외에서 쉽게 생장할 수 있다. ② 접촉억제현상이 상실되어 세포가 여러 층으로 자랄 수 있으며, 세포가 생장해 가는 방향이 다양해진다. 따라서 세포가 정상세포보다 훨씬 빨리 자란다. ③ 세포 표면에 여러 변화가 생긴다(이온들의 통과능력 증가, 독소 호르몬 결합능력 상실, 새로운 항원생성 등). ④ 염색체 이상이 일어난다. ⑤ 인터페론이라는 항바이러스제가 생성된다. ⑥ 세포의 노화현상이 일어나지 않는다. ⑦ cyclic AMP 증가로 정상세포 생성순환을 중지시키고 변형된 세포생성순환으로 유도한다. 이와 같이 바이러스가 일단 감염되고 나면 세포내 바이러스 퇴치가 어려워지므로 바이러스를 원천 제거할 필요성이 있겠다.
바이러스의 구조는 유전물질을 전달하는 핵산으로 구성된 중심부(core)와, 이것을 싸고 있는 단백외각(蛋白外殼:capsid)이 있고, 또 어떤 종류의 바이러스는 그 단백외각 밖을 싸고 있는 지방질로 된 외피(envelope)로 구성되어 있다. 핵산은 DNA나 RNA의 어느 한 가지만을 가지고 있고, 이것을 기준으로 DNA바이러스, RNA바이러스로 나눌 수 있다.
단백외각은 외각단위단백체(外穀單位蛋白體:capsomer)가 질서정연하게 결합되어 여러 가지 형태를 이루고 있다.
본 발명에 따른 신항균물질은 단백외각를 파괴하여 바이러스가 그 몸체를 유지할 수 없게 하여, 사멸(死滅)시킬 뿐만 아니라 DNA와 RNA에도 손상을 초래하여 바이러스를 확실하게 박멸하게 된다.
〈실 시 예〉
10㎠의 종이위에 본 발명에 따른 항균물질을 0.5㎜ 두께로 도포하고 여기에 포도상구균 약 10,000 cells을 투입한 결과 [ 표2 ] 와 같은 결과가 나왔다.

상술한 방법으로 제조한 항균물질을 사용할 경우 짧은 시간내에 각종 질병의 원인인 세균과 바이러스의 외벽을 파괴하고 유전자(DNA,RNA)에 치명적 손상을 초래하여 세균과 바이러스 박멸의 기능을 수행한다.
다시말해 높은 구제효율로 세균과 바이러스를 멸균, 차단할 수 있다.
또한 저렴한 가격으로 대량생산이 가능한 잇점을 지니고 있다.
본 발명은 기재된 구체예에 대해서만 상세히 설명되었지만 본 발명의 사상과 범위내에서 변형이나 변경할 수 있음은 본 발명이 속하는 분야의 당업자에게는 명백한 것이며, 그러한 변형이나 변경은 첨부한 특허등록청구범위에 속한다 할 것이다.

- 산화아연(ZnO) 40-45%중량, 지르코니아(ZrO2 ) 20-35%중량, 유황 15-20%중량 , 각섬석 20-30%중량을 포함하는 항균성 기능을 지닌 혼합물.
