Abstract
PURPOSE: A microorganism producing poly-beta-hydroxy butyric acid (PHB) and a process for producing PHB using the same are provided, thereby maximizing the growth rate of microorganism and PHB accumulation, and minimizing physical property difference between lots. CONSTITUTION: The microorganism Ralstonia eutropha(KCTC 10351BP) produces and accumulates 80% or more of poly-beta-hydroxy butyric acid (PHB) in a medium containing high fructose syrup as a sole carbon source, wherein Ralstonia eutropha(KCTC 10351BP) is produced by irradiating UV(ultraviolet) to Ralstonia eutropha strain ATCC 17697, spreading the UV treated microorganism on agar medium with high C/N ratio, culturing the microorganism for 48 to 72 hours, and isolating milky white colonies grown and selecting one microorganism capable of using glucose and accumulates PHB. The process for producing PHA(polyhydroxy alkanoate) comprises fed-batch fermenting Ralstonia eutropha(KCTC 10351BP) in a medium containing ammonia water as a nitrogen source and a pH regulating agent, wherein the carbon source is glucose, fructose and high fructose syrup; and the C/N ratio is maintained at 15 to 25 up to 48 hours during the fed-batch fermentation, and the C/N ratio is maintained at 25 to 35 and the pH regulating agent is substituted by sodium hydroxide after 48 hours.
