Claims (22)
- 서열 3 내지 7 및 서열 8 내지 29로 구성된 군으로부터 선택된 서열과 98% 이상 동일한 아미노산 서열로 이루어진 부테닐-스피노신 생합성 효소를 코딩하며, 상기 아미노산 서열이 선택된 서열에 100% 미만으로 동일한 경우에는 이러한 차이가 코딩된 효소의 기능적 특성에 실질적으로 영향을 미치지 않는 것인 DNA 서열을 포함하는 단리된 DNA 분자.
- 제1항에 있어서, 상기 DNA 서열이 각각 서열 1의 염기 1-13032, 13059-19505, 19553-29053, 29092-43890, 43945-60636, 62090-63937, 65229-66602 및 68762-69676 및 서열 2의 염기 114-938, 1389-2558, 2601-3350, 3362-4546, 4684-6300, 6317-7507, 7555-8403, 8640-9569, 9671-10666, 10678-12135, 12867-14177, 14627-15967, 16008-17141, 17168-17914, 18523-19932, 19982-20488, 20539-21033, 21179-21922, 22674-23453, 23690-24886, 26180-26923 및 27646-28473으로 기재된 busA, busB, busC, busD, busE, ORF RI, ORF RII, ORF RIII, busF, busG, busH, busI, busJ, busK, busL, busM, busN, busO, busP, busQ, busR, busS, ORF LI, ORF LII, ORF LIII, ORF LIV, ORF LVI, ORF LVII, ORF LVIII 및 ORF LIX로 구성된 유전자 군으로부터 선택된 것인 단리된 DNA 분자.
- 각각 서열 3의 아미노산 7-423, 528-853, 895-977, 998-1413, 1495-1836,1846-2028, 2306-2518, 2621-2710, 2735-3160, 3241-3604, 3907-4086 및 4181-4262로 기재된 KSi, ATi, ACPi, KSb, ATb, KRb, DHb, ACPb, KS1, AT1, KR1 및 ACP1로부터 선택된 부테닐-스피노신 PKS 도메인을 코딩하는 DNA 서열을 포함하는 단리된 DNA 분자.
- 제3항에 있어서, 상기 DNA 서열이 서열 1의 염기 16-1269, 1582-2559, 2683-2931, 2992-4239, 4483-5508, 5538-6084, 6916-7554, 7861-8130, 8203-9480, 9721-10812, 11719-12258 및 12541-12786으로 구성된 군으로부터 선택된 것인 단리된 DNA 분자.
- 각각 서열 4의 아미노산 1-421, 534-964, 990-1075, 1336-1681, 1685-1864 및 1953-2031로 기재된 KS2, AT2, DH2, ER2, KR2 및 ACP2로부터 선택된 스피노신 PKS 도메인을 코딩하는 DNA 서열을 포함하는 단리된 DNA 분자.
- 제5항에 있어서, 상기 DNA 서열이 서열 1의 염기 13059-14321, 14658-15900, 16026-16283, 17064-18100, 18111-18650 및 18915-19151로 구성된 군으로부터 선택된 것인 단리된 DNA 분자.
- 각각 서열 5의 아미노산 1-421, 528-814, 1157-1335, 1422-1503, 1526-1949, 2063-2393, 2697-2875 및 2969-3049로 기재된 KS3, AT3, KR3, ACP3, KS4, AT4, KR4및 ACP4로부터 선택된 스피노신 PKS 도메인을 코딩하는 DNA 서열을 포함하는 단리된 DNA 분자.
- 제7항에 있어서, 상기 DNA 서열이 서열 1의 염기 19553-20815, 21143-22000, 23021-23557, 23816-24061, 24128-25399, 25739-26731, 27641-28183 및 28457-28699로 구성된 군으로부터 선택된 것인 단리된 DNA 분자.
- 각각 서열 6의 아미노산 1-422, 537-864, 891-1076, 1382-1563, 1643-1724, 1746-2170, 2281-2611, 2914-3093, 3186-3267, 3289-3711, 3823-4151, 4342-4636 및 4723-4804로 기재된 KS5, AT5, DH5, KR5, ACP5, KS6, AT6, KR6, ACP6, KS7, AT7, KR7 및 ACP7로부터 선택된 스피노신 PKS 도메인을 코딩하는 DNA 서열을 포함하는 단리된 DNA 분자.
- 제9항에 있어서, 상기 DNA 서열이 서열 1의 염기 29092-30357, 30700-31683, 31762-32319, 33235-33780, 34018-34263, 34327-35601, 35932-36924, 37831-38370, 38647-38892, 38956-40224, 40560-41544, 42115-42999 및 43258-43503으로 구성된 군으로부터 선택된 것인 단리된 DNA 분자.
- 각각 서열 7의 아미노산 1-424, 530-848, 885-1072, 1371-1554, 1650-1728, 1751-2175, 2289-2616, 2642-2775, 3131-3315, 3396-3474, 3508-3921, 4036-4366,4389-4569, 4876-5054, 5148-5229 및 5278-5531로 기재된 KS8, AT8, DH8, KR8, ACP8, KS9, AT9, DH9, KR9, ACP9, KS10, AT10, DH10, KR10, ACP10 및 TE10으로부터 선택된 스피노신 PKS 도메인을 코딩하는 DNA 서열을 포함하는 단리된 DNA 분자.
- 제11항에 있어서, 상기 DNA 서열이 서열 1의 염기 43945-45216, 45532-46488, 46597-47160, 48055-48606, 48892-49083, 49195-50469, 50809-51792, 51868-52269, 53335-53889, 54130-54366, 54466-55707, 56050-57042, 57109-57651, 58570-59106, 59386-59631 및 59776-60537로 구성된 군으로부터 선택된 것인 단리된 DNA 분자.
- 서열 3의 아미노산 6-977, 서열 3의 아미노산 998-2710, 서열 3의 아미노산 2735-4262, 서열 4의 아미노산 1-2031, 서열 5의 아미노산 1-1503, 서열 5의 아미노산 1526-3049, 서열 6의 아미노산 1-1724, 서열 6의 아미노산 1746-3267, 서열 6의 아미노산 3289-4804, 서열 7의 아미노산 1-1728, 서열 7의 아미노산 1751-3474 및 서열 7의 아미노산 3508-5531로 구성된 군으로부터 선택된 스피노신 PKS 모듈 (module)을 코딩하는 DNA 서열을 포함하는 단리된 DNA 분자.
- 제13항에 있어서, 상기 DNA 서열이 서열 1의 염기 16-2931, 2992-8130, 8203-12786, 13059-19151, 19553-24061, 24128-28699, 29092-34263, 34327-38892, 38956-43503, 43945-49083, 49195-54366 및 54466-60537로 구성된 군으로부터 선택된 것인 단리된 DNA 분자.
- 제1항 내지 제14항 중 어느 한 항의 DNA 서열을 포함하는 재조합 DNA 벡터.
- 제15항의 재조합 벡터로 형질전환된 숙주 세포.
- 1) 부테닐-스피노신 또는 부테닐-스피노신 전구체를 생산하는 생합성 경로에서 속도 제한 활성의 발현을 코딩하는 상기한 본 발명의 DNA 서열을 포함하는 재조합 DNA 벡터 또는 그의 일부를 사용하여, 생합성 경로에 의해 부테닐-스피노신 또는 부테닐-스피노신 전구체를 생산하는 미생물을 형질전환시키는 단계 및2) 상기 벡터로 형질전환된 미생물을 세포 성장 및 세포 분열, 상기 DNA 서열의 발현 및 스피노신의 생산에 적합한 조건하에서 배양하는 단계를 포함하는, 스피노신을 생산하는 미생물의 스피노신 생산력을 증가시키는 방법.
- 게놈 내에 작동되는 부테닐-스피노신 생합성 유전자를 보유하는 미생물의 게놈을 변형시켜 부테닐-스피노신 생합성 유전자 busA, busB, busC, busD, busE, busF, busG, busH, busI, busJ, busK, busL, busM, busN, busO, busP, busQ, busR 및 busS 중 하나 이상의 복제 (duplicate) 카피가 존재하는 미생물을 배양하는 단계를 포함하는, 부테닐-스피노신의 제조 방법.
- 게놈 내에 부테닐-스피노신 생합성 유전자를 보유하며, 상기 유전자들 중 하나 이상이 불활성화되어 있고, 나머지 유전자들은 작동하여, 파괴된 유전자가 작동할 경우에 생산되는 것을 제외한 부테닐-스피노신을 생산하는 미생물을 배양하는 단계를 포함하는, 부테닐-스피노신의 제조 방법.
- 형질전환되어 게놈 내에 작동되는 부테닐-스피노신 생합성 유전자를 함유하는 이종 미생물을 배양하는 단계를 포함하는, 부테닐-스피노신의 제조 방법.
- 게놈 내에 작동되는 부테닐-스피노신 생합성 유전자를 보유하며, 상기 유전자가 a) 적어도 하나 이상의 작동되는 PKS 모듈을 서열 1에 제시된 것보다 더 많이 또는 더 적게 포함하거나, 또는 b) KR, DH 또는 ER 도메인의 결실, 불활성화 또는 부가에 의하거나 AT 도메인의 치환에 의해 서열 1에 기재된 상응하는 모듈과는 다른 PKS 모듈을 포함하는 것인 미생물을 배양하는 단계를 포함하는, 부테닐-스피노신의 제조 방법.
- 부테닐-스피노신을 생산하는 미생물의 게놈 라이브러리를 생성하는 단계, 길이가 염기 20개 이상인 서열 1 또는 서열 2의 표지된 단편을 혼성화 프로브로서 사용하는 단계를 포함하는, 부테닐-스피노신 생합성 유전자의 단리 방법.
