Abstract
체내 동태(動態)가 개선된, 종양괴사 인자 활성을 가진 생리활성 물질을 제공함을 과제로 한다.
그 해결수단으로서는, 종양괴사 인자 활성을 가지는 단백질 부분과, 그 단백질의 N 말단에 인위적으로 결합시킨 고분자 부분으로 된 생리활성 복합체와, 이러한 생리활성 복합체를 유효 성분으로 하는 감수성 질환제를 제공함으로써 상기 과제를 해결한다.
Description
본 발명은 생리활성 물질에 관한 것으로서, 특히, 종양괴사 인자 활성을 가지는 생리활성 복합체에 관한 것이다.
앤가스 톰슨 편집 『더 사이토카인 핸드북』, 1994년, 아카데믹 프레스사 발행, 289 내지 304 페이지 등에 기재되어 있는 바와 같이, 종양괴사 인자는, 1975년, L. J. 올드 등에 의해, BCG 및 세균내 독소를 축차(逐次) 투여한 토끼나 마우스 등의 동물의 혈청 중에 분비되는 세포 장해성 인자로서 발견되었다. 그 후의 연구에 의해, 현재로서는, 종양괴사 인자는 분자량 약 17,0000 달톤, 등전점 5.6의 단백질이며, 주로 매크로파아지에 의해 산생(産生)되는 것이 알려져 있다.
종양괴사 인자는, 발견 당초부터, 종양 세포에 대하여 상당히 선택적으로 세포 장해 작용을 미치므로, 예를 들면, 일본국의 특공소62-45208호 공보나 특공평4-46928호 공보 등에 나온 바와 같이, 악성 종양에 대한 치료제로서 주목되어 개발이 진척되어 왔다. 그러나 종양괴사 인자는 생체에 있어서의 체류 시간이 짧고, 예를 들면, 정맥경로 등에 의해 생체에 투여하면, 혈액중의 프로테아제에 의해 신속하게 분해되거나 극히 단시간에 오줌 중에 배설되어버려 소기의 혈중 레벨을 장시간 유지할 수 없다. 그렇다고 해서, 혈중 레벨을 높이기 위해 투여량을 올리면 종양괴사 인자에 의한 세포 장해 작용이 정상세포에 까지 미치게 된다.
이와 같은 상황을 감안하여, 본 발명은, 체내 동태가 개선된, 종양괴사 인자 활성을 가지는 안정한 생리활성 물질을 제공함에 있다.

본 발명자가 예의 연구하고, 검색한 결과, 본질적으로 종양괴사 인자 활성을 가지는 단백질 부분과, 그 단백질의 N 말단에 인위적으로 결합시킨 고분자 부분으로 된 생리활성 복합체는, 고분자 부분을 가지지 않은 것과 비교하여 안정하게 생체에 있어서의 체류 시간이 유의(有意)하게 길고, 그 결과로서, 적은 용량을 투여해도 소기의 혈중 레벨이 보다 오래 지속하는 것이 판명되었다.
즉, 본 발명은, 종양괴사 인자 활성을 가지는 단백질 부분과, 그 단백질의 N 말단에 인위적으로 결합시킨 고분자 부분으로 된 생리활성 복합체를 제공함으로써 상기 과제를 해결하는 것이다.
더욱이 본 발명은, 종양괴사 인자 활성을 가지는 단백질 부분과, 그 단백질의 N 말단에 인위적으로 결합시킨 고분자 부분으로 된 생리활성 복합체를 유효성분으로 하는 감수성 질환제를 제공함으로써 상기과제를 해결하는 것이다.
이하, 본 발명의 실시의 형태에 대해서 설명하면, 앞서 설명한 바와 같이, 본 발명에서 말하는 생리활성 복합체라 함은, 종양괴사 인자 활성을 가지는 단백질 부분과, 그 단백질의 N 말단에 인위적으로 결합시킨 고분자 부분으로 된 것이다. 본 발명의 생리활성 복합체를 구성하는 단백질은, 예를 들면, 단백질 공학의 수법에 의해, 종양괴사 인자에 있어서의 아미노산 서열의 대부분을 변경함이 없이 종양괴사 인자 활성이 실질적으로 소실하지 않는 범위에서, 종양괴사 인자를 구성하는 아미노산 중에서 유리 아미노기를 가지는 리신 등의 아미노산에 대해, 단백질의 N 말단에 위치하는 것을 제외하고 그 이외를 유리 아미노기를 가지지 않은 것, 바람직하게는 아스파라긴, 알라닌, 아르기닌, 세린, 트레오닌, 프롤린, 메티오닌 또는 로이신 중의 어느 하나로 치환함으로써 얻을 수 있다.
주지하는 바와 같이, 종양괴사 인자는 기원에 따라 아미노산 서열이 약간 다르고, 예를 들면, 사람의 종양괴사 인자는 157개의 아미노산으로 된, 서열표의 서열 번호 1로 나타내어지는 아미노산 서열을 가진다. 아래에서 설명하는 감수성 질환제에 배합하는 생리활성 복합체의 구체예로서는, 단백질 부분으로서, 예를 들면, 서열표에 있어서의 서열 번호 2로 나타내어지는 아미노산 서열을 가지는 것, 바람직하게는 서열표에 있어서의 서열 번호 3으로 나타내어지는 아미노산 서열을 가지는 것을 들 수 있다.
이와 같은 단백질은, 아미노산 서열에 있어서의 제11번째, 제65번째, 제90번째, 제98번째, 제112번째 및 제128번째의 리신이 모두 아스파라긴, 알라닌, 아르기닌, 세린, 트레오닌, 프롤린, 메티오닌 또는 로이신 중의 어느 하나로 치환되어 있는 점에서 종래 공지의 사람의 종양괴사 인자와는 상위하지만, 종양 일반에 대하여 사람의 종양괴사 인자와 동등 이상의 세포 장해 작용을 미친다.
이와 같은 단백질은, 앞서 설명한 바와 같이, 단백질을 구성하는 아미노산의 1 또는 복수를 소망의 아미노산으로 치환하는 단백질 공학의 수법에 의해 얻을 수 있다. 예를 들면, 종양괴사 인자를 코드하는 DNA에 있어서의 유리 아미노기를 가지는 아미노산을 코드하는 코돈을 NNS 서열로 치환한 올리고뉴클레오티드를 사용해서 PCR 반응시키고, 얻어진 PCR 산물의 존재하에서, 종양괴사 인자를 코드하는 DNA를 PCR 반응시킴으로써, 종양괴사 인자에 있어서의 유리 아미노기를 가지는 아미노산이 랜덤(random)한 아미노산으로 치환된 단백질을 코드하는 DNA의 라이브러리를 얻는다. 그 후, 「파아지 디스플레이법」등에 의해, 이 라이브러리의 DNA를 해서 각각 단백질을 발현시킨 후, 통상적인 시퀀스 해석과 함께, 예를 들면, 종양괴사 인자에 대한 항체를 이용하는 고상 효소 면역법, 종양괴사 인자에 대한 항체 혹은 수 용체 단백질을 이용하는 파닝법, 더욱이는, 종양괴사 인자의 표적세포를 이용하는 바이오어세이 등을 조합해서 적용함으로써, 종양괴사 인자 활성을 유지하면서 종양괴사 인자에 있어서의 유리 아미노기를 가지는 아미노산의 중에서 N 말단의 아미노산을 제외하고, 그 이외의 것이 유리 아미노기를 가지지 않은 아미노산으로 치환된 단백질을 코드하는 DNA를 얻는다.
이와 같은 DNA의 선별에 있어서, 파아지 디스플레이법은 극히 유용하며, 이방법과 위에 나온 방법의 1 또는 복수를 병용할 때에는, 종양괴사 인자에 있어서의 유리 아미노기를 가지는 아미노산 중에서 N 말단에 위치하는 것을 제외하고, 그 나머지가 유리 아미노기를 가지지 않은 기타의 아미노산으로 치환되고, 또한, 종양괴사 인자의 바람직한 생리활성을 고레벨로 유지하는 일군(一群)의 단백질을 신속하게 해서 망라적으로 선별할 수 있게 된다.
본 발명의 생리활성 복합체를 구성하는 단백질은, 이렇게 하여 얻어진 DNA를, 필요에 따라 PCR 반응에 의해 증폭한 후, 플라스미드 벡터 등을 통해서 대장균 등의 숙주에 도입해서 형질 전환하고, 생성한 형질 전환체로부터 목적으로 하는 단백질을 산생하는 클론을 채취하여 배양함으로써 소망량을 얻을 수 있다. 형질 전환체의 배양물로부터 단백질을 채취하기 위해서는, 배양물에, 예를 들면, 투석, 염석, 여과, 농축, 원심분리, 분별 침전, 겔 여과 크로마토그래피, 이온 교환 크로마토그래피, 소수 크로마토그래피, 어피니티 크로마토그래피, 크로마토포커싱, 겔 전기영동, 등전점 전기영동 등의 단백질을 정제하기 위한 범용(汎用)의 방법을 적용하면 좋고, 필요에 따라 이들 방법을 적당히 조합해서 적용한다.
본 발명의 생리활성 복합체는, 이렇게 해서 얻어진 종양괴사 인자 활성을 가지는 단백질의 N 말단에 고분자를 인위적으로 결합시킴으로써 얻을 수 있다. 본 발명에서 사용하는 고분자는 실질적으로 수용성이며, 생체에 대해 무해한 것, 특히, 생체에 대해 항원이 되기 어려운 비단백질성의 것이 바람직하다. 각각의 고분자로서는, 예를 들면, 폴리비닐 알코올, 폴리에틸렌 글리콜, 폴리비닐피롤리돈, 폴리프로필렌 글리콜 등의 단독 중합체, 에틸렌 글리콜과 비닐 알코올, 프로필렌 글리콜등과의 공중합체 또는 그것들의 유도체 등의 합성 고분자나, 엘시난, 덱스트란, 히드록시에틸 셀룰로오스, 풀룰란, 메틸 셀룰로오스 등의 천연 고분자를 들 수 있고, 이 중, 분자량이 균일한 조제물을 입수하기 쉽다는 점에서 폴리에틸렌 글리콜의 단독 중합체, 공중합체 및 그것들의 유도체가 바람직하다. 또한, 고분자의 분자량은, 평균 분자량으로서, 통상, 500 내지 50,0000 달톤, 바람직하게는 1,000 내지 10,0000 달톤의 범위에서 가감하고, 분자량이 불균일할 경우에는, 반응에 앞서, 예를 들면, 분별 침전, 겔 여과 크로마토그래피 등의 범용의 방법에 의해 분획한다. 고분자의 종류나 생리활성 복합체의 최종 용도에도 따르지만, 분자량이 이 범위를 하회하면 체내 동태를 개선하기 어려워지고, 반대로, 이 범위를 상회하면, 복합체의 수용성이 저하하여 의약품으로서 안전하게 사용할 수 없게 될 경우가 있다.
종양괴사 인자 활성을 가지는 단백질의 N 말단에 이와 같은 고분자를 결합시키기 위해서는, 유리 아미노기에 특이적으로 반응해서 공유결합을 형성하는 시약에 의해 별도로 활성화시켜 둔 고분자를 단백질에 반응시키거나, 혹은, 유리 아미노기에 특이적으로 반응하는 관능기를 가지는 다관능 시약에 의해 단백질과 고분자를 가교(架橋)하면 좋다. 각각의 반응 방법으로서는, 이 분야에서 범용되는 방법, 예를 들면, 일본국 특개소62-289522호 공보 등에 기재된 에스테르 결합법, 아미드 결합법 등을 들 수 있고, 이 중에서 단백질과 고분자의 사이에 형성되는 공유결합의 안정성의 점에서 아미드 결합법이 바람직하다.
반응 방법에도 따르지만, 반응 시작시에 있어서의 단백질과 고분자의 비율은, 몰비로 1:0.1 내지 1:100, 바람직하게는 1:0.5 내지 1:50의 범위에서 가감한다. 일반적으로 이 범위를 하회하면 단백질끼리의 결합이 현저해지고, 반대로, 이 범위를 상회하면 고분자끼리의 결합이 현저해진다. 어떻든간에, 반응과 반응후의 정제(精製)의 효율을 저하시키는 것이 되므로, 통상, 상기의 범위에서 가감하는 것이 바람직하다. 반응시의 온도, pH 및 반응 시간은, 종양괴사 인자 활성을 가지는 단백질이 실활하거나 분해하기 어렵고, 또한, 바람직하지 않은 부반응이 최소가 되도록 설정되는데, 구체적으로는, 반응 온도를 0 내지 100℃, 바람직하게는 20 내지 40℃, pH를 0.1 내지 12, 바람직하게는 pH 5 내지 9로 해서 0.1 내지 50시간, 바람직하게는 10시간 이내에 반응을 완결시킨다. 이렇게 하여 얻어진 생리활성 복합체는, 종양괴사 인자 활성을 가지는 단백질을 정제할 경우와 마찬가지의 방법에 의해 정제할 수 있고, 최종사용 형태에 따라, 예를 들면, 농축, 염석, 원심분리, 동결 건조 등의 방법에 의해 액상 또는 고상으로 한다.
본 발명의 생리활성 복합체는, 감수성 질환을 치료 및/또는 예방하기 위한 약제로서 극히 유용하다. 본 발명에서 말하는 감수성 질환이라 함은, 생리활성 복합체를 단독으로 투여하거나, 혹은, 다른 약제와 함께 투여함으로써 치료 및/또는 예방할 수 있는 질환 전반을 의미하고, 각각의 질환으로서는, 예를 들면, 결장암, 직장암, 위암, 갑상선암, 설암, 방광암, 융모암, 간암, 자궁암, 인두암, 폐암, 유방암, 악성 흑색종, 신경 아세포종, 난소 종양, 고환 종양, 골육종, 췌장암, 부신종, 갑상선종, 뇌종양, 악성 흑색종, 균상 식육증 등의 고형 종양, 백혈병, 림프종 등의 혈액계 종양, 더욱이는 바이러스성 질환, 세균 감염증, 면역 질환 등을 들 수 있다. 따라서, 본 발명의 감수성 질환제는 이와 같은 질환을 치료·예방하기 위한,예를 들면, 항종양제, 항바이러스제, 항감염증제, 면역 질환제 등으로서 다종다양한 용도를 가지는 것이 된다.
적용 대상이 되는 감수성 질환의 종류나 증상에도 따르지만, 본 발명에 의한 감수성 질환제는, 투여 경로에 따라, 생리활성 복합체를 0.1 ng/kg 체중/회 이상, 바람직하게는 1 내지 1,000 ng/kg 체중/회 함유시키며, 용도에 따라, 엑기스제, 엘릭시르제, 하기도(下氣道) 흡입제, 캡슐제, 과립제, 안과용 서방제, 환제, 안연고제, 구강점막 첩부제, 현탁제, 에멀젼제, 경고제, 좌제, 가루약, 정제, 시럽제, 침지제, 전제(煎劑), 주사제, 용액제, 점안제, 점이제(点耳劑), 점비제(点鼻劑), 트로치제, 연고제, 팝제, 방향수제, 코용 분무제, 리니멘트제, 리모나데제, 유동 엑기스제, 로션제 등으로 조제된다.
본 발명의 감수성 질환제는, 소위, 투약 단위 형태의 약제도 포함하는 것으로 하고, 그 투약 단위 형태의 약제라 함은, 생리활성 복합체를, 예를 들면, 1회당의 용량 또는 그 정수배 (4배까지) 혹은 약수 (1/40까지)에 상당하는 양을 함유해서 되며, 투약에 적합한 물리적으로 분리한 일체의 제형으로 되어 있는 약제를 의미한다. 이러한 투약 단위 형태의 약제로서는, 예를 들면, 캡슐제, 과립제, 환제, 좌제, 가루약, 정제, 주사제, 팝제 등을 들 수 있다.
본 발명의 감수성 질환제는, 유효성분으로서의 생리활성 복합체 이외의, 예를 들면, 부형제, 연고 기제(基劑), 용해제, 교미제(矯味劑), 교취제(矯臭劑), 착색제, 유화제 등의 약제 일반에 범용되는 적당한 조제용 약을 배합하여도 좋다. 또한, 본 발명의 목적을 일탈하지 않는 범위에서, 생리활성 복합체와 함께 다른 유효성분으로서, 예를 들면, 외피용 살균 소독제, 창상 보호제, 소염제 등의 외피용 약, 비타민 A제, 비타민 B제, 비타민 C제, 비타민 D제, 비타민 E제, 비타민 K제 등의 비타민제, 칼슘제, 무기질 제제, 당류제, 유기산 제제, 단백 아미노산 제제, 장기 제제 등의 자양 강장약, 클로로필 제제, 색소 제제 등의 세포 부활용(賦活用) 약, 알킬화제, 대사 길항제, 항종양성 항생물질 제제, 항종양성 식물성분 제제 등의 항종양약, 항히스타민제 등의 알레르기용 약, 항결핵제, 합성 항균제, 항바이러스제 등의 화학요법제, 더욱이 호르몬제, 항생물질 제제, 생물학적 제제 등의 생리활성 복합체 이외의 약제를 1 또는 복수 배합해도 좋다.
또한, 본 발명의 생리활성 복합체는, 면역 조성제로서, 예를 들면, 악티노마이신 D, 아세그라톤, 이호스파미드, 우베니메쿠스, 에토포시드, 에노시타빈, 염산 아쿠랄비신, 염산 이달비신, 염산 이리노테칸, 염산 에피루비신, 염산 겜시타빈, 염산 다우노루비신, 염산 독소루비신, 염산 나이트로젠 마스터드-N-옥사이드, 염산 님스틴, 염산 피라루비신, 염산 파도졸 수화물, 염산 블레오마이신, 염산 브로카르바진, 염산 미토크산트론, 카르보콘, 카르보프라틴, 카로모풀, 시트르산 타목시펜, 시트르산 토레미펜, 쿠레스틴, 아세트산 메드록시푸로게스트론, 시클로포스파미드, 시스플라틴, 시조피란, 시타라빈, 시타라빈옥포스페이트, 디노스탄틴스티마라마, 타르타르산 비노레루빈, 소부족산, 다카르바진, 티오테파, 데카풀, 데카풀·우라실, 데타풀·기메스타트·오타스타트칼륨, 독시플루리신, 도세타키셀 수화물, 트레티노인, 네오카르티노스타틴, 네다플라틴, 바크리닥키셀, 비카르타미드, 피시바닐, 히드록시카르바미드, 부술판, 플루오로우라실, 플루타미드, 벤토스타틴, 폴피마나트륨, 마이토마이신 C, 미토브리니톨, 메토트렉세이트, 메르캅토푸린, 6-메르캅토푸린리보시드, 황산 블레오마이신, 황산 빈크리스틴, 황산 빈데신, 황산 빈브라스틴, 황산 페프로마이신, 렌티난 등의 항종양약과 조합해서 사용하면, 어느 한쪽만으로는 용이하게 달성할 수 없는, 상승적으로 높은 항종양 효과를 얻을 수 있다. 게다가, 이와 같은 병용은 항종양약의 용량을 저감할 수 있고, 그 결과로서, 항종양약에 의한 부작용을 현저하게 경감할 수 있는 실익이 있다.
본 발명의 감수성 질환제는, 경구경로에 적용해도, 그리고 비경구경로에 적용해도 감수성 질환의 치료·예방에 효과를 발휘한다. 감수성 질환의 종류나 증상에도 따르지만, 구체적으로는, 예를 들면, 환자의 증상이나 적용후의 경과를 관찰하면서, 체중 1 kg당 0.1 내지 1,000 ng/일, 바람직하게는 1 내지 100 ng/일의 생리활성 복합체를, 필요에 따라 복수회로 나누어 1 내지 7회/주의 빈도로 1주간 내지 반년간에 걸쳐서 경구경로에 적용하거나, 혹은, 피내(皮內), 피하, 근육내, 정맥내, 비강내, 직장내, 복강내 등의 비경구 경로에 의해 적용한다. 본 발명의 생리활성 복합체는 안정하며, 혈액중의 프로테아제 등에 의해 분해되기 어렵고, 게다가, 생체에 있어서의 체류 시간이 종양괴사 인자보다 유의하게 길며, 투여 경로에 따라서는 2배 이상이나 달하므로, 동일한 감수성 질환에 대하여 동일한 경로로 적용할 경우 투여량을 유의하게 적게 할 수 있고, 그 결과로서, 정상세포에 대한 세포 장해성 등에서 기인하는 부작용을 보다 적게 할 수 있는 실익이 있다.
이하, 본 발명의 실시의 형태에 대해, 실시예에 근거해서 설명한다.
[실시예 1]
<종양괴사 인자 활성을 가지는 단백질의 조제>
통상적인 방법에 따라 서열표에 있어서의 서열 번호 4 및 서열 번호 5로 나타내어지는 올리고뉴클레오티드를 프라이머로서 이용하고, Taq 폴리메라제에 의해 60℃에서 PCR 반응시킨 후, 반응 혼합물을 정제하여, 서열표에 있어서의 서열 번호 1로 나타내어지는 사람의 종양괴사 인자의 제65번째, 제90번째 및 제98번째의 리신이 랜덤한 아미노산으로 치환된 162 염기쌍의 PCR 산물을 채취하였다.
이어서 서열표에 있어서의 서열 번호 6 및 서열 번호 7로 나타내어지는 프라이머로서의 올리고뉴클레오티드의 존재하에, 위에서 얻어진 PCR 산물을 Taq 폴리메라제에 의해 60℃에서 다시 PCR 반응시킨 후, 반응 혼합물을 정제하여, 서열표에 있어서의 서열 번호 1로 나타내어지는 사람의 종양괴사 인자의 제65번째, 제90번째, 제98번째, 제112번째 및 제128번째의 리신이 모두 랜덤한 아미노산으로 치환된 251 염기쌍의 PCR 산물을 채취하였다.
별도로, 통상적인 방법에 따라 파아지미드 벡터(상품명 『pCantab5e』, Amersham Pharmacia사 제조)에 서열표에 있어서의 서열 번호 1로 나타내어지는 사람의 종양괴사 인자를 코드하는 DNA를 도입하고, 이것을 주형으로 하여, 위에서 얻은 프라이머로서의 251 염기쌍의 PCR 산물과 함께, 서열표에 있어서의 서열 번호 8 및 서열 번호 9로 나타내어지는 올리고뉴클레오티드의 존재하에 Accutaq DNA 폴리메라제에 의해 증폭하여, 서열표에 있어서의 서열 번호 1로 나타내어지는 아미노산 서열에 있어서의 제11번째, 제65번째, 제90번째, 제98번째, 제112번째 및 제128번째의 리신이 모두 랜덤한 아미노산으로 치환된 463 염기쌍의 PCR 단편을 얻었다.
그 후, 범용의 파아지 디스플레이법에 의해, 이 PCR 단편을 파아지미드 벡터에 도입하여, 6개의 리신의 일부 또는 전부가 랜덤한 아미노산으로 치환된 사람의 종양괴사 인자의 변이체를 발현한 파아지 라이브러리를 얻었다. 이 라이브러리에 대하여, 종양괴사 인자에 대한 항체를 센서 칩에 고정화한 시판의 표면 플라즈몬 공명 장치(상품명 『BIACORE 2000』, 비아코아 주식회사 판매)를 이용하는 파닝법을 반복하여 적용함으로써 항체에 결합하는 클론을 검색하고, 또한, 이렇게 하여 검색된 클론에 대하여, 표적세포로서 마우스 결합 조직 유래의 세포인 L-M 세포(ATCC CCLl.2)를 이용해서 세포 장해성을 측정하는 바이오어세이를 적용함으로써 종양괴사 인자 활성을 가지는 단백질을 코드하는 클론을 선별한 결과, 서열표에 있어서의 서열 번호 1로 나타내어지는 아미노산 서열에 있어서, 제11번째, 제65번째, 제90번째, 제98번째, 제112번째 및 제128번째의 리신이 아스파라긴, 아르기닌, 알라닌, 세린, 트레오닌, 프롤린, 메티오닌 또는 로이신 중의 어느 하나로 치환된 단백질이 높은 종양괴사 인자 활성을 유지하고 있는 것이 밝혀졌다.
따라서, 이와 같은 단백질을 코드하는 클론 하나에 대해, 가장 높은 종양괴사 인자 활성을 가지는 단백질을 코드하는 DNA의 부분을 프라이머로서의 서열표에 있어서의 서열 번호 10 및 서열 번호 11로 나타내어지는 올리고뉴클레오티드의 존재하에서 증폭한 후, 얻어진 서열표에 있어서의 서열 번호 12로 나타내어지는 염기서열을 가지는 DNA를 플라스미드 벡터(상품명 『pUC18』, 일본국의 寶酒造 주식회사 제조)에 도입하였다. 그 후, 통상적인 방법에 따라 이 플라스미드를 대장균 BL21DE3주에 도입하고, 얻어진 형질 전환체를 배양하고, 배양물을 이온 교환 크로마토그래피, 겔 여과 크로마토그래피 등을 이용하는 방법에 의해 정제해서 서열표에 있어서의 서열 번호 3으로 나타내어지는 아미노산 서열을 가지는 단백질을 채취하였다.
이렇게 하여 얻어진 단백질의 일부를 취하여, 2-메르캅토에탄올 존재하에서 SDS-PAGE 한 결과, 분자량 17,0000 달톤 부근에서 주된 밴드가 관찰되었다. 통상적인 플루오레스카민법에 의해 유리 아미노기를 측정한 결과, 본 실시예의 단백질은 1분자당, 유리 아미노기를 1 잔기 가지고 있었다. 그리고 통상적인 방법에 따라 L-M 세포를 표적세포로 하는 바이오어세이에 의해 세포 장해성을 조사한 결과, 대조로서의 사람의 재조합형 종양괴사 인자의 IC50 (표적세포의 생존을 50% 저해하는 시료의 농도)이 0.22 ng/ml이었던 것에 대해서, 본 실시예의 단백질은 그것과 거의 동등한 0.23 ng/ml이었다. 이들 결과는, 본 실시예의 단백질에 있어서는, 종양괴사 인자에 있어서의 유리 아미노기를 가지는 7개소(個所)의 아미노산 중에서, N 말단에 위치하는 아미노산을 제외하고 6개소의 리신 모두가 유리 아미노기를 가지지 않은 기타의 아미노산으로 치환되어 있는 것과, 그리고 이렇게 하여 창출된, 종양괴사 인자와는 구성 아미노산의 몇개가 상위한 본 실시예의 단백질이, 표적세포에 대하여 종양괴사 인자와 동등한 세포 장해 작용을 미치는 것을 나타내고 있다.
[실시예 2]
<생리활성 복합체의 조제>
실시예 1의 방법에 따라 얻어진 종양괴사 인자 활성을 가지는 단백질을 인산완충 생리 식염수 (pH 8.5)에 농도 0.1 내지 1 mg/ml이 되도록 용해한 후, 고분자로서 모노메톡시폴리에틸렌 글리콜 N-숙신이미딜 프로피오네이트에 의해 활성화시킨 메톡시폴리에틸렌 글리콜 (평균 분자량 5,0000 달톤)을 몰비로 단백질의 5배가 되도록 첨가하고 37℃ 에서 30분간 반응시켰다. 이어서, 반응 혼합물에 ε-아미노카프론산을 몰비로 고분자의 10배 가하고, 잠시 정치해서 반응을 정지시킨 후, 반응 혼합물을 겔 여과 크로마토그래피를 이용하는 범용의 방법에 의해 정제하여, 단백질 부분의 N 말단 부분에만 폴리에틸렌 글리콜이 결합해서 된, 분자 레벨의 불균일성을 가지지 않은 균질한 생리활성 복합체를 얻었다.
이렇게 하여 얻어진 복합체는, 2-메르캅토에탄올 존재하의 SDS-PAGE에 있어서 24,000 달톤 부근에서 주된 피크를 나타내었다. 표적세포로서 L-M 세포를 이용하는 앞서 나온 방법에 의해 세포장해 활성을 측정한 결과, 본 실시예의 복합체도, 그리고 대조로서의 사람의 재조합형 종양괴사 인자도, IC50은 함께 0.34 ng/ml이며, 양자는 동등한 활성을 유지하고 있었다. 즉, 폴리에틸렌 글리콜의 평균 분자량을 40,000 달톤으로 변경해서 조제한 다른 생리활성 복합체에 대해 상기와 마찬가지로 해서 L-M 세포에 대한 세포 장해성을 시험한 결과, 그 IC50은 0.43 ng/ml이며, 대조의 종양괴사 인자의 활성을 약 70% 유지하고 있었다.
[실시예 3]
<항종양 작용>
통상적인 방법에 따라 마우스 육종세포의 일종인 methA (ATCC63181)를 이식한 한 무리의 BALB/c 마우스 (암컷, 4주령, 체중 20 g, 3마리/군)에 대하여, 이식으로부터 7일 후에, 꼬리정맥에 실시예 2의 방법으로 얻은 생리활성 복합체, 실시예 1의 방법으로 얻은 종양괴사 인자 활성을 가지는 단백질 또는 사람의 재조합형 종양괴사 인자 중의 어느 하나의 생리 식염수 용액을 표 1에 나타내는 투여량으로 주사 투여하였다.
투여로부터 24시간 후에 출혈 괴사와 돌연사의 유무를 조사함과 아울러, 계속하여 종양 지름을 경시적으로 측정하여, 케이스케·하라나카 등의 『인터내셔널·저널·오브·메디슨』, 제34권, 263 내지 267 페이지 (1984년)에 기재된 방법에 따라 종양체적을 계산하였다. 그리고 종양 이식후 20일째의 종양체적을 생리 식염수만을 주사 투여하는 대조의 계와 비교함으로써 항종양 효과(%)를 산출하였다. 그 결과를 표 1에 나타낸다.

표 1의 결과에서 나온 바와 같이, 실시예 1의 방법으로 얻은 단백질 또는 사람의 재조합형 종양괴사 인자 중의 어느 하나를 투여한 군은 모두, 10 ㎍/마리 투여하면 투여로부터 24시간 이내에 출혈 괴사를 초래하여 모두가 돌연사하였다. 투여량을 단계적으로 감소한 결과, 돌연사는 회피할 수 있었지만, 종양의 완전 축퇴(縮退)는 나타나지 않고 대다수가 40일 이내에 사망하였다. 한편, 본 발명에 의한 생리활성 복합체를 투여한 군에서는, 10 ㎍/마리 투여해도 돌연사는 전혀 나타나지 않고, 또한, 3 ㎍/마리의 투여에 의해서도 현저한 항종양 효과가 확인되었다.
이 것은 본 발명의 생리활성 복합체가 종양괴사 인자 활성을 가진 단백질의 항종양 효과를 효과적으로 나타내면서 그 부작용을 현저하게 저감하는 것을 나타내고 있다.
[실시예 4]
<체내 동태>
BALB/c 마우스로부터 채취한 혈청과, 생리 식염수에 의해 3.3 ㎍/ml로 희석한 실시예 2의 방법으로 얻은 생리활성 복합체를 체적비 3:1의 비율로 혼합하고, 일정간격을 두어 샘플링 하면서 37℃에서 9시간 인큐베이트하였다. 채취한 샘플을 즉시 L-M 세포를 표적세포로 하는 세포 장해성 활성의 측정에 사용하여 잔존하는 세포장해 활성을 판정함으로써 안정성의 목표로 하였다. 이와 병행하여 생리활성 복합체 대신에 사람의 재조합형 종양괴사 인자 또는 실시예 1의 방법으로 얻은 단백질을 투여하는 계를 각각 마련하고, 이들을 상기와 마찬가지로 처치해서 대조로 하였다. 그 결과를 도 1에 나타낸다. 그리고 도 1에 있어서, 생리활성 복합체, 사람의 재조합형 종양괴사 인자 및 실시예 1의 방법으로 얻은 단백질을 투여한 계를, 각각, 「」, 「」 및 「△」로서 표시하였다.
도 1에 나온 바와 같이, 대조에 있어서는, 종양괴사 인자 및 단백질이 용이하게 분해되고, 9시간 인큐베이트하면, 세포 장해성 활성이 3% 전후까지 저하하였다. 이것에 대하여, 본 발명의 생리활성 복합체는, 마찬가지로 9시간 인큐베이트해도 여전히 20% 이상의 세포장해 활성을 유지하고 있었다. 이 것은, 본 발명의 생리활성 복합체가 혈액중의 프로테아제 등에 의해 분해되기 어렵고 안정하다는 것을 나타내고 있다.
더욱이 실시예 2의 방법으로 얻은 생리활성 복합체를 농도 5 ㎍/ml이 되도록 생리 식염수로 희석하고, BALB/c 마우스의 꼬리 정맥에 200 ㎕/마리의 용량으로 주사 투여하였다. 그 후, 3시간에 걸쳐서 마우스로부터 일정간격을 두고 혈액을 채취하고, 중화 항체를 사용해서 복합체의 혈청 레벨을 측정하였다. 이와 병행하여 생리활성 복합체 대신에 사람의 재조합형 종양괴사 인자 또는 실시예 1의 방법으로 얻은 단백질을 투여하는 계를 마련하고, 이들을 상기와 마찬가지로 처치해서 대조로 하였다. 그 결과를 도 2에 나타낸다. 또한, 도 2에 있어서, 생리활성 복합체, 사람의 재조합형 종양괴사 인자 및 실시예 1의 방법으로 얻은 단백질을 투여한 계를, 각각, 「」, 「」 및 「△」로서 표시하였다.
도 2의 결과에 나온 바와 같이 대조에 있어서는, 투여 직후로부터 종양괴사 인자 및 단백질의 혈청 레벨이 급격하게 저하하고, 3시간 경과한 시점에서 당초의 10% 이하로까지 저하하고 있었던 것에 대해서, 본 발명의 생리활성 복합체는 당초의 혈청 레벨의 50%강(强)을 유지하고 있었다. 이 것은, 본 발명의 생리활성 복합체가 종래 공지의 종양괴사 인자나 N 말단에 고분자 부분을 가지지 않은 종양괴사 인자의 변이체에 비하여 체내 동태가 유의하게 우수하고, 주사 투여하여도 생체에의해 길게 체류할 수 있는 것을 나타내고 있다.
[실시예 5]
<급성 독성>
통상적인 방법에 따라 8주령의 마우스에 실시예 2의 방법으로 얻은 생리활성 복합체를 경피, 경구 또는 복강 경로로 주사 투여한 결과, 생리활성 복합체의 LD50은 어떠한 투여 경로에 의해서도 1 mg/kg 체중 이상이었다. 이 것은, 본 발명의 생리활성 복합체가 사람에 대한 투여를 전제로 하는 의약품에 배합해서 안전하다는 것을 나타내고 있다.
[실시예 6]
<액제>
안정제로서 1%(w/w) 사람혈청 알부민을 함유하는 생리 식염수에 실시예 2의 방법으로 얻은 생리활성 복합체를 1 mg/ml이 되도록 용해하고, 통상적인 방법에 따라서 정밀여과에 의해 멸균해서 액제를 얻었다.
안정한 이 제품은, 악성 종양, 바이러스성 질환, 세균 감염증 및 면역질환을 비롯한 감수성 질환을 치료·예방하기 위한 주사제, 점안제 및 점비제 등으로서 유용하다.
[실시예 7]
<건조 주사제>
안정제로서 1%(w/w) 정제 젤라틴을 함유하는 생리 식염수 100 ml에 실시예 2의 방법으로 얻은 생리활성 복합체를 100 mg 용해하고, 통상적인 방법에 따라서정밀여과에 의해 멸균하여 바이알 병에 1 ml 씩 나누어 넣고, 동결 건조시킨 후, 마개로 밀봉하여 건조 주사제를 얻었다.
안정한 이 제품은, 악성 종양, 바이러스성 질환, 세균 감염증 및 면역질환을 비롯한 감수성 질환을 치료·예방하기 위한 건조 주사제로서 유용하다.
[실시예 8]
<연고제>
멸균 증류수에 카르복시비닐 폴리머 (상품명 『하이비스와코』, 일본국의 주식회사 和光純藥공업 제조)와, 파이로젠을 제거한 고순도 트레할로오스 (상품명 『Treha』, 일본국의 주식회사 林原 제조)를 각각 농도 1.4%(w/w) 및 2.0%(w/w)이 되도록 용해한 후, 실시예 2의 방법으로 얻은 생리활성 복합체 적당량을 균일히 혼합하여 pH 7.2로 조제하여 1 g당 생리활성 복합체를 약 5 ㎍ 함유하는 페이스트상 연고제를 얻었다.
연전성(延展性)과 안정성이 우수한 이 제품은, 악성 종양, 바이러스성 질환, 세균 감염증 및 면역질환을 비롯한 감수성 질환을 치료·예방하기 위한 연고제로서 유용하다.
[실시예 9]
<정제>
무수 결정성 α-말토오스 분말 (상품명 『파인토스』, 일본국의 주식회사 林原 제조)에 실시예 2의 방법으로 얻은 생리활성 복합체 적당량을 균일히 혼합하고, 얻어진 혼합물을 통상적인 방법에 의해 타정(打錠)해서 제품 1정(錠) (약 200 mg)당 생리활성 복합체를 약 1 ㎍ 함유하는 정제를 얻었다.
섭취성, 안정성이 우수한 이 제품은, 악성 종양, 바이러스성 질환, 세균 감염증 및 면역질환을 비롯한 감수성 질환을 치료·예방하기 위한 정제로서 유용하다.
이상 설명한 바와 같이, 종양괴사 인자 활성을 가지는 단백질 부분과, 그 단백질의 N 말단에 인위적으로 결합시킨 고분자로 된 본 발명의 생리활성 복합체는, 체내 동태가 우수하고, 주사 투여해도 당초의 혈청 레벨을 장시간 지속한다. 따라서 이와 같은 생리활성 복합체를 유효성분으로 하는 본 발명의 감수성 질환제는 의약품 분야에 있어서, 예를 들면, 항종양제, 항바이러스 질환제, 항감염증제, 면역 질환제 등으로서 다종 다양한 용도를 가진다.
더욱이 본 발명의 생리활성 복합체는, 종양괴사 인자 활성을 가지는 단백질의 N 말단에만 고분자가 결합해서 되는 것이므로, 원리상, 림포카인 활성을 가지는 종래 공지의 복합체에서 우려되는 분자 레벨의 불균일성을 가지지 않는다고 하는 특징이 있다. 또한, 본 발명의 생리활성 복합체를 구성하는 단백질로서, 종양괴사 인자에 있어서의 유리 아미노기를 가지는 아미노산 중에서 N 말단에 위치하는 것을 제외하고, 그 이외가 유리 아미노기를 가지지 않는 기타의 아미노산으로 치환된 단백질을 사용할 때에는, N 말단에만 고분자가 결합한 생리활성 복합체를 극히 고수율로 얻을 수 있다. 이 때, 유리 아미노기를 가지지 않은 아미노산으로서, 예를 들면, 아스파라긴, 알라닌, 아르기닌, 세린, 트레오닌, 프롤린, 메티오닌, 로이신 등을 채용할 때에는, 종양괴사 인자의 바람직한 생리작용을 그대로 유지하거나, 극히 고레벨로 유지하는 생리활성 복합체를 거의 이론대로의 수율로 얻을 수 있다.
이렇게도 현저한 효과를 나타내는 본 발명은, 이 분야에 공헌하는 바 실로 다대한 의의가 있는 발명이라고 할 수 있다.
Claims (5)
- 종양괴사 인자 활성을 가지는 단백질 부분과, 그 단백질의 N 말단에 인위적으로 결합시킨 고분자 부분으로 된 생리활성 복합체.
- 제1항에 있어서, 단백질 부분이 서열표에 있어서의 서열 번호 2로 나타내어지는 아미노산 서열을 가지는 생리활성 복합체.(서열 번호 2로 나타내어지는 아미노산 서열에 있어서, Ⅹaa는 아스파라긴, 알라닌, 아르기닌, 세린, 트레오닌, 프롤린, 메티오닌 또는 로이신 중의 어느 하나인 것으로 한다.)
- 제1항 또는 제2항에 있어서, 고분자 부분이 폴리에틸렌 글리콜의 단독 중합체 혹은 공중합체 또는 이들의 유도체인 생리활성 복합체.
- 청구항 1 내지 3 중의 어느 하나에 기재한 생리활성 복합체를 유효 성분으로 하는 감수성 질환제.
- 제4항에 있어서, 항(抗)종양제로서의 감수성 질환제.
