Abstract
본 발명은 신규한 아폽토시스 유도제에 관한 것으로, 보다 상세하게는 육산화사비소로 이루어진 아폽토시스 유도제 및 이를 함유하는 약제학적 조성물에 관한 것이다. 본 발명에 따르는 아폽토시스 유도제는 고형암은 물론 백혈병을 포함하는 각종 혈액암에 세포사멸을 유도함으로써 이들 질병의 치료제로서 사용할 수 있다.
Description
본 발명은 육산화사비소(solid As4O6)의 아폽토시스 유도제 및 항암제로서의 새로운 용도 및 그 약학적 조성물에 관한 것으로, 보다 상세하게는 종래에 천지산이라는 상표명으로 알려진 육산화사비소가 아폽토시스라는 세포의 능동적 자살기전을 유도함으로써 백혈병과 같은 혈액암 치료제로서 사용할 수 있는 새로운 용도에 관한 것이다.
항암제의 개발은 아폽토시스에 관한 연구가 진행되기 이전부터 실시되어 왔으며 최근 항암제가 암세포의 아포토시스를 유도하여 항암작용을 나타낸다는 것이 공론화 되어있다. 그러나 항암제에 의한 아폽토시스의 정확한 기전이 밝혀지지 않은 상태에 있을 뿐 아니라 기존 항암제에 대한 저항성 암세포의 출현 및 항암제 자체의 독성으로 인하여 새로운 항암제의 개발 및 항암제 유도 아폽토시스 기전을 파악하는 것이 필수 불가결한 시점에 있다.
세포사(細胞死)로 생각되던 괴사(necrosis)는 화상, 허혈, 독극물 등의 자극에 일어나는 수동적 세포붕괴과정으로 세포의 팽화, 용해에 의해 세포질 성분의 유출로 인하여 염증반응을 일으킨다. 반면에 아폽토시스(apoptosis)는 세포의 능동적 자살기전으로 괴사와는 달리 세포의 축소, 핵의 농축 및 DNA의 단편화 등이 발견되며 사멸체 (apoptotic body)라는 포낭을 형성하여 식세포 작용에 의해 제거되기 때문에 생체내에서는 염증반응을 일으키지 않는다. 이러한 아포토시스는 다세포 생명체의 발달, 분화 및 항상성(homeostasis)을 유지하는 데 필수적이다.
전통적으로 동양의학에서는 비소화합물을 항암제로 사용되어 왔으며 1970년대에 들어 비소화합물의 약효 주성분은 삼산화비소(As2O3)로 확인되었다. 최근 임상적으로 적용가능한 농도(0.5-2μmol/L)의 비소투여로 트랜스형 레티노산(all-trans retinoic acid)에 저항성이 있는 급성 전골수세포성 백혈병(acute promyelocytic leukemia; APL) 환자를 치료하는 데 매우 효과적이라는 것이 밝혀졌다 (Gallagher, R. Blood, 88: 1052-1061, 1996). 비소에 의한 급성 전골수세포성 백혈병 치료의 성공은 새로운 개념의 항암제의 등장이라는 획기적인 일로 받아들여지고 있으며 기타 혈액암과 고형암에 그 적용범위를 넓히려는 연구가 활발히 진행중에 있다.
그러나 기존의 비소화합물인 삼산화비소로 백혈병 환자를 치료하는데 있어 제한이 있다. 즉, 장시간 처리시 저항성 세포가 돌출 (Gianni, M., Koken, M. H. M., Chelbi-Alix, M. K., Benoit, G., Lanotte, M., Chen, Z., de The, H. 육산화사비소(고체 As4O6)는 삼산화비소 및 대부분의 다른 비소 화합물을 제조하기 위한 기초원료 물질로서 사용되어 왔으며 살충제나 유리 제조시 탈색제 또는 가죽제품의 보존제로서 사용된다. 이러한 육산화사비소는 삼산화비소와 마찬가지로 암을 유발하는 독성 물질로서만 알려져왔으며, 그 화학적 구조에 관한 것만이 화학자들의 관심의 대상이 되어 왔다.(Becker K.A., Plieth K., and Stranski I.N. 한편, 본 발명자는 자연산 신석으로부터 육산화사비소를 분리정제하여 항암효과를 확인하여 대한민국 특허출원 제1998-16486호로 특허출원하였으며, 1999년에는 이 육산화사비소의 항암제로서의 용도에 대하여 일본 특허청으로부터 특허(일본특허 제3007627호)를 받은 바 있다.
특히 삼산화비소는 경구투여할 경우 심각한 위장관 및 간 부작용이 보고된 바 있으나(Shen ZX, Chen GQ, Ni JH, Li XS, Xiong SM, Qiu QY, Zhu J, Tang W, Sun GL, Yang KQ, Chen Y, Zhou L, Fang ZW, Wang YT, Ma J, Zhang P, Zhang TD, Chen SJ, Chen Z, Wang ZY Use of arsenic trioxide (As2O3) in the treatment of acute promyelocytic leukemia (APL): II. Clinical efficacy and pharmacokinetics in relapsed patients. Blood 1997 May 1;89(9):3354-60), 이건 발명자는 상기 특허를 통해서 약 200g의 스프라그 돌리(Sprague-Dawley) 랫트에 고용량, 즉 약 100mg/kg 용량의 육사화사비소를 28일간 경구투여한 경우 신장에만 미세한 조직학적 변화를 일으켰을 뿐 위장관 및 간에 부작용을 일으키지 않았으며, 중간용량(약 10mg/kg) 및 저용량(1mg/kg)에서는 전혀 부작용이 나타나지 않는다는 것을 확인하였다.
따라서 본 발명의 목적은 육산화사비소로된 아폽토시스 유도제를 제공함으로서, 백혈병을 포함하는 각종 혈액암 및 고형암들을 치료할 수 있는 육산화사비소의새로운 용도를 제시하는 것이다.

상기 목적을 달성하기 위하여 본 발명자는 첨부된 도 1과 같은 구조를 갖는 육산화사비소를 이용하여 백혈병 세포인 U937 세포주를 대상으로 아폽토시스 유도 및 그 기전을 관찰한 결과, 육산화사비소가 상대적으로 낮은 농도에서 아폽토시스를 유도하는 사실을 확인하였다.
본 발명자가 백혈병 세포에서 DNA 단편화 및 FITC-아넥신 V로 DNA를 염색하여 확인한 결과, 본 발명에 따르는 육산화사비소는 미토콘드리아 막으로부터 시토크롬 c 유출을 유도하여 카스파제(caspase)를 활성화시킴으로써 아폽토시스를 일으킨다는 것을 확인하였다. 이러한 일련의 아폽토시스 기전은 활성산소, 특히 과산화수소의 발생을 저해시켰을 때에 완전히 저해되었을 뿐만 아니라, 육산화사비소 처리후 과산화수소 발생을 측정한 결과 30분 경과후부터 과산화수소 발생의 증가가 관찰되었다. 이로부터 본 발명에 따르는 육산화사비소는 암세포로부터 과산화수소의 과량발생을 포함하는 활성산소의 발생을 유발시킴으로써 아폽토시스를 일으킨다는 것을 확인할 수 있었으며, 이로부터 백혈병을 포함하는 각종 혈액암을 치료할 수 있는 효능을 가짐을 알 수 있었다.
본 발명자가 실험한 U937 백혈병 세포는 종래의 백혈병 치료효과가 있는 것으로 알려진 삼산화비소에 대해 저항성이 있는 세포주일 뿐만 아니라 PML-RARα 퓨전 단백질을 발현하지 않는 세포주이다. 따라서, PML 환자 이외에 다른 백혈병 환자의 치료에 대해서도 효능이 우수할 것으로 생각된다.
본 발명에 따르는 육산화사비소는 일명 천지산으로 알려져 있는 물질로서, 특히 그 제조방법에 대해서는 본 발명자의 특허출원 제1998-16486호를 통해 알려져 있다. 즉, 본 발명에 따르는 아폽토시스 유도 효과를 갖는 육산화사비소는 비소를 함유하는 천연산 신석 및 시약용 비소 등을 다단계의 가열처리를 통하여 백색의 천연 화학물질인 육산화사비소를 제조할 수 있으며, 본 발명에서는 보다 상세한 제조방법에 대한 기술은 생략하겠다.
또한, 본 발명에 따르는 아폽토시스 유도 효과를 갖는 육산화사비소는 상기 제조방법 이외에 다른 제조방법에 의해 제조된 것이라도 첨부된 도 1의 구조를 갖는 것이라면 사용할 수 있다.
본 발명에 따르는 육산화사비소는 인체에 적용할 경우에는 질환의 종류, 환자의 연령, 성별, 건강상태 등에 따라 그 용량을 증감 조절할 수 있다.
본 발명에 따르는 육산화사비소는 단독으로 또는 다른 항암제와 복합제제로서 또는 병행하여 사용될 수 있으며, 부형제, 붕해제, 방향제, 활택제 등 기타 약제학적으로 허용되는 각종 첨가물과 혼합하여 산제, 정제, 액제 등 각종 경구 또는 비경구용 제제로 제제화할 수 있다.
본 발명에 따르는 육산화사비소는 아폽토시스와 관련있는 것으로 알려져 있는 각종 질환, 예컨대 백혈병을 포함한 각종 혈액암은 물론 고형암에도 치료제로서 적용이 가능하다. 이 경우에는 본 발명에 따르는 육산화사비소는 해당 질환과 관련된 다른 치료제와 병행 또는 혼합하여 사용할 수 있다.
이하 실시예를 통해 본 발명에 따르는 육산화사비소로된 아폽토시스 유도제에 대하여 보다 상세하게 설명하며 본 발명의 권리범위는 이들 실시예에만 한정되는 것은 아니다.
실시예 1: 아폽토시스 측정실험
본 발명에 따르는 육산화사비소의 아폽토시스 유도 효과 확인 실험을 하였다. 본 실시예를 포함한 이하 실시예에서 사용한 시약 및 세포는 아래와 같은 방법으로 입수하였다.
1) 시약 및 세포배양
육산화사비소는 주식회사 천지산(Seoul, Korea)으로부터 제공받아 1 M NaOH 용액에 녹인 후 이를 희석하여 실험에 사용하였다. Z-DEVD-FMK와 N-아세틸-N-시스테인(NAC)은 캘바이오켐사(Calbiochem, SanDiego, CA, USA)에서 카탈라제는 시그마(Sigma Co.)에서 카스파제-3와 시토크롬 c 항체는 파민젠사(Pharmingen, San Diego, CA, USA)에서 구입하여 웨스턴 블랏팅(Western blotting)에 사용하였다. 미토콘드리아 막 전위 손실을 관찰하기 위한 3, 3'-디헥실옥사카르보시아닌 요다이드(3,3'-dihexyloxacarbocyanine iodide)와 과산화수소 발생을 측정하기 위한 2',7'-디클로로디하이드로플루오레세인 디아세테이트(2',7'-dichlorodihydrofluorescein diacetate)는 몰레큘러 프로브사(Molecular Probes, Eugene, OR, USA)에서 구입하였다.
U937 세포주는 골수성 백혈병 세포주로 한국 세포주은행(Cancer ResearchCenter, Seoul National Medical College, Seoul, Korea)에서 분양받았으며, 10% 소태아혈청(fetal calf serum, GIBCO, Grand Island, NY, USA) 및 페니실린/스트렙토마이신이 첨가된 RPMI-1640 배지(GIBCO)로 가습시킨 5% CO2, 37℃ 배양기에서 계대 배양하며 실험에 사용하였다.
이하 실시예에서는 본 발명의 육산화사비소의 아폽토시스 효능을 검증하기 위하여 U937세포주에 일정농도의 육산화사비소를 첨가하여 10% 소태아혈청(fetal calf serum, GIBCO, Grand Island, NY, USA) 및 페니실린/스트렙토마이신이 첨가된 RPMI-1640 배지(GIBCO)로 가습시킨 5% CO2, 37℃에서 배양하였는바, 이하 이러한 과정을 육산화사비소로 처리한다고 표현한다.
2) 아폽토시스의 측정
U937세포주 1 X 105세포에 육산화사비소를 0, 0.5, 1.0, 2.5, 5.0μM의 농도로 처리하여 18시간 배양후 아폽토시스를 관찰하기 위하여 PBS로 수세한 후 결합 완충액(binding buffer)[10 mM HEPES/NaOH(pH 7.4), 140 mM NaCl, 2.5 mM CaCl2]에 부유시켰다. 그리고 나서, 5μl의 아넥신(annexin) V-FITC와 프로피듐 요다이드(propidium iodide: PI)(PharMingen, SanDiego, CA, USA)를 첨가한 후 15분 동안 실온에서 반응시킨 다음, 세포유동분석법으로 측정하였다. 그 결과를 도 2a 및 2b에 그래프로 도시하였다.
도 2a에 나타낸 바와 같이 본 발명에 따르는 육산화사비소의 아폽토시스 유도는 0.5~5μM의 농도에서 농도 의존적인 것으로 확인되었다. 또한, 도 2b로부터 확인되는 바와 같이 대조군에서는 5% 세포만이 아넥신 V-FITC/PI 염색이 관찰되는 반면, 1μM의 육산화사비소에서는 53%의 세포가 염색되어 더 많은 세포가 사멸된 것을 알 수 있었다.
3) DNA 단편화 분석
1 X 105세포에 육산화사비소를 0, 0.5, 1, 2.5 및 5μM의 농도로 처리한 다음, PBS로 수세한 후 10 mM Tris, pH 8.0, 1 mM EDTA, 1% SDS로 구성된 분해 완충액(lysis buffer)로 처리하였다. 세포 파쇄액을 DN아제-프리 프로테인아제 K(DNase-free proteinase K)로 처리한 후 55℃에서 1시간 동안 반응시켰다. 반응 후 페놀/클로로포름으로 처리하여 5M NaCl과 이소프로필알콜로 DNA를 침전시킨 후 증류수에 녹였다. DN아제-프리 RN아제(10 units)로 RNA를 제거한 후 1.8% 아가로스 겔에서 전기영동하여 에틸브로마이드(C2H5Br)로 염색하여 UV에서 DNA를 확인하였다.
그 결과 도 2c로부터 확인되는 바와 같이 1μM의 농도에서 DNA 단편화가 관찰되었다.
이상 실시예 1의 실험 결과로부터 육산화사비소가 아폽토시스를 유도하는 것으로 판단되었다.
실시예 2: 미토콘드리아 막 전위 손실의 측정
아폽토시스 유도과정에는 미토콘드리아가 긴밀하게 관여되어 있음이 알려져 있는 바, 본 발명에 있어서 육산화사비소에 의한 아폽토시스에서 미토콘드리아의 막전위 손실이 유도되는지를 관찰하기 위하여 U937 세포주를 2.5μM의 육산화사비소로 처리한 후 시간별로 미토콘드리아의 막 전위 손실을 관찰하였다.
육산화사비소를 U937 세포주 1 X 105세포에 2.5μM의 농도로 처리한 후 미토콘드리아에 축적되는 형광성 물질인 3,3'-디헥실로사카보시아닌 요다이드를 50 nM 농도로 세포배양액에 첨가하여 37℃에서 15분동안 반응시킨 다음, PBS로 수세한 후 세포유동분석법으로 분석하여 시간대별로 측정한 결과를 도 3에 나타냈다.
도 3을 살펴보면 2.5μM 농도로 육산화사비소를 첨가한 후 6시간째부터 미토콘드리아의 막 전위가 손실되는 것을 확인할 수 있다.
실시예 3: 시토크롬 c의 세포질내 유출 측정
막전위 손실에 의해 미토콘드리아의 막이 와해되면 시토크롬 c가 미토콘드리아로부터 유출되어 아폽토시스 신호전달을 초기화시키는 것으로 보고되고 있다. 본발명에 따르는 육산화사비소의 아폽토시스 유도에서도 시토크롬 c의 세포질내 유출이 관계되는지 확인하는 실험을 하였다.
U937세포주 1x105세포에 육산화사비소를 2.5μM 농도로 일정시간 동안 처리한 다음, 1200 rpm에서 5분간 원심분리하여 세포 펠릿을 만들었다. 220 mM 만니톨, 70 mM 수크로즈, 50 mM Pipes-KOH (pH 7.4), 50 mM KCl, 5 mM EGTA, 2 mM MgCl2, 1mM EDTA, 1 mM 디티오트레오톨과 다종의 프로테아제 저해제가 포함된 용액에 상기 세포 pellet를 용해한 후 균질화(homogenization)하여 원심분리하고 상등액을 취하여 웨스턴 블랏팅을 실행하였다.
웨스턴 블랏팅은 시료들을 트리스-글리신 완충액(Tris-glycine running buffer)에서 10-12% SDS-PAGE(sodium dodecyl sulfate-polyacrylamide electrophoresis)를 실행하고 0.45μM 프로트론 니트로셀룰로스(protron nitrocellulose, Schleicher & Schuell, Germany)에 전기영동 시켰다. 전기영동된 멤브레인을 30 분간 블로킹 완충액(5% skim milk)에서 블로킹하고 분석하고자 하는 단백질의 1차 항체를 1;1,000 비율로 블로킹 완충액에 희석하여 1 시간 동안 반응시켰다. 1차 항체가 결합된 멤브레인을 PBST로 20 분간 3회 반복 수세하고 양고추냉이 퍼옥시다제(horse-radish peroxidase)가 접합된 2차 항체를 1;3,000 비율로 블로킹 완충액에 희석하여 30 분 동안 반응시켰다. 반응 후 PBST로 20 분간 3회 반복 수세 후 ECL 시스템(Amershame, Arlington Heights, IL, USA)을 이용하여 발색시켰다. 결과 사진을 도 4에 나타냈다.
도 4로부터 확인되는 바와 같이 2.5μM 의 육산화사비소로 처리후 세포질내의 시토크롬 c가 관찰되었다. 이러한 결과로부터 육산화사비소 유도 아폽토시스는 미토콘드리아 막 전위 손실에 따른 시토크롬 c의 유출이 관여되는 것으로 확인되었다.
실시예 4: 카타파제-3 활성화
1) 카타파제-3 활성화 측정
카타파제-3의 활성화가 육산화사비소에 의해 유도된 아폽토시스에 관여하는 지를 관찰하기 위하여, 활성화된 카타파제-3과 공유결합하여 형광성을 나타내는 Z-DEVD-FMK 유도체인 FAM-DEVD-FMK를 육산화사비소로 처리한 세포배양액에 첨가하여 형광성을 관찰하였다.
카타파제-3의 활성화를 측정하기 위한 시약으로는 CaspaTagTM카타파제 활성 키트(Intergen, Purchase, NY, USA)를 사용하였다. 제조사의 프로토콜에 따라 1 X 105세포에 육산화사비소를 2.5μM 농도로 18시간동안 처리한 다음, 10μl의 FAM-DEVD-FMK를 배양액에 첨가한 후 1시간동안 배양기에서 반응시켰다. 수세 용액으로 수세한 후 세포유동분석법으로 분석하였다.
도 5a은 이상과 같이 육산화사비소를 처리한 세포와 처리하지 않은 세로(대조군)을 FAM-DEVD-FMK로 표식하여 세포유동분석법으로 분석한 그래프이다. 이 그래프에서 카타파제-3이 활성화되지 않은 세포는 X축의 첫 구획(M1)에 나타나고, 카타파제-3 활성화된 세포는 뒷 구역(M2)에 나타난다. 도 5a로부터 확인되는 바와 같이 2.5μM의 육산화사비소를 18시간 처리한 경우 세포의 19%가 FAM-DEVD-FMK가 염색되었다. 이로부터 U937 세포에 육산화사비소에 의해 유도되는 아폽토시스는 카스파제-3의 활성화와 관련이 있는 것을 알 수 있다.
또한, 카타파제-3의 활성을 형광현미경으로 관찰하기 위하여 훽스트 3342 (Hoechst 33342)와 PI를 첨가하여 5분동안 추가로 반응시켜 핵을 염색한 후 형광현미경으로 관찰하였다. 그 결과 사진을 도 5b로 나타냈으며, 카타파제-3가 활성화가 있는 세포는 초록의 형광성을 나타내고 PI 염색된 죽은 세포들은 붉은 형광성을 나타낸다.
2) 웨스턴 블랏팅 분석
육산화사비소를 농도별로 18시간 동0, 0.5, 1, 2.5 및 5μM농도로 처리한 세포 시료들을 카타파제-3 활성 검출 키트를 사용하여 상기 실시예 3과 같은 방법으로 웨스턴 블랏팅을 하여 찍은 사진을 도5c에 나타냈다.
도 5c로부터 확인되는 바와 같이 천지산 처리 농도가 높아질수록 카타파제-3 가 활성화됨으로써 기질이 분해되어 밴드가 약하게 나타나는 것을 확인할 수 있다.
실시예 5: 카스파제-3 의존성 확인 실험
카스파제-3 저해제인 Z-DEVD-FMK를 이용하여 육산화사비소에 의한 아폽토시스 유발작용이 카스파제-3의 활동차단 여부에 따라 영향을 받는지 여부를 검사하였다. 육산화사비소 1μM로 처리한 군과 육산화비소 1μM와 Z-DEVD-FMK로 아폽토시스를 억제한 군 및 정상 대조군에 대하여 실시예 1의 2)의 방법과 같이 아넥신-FITC V/PI 염색을 하여 세포유동분석법으로 사멸된 세포수를 측정하였다.
또한, 상기 3개의 실험군에 대하여 실시예 2과 동일한 방법을 사용하여 미토콘드리아 막전위 손실을 측정하여 도 6에 나타냈다.
도 6으로부터 확인되는 바와 같이, 대조군에 비하여 육산화사비소로 처리한군에서는 39.21%의 세포가 사멸되고, 미토콘드리아 막전위가 손실되는 것을 알 수 있다. 그리고, 카스파제-3 억제제인 Z-DEVD-FMK를 200μM까지 처리했으나 아폽토시스가 저해되지 않는 것으로 보아 육산화사비소의 아폽토시스 유도는 카스파제-3에 비의존적인 것으로 판단된다.
실시예 6: 활성산소와의 연관성 실험
1) 항산화제에 의한 아폽토시스 저해
U937 세포에서 육산화사비소 유도 아폽토시스 과정중 미토콘드리아 막 전위 손실이 세포내 활성산소(reactive oxygen species) 발생과 관련이 있는지를 관찰하기 위하여 항산화제인 N-아세틸-L-시스테인(NAC)을 전처리하여 육산화사비소 유도 아폽토시스의 정도를 관찰하였다.
육산화비소를 넣기 30분 전에 NAC로 전처리한 U937세포 (1 X 105세포)에 2.5μM의 육산화사비소로 처리한 다음, 실시예 1의 2)의 방법과 같이 아넥신-FITC V/PI 염색을 하여 세포유동분석법으로 사멸된 세포수를 측정하였다. 실험을 3회 반복하여 대조군의 사멸세포수를 1로 하였을 때의 비율을 환산하여 도 7a로 나타냈다.
또한, 상기 처리된 세포에 대하여 미토콘드리아 막전위 소실을 실시예 2와 동일한 방법으로 측정하여 도 7b에 나타냈다.
도 7a 및 도 7b에서 보는 바와 같이 25 mM의 NAC에 의해서 육산화사비소 유도 아폽토시스가 완전히 저해되었다. 이 결과로부터 활성산소의 발생이 육산화사비소 유도 아폽토시스와 관련이 있을 것으로 판단된다.
2) 과산화수소 발생 확인실험
육산화사비소 유도 아폽토시스가 과산화수소의 발생과 관련이 있는지를 확인하기 위하여 U937세포를 육산화사비소로 처리한 후 과산화수소의 발생을 측정하였다.
세포내 과산화수소의 발생을 측정하기 위하여 과산화수소에 특이적으로 결합하여 형광성을 나타내는 물질인 2',7'-디클로로디하이드로플루오레세인 디아세테이트(Molecular Probes, Eugene, OR, USA)를 사용하였다. U937 세포주에 2.5μM의 육산화사비소를 각각 0.5, 1, 2, 3, 5 시간 처리한 군 각각에 20μM의 2',7'-디클로로디하이드로플루오레세인 디아세테이트를 배양액 내에 첨가한 후 30분 동안 반응시킨 다음 세포유동분석법으로 분석하였다. 그 결과를 나타낸 도 7c로부터 확인되는 바와 같이 대조군에 비하여 30분째부터 과산화수소의 발생이 증가되었으며, 1시간째에는 과산화수소의 발생이 약 2배로 증가되었다.
3) 카탈라제에 의한 아폽토시스 차단효과
과산화수소 발생이 육산화사비소 유도 아폽토시스에 필수적인지를 관찰하기 위하여 과산화수소를 물과 산소로 분해하는 효소인 카탈라제를 전처리하여 육산화사비소 유도 아폽토시스를 관찰하였다.
육산화사비소를 넣기 30분 전에 카탈라제로 전처리한 U937세포(1 X 105세포)에 2.5μM의 육산화사비소로 처리한 다음, 실시예 1의 2)의 방법과 같이 아넥신-FITC V/PI 염색을 하여 세포유동분석법으로 사멸된 세포수를 측정하였다. 실험을 3회 반복하여 대조군의 사멸세포수를 1로 하였을 때의 비율을 환산하여 도 7d로 나타냈다.
그 결과, 도 7d로부터 확인되는 바와 같이 500 U/ml의 카탈라제로 전처리한 세포에서 육산화사비소 유도 아폽토시스가 거의 완전히 저해됨이 관찰되었다.
이상의 실시예 결과로부터 U937 세포에서 육산화사비소 유도 아폽토시스는 과산화수소 발생으로 인한 미토콘드리아 막 전위 손실, 시토크롬 c의 세포질내로의 유출, 카스파제-3의 활성화에 의한 것으로 확인되었다.
또한, 상기 실시예를 통해 육산화사비소에 의해 과산화수소가 증가되는 것이 확인됨으로써, 육산화사비소 유도 아폽토시스의 기전은 과산화수소 발생에 의한 것으로 확인되었다. 특히, 항산화제인 NAC 및 카탈라제를 처리한 결과 아폽토시스가 완전히 저해되는 것으로 보아 육산화사비소 유도 아폽토시스는 과산화수소의 발생에 의한 것으로 판단된다.
따라서, 육산화사비소에 의해 증가된 과산화수소는 미토콘드리아의 막 전위 손실을 유도하고 막 전위 손실은 다시 시토크롬 c의 유출을 유도하였으며 그후 연속적으로 카스파제-3의 활성화를 유도하여 궁극적으로 U937 세포에서 아폽토시스를일으키는 것으로 판단된다.
이상에서 살펴본 바와 같이, 본 발명에 의해 육산화사비소가 아폽토시스 유도를 위한 용도로 사용할 수 있어, 아폽토시스 기전과 연관된 각종 질환의 예방 및 치료에 유용하게 사용할 수 있음을 알 수 있다. 그리고, 삼산화비소에 저항성이 있는 것으로 알려진 U937 백혈병 세포주에서 낮은 농도에서도 효과적으로 아폽토시스를 유발시켰다는 점에서, 육산화사비소가 각종 고형암은 물론 백혈병을 포함하는 각종 혈액암에 광범위하게 적용될 수 있는 매우 유용한 발명임을 뜻한다.

- 제 1 항에 있어서,상기 아폽토시스는 활성산소를 발생을 통해 유도되는 것을 특징으로 하는 아폽토시스 유도제.
- 육산화사비소를 함유하는 것을 특징으로 하는 아폽토시스 유도능을 갖는 약제학적 조성물.
- 육산화사비소를 함유하는 것을 특징으로 하는 혈액암 치료제.
- 제 4 항에 있어서,혈액암이 백혈병인 것을 특징으로 하는 혈액암 치료제.
