Abstract

PURPOSE: A method for diagnosing and identifying the species of Mycobacteria causing tuberculosis, and probes and primers used therefor are provided which improve diagnosis and identification for Mycobacteria in species level. Therefore, a number of species of Mycobacteria can be diagnosed and identified simultaneously by using the method. CONSTITUTION: The probes and primers for identifying Mycobacteria are designed based on the internal transcription (ITS) base sequence of Mycobacteria. The ITS base sequence is useful in distinguishing genotypes since it has more numerous polymorphism regions as well as conservative regions than 16S rRNA commonly used as a target DNA. The base sequences of M. fortuitum and M. chelonae are identified, and probes and primers are synthesized using their base sequences. The probes and primers for other Mycobacteria are designed by analyzing the base sequence obtained from public domain(Genbank) with multialignment and blast and by selecting the distinguishable regions by polymorphism. The method comprises the following steps of: a)culturing Mycobacteria in Ogawa medium and isolating genomic DNA therefrom by vortexing and centrifuging; b)constructing primers for amplifying ITS; c)performing PCR and confirming PCR products by agarose gel electrophoresis; d)cloning ITS of M. fortuitum and M. chelonae with pGEM-T vector and transforming E.coli JM 109 therewith by Heat-shock; e)DNA sequencing using a DNA auto sequencer and universal primer M13 by dye terminator; f)designing primers and probes from the ITS sequences.

Heat-shock을 이용한 유전형질 도입방법으로 숙주인 E. coli JM 109내로 유도시켜 형질전환체들을 준비한 X-gal/IPTG/Amp+/LB 한천 배지에 도말하여 37℃ 배양기에 16-18시간 배양하였다. 배양 후 흰색의 균락만을 골라내어 LB(Amp+) 액체배지에 키워 플라스미드를 분리, 정제하였다. 플라스미드 DNA는 QIAprep kit로 분리하고 제한효소로 잘라 삽입된 DNA 절편을 확인하였다(제3도).After confirming the PCR reaction, the reactions of ITS of M. fortuitum and M. chelonae, whose ITS sequences were not disclosed, were purified by PCR purification kit (QIAGEN) and used for cloning. 본 발명에 따른 올리고뉴클레오타이드 프로브를 프라이머로 제작하여 각 균주를 특이적으로 동정이 가능한지에 대한 실험을 PCR의 증폭여부로 확인하였다.TB complex 동정을 위해서 서열번호 3의 과 10의 염기서열을 선택, MOTT균주중 MAC의 동정을 위해서 서열번호 21과 18의 염기서열을 선택, M. fortuitum의 동정을 위해서는 서열번호 31과 23의 염기서열을 선택, M. chelonae의 동정을 위해서는 서열번호 34와 37의 염기서열을 선택, M. gordonae의 동정을 위해서는 서열번호 40과 42의 염기서열을 선택, M. terra의 동정을 위해서는 서열번호 47과46의 염기서열을 선택, M. scrofulaceum의 동정을 위해서는 서열번호 51과 52의 염기서열을 선택하여 첫번째 염기서열의 센스 스트랜드(sense strand)를 가진 프라이머와 두번째 염기서열의 안티센스 스트랜드(antisense strand)를 가진 프라이머를 제작하여 마이코박테리아의 균종과 중합반응시켜 전기영동한 결과 도 7과 효소중합연쇄반응이 일어났음을 확인할 수 있어, 각각의 프라이머쌍에 의한 PCR반응으로 마이코박테리아의 각 균종이 진단가능함을 알 수 있다.The oligonucleotide probe according to the present invention was prepared as a primer to determine whether each strain can be specifically identified by PCR amplification.
