Abstract
The present invention relates to a composition for preserving body fluids, and a method for preparing the same. It is very important to preserve cells in saliva without degeneration in order to analyze the correct genotype from saliva samples. However, the saliva contains a number of proteases, nucleases, nasopharyngeal secretions, food residues, bacteria, and viruses, which degrade cells, which reduces the accuracy of genotyping. Therefore, the necessity of developing a composition for preserving cells in saliva without degeneration from the moment of saliva collection until the genotyping is actually performed is remarkable.
Accordingly, the present invention is conceived to solve the problems of the prior art as described above, and relates to a composition for preserving body fluids, and a method of manufacturing the same. Since the composition of the present invention was developed to stably preserve cells by reflecting the characteristics of saliva, it is expected to be widely used in the fields of medicine and health.
Description
The present invention relates to a composition for preserving body fluids, and a method for preparing the same.
Saliva is secreted from about 1 to 1.5 ℓ per day from the salivary glands. Most of saliva consists of water (99.3%), and the rest consists of inorganic and organic substances such as mucin (0.3%), amylase, and amino acids. Saliva is secreted from large and small salivary glands such as the parotid gland, submandibular gland, and sublingual gland. Saliva is very important as a gene detection sample. Recently, the number of saliva samples as evidence in the field of predicting an individual's genetic disease or as evidence has increased significantly. Since the oral epithelial cells are always present in the saliva purely obtained without going through the purification process, and genetic analysis is possible even in a small amount of samples, genotype can be analyzed from the oral epithelial cells present in the saliva. In addition, saliva samples are non-invasive and can be conveniently collected regardless of time/space. Therefore, it would be said that saliva is of great value as a genotyping sample.
Therefore, the necessity of developing a composition for preserving cells in saliva without degeneration is remarkable from the moment of saliva collection until the genotyping is actually performed. Regarding the preservation of cell samples, the invention of “antioxidant preservation solution for biological samples” was disclosed in Korean Patent No. 10-0763686, and “Composition for preservation and stabilization of cells or macromolecules” in Korean Patent No. 2010-0015578 "Although the invention has been disclosed, there is a problem that all of the prior documents did not properly reflect the characteristics of the saliva sample.

Example 2: Analysis of the effect of the composition for preserving body fluids
Saliva samples were collected with the compositions of Preparation Examples 1 to 5 of Example `1. As a control for the compositions prepared in the present invention, a product of Oragen (Oragene·DNA OG-510, DNA Genotek Inc.) sold by Genotech was used. The same amount of saliva was added to each saliva preserving composition, and the sample was stored at room temperature. On the 1st, 3rd, 5th, 7th, or 14th day from saliva collection, DNA was extracted from the sample using a DNA extraction kit (AccuPrep® Genomic DNA Extraction Kit, Bioneer) according to the manufacturer's recommended protocol. Extracted. The extracted DNA was measured for absorbance at A260/280 wavelength to evaluate the purity and concentration of the DNA. The results are shown in FIGS. 1 to 4.
특히 제조예 5의 경우 대조군과 비교하여 DNA 검출 농도가 약 57% 증가하여, 타액 보존용 조성물로서 매우 우수한 것을 알 수 있었다(도 4).As a result of the experiment, it was found that the compositions of Preparation Examples 1 to 5 and the control group of the present invention all showed a DNA purity of 1.8 to 2.0 under conditions of 1 to 14 days, so that all compositions had a cell preservation effect at a level suitable for genotyping. Could be (Fig. 2). However, in the detection of DNA concentration, there was a large difference in the detected DNA concentration for each composition. In the results of 1 day, most similar DNA concentrations were detected except for Preparation Example 4, but after 14 days from saliva collection, the composition in which the lowest DNA concentration was detected (Preparation Example 4) and the composition in which the highest DNA concentration was detected (Production Example 5) There was a large gap. In particular, in the case of Preparation Example 5, the DNA detection concentration was increased by about 57% compared to the control, and it was found that it is very excellent as a composition for preserving saliva (FIG.
