Abstract
일 양상에 따른 반응성 성상세포를 유도하는 형질전환 동물, 이를 제조하는 방법, 및 형질전환 동물을 이용하여 뇌질환 치료용 물질을 스크리닝하는 방법을 제공한다. 이에 따르면, 성상세포의 반응성을 조절할 수 있고, 국소적 동물 모델을 제조할 수 있다. 또한, 뇌염증, 퇴행성 뇌질환, 치매, 뇌졸중 등의 반응성 성상세포가 연관된 다양한 질병을 치료하기 위한 물질을 스크리닝하는데 이용할 수 있다.
Description
반응성 성상세포를 유도하는 형질전환 동물, 이를 제조하는 방법, 및 형질전환 동물을 이용하여 뇌질환 치료용 물질을 스크리닝하는 방법에 관한 것이다.
퇴행성 뇌질환은 나이가 들어감에 따라 발병하는 퇴행성 질환 중 뇌에서 발생하는 질환이다. 퇴행성 뇌질환은 뇌와 척수의 특정 뇌세포가 서서히 기능이 변형되거나 기능을 잃고, 뇌세포의 수가 감소하여, 인지 장애, 운동 장애 등의 기능장애가 진행되는 것으로 알려져 있다. 치매(dementia)는 기억력 장애, 판단력 상실 등 전신기능의 전반적인 장애를 동반하는 질환이다. 치매의 발병 형태는 매우 다양한데, 약 50% 정도는 알츠하이머형 치매(dementia of Alzheimer type), 20 내지 30%는 혈관성 치매(dementia of Vascular type)이고, 알코올성 치매 및 파킨슨병 치매 등이 있으며, 약 15∼20%는 알츠하이머형 및 혈관성 치매 양쪽성으로 발병하는 것으로 알려져 있다.
성상세포(astrocyte)는 성상교세포(astroglia)로도 알려져 있는 세포로서, 뇌 및 척수에서 별 모양의 교세포이다. 성상세포는 혈관-뇌 장벽을 형성하는 내피 세포의 생화학적 지지, 신경 조직에 영양의 제공, 세포외 이온 균형의 유지, 외상성 장해 후 뇌와 척수의 회복(repair) 등에 기능을 한다. 그러나, 반응성(reactive) 성상세포는 성상세포가 변형된 것으로 신경세포에 독성을 가지고, 신경세포를 사멸시킬 수 있는 신호를 방출할 수 있는 것으로 알려져 있다(Liddelow et al., Nature. 541 (7638): 481-487).
따라서, 반응성 성상세포를 유도하는 동물 모델을 개발하고, 이를 이용하여 뇌질환 치료용 물질을 스크리닝 방법을 개발할 필요가 있다.

신경교 섬유질 산성 단백질(glial fibrillary acidic protein: GFAP) 프로모터 서열과 Cre 재조합효소-에스트로겐 수용체 T2(Cre recombinase-estrogen receptor T2: Cre-ERT2) 폴리펩티드를 암호화하는 뉴클레오티드 서열이 작동가능하게 연결된 제1 유전자 카세트; 및
두 개의 LoxP 뉴클레오티드 서열 사이에 전사종결 뉴클레오티드 서열이 위치하고, 어느 하나의 LoxP 뉴클레오티드 서열의 하류에 디프테리아 독소 수용체(Diphtheria Toxin Receptor: DTR)를 암호화하는 뉴클레오티드 서열을 포함하는 제2 유전자 카세트가 형질전환되고,
에스트로겐 길항제의 투여에 의해 CreERT2-LoxP 시스템이 작동되어 반응성 성상세포 특이적으로 DTR의 발현이 유도되는 형질전환 동물을 제공한다.
용어 "성상세포(astrocyte)"는 신경교세포의 하나로서, 성상교세포(astroglia)로도 불리고, 척추동물의 중추신경계에 존재하는 별 모양의 뇌세포를 말한다. 성상세포는 신경세포에 영양소 공급, 혈관-뇌 장벽에서 내피 세포의 지지, 신경전달물질 흡수 및 전달, 세포외 공간에서 이온 농도의 조절, 시냅스 전달의 조절, 신경계 회복, 또는 이들의 조합의 기능을 수행할 수 있다. 뇌에 상해를 입었을 경우, 성상 세포는 증식이 활발해지고(astrocytosis), 팽윤(swelling)이 일어나고, 성상교세포증(astrogliosis)이 나타날 수 있다.
용어 "반응성 성상세포(reactive astrocyte)"는 성상세포가 변형되어 비정상적으로 증식하는 성상세포를 말한다. 반응성 성상세포는 중추신경계 외상, 감염, 허혈, 뇌졸중, 자가면역 반응, 신경퇴행성 질환 등에 의해 성상세포의 수가 비정상적으로 증식할 수 있다. 반응성 성상세포는 신경세포의 재생을 억제하거나 독성 물질을 분비하여 다른 성상세포나 신경세포의 사멸을 유도할 수 있다.
용어 "형질전환 동물(transgenic animal)"은 외래의 표적 유전자를 동물에 형질전환시켜 표적 유전자의 발현을 인위적으로 조절할 수 있는 동물을 말한다. 상기 동물은 인간을 제외한 포유동물, 예를 들어, 마우스, 래트, 기니 피그, 개, 돼지, 소, 염소, 말, 양, 고양이, 및 유인원일 수 있다.
상기 형질전환 동물은 신경교 섬유질 산성 단백질(glial fibrillary acidic protein: GFAP) 프로모터 서열과 Cre 재조합효소-에스트로겐 수용체 T2(Cre recombinase-estrogen receptor T2: Cre-ERT2) 폴리펩티드를 암호화하는 뉴클레오티드 서열이 작동가능하게 연결된 제1 유전자 카세트가 형질전환된 것이다.
상기 신경교 섬유질 산성 단백질(glial fibrillary acidic protein: GFAP)은 성상세포 및 뇌실막세포(ependymal cell)를 포함한 중추신경계의 다양한 세포에서 발현되는 중간 섬유(intermediate filament) 단백질이다. GFAP는 성상세포의 기계적 강도를 유지하는데 기능을 하는 것으로 알려져 있다.
상기 GFAP 프로모터는 GFAP의 발현에 필요한 조절 영역으로서, 전사가 개시되는 DNA 영역일 수 있다. GFAP 프로모터는 GFAP 전사 개시점으로부터 상류, 즉 5'방향에 존재하는 뉴클레오티드 서열일 수 있다. 상기 GFAP 프로모터 서열은 인간 유래 서열일 수 있다. 상기 GFAP 프로모터는 반응성 성상세포 특이적인 프로모터일 수 있다.
Cre 재조합효소(recombinase)는 P1 박테리오파지 유래의 티로신 재조합 효소이다. Cre 재조합효소는 두개의 DNA 인식 부위(LoxP 부위) 사이에 부위 특이적 재조합을 촉매하는 효소이다.
에스트로겐 수용체 T2(estrogen receptor T2)는 G400V, M543A, 및 L544A의 돌연변이를 가진 에스트로겐 수용체일 수 있다.
상기 형질전환 동물은 두 개의 LoxP 뉴클레오티드 서열 사이에 전사종결 뉴클레오티드 서열이 위치하고, 어느 하나의 LoxP 뉴클레오티드 서열의 하류에 디프테리아 독소 수용체(Diphtheria Toxin Receptor: DTR)를 암호화하는 뉴클레오티드 서열을 포함하는 제2 유전자 카세트가 형질전환된 것이다.

디프테리아 독소 수용체(Diphtheria Toxin Receptor: DTR)는 디프테리아 독소에 특이적으로 결합하는 수용체로서, 수용체-매개 내포작용(endocytosis)에 의해 독소가 세포 내로 도입시킬 수 있다.
제2 유전자 카세트는 ROSA26 폴리뉴클레오티드를 더 포함할 수 있다. ROSA26 폴리뉴클레오티드는 구성적(constitutive) 또는 편재성(ubiquitous) 유전자 발현에 이용될 수 있는 뉴클레오티드 서열 또는 로커스(locus)일 수 있다.
상기 형질전환은 형질전환, 형질도입, 형질주입, 및 바이러스 감염을 포함할 수 있다.
상기 형질전환 동물은 에스트로겐 길항제의 투여에 의해 CreERT2-LoxP 시스템이 작동되어 반응성 성상세포 특이적으로 DTR의 발현이 유도될 수 있다.
상기 에스트로겐 길항제는 타목시펜, 4-히드록시타목시펜, 클로미펜(clomifene), 라록시펜(raloxifene), 또는 이들의 조합일 수 잇다. 상기 에스트로겐 길항제는 1일 내지 2주일, 1일 내지 1주일, 또는 1일 내지 5일 동안 투여될 수 있다. 상기 에스트로겐 길항제는 1일 1회, 또는 2일 내지 1주일에 1회 투여될 수 있다.

상기 디프테리아 독소는 0.1 ㎍/체중 kg/일 내지 1 mg/체중 kg/일, 1 ㎍/체중 kg/일 내지 500 ㎍/체중 kg/일, 5 ㎍/체중 kg/일 내지 250 ㎍/체중 kg/일, 10 ㎍/체중 kg/일 내지 100 ㎍/체중 kg/일, 10 ㎍/체중 kg/일 내지 75 ㎍/체중 kg/일, 또는 12 ㎍/체중 kg/일 내지 50 ㎍/체중 kg/일의 용량으로 투여될 수 있다.
상기 디프테리아 독소는 1일 내지 1 개월, 1일 내지 25일, 1일 내지 20일, 1일 내지 16일, 또는 2일 내지 16일 동안 투여될 수 있다.
상기 형질전환 동물에 디프테리아 독소의 투여에 의하여 반응성 성상세포가 유도될 수 있다. 반응성 성상세포의 유도에 의해 뇌질환이 유발될 수 있다. 상기 뇌질환은 뇌염증, 퇴행성 뇌질환, 치매, 뇌졸중, 또는 이들의 조합일 수 있다. 상기 퇴행성 뇌질환은 알츠하이머병, 파킨슨병, 헌팅톤병, 다발성 경화증, 근위축성 측삭 경화증(amyotrophic lateral sclerosis: ALS), 또는 이들의 조합일 수 있다.
상기 형질전환 동물은 신경세포의 사멸, 기억 손실, 기억 능력의 손상, 뇌 크기의 감소, 생존율 감소, 또는 이들의 조합의 가질 수 있다. 상기 기억 손실은 기존 기억의 손실일 수 있다. 상기 기억 능력의 손상은 기억할 수 있는 능력의 손상일 수 있다.
상기 반응성 성상세포는 모노아민 옥시다제 B(monoamine oxidase B: MAO-B), 유도성 산화질소 합성효소(inducible nitric oxide synthase: iNOS), p67[hpox], 또는 이들의 조합에 의해 유도될 수 있다.

상기 바이러스는 해마(hippocampus)에 국소적으로 감염된 것일 수 있다. 상기 해마는 해마 CA1 영역, CA2 영역, CA3 영역, CA4 영역, 또는 이들의 조합일 수 있다. 예를 들어, 상기 해마는 해마 CA1 영역이다.
상기 바이러스는 아데노-연관 바이러스(adeno-associated virus: AAV)일 수 있다.

실시예
1. 반응성 성상세포를 특이적으로 유도할 수 있는 실험 동물 모델의 제작
1. 성상세포-특이적 독소 수용체 모델의 제작
성상세포에서 특이적으로 작용하는 인간 신경교 섬유질 산성 단백질(human glial fibrillary acidic protein: hGFAP) 프로모터 서열과 Cre 재조합효소-에스트로겐 수용체 T2(Cre recombinase-estrogen receptor T2: Cre-ERT2) 융합 단백질을 암호화하는 뉴클레오티드 서열이 작동가능하게 연결된 GFAP-cre/ERT2 형질전환 마우스(Jackson Laboratory, stock number 012849)를 준비하였다.
ROSA26 로커스(locus), STOP 카세트, 및 디프테리아 독소 수용체(Diphtheria Toxin Receptor: DTR)를 암호화하는 뉴클레오티드 서열이 작동가능하게 연결된, 유도성 디프테리에 독소 수용체(inducible Diphtheria Toxin Receptor: iDTR) 형질전환 마우스(Jackson Laboratory, stock number 007900)를 준비하였다.
준비된 GFAP-cre/ERT2 형질전환 마우스와 iDTR 형질전환 마우스를 교배하여 성상세포-특이적 독소 수용체 동물 모델을 제작하고, 이를 "GiD" 마우스라고 명명하였다(도 1a). 음성 대조군으로 GFAP-cre/ERT2 형질전환 마우스를 사용하였고, "Gcon" 마우스로 명명하였다.

GiD
마우스 모델에서 반응성 성상 세포의 선택적 유도
1.에 기재된 바와 같이 제작된 GiD 마우스 또는 Gcon 마우스에 타목시펜(Sigma, T5648)을 100 mg/체중 kg/일의 용량으로 5일간 복강 주사하였다. 10일 후에, 디프테리아 독소(DT)를 50 ㎍/체중 kg/일로 2일간 복강 주사하였다. DT를 주입한 지 1주일 후에, 각 마우스를 심장관류 및 동결절단(cryosection)(30 ㎛ 두께)하였다(도 1b).
동결절단된 뇌 조직은 항-GFAP 항체(Millipore ab5541)를 사용하여 면역염색하고, DAPI(4',6-디아미디노-2-페닐인돌)로 세포의 핵을 염색하였다. 면역염색 및 핵 염색 결과를 도 1c에 나타내었다(초록색: GFAP, 파란색: DAPI).
도 1c에 나타난 바와 같이, GiD 마우스는 Gcon 마우스에 비해 DT-유도성 GFAP가 강하게 염색됨을 확인하였다. 따라서, GiD 마우스는 성상세포에 특이적으로 발현시킨 DTR에 의해, 반응성 성상세포를 선택적으로 유도할 수 있음을 확인하였다.

GiD
마우스 모델에서 경증 및 중증 반응성 성상세포의 유도 조건 확립
GiD 마우스 또는 Gcon 마우스에 타목시펜을 100 mg/체중 kg/일의 용량으로 5일간 복강 주사하였다. 10일 후에 디프테리아 독소(DT)를 50 ㎍/체중 kg/일의 용량으로 2일간(경증(mild: m) 조건) 또는 12 ㎍/체중 kg/일의 용량으로 16일간(중증(severe: s) 조건) 복강 주사하였다. DT를 주입한 지 1주일 또는 1개월 후에, 각 마우스를 심장관류 및 동결절단(30 ㎛ 두께)하였다. 경증 1주일 조건을 GiDm, 중증 1개월 조건을 GiDs로 명명하였다.
동결절단된 조직에 대하여 2.에 기재된 기재된 바 같이 항-GFAP 항체로 면역염색하고 DAPI로 핵을 염색하였다. 염색 이미지 및 각 군에서 GFAP 형광 강도의 그래프를 도 2a에 나타내었다(초록색: GFAP, 파란색: DAPI, AU: 임의 단위(arbitrary unit), ***: p<0.001).
또한, 면역염색 결과에 대하여 숄(sholl) 분석을 수행하고 그 결과를 도 2b에 나타내었다.
도 2a 및 도 2b에 나타난 바와 같이, 경증 조건으로 DT를 투여한 GiD 마우스의 경우, 약 1주일 후 비대해진 GFAP 신호가 약 1개월 후에 다시 원래의 상태로 돌아왔다. 중증 조건으로 DT를 투여한 GiD 마우스의 경우, 성상세포의 반응성이 1달까지 점차 증가하고, 교세포의 숫자도 증가하였다. 따라서, GiD 마우스에서 DT의 주입 농도 및 횟수에 따라 성상세포의 반응성을 조절할 수 있는 조건을 확립하였다.
4.
GiD
중증 모델에서 신경세포 사멸의 유도
3.에 기재된 바와 같이 Gcon 마우스 및 GiD 중증 모델을 준비하였다.
GiD 중증 모델(GiDs)에 과산화수소(H2O2) 소거물질인 AAD-2004(2-히드록시-5-[2-(4-트리플루오로메틸페닐)-에틸아미노벤조산)(GNTPharma)을 DT처리 시작점부터 16일간 3.3 mg/체중 kg/일의 용량으로 복강 투여하였다. 2.에 기재된 바와 같이 마우스의 뇌조직을 준비하였다. 그 후 신경세포의 바이오마커인 NeuN 및 MAP2를 검출하기 위해, 항-NeuN 항체(Millipore mab377) 및 항-MAP2 항체(Abcam, ab5392)을 사용하여 면역조직화학법(immunohistochemistry)을 수행하였다. 면역조직화학 이미지를 분석하여 NeuN 강도 및 MAP2 강도를 산출하고, 면역조직화학 이미지 및 NeuN 강도 및 MAP2 강도를 도 3a 및 도 3b에 나타내었다(****, ++++, p<0.0001, N=number of ROI).
도 3a 및 도 3b에 나타난 바와 같이, GiDs 마우스에서 NeuN 및 MAP2의 강도가 유의미하게 감소하였다. 또한, AAD-2004를 처리하였을 때, NeuN 및 MAP2의 강도이 감소하는 것이 억제되었다. 따라서, GiDs 모델에서 H2O2 의존적으로 중증 반응성 성상세포에 의해 신경세포가 사멸한다는 것을 확인하였다.
5.
GiD
중증 모델에서 기존 기억의 손실 및 영구적인 기억 능력의 손상
Gcon 마우스 및 GiD 마우스에 대하여, 수동 회피반응 시험(passive avoidance test)을 수행하였다.
밝은 방보다 어두운 방을 더 선호하는 성질을 보이는 마우스의 특징을 이용하여, 밝은 방에 넣은 Gcon이나 GiDs 마우스가 어두운 방으로 들어가면, 바닥의 전기 쇼크를 주어, 어두운 방에 대한 공포 기억을 형성시켰다(Acquisition). 다음날부터 밝은 방에 넣었을 때 어두운 방으로 들어가기까지의 시간(지연 시간)을 측정하여, 형성된 공포 기억에 대한 기억력을 측정하였다(Retrieval). 첫번째 기억력 측정 후, DT를 중증 조건으로 주입하였다. AAD-2004 투여군의 경우, AAD-2004를 DT 처리 시작점부터 16일간 3.3 mg/체중 kg/일의 용량으로 복강 투여하였다. 30일째, 두번째 기억력 측정을 수행하였다. 31일째에 마우스에 전기 쇼크를 다시 주어서, 공포 기억을 재형성(reinforcement)시켰다. 수동 회피반응 시험의 모식도를 도 4a에 나타내고, 마우스의 지연 시간을 도 4b에 나타내었다(Acq.: 기억 형성, Ret.1: 측정 1, Ret.2: 측정 2, Ret.3: 측정 3, Ret.4: 측정 4).
도 4b에 나타난 바와 같이, GiDs에서 지연 시간이 유의하게 감소하여 형성되었던 공포 기억이 사라짐을 확인하였다. 또한, 공포 기억의 재형성 이후에도 새로운 기억이 형성되지 않았다. AAD-2004 투여군은 기억 손상이 유의미하게 억제되었고, 공포 기억의 재형성 이후에 공포 기억이 잘 형성됨을 확인하였다. 따라서, 중증 반응성 성상세포가 H2O2 의존적으로 기억의 비가역적 손상을 유도한다는 것을 확인하였다.

GiD

생존률

GiDs 마우스에 AAD-2004를 DT 처리 시작점부터 16일간 3.3 mg/체중 kg/일의 용량으로 복강 주사한 후, 쥐의 뇌를 적출하였다. 뇌의 크기를 측정한 결과를 도 5a에 나타내었다. 또한, 쥐의 생존율을 산출하여 도 5b에 나타내었다(**, ++, p<0.01, *, p<0.05). 도 5a 및 도 5b에 나타난 바와 같이, GiDs 마우스에서 뇌의 크기와 생존율이 유의하게 감소하였다. 반면에, H2O2 소거물질인 AAD-2004를 처리하였을 때, 뇌 수축이 억제됨과 동시에 생존율도 회복되는 것을 확인하였다.
7. 반응성 성상세포의 유도에 대한 메커니즘
반응성 성상세포가 MAO-B에 의한 활성 산소종에 의해 유도되는지 여부를 확인하기 위해 선택적 MAO-B 억제제인 세레길린 및 과산화수소 소거물질인 AAD-2004를 사용하였다.
GiDm 마우스에 DT처리 시작점부터 7일간 10 mg/체중 kg/일의 용량의 세레길린(selegiline)(Sigma) 및 3.3 mg/체중 kg/일의 용량의 AAD-2004를 복강 주사하였다. 그 후, 1.에 기재된 바와 같이, 항-GFAP 항체(Millipore ab5541) 및 항-GABA 항체(Millipore
ab175)를 사용하여 면역조직화학을 수행하였다.
면역조직화학의 이미지를 도 6a에 나타내고, GFAP 강도(변화 배수)와 GFAP+ 세포에서 GABA의 강도(변화 배수)를 나타내는 그래프를 도 6b에 나타내었다.
도 6a 및 도 6b에 나타난 바와 같이, GiDm 마우스에서 GFAP 및 GABA가 유의하게 증가하였다. 세레길린을 처리한 경우 GFAP과 GABA의 증가가 억제되었다. AAD-2004를 처리한 경우 GFAP의 증가는 억제된 반면, GABA의 증가는 유지되었다. 따라서, 반응성 성상세포의 구조 변화가 MAO-B에 의한 활성산소종에 의해 매개된다는 것을 확인하였다.
8. 중증 반응성 성상세포의
질산화적
스트레스
중증 반응성 성상세포에서 질산화적(nitrosative) 스트레스가 유도되는지 여부를 확인하기 위해 활성산소종을 생산하는 효소인 유도성 NO 생성효소(inducible NO synthase: iNOS) 및 p67[phox]를 검출하였다.
GiDs 마우스에 DT 처리 시작점부터 16일간 3.3 mg/체중 kg/일의 용량으로 AAD-2004를 복강 주사하였다. 마우스의 뇌 조직을 수득하고, 항-iNOS 항체(BD, 610332), 항-p67[phox] 항체(BD, 610913), 항-lba1 항체(Wako 019-19741), 및 항-GFAP 항체를 사용하여 면역조직화학을 수행하고, DAPI로 핵을 염색하였다. 염색 이미지를 도 7a에 나타내고, GFAP 강도(변화 배수), iNOS 강도, p67[phox] 강도, 및 lba1 강도를 도 7b에 나타내었다(AU: 임의 단위, ****, p<0.0001; ***, +++, p<0.001)
도 7a 및 도 7b에 나타난 바와 같이, GiDs 마우스에서 iNOS 및 p67[phox]의 강도가 유의하게 증가하였다. iNOS는 반응성 성상세포에서 및 p67[phox]는 활성화된 미세아교세포에 특이적으로 발현이 증가하였다. 또한, AAD-2004를 처리하였을 때, 발현의 증가가 억제됨을 확인하였다. 따라서, GiDs 모델에서 H2O2 의존적으로 iNOS 및 p67[phox]의 발현이 유도된다는 것을 확인하였다.
9. 국소적인 반응성 성상세포 유도 모델의 확립
유도성 디프테리아 독소 수용체(inducible diphtheria toxin receptor: iDTR) 마우스((Jackson Laboratory, stock number 007900)의 해마 CA1 영역에 AAV-GFAP-Cre 바이러스(KIST virus facility)를 주입하였다. 그 후, 마우스에 DT를 50 ㎍/kg/일 또는 12 ㎍/체중 kg/일의 용량으로 각각 2일간 또는 16일간 복강 주사하였다. 50 ㎍/체중 kg/일의 용량으로 2일간 주입한 경우를 fGiDm(경증 국소적(focal) GiD)으로 명명하고, 2 ㎍/체중 kg/일의 용량으로 16일간 주입한 경우를 fGiDs(중증 국소적 GiD)로 명명하였다.
DT를 주입한지 1주일 또는 1개월 후에, 각 마우스를 2.에 기재된 바와 같이 희생시키고, 면역조직화학을 수행하였다.
국소적인 반응성 성상세포 유도 모델(fGID) 마우스를 제작하는 과정의 모식도를 도 8a에 나타내고(초록색: GFAP, 파란색: DAPI), 염색 결과를 도 8b에 나타내고, GFAP 강도(AU: 임의 단위), DAPI 염색 세포수(50 ㎛×50 ㎛ 관심 영역(region of interest: ROI) 중 개수), 및 CA1 크기를 도 8c에 나타내었다(**, p<0.01; ***, p<0.001, ****, p<0.0001)
fGiDm와 fGiDs에서 GFAP의 강도가 점차 증가하였고, fGiDs에서 교세포의 개수도 증가함을 확인하였다. fGiDs에서는 CA1 영역의 위축(atrophy)도 확인하였다. 따라서, GiD 시스템을 국소적으로 적용하여, 뇌의 위치 별로 성상세포의 반응성을 조절할 수 있는 조건을 확립하였다.
Claims (18)
- 반응성 성상세포를 유도하는 형질전환 동물에 대해 성상세포의 반응성을 조절하는 방법으로서,
신경교 섬유질 산성 단백질(glial fibrillary acidic protein: GFAP) 프로모터 서열과 Cre 재조합효소-에스트로겐 수용체 T2(Cre recombinase-estrogen receptor T2: Cre-ERT2) 폴리펩티드를 암호화하는 뉴클레오티드 서열이 작동가능하게 연결된 제1 유전자 카세트; 및
두 개의 LoxP 뉴클레오티드 서열 사이에 전사종결 뉴클레오티드 서열이 위치하고, 어느 하나의 LoxP 뉴클레오티드 서열의 하류에 디프테리아 독소 수용체(Diphtheria Toxin Receptor: DTR)를 암호화하는 뉴클레오티드 서열을 포함하는 제2 유전자 카세트가 형질전환되고,
에스트로겐 길항제의 투여에 의해 CreERT2-LoxP 시스템이 작동되어 반응성 성상세포 특이적으로 DTR의 발현이 유도되고, 디프테리아 독소(DT)의 주입 농도 및 횟수에 따라 성상세포의 반응성을 조절할 수 있는, 형질전환 동물에 대해 디프테리아 독소(DT)를 50 ㎍/체중 kg/일의 용량으로 2일간 또는 12 ㎍/체중 kg/일의 용량으로 16일간 투여함으로써 성상세포의 반응성을 조절하는 방법. - 청구항 1에 있어서, 상기 GFAP 프로모터 서열은 인간 유래 서열인 것인 방법.
- 청구항 1에 있어서, 제2 유전자 카세트는 ROSA26 폴리뉴클레오티드를 더 포함하는 것인 방법.
- 청구항 1에 있어서, 상기 에스트로겐 길항제는 타목시펜, 4-히드록시타목시펜, 클로미펜(clomifene), 라록시펜(raloxifene), 또는 이들의 조합인 것인 방법.
- 청구항 1에 있어서, 상기 형질전환 동물에 디프테리아 독소의 투여에 의하여 뇌질환이 유발된 것인 방법.
