Real-time polymerase chain reaction was carried out on a real-time PCR machine using a mixture of the synthesized cDNA with the target protein MMP-1 primer and the cyanine dye cyberrin (SYBR Green supermis, Applied Biosystems, USA). Finally, the expression level of the MMP-1 gene was confirmed. The expression level of the gene was finally analyzed through correction for the β-actin gene and the analysis results are shown in FIG. 1.
도 1에 나타낸 바와 같이, UVB 조사군에서 MMP-1 발현량이 증가한 것에 반해, 2-푸코실락토오스 처리군에서 MMP-1 유전자 발현량이 유의적으로 감소함을 확인하였다. 따라서, 푸코실락토오스는 콜라겐을 분해하여 주름 형성을 가속화시키는 MMP-1의 발현을 감소시킴으로써 주름 개선 효과 및 노화 방지 효과를 나타냄을 알 수 있다.Therefore, it can be seen that fucosyllactose exhibits wrinkle improvement and anti-aging effects by reducing expression of MMP-1, which degrades collagen to accelerate wrinkle formation.
실시예Example 2: 2: 푸코실락토오스의Fucosylactose IL- IL- 6 및6 and IL-8 저해 효과 측정 Measurement of IL-8 Inhibitory Effect
2-푸코실락토오스의 IL-6, IL-8 저해 효과를 평가하기 위해 상기 실시예 1에 기재한 것과 동일한 세포를 사용하여 동일 조건으로 실험을 진행하였다. 다만 최종적으로 실시간 중합효소 연쇄반응을 진행하는 과정에서 MMP-1 프라이머 대신 IL-6, IL-8 프라이머를 사용하였다. 유전자의 발현량은 β-actin 유전자에 대한 보정을 통해 최종적으로 분석하고 분석 결과를 각각 도 2 및 도 3에 나타내었다.In order to evaluate the IL-6 and IL-8 inhibitory effects of 2-fucosyllactose, experiments were conducted under the same conditions using the same cells as described in Example 1 above. 실시예Example 3: 3: 푸코실락토오스의Fucosylactose 콜라겐 생성 촉진 효과 측정 Measurement of collagen production promoting effect
2-푸코실락토오스의 피부세포에서의 콜라겐 생성 촉진 효과를 측정하기 위해 상기 실시예 1 및 2에 기재한 것과 동일 세포를 사용하여 실험을 진행하였다. Experiments were conducted using the same cells as described in Examples 1 and 2 to determine the effect of promoting the collagen production in skin cells of 2-fucosyllactose.
상기 실시예 1의 세포 배양 조건으로 동일한 방법으로 세포를 96 웰 플레이트에 분주한 후 37℃, 5% CO2 조건의 배양기에서 24시간 동안 배양하였다. 이후 2-푸코실락토오스 (10㎍/㎖ 또는 100㎍/㎖) 또는 TGF-β를 포함하는 배지와 효능 원료를 포함하지 않은 배지로 교체해 준 다음 24시간 동안 추가로 배양하였다. 24시간 이후 배양 배지를 회수하여 PIP EIA Kit(TAKARA, MK101)를 이용하여 배지로 분비된 프로콜라겐 Ⅰ의 생성량을 확인하였다. 그 결과를 도 4에 나타내었다.Cells were cultured in 96 well plates in the same manner as the cell culture conditions of Example 1, and then cultured for 24 hours in an incubator at 37 ° C. and 5% CO 2 . 푸코실락토오스를 함유하는 토너제를 하기의 표 1에 나타낸 조성성분 및 조성비에 따라 통상적인 방법으로 제조하였다. Toner preparations containing fucosylactose were prepared in a conventional manner according to the compositional components and the composition ratios shown in Table 1 below.
제제예Formulation example 2: 로션제의 제조 2: Preparation of Lotion
푸코실락토오스를 함유하는 로션제를 하기의 표 2에 나타낸 조성성분 및 조성비에 따라 통상적인 방법으로 제조하였다. A lotion agent containing fucosylactose was prepared in a conventional manner according to the composition ingredients and composition ratios shown in Table 2 below.
제제예Formulation example 3: 크림제의 제조 3: Manufacture of cream
푸코실락토오스를 함유하는 크림제를 하기의 표 3에 나타난 조성성분 및 조성비에 따라 통상적인 방법으로 제조하였다. Cream containing fucosyllactose was prepared by a conventional method according to the composition and the composition ratio shown in Table 3 below.

The cosmetic composition of claim 1, wherein the skin wrinkles are caused by an exogenous factor or an endogenous factor.
The cosmetic composition according to claim 1, wherein the 2-fucosyllactose is derived from breast milk.
The cosmetic composition of claim 1, wherein the 2-fucosyllactose inhibits the expression of MMP-1, IL-6, or IL-8.
The method of claim 1, wherein the cosmetic composition skin lotion, skin softener, skin toner, astringent, lotion, milk lotion, moisturizing lotion, nutrition lotion, massage cream, nutrition cream, moisturizing cream, hand cream, foundation, essence, nutrition essence, Cosmetic composition having at least one formulation selected from the group consisting of a pack, soap, cleansing foam, cleansing lotion, cleansing cream, body lotion, and body cleanser.
