Description
도 2는 멸균시킨 고삼(Sophora flavescens) 식물체의 종자를 발아 배지에서 발아시켜, 줄기, 잎 및 뿌리를 유도하는 단계를 나타낸 것이다. (a) 발아처리 후 3일, (b) 발아처리 후 5일, (c) 발아처리 후 7일, (d) 발아처리 후 10일, (e) 발아처리 후 14일, (f) 기내배양 상태로 유지 중인 식물체.
도 3은 고삼 식물체 조직 유래별 캘러스의 종류를 나타낸다. A: 덩어리를 형성하며 증식, 물기를 많이 함유, B: 노란색을 넘어 연두색으로 변하며 작은 덩어리를 이루며 증식, C: 쉽게 부스러지기 쉬운 보푸라기 형태로 증식, D: 노란색을 띠며 알갱이로 분리되기 쉬운 캘러스 형태.

(c) 상기 증식된 잎 유래의 캘러스를 MS 기본 배지에 0.4~0.6㎎/ℓ 벤질아데닌 및 1.8~2.2㎎/ℓ 피클로람이 첨가된 캘러스 증식 배지에 계대배양하여 각각 증식시키는 단계; 및
(d) 상기 계대배양하여 증식시킨 잎 유래의 캘러스를 대상으로 마키아인 함량을 각각 측정하여 마키아인 생산성이 우수한 캘러스를 선발하는 단계를 포함하는 고삼 식물체 유래의 마키아인(maackiain) 대량 생산용 캘러스의 제조방법. - 삭제
