Abstract
Description
본 발명은 해양 유래 바실러스 리케니포르미스(Bacillus licheniformis)가 생산하는 진균 및 세균에 대한 우수한 활성을 보이는 신규한 글리코리피드 화합물에 관한 것이다.
해양 생태계는 다양한 생물학적 활성을 가진 방대한 화학물질을 생산할 수 있는 광대한 원천으로서의 역할을 하고 있다. 그러한 배경에서, 해양 천연물 화학은 전례가 없는 속도로 진행되어 왔고, 육상에서는 통상적으로 발견되지 않는 다양한 탄소골격 및 화합물들이 해양에서 발견되어 왔다.
사실, 해양미생물은 해양환경에서 어느 곳에서나 존재하며, 고온, 고압, 고염도, 높은 pH 등과 같은 악조건에서도 생존할 수 있으며, 해양미생물은 육상미생물과는 다른 독특한 대사 경로를 가지고 있다.
특히 바실러스 속 미생물은 다양한 환경에서 성장할 수 있으며, 널리 분포되어 있는 세균의 한 예이며, 상당한 유전적 다양성을 나타낸다. 바실러스 균주에 대한 게놈 연구에 따르면, 바실러스 게놈의 약 8%가 항생제를 합성하는데 이용되는 것으로 나타났다. 반면, 인도양에서 분리된 해양유래 바실러스 서브틸리스 아종 스피지세니(B. subtilis subsp. spizisenii) 균주 gtP20b에 대한 게놈 연구를 통해 이차 대사물의 생합성을 위한 막대한 수의 유전자가 존재한다는 것이 밝혀졌다.
결론적으로, 해양 유래 바실러스 종에 대한 많은 천연물 탐색연구 결과, 당지질류, 지질펩타이드류, 폴리펩타이드류, 마크로락톤류, 지방산류, 폴리케타이드류, 리포아미드류 및 이소쿠마린류를 포함하는 다양한 종류의 이차 대사산물을 발견하게 되었다.
이러한 화합물 중 글리코리피드(당지질)는 해양 세균 및 진균으로부터 유래된 다양한 종류의 이차 대사산물로서, 항미생물, 항암, 면역억제 및 항바이러스 등의 활성을 지닌 것으로 알려졌다. 따라서, 강력한 활성을 가지는 여러 가지의 당지질 유도체들이 합성되었다.
예를 들면, 잘 알려진 글리코리피드인 KRN7000은 암(흑색종, 췌장암 및 결장암), 말라리아, 소아 당뇨 및 B형 간염과 같은 다양한 종류의 질병에 대하여 강력한 활성을 나타낸다. 세균, 진균 및 바이러스가 일으키는 전염병은 약물과 농약의 많은 발전에도 불구하고 여전히 공중보건과 농작물 생산에 위협적이다. 전염병의 영향은 특히 상대적으로 효과적인 약물을 이용하지 못하고, 약물내성이 널리 퍼져있는 개발도상국에서 크다. 항생제에 내성을 가지는 병원균의 계속적인 진화 때문에 새롭고 효과적인 항미생물 활성을 나타내는 화합물의 발견과 개발에 대한 많은 요구가 있어왔다.
이를 위하여 본 발명자들은 항미생물 활성을 나타내는 신물질 발견을 위한 지속적인 연구의 일부로서, 해양 퇴적토 유래의 세균인 바실러스 리케니포르미스(Bacillus licheniformis)의 발효액으로부터 두 개의 새로운 항미생물 당지질펩타이드인 이어도글루코마이드 A와 B를 이전에 발표하였다(대한민국 공개특허공보 제10-2014-0020022호 참조).
그러나 항생제 내성 병원균에 의한 위협, 항진균 약물의 제한적 효능 및 최근의 바이오테러리즘의 등장에 맞설 신규 항미생물 제제가 여전히 필요한 실정이다.
따라서, 본 발명이 해결하고자 하는 과제는 항생제 내성 병원균에 의한 위협, 항진균 약물의 제한적 효능 및 최근의 바이오테러리즘의 등장에 맞설 신규 항미생물 제제를 제공하는 것이다.
상기 기술적 과제를 달성하기 위하여 본 발명은 하기 화학식 1 내지 2로 표시되는 화합물인 신규의 글리코리피드를 제공한다.
본 발명에 따른 상기 화학식 1 또는 화학식 2로 표시되는 화합물은 항진균 활성 및 항균 활성을 가지며, 구체적으로 상기 화학식 1 또는 화학식 2로 표시되는 화합물이 항균 활성을 보이는 세균이 그람 양성 세균인 포도상구균(Staphylococcus aureus), 고초균(Bacillus subtilis) 및 세레우스균(Bacillus cereus)과 그람 음성 세균인 티푸스균(Salmonella typhi), 대장균(Escherichia coli) 및 녹농균(Pseudomonas aeruginosa)이며, 상기 화학식 1 또는 화학식 2로 표시되는 화합물이 항진균 활성을 보이는 진균이 식물 병원 진균인 아스페르길루스 니게르(Aspergillus niger), 리족토니아 솔라니(Rhizoctonia solani), 콜렉토트리쿰 아큐타툼(Colletotrichum acutatum), 보트리티스 세네레아(B. cenerea)과 인간 병원균인 칸디다 알비칸스(Candida albicans)인 것을 특징으로 한다.
또한 본 발명에 따른 화학식 1 또는 화학식 2로 표시되는 화합물은 기탁번호 KCTC 12254BP로 기탁된 미생물에 의해 제조되는 것을 특징으로 한다.
본 발명에 따르면 병원성 진균에 대한 항진균 활성 및 세균에 대한 항균 활성이 우수한 신규의 글리코리피드 화합물이 제공된다.

본 발명자들은 해양 유래 세균인 바실러스 리케니포르미스(Bacillus licheniformis)의 발효액의 에틸아세테이트 추출물을 각종 컬럼크로마토그래피 및 HPLC를 통한 분리, 정제과정을 거쳐 두 개의 새로운 항미생물 당지질펩타이드인 이어도글루코마이드 C(화합물 1, 화학식 1로 표시)와 이어도당지질(화합물 2, 화학식 2로 표시)을 분리하였으며(도 1, 상기 화학식 1 및 2 참조), 이 화합물 1과 2의 구조적 특성은 2D NMR 실험을 포함하는 광범위한 분광학적 분석을 통하여 규명하였으며, NMR 및 LC-MS 데이터 분석 및 화학적 유도체화 기술과 문헌 데이터 등의 종합적으로 분석하여 화합물 1과 2의 입체배열을 결정하였다.
이와 같은 화합물 1과 2는 포도상구균(Staphylococcus aureus), 고초균(Bacillus subtilis), 세레우스균(Bacillus cereus), 티푸스균(Salmonella typhi), 대장균(Escherichia coli) 및 녹농균(Pseudomonas aeruginosa)에 대하여 0.01~0.05의 MIC 값을 갖는 좋은 항생제의 특성을 보였다.
게다가, 화합물 1과 2의 항진균 활성을 식물 병원 진균인 아스페르길루스 니게르(Aspergillus niger), 리족토니아 솔라니(Rhizoctonia solani), 보트리티스 세네레아(Botrytis cenerea)와 콜렉토트리쿰 아큐타툼(Colletotrichum acutatum) 뿐만 아니라 인간 병원균인 칸디다 알비칸스(Candida albicans)에 대하여 평가한 결과, 화합물 1과 2는 0.03~0.05의 MIC 값으로 상기 병원균의 균사체 성장을 억제하는 것으로 보아, 화합물 1과 2는 새로운 항진균제 개발에 좋은 후보물질이 될 수 있음을 밝혀냈다.
이하 본 발명을 실시하기 위한 구체적인 내용을 설명하기로 한다.
먼저 화합물 1과 2의 화학 구조 분석 및 항미생물 활성 시험 등에 이용되었던 기구 및 조건 등의 과정에 대하여 설명한다.
핵자기 공명(NMR) 분광분석 데이터는 Varian Unity 500 분광계(McKinley, Spectra NJ)를 이용하여 얻었다. IR 스펙트럼을 JASCO FT/IR-4100(JASCO, Easton, MD, USA) 분광분석기를 사용하여 측정하였다. 선광값은 JASCO(DIP-1000) 디지털 편광계를 이용하여 측정하였다. UV 스펙트럼은 Shimadzu UV-1650PC(Shimadzu, Kyoto, Japan) 분광분석기를 사용하여 얻었다. 고해상도 ESIMS는 Shimadzu 하이브리드 이온트랩 비행시간 질량 분광분석기(Shimadzu, Kyoto, Japan)를 이용하여 측정하였다. HPLC는 RI101 Shodex 굴절률 검출기 및 M525 가변성 UV검출기(Shoko Scientific Co., Yokohama, Japan)가 장착된 PrimeLine 바이너리 펌프와 250mm × 4.6mm i.d, 5㎛, YMC-Pack-ODS-A 컬럼을 사용하여 수행되었다. 덱스트로즈, 아가와 같은 배양 배지는 Junsei Chemical(Tokyo, Japan)에서 구매하였으며, 트립톤, 효모 추출물, 쇠고기 추출물들은 Becton, Dickinson(Sparks, MD, USA)로부터 구매하였다. 유기용매들은 Duksan Pure Chemicals(Ansan, South Korea)로부터 구입하여 사용하였다. 사용된 모든 용매들은 스펙트럼 등급이거나 사용 전에 증류하였다. 천연 해수는 대한민국 동해의 20m 깊이에서 채집한 것을 사용하였다. 항미생물 활성시험을 위한 미생물들은 대한민국 한국생명공학연구원(KRIBB)의 미생물자원센터(KCTC)로부터 구입하여 이용하였다.
이어서, 균주 분리, 배양, 화합물 분리 및 구조 분석, 항미생물 활성의 시험에 대하여 상세하게 설명한다.
바실러스 리체니포르미스(Bacillus licheniformis)는 인간과 환경에 악영향이 적으면서, 아밀라제, 단백질분해효소, 항생제 및 특수 화학물질을 생성하는 발효 산업에 통상적으로 사용되어온 그람-양성 박테리아이다.육상에서 분리된 이 그룹의 몇 가지 균주들은 생물계면활성제, 항생제 및 항진균(폴리펩타이드) 화합물을 생성하기도 한다.
해양 환경은 생물학적 다양성이 육상 환경 보다 더 높기 때문에 해양세균은 독특한 구조적 특성을 지닌 생리활성 화합물의 잠재적 생산자로 각광을 받고 있다.따라서, 항미생물 활성을 나타내며 16s rRNA 시퀀싱(GenBank accession no. JQ349055)에 의해 바실러스 리체니포르미스로 동정된 균주 09IDYM23를 더욱 화학적으로 조사하였다. 이 동정된 09IDYM23에 대하여 기탁번호 KCTC 12254BP로 기탁하였다.
상기 균주는 대한민국의 남쪽 암초인 이어도에서 채집된 해양퇴적토 샘플로부터 변형된 베넷(Bennett)의 한천배지 상(염분 농도 18.3g/l인 1L 해수에 대하여, 1% 덱스트로즈, 0.2% 트립톤, 0.1% 효모 추출물, 0.1% 쇠고기 추출물, 1.8% 아가 및 pH 7.02)에서 분리되었다.
세균 바실러스 리케니포르미스는 염분농도와 해수의 pH가 각각 32.3g/L, 8.01인 이어도에서 채집된 퇴적토 샘플로부터 분리되었다. 미생물의 생리과정은 다양한 조건에서 달라지기 때문에, 세균의 성장조건은 다른 염분, pH, 온도를 조합한 배양을 통해서 최적화되었다. 균주의 최대 성장을 위한 최적의 염분, pH, 온도는 각각 18.3g/l, 7.02, 32℃임을 알았다. 대량배양을 위해 한천 상에서 배양한 단일 콜로니의 세균을 50ml 배넷의 배지(상기와 동일 조성물)가 담긴 세 개의 100ml 플라스크에 무균 접종하여, 종자 배양을 위해 32℃에서 2일간 종자배양하였다. 그 후에 종자는 1.2L의 멸균된 배양 배지가 들어있는 2L 플라스크(전체 42개 플라스크)에 접종하였고, 최적의 조건 하에서 7일간 배양하였다.
배양액을 수확한 후에, 연속원심분리기를 이용하여 6,000rpm에서 균체를 배양액으로부터 분리하였다. 발효액(50L)으로부터 얻어진 상등액을 EtOAc로 2번 추출하였다. EtOAc층을 40℃에서 감압하에 건조시켰다. 조추출물(5.6g)을 ODS를 이용한 개방형 컬럼크로마토그래피에 흡착시킨 후에, 용리액으로서 MeOH-H2O(v/v)(1:4, 2:3, 3:2, 4:1 및 5:0)를 단계별로 사용하여 구배 용출하였다. MeOH:H2O(4:1, v/v)로 용출된 분획물을 역상 MPLC 및 HPLC로 순차적으로 정제하여 순수한 화합물 1(6.6mg) 및 2(5.6mg)를 얻을 수 있었다. 화합물 1은 이어도글루코마이드 C로 명명하였으며 화학식 1로 표시되며, 화합물 2는 이어도당지질로 명명하였으며, 화학식 2로 표시된다.

화합물 1(3.9mg)을 4ml의 유리 바이알에 넣고, 600㎕ 1N HCl로 110℃에서 12시간 동안 가수분해하였다. 남아있는 HCl을 제거하고, 가수분해물을 n-hexane, EtOAc 및 MeOH를 이용하여 순차적으로 추출하였다.
n-hexane, EtOAc 및 수용성 MeOH 분획물로부터 각각 지방산(1.6mg), 알라닌(Ala, 0.6mg)으로 확인된 아미노산, 글루코스(Glc, 0.8mg)가 얻어졌으며, 1H NMR과 ESIMS 데이터 분석을 통해 확인하였다.
산 가수분해에 의해 얻은 알라닌(0.6mg)을 4ml의 유리 바이알에 넣고, 아세톤에 녹인 0.1% FDAA용액(Marfey 시약, 100㎕) 및 1N NaHCO3(20㎕)를 첨가하고, 40℃에서 1시간 동안 가열하였다. 반응혼합액을 냉각시키고, 1N HCl로 중화하였다. 유사한 방법으로, 표준 L-Ala와 D-Ala를 각각 FDAA시약으로 유도체화 하였다. 화합물 1에 있는 알라닌 및 표준 아미노산의 Marfey 유도체들을 역상 HPLC (YMC ODS-A column, 50, 2.0mm)를 이용하여 구배 용매 A(0.1% TFA를 포함하는 MeOH)와 용매 B(0.1% TFA를 포함하는 H2O)의 선형구배를 사용해 25℃에서 유동속도 0.5ml/min, 340nm에서의 UV검출기로 50분간 분석하였다. L-Ala와 D-Ala의 FDAA 유도체들의 유지 시간은 각각 39.9분, 41.3분이었으며, 화합물 1에 있는 알라닌의 FDAA 유도체의 유지 시간은 39.9분이었다.
화합물 1의 지방산(0.9mg)을 유리 바이알에 넣어 피리딘 120㎕에 용해하고, 실온에서 10분 동안 교반하였다. 상기 바이알에, (R)-(-)(MTPA-Cl) 20㎕을 첨가하고, (S)-MTPA 에스테르(1a)를 준비하기 위해 상기 혼합물을 실온에서 15시간동안 교반하였다. 반응의 완료는 ESIMS로 확인하였다. 상기 반응 혼합물을 진공에서 건조시켜 EtOAc에 재용해하고, 물로 씻은 후에 30% MeOH 수용액을 용리액으로서 사용하는 C18 HPLC로 정제하여 화합물 1a(0.5mg)를 얻었다.

비슷하게, (R)-MTPA 에스테르(1b)는 (S)-(+)-MTPA-Cl를 사용하여 준비하였다.
1b (0.6mg): 노란 오일; 1H NMR (CD3OD), δH 0.89 (d, H-16), 2.01 (m, H-15), 4.44 (m, H-14), 1.39 (H-13), 1.275 (brs. H-12), 3.51 (OCH3, s), 7.31.50 (15H, m), ESIMS m/z 511.57 [M + Na]+.
지방산 사슬 길이를 결정하기 위해, 화합물 2(2.6mg)를 HCl:MeOH(2:3)로 산 메탄분해 처리하였다. 반응 혼합물을 밀봉된 튜브에서 75℃로 16시간 동안 유지한 후에 얼음물로 냉각시켜 반응을 중지시키고, n-hexane로 분배하여 메틸 펜타데카노에이트를 얻었다. 메틸 펜타데카노에이트를 1H NMR과 ESIMS 데이터 분석으로 확인하였다.
메틸 펜타데카노에이트: 무색 오일 (1.7mg); 1H NMR (CD3OD), δH 0.87 (d, J = 6.5, H-14), 1.28 (brs., H-4 ~ H-14), 1.59 (m, H-3), 2.32 (t, J = 7.5, H-2), 3.64 (s, OCH3); ESIMS m/z 254.92 [MH]-).
화합물 1과 2에 있는 글루코스를 MeOH:EtOAc(1:4) 용매 시스템으로 TLC 분석을 하였다. 표준품의 α-와 β-D-Glc를 상기 용매 시스템을 사용하여 TLC 분석 후에 Rf 값을 기록하였다. 비슷하게, 화합물 1과 2의 산 가수분해와 메탄분해로부터 각각 얻어진 글루코스(Glc)을 상기와 같은 용매 시스템을 이용하여 분석하였다.
화합물 1과 2의 항미생물 활성은 세균 및 진균에 대하여 broth twofold dilution assay을 사용하여 평가하였다(표 2 및 3 참조). 항균 및 항진균 시험은 각각 nutrient broth 및 yeast maltose broth를 이용하여 수행하였다. 그 후에, MIC를 시각적으로 측정하였다. MIC는 미생물이 가시적 성장을 보이지 않는 샘플의 최소농도이다.
화합물 1은 노란 오일 상태로 분리되었고, 분자식은 1H, 13C NMR 데이터 및 HRESIMS(m/z 604.3337[M-H]-)를 기반으로 하여 C29H51NO12로 결정되었다. 화합물 1의 1H 및 13C NMR 데이터(표 1 참조) 및 HSQC 스펙트럼으로부터 2개의 메틸기, 15개의 메틸렌기, 산소와 질소에 연결된 각각 한개씩의 메틴기, 4개의 케톤 카보닐기 및 한 개의 글루코피라노스 분체 등으로 확인된 29개의 탄소 신호의 존재를 알 수 있었다. 3393cm-1 파장에서의 IR의 흡광도(brd)로 히드록시기의 존재를 알 수 있었다. 또한, 1630과 1537cm-1에서의 전형적인 IR 흡광도로부터 아미드 결합의 존재를 알 수 있었으며, 이것은 13C NMR 스펙트럼에서 질소와 연결된 탄소인 δc 49.0(C-2′) 신호와 δc 176.2(C-1)에서의 카르보닐기 신호로 확인할 수 있었다.
1H NMR 스펙트럼에서, δH 1.38(H-3′)의 이중 피크의 메틸기 신호, δH 4.36(H-2′)의 사중 피크의 메틴기 신호와 DMSO-d6에서 δH 8.05의 NH 신호 관찰로부터 확인된 아미드 작용기 등으로부터 알라닌의 존재를 알 수 있었다. δH 0.94(H-16)에서 공명하는 삼중 피크의 말단 메틸기, δH 1.55(H-15)의 sp2 메틸렌기, δH 3.58(H-14)의 산소와 연결된 메틴기와 긴 알킬 사슬 유래의 넓은 일중 피크를 나타내는 δH 1.29의 신호 등으로부터 지방산 unit [14-hydroxyhexadecanoic acid(HHDA)]의 존재를 알 수 있었다.
또한, 상기 HHDA unit은 화합물 1의 산 가수분해 후 ESIMS 분석 m/z 273.27[M-H-]으로 확인할 수 있었다. 1H NMR에서 δH 4.30(H-1′′′)의 아노머 양성자의 존재를 알 수 있었고, 13C NMR에서 δc 65.2, 71.9, 75.2, 78.1, 75.4와 103.6에서 공명하는 탄소 신호들로부터 β-Glc의 존재를 알 수 있었다. 또한, 화합물 1에 결합되어 있는 숙신산(SA)의 구조는 δc 174.1(C-1′′), 176.0(C-4′′)에서 공명하는 카르보닐 탄소의 존재와 up-field에 나타난 δc 29.9(C-3′′), 30.2(C-2′′)의 두 개의 메틸렌 탄소신호에 의해 결정할 수 있었다. HSQC 스펙트럼에서 δH 2.61(m, 중첩)의 수소와 상기 두 개의 메틸렌 탄소가 결합되어 있음을 알 수 있었다. 뿐만아니라, δH 2.61(H-2′′, H-3′′)의 양성자 신호 사이의 COSY 상관관계(도 2 참조)와 H-2′′와 C-1′′ 및 H-3′′와 C-4′′사이의 긴 범위(long range) HMBC 상관관계로 SA 분체의 존재를 확인 할 수 있었다. 상기 네 개의 부분구조를 HMBC 및 ROESY 실험에서 관찰된 상관관계를 기반으로 하여 연결할 수 있었다(도 2 참조). HHDA와 알라닌의 NH 사이의 연결은 알라닌의 NH 양성자(δH 8.05)와 HHDA의 C-1(δc 176.2) 사이의 긴 범위 HMBC 상관관계와 알라닌의 NH 양성자와 HHDA의 H-2(δH 2.22) 사이의 ROESY 상관관계로 확인할 수 있었다. HHDA unit의 δH 3.58(H-14)의 산소와 연결된 양성자는 δc 103.6(C-1′′′)의 Glc의 아노머 탄소와 HMBC 상관관계를 보였다.
Glc unit의 메틸렌 양성자들인 δH 4.19(H-6a′′′) 및 4.43(H-6b′′′)은 숙신산(SA) unit의 탄소인 δc 174.1(C-1′′)와 HMBC 상관관계를 나타내었다. 이와 같은 상세한 데이터 분석으로부터, 이어도글루코아마이드(ieodoglucoamide) C (1)의 완전한 구조를 결정할 수 있었다.
화합물 1의 알라닌과 HHDA의 C-14의 절대 입체화학은 각각 Marfey와 변형된 Mosher의 방법으로 결정하였다. 화합물 1(3.9mg)을 3N HCl을 사용하여 가수분해 하였으며, 반응 혼합물을 각각 n-hexane, EtOAc 및 methanol로 추출하여 각각 지방산, 알라닌 글루코스 잔기를 얻을 수 있었다. 알라닌은 Marfey의 시약으로 유도체화한 후에, 표준 L- 및 D-Ala의 Marfey 유도체와의 HPLC 비교분석에 의해 L-form으로 결정되었다.
HHDA의 C-14의 입체배열은 HHDA를 (R)-과 (S)-(MTPA-Cl)로 각각 아실화하여 (S)-와 (R)-MTPA 에스테르 화합물 1a와 1b를 얻어 결정하였다(도 3 참조).
화합물 1a와 1b의 1H NMR과 COSY 스펙트럼 분석으로 두 에스테르 화합물의 에스테르화된 탄소에 인접한 양성자들의 화학이동(chemical shift) 값을 정할 수 있었다. △ δS-R 값의 계산에 의해 HHDA의 C-14의 절대배열은 R 임을 알 수 있었다. Methanol 분획물에서 얻어진 글루코스 잔기는 표준품 샘플의 선광도 값[0.6mg, [α]23 D + 27° (c 0.45, H2O)]과 Rf 값(0.58)을 비교하여 β-D-Glc으로 결정하였다.
화합물 2는 무정형의 고체로서 분리되었고, 분자식은 1H, 13C NMR 데이터와 HRESIMS(m/z 663.3563[M+Na]+)에 근거하여 C30H56O14로 결정하였다. 화합물 2는 1H, 13C NMR 데이터(표 1 참조) 및 HSQC 스펙트럼에서 δH 0.87(d, J=6.5)의 메틸 신호, δH 1.29 (brs)의 긴 사슬 메틸렌 신호, δc 175.6의 에스테르 카르보닐 신호 및 산소와 연결된 하나의 메틴과 δH 3.72와 4.14 사이의 두 개의 메틸렌 신호를 보였으며, 이들 신호들로부터 글리세롤과 지방산인 펜타데카노 산(PDA)을 가지고 있음을 알 수 있었다. δH 4.29와 δH 4.36에서 공명하는 아노머 양성자들은 δc 105.0와 δc 104.9의 탄소와 각각 HSQC 교차 피크를 보였는데, 이것으로 두 개의 β-glucose의 존재를 알 수 있었다. 화합물 2에 대한 1H-1H COSY와 HMBC 스펙트럼의 광범위한 해석으로 글리세롤과 글루코스 분체에 대한 모든 13C와 1H NMR 신호를 규명할 수 있었다 (도 2 참조). 뿐만 아니라, 글루코스 unit의 아노머 신호인 δH 4.29(H-1′′)의 수소는 글리세롤의 δc 72.9(C-3′)의 탄소와 긴 범위 HMBC 상관관계를 보여 글루코스와 글리세롤이 연결됨을 알 수 있었다. 또한, 글리세롤과 PDA 사이의 연결은 H-1′(δH 4.14)와 C-1(δc 175.6) 사이의 HMBC 상관관계로부터 알 수 있었다. 지방산 사슬의 길이는 화합물 2를 메탄분해 후에 LC-MS 데이터 분석으로 결정하였다.
화합물 2의 메탄분해 생성물을 n-hexane으로 추출하여 메틸 에스테르를 얻었으며, 1H NMR 데이터와 ESIMS(m/z 254.92 [M-H]-)(SI-18)를 근거로 하여 분자식은 C14H29COOCH3로 결정하였고, 이 지방산은 메틸 펜타데카노에이트로 확인되었다. 수용성 메탄올층을 증발 건조시켜서 글루코스 분체를 얻을 수 있었다. 화합물 1과 유사하게, 화합물 2의 글루코스 분체는 표준품과의 TLC의 비교 분석으로 β-D-Glc임을 알 수 있었다. 화합물 2의 절대 배열은 김주선 등에 의해 발표된 모노아실다이갈락토실 글리세롤(MADG)과 동일한 구조적 특성 및 선광도 값 {[α]23 D +68.0° (c 0.3, MeOH)}을 갖기 때문에 MADG와 유사한 것으로 결정되었다.
또한 화합물 1과 2를 표준 배양액 희석법으로 세균과 진균에 대하여 항미생물 활성을 평가하여 그 결과를 각각 표 2 및 표 3에 나타냈다.
상기 표 2 및 표 3을 보면 이 화합물들은 그람 양성과 음성 세균 및 진균에 대하여 좋은 활성을 나타낸 것을 확인할 수 있다.
새로운 당지질 펩타이드인 이어도글루코마이드 C(화합물 1)는 세균 바실러스 리케니포르미스 발효액의 EtOAc 추출물로부터 얻어진 80% MeOH 분획물로부터 얻을 수 있었다. 상세한 NMR 데이터 분석에 의해, 화합물 1은 아미노산, 지방산, 글루코스 및 숙신산 unit으로 구성되어 있음을 알 수 있었다. 아미노산은 Marfey의 유도체 분석으로 L-Ala임을 알 수 있었다. 충분한 NMR 자료를 획득한 후에, 화합물 1의 가수분해와 ESIMS 데이터 분석으로부터 화합물 1에 포함되어 있는 지방산은 HHDA 임을 확인하였다. 또한, HHDA의 C-14의 절대 입체화학은 R-배열로 결정하였다. 표준품과의 TLC 분석과 1D와 2D의 상세한 NMR데이터 분석을 통해, 글루코스 unit은 β-D-Glc임을 알 수 있었다. 이와 유사하게, 화합물 1의 숙신산 unit은 HMBC 스펙트럼을 이용하여 각 원자들의 상관관계로 결정할 수 있었다. 상기 네 가지 unit을 HMBC와 ROSEY의 상관관계에 의해 연결할 수 있었으며, 새로운 화합물인 이어도글루코마이드 C(화합물 1)의 구조를 결정할 수 있었다.
새로운 당지질 유도체인 이어도당지질(화합물 2)을 세균 바실러스 리케니포르미스 발효액으로부터 얻어진 EtOAc 추출물의 100% MeOH 분획물로부터 분리하였다. 상세한 NMR 데이터의 해석으로 상기 화합물은 글리세롤, 지방산 및 글루코스 unit으로 구성되어 있음을 알 수 있었다. 표준품과의 TLC 분석에 의한 Rf 값을 비교하여 글루코스 잔기는 β-D-Glc 임을 알 수 있었다. 화합물 2의 메탄분해 생성물의 hexane 분획물을 ESIMS 데이터로 분석한 결과, 화합물 2의 지방산은 메틸 펜타데카노에이트로 결정할 수 있었다. 이와 같이 화합물 2의 완전한 구조를 광범위한 1D와 2D NMR의 데이터 분석으로 결정할 수 있었다.
표준물질[양성 대조군으로 세균에 대해서 아지쓰로마이신(azithromycin)을 사용하였으며 진균에 대해서 암포테리신 B(amphotericin B)을 이용]과 비교하였을 때, 화합물 1과 2는 좋은 항미생물 활성을 보였다(표 2,3 참조).
이번 연구에서, 이어도글루코마이드 C는 그람 양성 세균[포도상구균(Staphylococcus aureus), 고초균(Bacillus subtilis) 및 세레우스균(Bacillus cereus)]과 그람 음성 세균[티푸스균(Salmonella typhi), 대장균(Escherichia coli) 및 녹농균(Pseudomonas aeruginosa)]에 대하여 활성(MIC 값 범위: 0.01~0.05)이 있었으며, 이어도당지질(화합물 2) 보다 더 나은 활성이 있음을 알 수 있었다.
또한, 화합물 1과 2는 식물 병원 진균인 아스페르길루스 니게르(Aspergillus niger), 리족토니아 솔라니(Rhizoctonia solani), 콜렉토트리쿰 아큐타툼(Colletotrichum acutatum)과 보트리티스 세네레아(B. cenerea) 뿐만 아니라 인간 병원균인 칸디다 알비칸스(Candida albicans)에 대하여 0.03~0.05의 MIC 값을 가지는 좋은 활성을 나타냈다. 뿐만 아니라, 화합물 2는 화합물 1보다 칸디다 알비칸스에 대하여 더 좋은 활성이 관찰되었다. MADG 및 다이아실다이갈락토실 글리세롤(DADG)은 잘 알려진 당지질이다. DADG는 MADG 보다 자연계에 더 풍부하게 존재하고, 표면 활성을 가지는 성질 때문에 제약 및 화장품 분야에서 큰 잠재력을 가지고 있다.
결론적으로, 이와 같은 항미생물 결과들로 미루어보아, 당지질 화합물 1 과 2 또는 이들의 변형된 형태의 화합물들은 KRN7000과 비슷하거나, 더 좋은 활성을 가지는 유용한 살균제 및 약물을 개발하기 위한 선도물질이 될 수 있을 것으로 사료되며, 또한 신약 개발 및 농업 부문에서도 사용될 수 있다.

- 제 1항에 있어서, 상기 화학식 1 또는 화학식 2로 표시되는 화합물이 항진균 활성 및 항균 활성을 가지는 것을 특징으로 하는 글리코리피드.
- 제 2항에 있어서, 상기 화학식 1 또는 화학식 2로 표시되는 화합물이 항균 활성을 보이는 세균이 그람 양성 세균인 포도상구균(Staphylococcus aureus), 고초균(Bacillus subtilis) 및 세레우스균(Bacillus cereus)과 그람 음성 세균인 티푸스균(Salmonella typhi), 대장균(Escherichia coli) 및 녹농균(Pseudomonas aeruginosa)이며, 상기 화학식 1 또는 화학식 2로 표시되는 화합물이 항진균 활성을 보이는 진균이 식물 병원 진균인 아스페르길루스 니게르(Aspergillus niger), 리족토니아 솔라니(Rhizoctonia solani), 콜렉토트리쿰 아큐타툼(Colletotrichum acutatum), 보트리티스 세네레아(B. cenerea)과 인간 병원균인 칸디다 알비칸스(Candida albicans)인 것을 특징으로 하는 글리코리피드.
- 제 1항 내지 3항 중 전술한 중 어느 한 항에 있어서, 상기 화학식 1 또는 화학식 2로 표시되는 화합물이 기탁번호 KCTC 12254BP로 기탁된 미생물에 의해 제조되는 것을 특징으로 하는 글리코리피드.
