Abstract
본 발명은 발아현미의 수용성 GABA성분 추출 방법 및 이를 이용한 건강기능 조성물에 관한 것이다. 본 발명은 건조된 발아현미를 파쇄한 분말에 75℃에서 100℃사이의 물을 발아현미 중량(ℓ/㎏)비에 10 ∼ 15배 투입하여 2 ∼ 6시간 동안 교반하여 수용성 발아현미 추출액을 얻은 다음 추출액의 온도를 70℃까지 냉각시키는 제 1 공정; 발아현미의 건조 중량 대비 3% 중량에 해당하는 글루탐산 탈탄산 효소와 발아현미의 건조 중량 대비 3% 중량에 해당하는 당 분해 효소인 α-아밀라제를 추출탱크에 투입하여 발효온도 70℃, 발효시간 6시간으로 추출액을 발효시키는 제 2 공정; 발아현미 발효 추출액에서 고형분을 제거하기 위해 원심 탈수기나 필터프레스를 이용하여 고형분을 분리한 후 추출발효액 내 불용성 미립자, 단백질 및 거대 탄수화물을 제거하기 위하여 초고속 원심분리기를 이용하여 발아현미 수용성 1차 정제액을 추출하는 제 3 공정; 추출된 1차 정제액의 비활성 고분자 탄수화물과 세균성 미생물을 제거하기 위하여 TFF(Tangential Flow Filtration) 방식의 Membrane이 장착된 Ultra filtering system을 retentate와 permeate로 분리한 후, permeate되는 발아현미 2차 정제액을 추출하는 제 4 공정; 추출된 발아현미 2차 정제액을 감압 농축기에 투입하여 감압 후 농축 정도가 20brix이상 도달될 때까지 농축하거나 65brix이상 고농축하여 GABA 정제액을 추출하는 제 5 공정을 포함한다.
