Abstract
본 발명은 Nutlin-3a 화합물을 유효성분으로 포함하는 면역질환의 예방 또는 치료용 조성물에 관한 것이다. 본 발명의 유효성분인 Nutlin-3a 화합물은 p53 과발현을 통해 IL-17의 발현을 억제시킴으로써 관절염 동물 모델에서 관절염을 개선시키는 효과 있으므로, 이를 유효성분으로 포함하는 본 발명의 조성물은 면역질환 예방 또는 치료에서 유용하게 사용될 수 있다.
Description
본 발명은 면역질환의 예방 또는 치료용 조성물에 관한 것으로서, 보다 구체적으로는 Nutlin-3a 화합물을 유효성분으로 포함하는 면역질환의 예방 또는 치료용 조성물에 관한 것이다.
면역 반응은 생체의 내부 환경이 외부 인자에 대하여 방어하는 현상을 말한다. 이러한 면역 시스템은 박테리아 및 바이러스와 같은 병원성 외래 미생물의 인식 및 제거에 의한 감시 및 방어 메카니즘과 함께 발달되어 왔다. 따라서 생체는 자신의 세포 또는 조직(자기-항원)을 외래 미생물(비자기-항원)과 구별하고, 자기-항원에 대해서 반응하지 않거나, 또한 반응하였다 하여도 면역 기능을 나타내지 않는다(면역관용).
그러나 일련의 면역 반응에서 비정상적 반응이 일어날 때, 림프구, 특히 T 세포는 자기-항원에 강력하게 반응하고, 조직손상을 일으킬 수 있다. 이러한 상태를 자가면역질환이라고 한다. 비정상적인 면역반응을 유발하는 원인 중 하나는 바이러스 또는 박테리아에 대한 정상적인 면역반응이 어떠한 종류의 시스템에 의해 자기-항원에 대한 면역반응으로 변화되기 때문이다. 또한, 장기이식 또는 골수(조혈 간세포)이식의 경우에 피이식체에 있어서 이식조직에 대하여 거부면역 반응이 발생하는데, 이러한 면역반응은 정상적인 면역반응이라 할지라도 억제하는 것이 치료적으로 바람직하다.
이러한 자가면역질환은 대부분 면역 매개성 만성 염증 질환이다. 따라서 자가면역질환의 치료제로 개발된 많은 약제들은 염증의 유발과 진행을 억제하는 것에 초점이 맞춰져 있으며, 또한 T 세포의 활성화를 억제하는 화합물 면역억제제 투여법도 널리 사용되고 있다. 현재까지 자가면역질환 치료를 위해 화합물 면역억제제가 많이 개발되어 있으나 사이클로스포린에이가 가장 우수한 임상효과를 나타내고 있으며 급성 이식편대숙주병을 포함하여 자가면역질환, 장기이식거부반응, 다양한 염증질환에 널리 쓰이고 있다. 사이클로스포린에이는 고용량으로 사용할 경우 T 세포의 활성화를 완벽하게 억제하여 질병을 치료할 수 있으나 신장독성을 포함하여 상당한 부작용을 나타내는 단점을 가지고 있어, 저용량으로 사용하는 것을 권장하고 있다.
한편, Nutlin-3은 MDM2에 결합해 MDM2와 p53 단백질 간 결합을 저해하는 소형분자 화합물로서 고형암 등의 치료를 위해 현재 임상에서 개발 중인 항암 약물이다. 자세하게는, MDM2 단백질은 암을 억제하는 유전자로 알려진 p53의 전이활성(transactivation) 부위에 결합하여 활성화 능력을 차단시키는 역할을 하는데, Nutlin-3 화합물의 경우 p53 과 유사한 분자 구조를 가지기 때문에 MDM2 단백질과 결합을 형성하며, 이로 인해 p53 단백질이 MDM2 단백질과 결합하지 못함에 따라 활성화 능력이 유지되어 p53이 과발현되게 되는 것이다.
그러나 상기 Nutlin-3 화합물은 현재 임상에서 개발 중인 항암 약물로만 알려져 있으며, 면역질환 치료와 관련하여 효과를 가진다는 내용에 대해서는 전혀 보고된 바가 없다.
이에 본 발명자들은 Nutlin-3a 화합물이 IL-17 발현을 억제시키는 기작을 통하여, 관절염을 개선시킬 수 있음을 확인함으로써 본 발명을 완성하게 되었다.
따라서 본 발명의 목적은 다양한 면역질환을 예방 또는 치료할 수 있는 면역질환 예방 또는 치료용 조성물을 제공하는 것이다.
상기와 같은 본 발명의 목적을 달성하기 위해서, 본 발명은 Nutlin-3a 화합물을 유효성분으로 포함하는 면역질환의 예방 또는 치료용 조성물을 제공한다.
본 발명의 일실시예에 있어서, 상기 Nutlin-3a 화합물은 IL-17 생성을 억제시킬 수 있다.
본 발명의 일실시예에 있어서, 상기 IL-17 생성 억제는 p53 과발현을 통해 이루어질 수 있다.
본 발명의 일실시예에 있어서, 상기 면역질환은 자가면역질환; 염증성질환; 및 세포, 조직 또는 기관의 이식거부(transplantation rejection)질환으로 이루어진 군으로부터 선택될 수 있다.
본 발명의 일실시예에 있어서, 상기 자가면역질환 또는 염증성 질환은 류마티스 관절염 (Rheumatoid Arthritis), 천식 (Asthma), 피부염 (Dermititis), 건선 (Psoriasis), 낭섬유증 (Cystic Fibrosis), 다발성 경화증 (Multiple Sclerosis), 전신성 홍반성 루푸스(systemic lupus erythematosus), 쇼그렌 증후군(Sjogren syndrome), 하시모토 갑상선(Hashimoto thyroiditis), 다발성근염(polymyositis), 경피증(scleroderma), 아디슨병(Addison disease), 백반증(vitiligo), 악성빈혈(pernicious anemia), 사구체신염(glomerulonephritis), 폐섬유증(pulmonary fibrosis), 염증성장질환 (Inflammatory Bowel Dieseses), 자가면역성 당뇨 (Autoimmune Diabetes), 당뇨 망막증 (Diabetic retinopathy), 비염 (Rhinitis), 혀혈-재관류 손상 (Ischemia-reperfusion injury), 혈관성형술후 재협착 (Post-angioplasty restenosis), 만성 폐색성 심장 질환 (Chronic obstructive pulmonary diseases; COPD), 그레이브병 (Graves disease), 위장관 알러지 (Gastrointestinal allergies), 결막염 (Conjunctivitis), 죽상경화증 (Atherosclerosis), 관상동맥질환 (Coronary artery disease), 협심증 (Angina) 및 소동맥 질환으로 이루어진 군으로부터 선택될 수 있다.
본 발명의 유효성분인 Nutlin-3a 화합물은 p53 과발현을 통해 IL-17의 발현을 억제시킴으로써 관절염 동물 모델에서 관절염을 개선시키는 효과 있으므로, 이를 유효성분으로 포함하는 본 발명의 조성물은 면역질환 예방 또는 치료에서 유용하게 사용될 수 있다.

본 발명은 Nutlin-3a 화합물이 p53 과발현시켜 IL-17 생성을 억제하는 작용을 통해 면역질환을 예방 또는 치료할 수 있는 효과가 있음을 최초로 규명하였으며, 따라서 본 발명은 Nutlin-3a 화합물을 유효성분으로 함유하는 면역질환의 예방 또는 치료용 조성물을 제공함에 그 특징이 있다.
일반적으로 ‘Nutlin-3a’는 MDM2에 결합해 MDM2와 p53 단백질 간 결합을 저해하는 소형분자 화합물로서, 항암 효과가 알려져 있으며, 화학구조는 하기 도 1과 같다
한편, 본 발명에서는 항암 활성이 있는 것으로 알려진 Nutlin-3a 화합물이 면역질환을 치료할 수 있다는 사실을 확인하였는데, 본 발명의 하기 실시예 1에 따르면, 마우스로부터 분리한 비장세포를 Th17 분화조건에서 Nutlin-3a 화합물을 처리하는 경우 병원성 세포인 Th17 세포로부터 생성되는 염증성 사이토카인인 IL-17 생성이 억제되는 것으로 나타났다.
또한 본 발명의 하기 실시예 2에 따르면, 관절염 마우스 동물 모델에 Nutlin-3a 화합물을 주입한 경우 관절염 수치, 염증 수치 및 연골 손상 정도가 유의적으로 감소되는 것으로 나타나 관절염이 개선되는 효과가 있음을 확인할 수 있었다.
또한 본 발명의 하기 실시예 3에 따르면, 관절염 마우스 동물 모델에 Nutlin-3a 화합물을 주입한 후 마우스에서 비장세포를 분리하여 IL-17 발현 정도를 측정한 결과, 염증성 사이토카인인 IL-17 발현이 억제되는 것을 확인할 수 있었다.
따라서 Nutlin-3a 화합물은 염증성 사이토카인인 IL-17 발현(생성)을 억제시킴으로써 관절염 동물 모델에서 관절염을 개선시키는 효과 있으므로, 이를 유효성분으로 포함하는 본 발명의 조성물은 면역질환 예방 또는 치료에서 유용하게 사용될 수 있다.
본 발명에서 상기 ‘면역질환’은 포유류 면역계의 구성성분들이 포유류의 병리상태를 야기하거나, 매개하거나 또는 기타 공헌하는 질환을 의미한다. 또한, 면역 반응의 자극 또는 중단이 그 질병의 진행에 보상적인 효과를 갖는 질환을 모두 포함할 수 있는데, 본 발명에서는 과민성 면역반응으로 인해 야기되는 질환들을 포함할 수 있다.
본 발명에서 상기 '치료'란, 달리 언급되지 않는 한, 상기 용어가 적용되는 질환 또는 질병, 또는 상기 질환 또는 질병의 하나 이상의 증상을 역전시키거나, 완화시키거나, 그 진행을 억제하거나, 또는 예방하는 것을 의미하며, 본원에서 사용된 상기 '치료'란 용어는 '치료하는'이 상기와 같이 정의될 때 치료하는 행위를 말한다. 따라서 포유동물에 있어서 면역질환의 "치료" 또는 "치료요법" 은 하기의 하나 이상을 포함할 수 있다:
(1) 면역질환의 성장을 저해함, 즉, 그 발달을 저지시킴,
(2) 면역질환의 확산을 예방함, 즉, 전이를 예방함,
(3) 면역질환의 경감시킴.
(4) 면역질환의 재발을 예방함, 및
(5) 면역질환의 증상을 완화함(palliating)
본 발명에서 제공하는 면역질환의 예방 또는 치료용 조성물은 약제학적 조성물로 사용될 수 있다.
본 발명의 약제학적 조성물은 유효성분인 Nutlin-3a 화합물 이외에, 당업계의 공지된 담체 및 희석제와 함께 적절한 제형으로 투여될 수 있으며, 목적하는 방법에 따라 비경구 투여, 예를 들면, 정맥 내 주사, 근육 내 주사, 복강 내 주사, 피하주사, 좌제 등의 제형을 가질 수 있다. 본 발명의 치료제가 경구로 투여될 때, 치료제는 정제, 캡슐, 입자, 분말, 환제, 트로키, 내액제, 현탁제, 유화제, 또는 시럽 등의 제형으로 투여될 수 있다.
상기 제형들은 약제학적 조성물에 통상적으로 사용되는 적당한 부형제, 충전물, 결합제, 습윤제, 분해제, 윤활제, 계면활성제, 분산제, 완충액, 방부제, 용해보조제, 소독제, 감미료, 향신료, 진통제, 안정화제, 등장액제 등을 이용한 통상적인 방법에 의해 제조될 수 있다.
상기 기술된 각각의 제형은 약학적으로 허용되는 담체 또는 첨가제를 포함할 수 있다. 상기 담체 또는 첨가제의 구체적인 예로는 물, 약학적으로 허용되는 유기용매, 콜라겐, 폴리비닐 알코올, 폴리비닐피롤리딘, 카복시비닐 중합체, 알긴산 나트륨, 수용성 덱스트란, 카복시메틸 나트륨 녹말, 펙틴, 잔탄 고무, 아라비아 고무, 카제인, 겔라틴, 한천, 글리세롤, 프로필렌 글리콜, 폴리에틸글리콜, 바셀린, 파라핀, 스테아릴 알코올, 스테아린산, 인간 혈청 알부민, 만니톨, 소비톨 및 젖산 등이 있다. 한 개 또는 그 이상의 첨가제가 조제 형태에 따라 선택되거나 또는 적절히 조합될 수 있다. 더 나아가, 유전자요법 치료제 투여방법으로는, 정맥내, 동맥내 투여와 같은 통상적인 전신투여 이외에 표적세포로의 국소적 투여가 행해질 수 있으며, 카테터 기술 및 외과적 수술과 조합된 투여 방법이 사용될 수 있다.
본 발명의 약제학적 조성물이 치료효과를 나타낼 수 있는 면역질환으로는, 자가면역질환; 염증성질환; 및 세포, 조직 또는 기관의 이식거부(transplantation rejection)질환을 포함할 수 있다.
본 발명에서 상기 ‘자가면역질환’이란 자기항원에 대하여 면역반응이 일어나, 이로 인하여 자신의 조직을 공격하는 현상이 발생하는데 이러한 과정에 의해 발생되는 질환을 의미한다.
본 발명에서 상기 ‘염증성질환’이란 염증유발인자 또는 방사선조사 등 유해한 자극으로 인해 인체 면역체계를 과도하게 항진시켜 대식세포와 같은 면역세포에서 분비되는 TNF-α(tumor necrosis factor-α), IL-1(interleukin-1), IL-6, 프로스타글란딘(prostagladin), 루코트리엔(luecotriene) 또는 산화질소(nitric oxide, NO)와 같은 염증유발물질(염증성 사이토카인)에 의해 유발되는 질환을 말한다.
상기 자가면역질환 또는 염증성 질환의 예로는 류마티스 관절염 (Rheumatoid Arthritis), 천식 (Asthma), 피부염 (Dermititis), 건선 (Psoriasis), 낭섬유증 (Cystic Fibrosis), 다발성 경화증 (Multiple Sclerosis), 전신성 홍반성 루푸스(systemic lupus erythematosus), 쇼그렌 증후군(Sjogren syndrome), 하시모토 갑상선(Hashimoto thyroiditis), 다발성근염(polymyositis), 경피증(scleroderma), 아디슨병(Addison disease), 백반증(vitiligo), 악성빈혈(pernicious anemia), 사구체신염(glomerulonephritis) 및 폐섬유증(pulmonary fibrosis), 염증성장질환 (Inflammatory Bowel Dieseses), 자가면역성 당뇨 (Autoimmune Diabetes), 당뇨 망막증 (Diabetic retinopathy), 비염 (Rhinitis), 혀혈-재관류 손상 (Ischemia-reperfusion injury), 혈관성형술후 재협착 (Post-angioplasty restenosis), 만성 폐색성 심장 질환 (Chronic obstructive pulmonary diseases; COPD), 그레이브병 (Graves disease), 위장관 알러지 (Gastrointestinal allergies), 결막염 (Conjunctivitis), 죽상경화증 (Atherosclerosis), 관상동맥질환 (Coronary artery disease), 협심증 (Angina) 및 소동맥 질환 등을 들 수 있되, 이에 한정되는 것은 아니다.
본 발명의 약제학적 조성물은 Nutlin-3a 화합물을 약제학적으로 허용되는 담체와 함께 약제학적으로 유효한 양으로 포함할 수 있다.
본 발명에서 '약제학적으로 유효한 양'이란, 치료하고자 하는 면역질환에 대해 완화, 억제, 호전 및/또는 완치효과를 나타내는 유효성분의 양을 말한다. 본 발명의 Nutlin-3a 화합물의 투여량은 질환의 종류, 질환의 중증도, 조성물에 함유된 유효성분 및 다른 성분의 함량, 제형의 종류, 및 환자의 연령, 체중, 일반 건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료기간, 동시 사용되는 약물을 비롯한 다양한 인자에 따라 조절될 수 있다. 예를 들면, 치료적으로 유효한 투여량은, 초기에는 세포배양을 통한 시험관내 분석을 사용하여 결정할 수 있으며, 동물모델에서 실험을 수행하여 혈중 Nutlin-3a 화합물의 농도 범위가 세포배양에서 결정된 IC50(시험관내 분석, 약물 처리 시, 배양세포의 50%에 대한 치사량의 테스트 화합물의 농도)을 통하여 결정할 수 있다. 당 분야에서 과도한 실험을 거치지 않고도 치료에 유효한 양을 결정할 수 있을 것이며, 이러한 정보를 이용하여 인간에서 유용한 투여량을 더욱 정확하게 결정 할 수 있다.
본 발명은 또한 면역질환의 예방 또는 치료용 의약의 제조를 위한 상기 Nutlin-3a 화합물을 유효성분으로 포함하는 조성물의 용도를 제공한다. Nutlin-3a 화합물을 유효성분으로 포함하는 본 발명의 조성물은 면역질환의 예방 또는 치료용 의약의 제조를 위한 용도로 이용될 수 있다.
또한 본 발명은 포유동물에게 치료상 유효량의 본 발명의 약제학적 조성물을 투여하는 것을 포함하는 면역질환의 예방 또는 치료방법을 제공한다.
여기에서 사용된 용어 ‘포유동물’은 치료, 관찰 또는 실험의 대상인 포유동물을 말하며, 바람직하게는 인간을 말한다.
여기에서 사용된 용어 ‘치료상 유효량’상기 약제학적으로 유효한 양과 동일한 개념이다.
본 발명의 치료방법에서 본 발명의 Nutlin-3a 화합물을 유효성분으로 포함하는 조성물은 경구, 직장, 정맥내, 동맥내, 복강내, 근육내, 흉골내, 경피, 국소, 안구내 또는 피내 경로를 통해 통상적인 방식으로 투여할 수 있다.
본 발명의 이점 및 특징, 그리고 그것들을 달성하는 방법은 상세하게 후술되어 있는 실시예들을 참조하면 명확해질 것이다. 이하, 본 발명을 실시예에 의해 상세히 설명하기로 한다. 그러나 이들 실시예들은 본 발명을 구체적으로 설명하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.

-17 생성억제 활성 분석
본 발명자들은 Nutlin-3a 화합물이 병원성 세포인 Th17세포에서 염증성 사이토카인인 IL-17의 생성을 억제하는 활성이 있는지를 확인하기 위하여, FACS, ELISA 및 RT-PCR을 통해 IL-17의 발현 정도를 측정하였다.

마우스로부터 적출된 비장은 유리 슬라이드를 이용하여(teasing slide) 비장 조직을 잘게 갈은 후 적혈구 용혈 용액으로 비장내의 적혈구를 제거한 후, PBS 완충 용액을 첨가하고 원심 분리하여 세척함으로써 비장(spleen)세포를 수득하였다.
상기 수득된 비장세포 1×106을 anti-CD3 항체가 1μg/mL로 코팅(coating) 처리된 24 웰 플레이트에 분주하고, Th17세포를 자극시킬 수 있는 조건인 anti-CD28 항체 1μg/mL, TGF-β 2ng/ml, IL-6 20ng/ml, anti-IL-4 10 μg/ml, anti-IFNγ 10 μg/ml를 동시에 함께 처리한 후, 3일 동안 세포를 배양하여 Th17 세포 분화를 유도하였다. 이때, 상기 세포에 Nutlin-3a 화합물을 각각 50nM, 100nM, 200nM의 농도로 처리하여 배양하였다.

ELISA
분석
생성된 IL-17 사이토카인의 양을 측정하기 위하여, 세포가 배양된 상층액을 모아 IL-17 및 sandwich ELISA를 이용하여 IL-17 발현 정도를 조사하였다. 이를 위해 먼저 96웰 플레이트에 단클론성 항-IL-17을 2μg/mL으로 4℃에서 밤새 반응시킨 후, 차단용액(1% BSA/PBST)으로 비특이적 결합을 차단시켰다. IL-17 recombinant를 1/2씩 연속 희석하여 standard로 사용하였으며, 세포배양 상층액을 넣고 실온에서 2시간 동안 반응시켰다. 이후, biotinylated 항-IL-17을 2시간 동안 실온에서 반응시킨 후 4회 세척한 다음, ExtraAvidin-Alkaline Phosphatase conjugate를 희석하여 첨가하고 실온에서 2시간 동안 반응시켰다. 이후, PNPP/DEA 용액을 넣고 발색한 후 405 nm 파장에서 흡광도를 측정하였다.
그 결과 도 2에 나타낸 바와 같이, 병원성 세포인 Th17 세포로부터 생성되는 염증성 사이토카인인 IL-17은 본 발명의 Nutlin-3a 화합물에 의해 생성이 억제되는 것으로 나타났다.

Th17세포로 분화한지 3일 후에 세포를 모으고 FACs 버퍼로 세척한 후, 비특이성 결합을 억제하기 위해 블로킹을 4℃에서 5분간 반응시킨 다음, anti CD4 percp cy5.5를 넣고 4℃에서 30분간 반응시키고 FACs 버퍼로 세척하였다. 세포내 염색을 위하여 permeabilization buffer로 30분간 반응시킨 후, perm wash buffer로 세척하였다. 그 후 Anti IL-17 PE를 넣고 4℃에서 30분간 반응시킨 후 perm wash buffer로 세척하였다. 염색이 끝난 세포는 FACS 버퍼로 세척한 후, 유세포분석기(FACS,fluorescent-activated cell sorter)를 이용하여 IL-17 발현 정도를 분석하였다.
그 결과 도 3에 나타낸 바와 같이, Nutlin-3a 화합물을 농도별(50nM, 100nM, 200nM)로 처리한 경우 IL-17의 발현은 각각 2.62, 2.7, 2.72로 나타난 반면, 처리하지 않은 대조군은 IL-17의 발현이 6.09로 나타났다. 이러한 결과를 통해 본 발명의 Nutlin-3a 화합물의 처리가 IL-17의 발현은 감소시킨다는 사실을 확인할 수 있었다.

본 발명의 Nutlin-3a 화합물이 IL-17 발현을 억제시키는 작용이 있는지를 조사하기 위해, 상기 실시예<1-1>의 Th17 분화 과정을 거친 세포에서 IL-17 발현 정도를 RT-PCR을 수행하여 분석하였다.
Real-time PCR은 StepOnePlus™ Real-Time PCR System 을 이용하여 수행하였고, 이를 위한 반응 화합물은 1μg으로 합성한 cDNA를 1/10로 희석한 것 5 μl, Taqman GEx master mix 10 μl, Taqman probe (Applied biosystems) 1μl를 혼합한 후 증류수로 최종 부피를 20μl로 만들었다. 반응 조건은 50℃에서 10분, 95℃ 10분, 95℃ 15초, 60℃ 1분으로 총 50회 반응시켰다. Cycle threshold (Ct)값을 분석하여 p53의 mRNA 발현양을 GAPDH mRNA 발현양과의 상대정량으로 나타내었다.

그 결과 도 4에서 나타낸 바와 같이, 본 발명의 Nutlin-3a 화합물 50nM의 농도로 처리하였을 때 세포에서 IL-17의 발현이 처리하지 않은 대조군에 비해 현저히 감소하는 것으로 나타났다.

-3a 화합물의 처리에 따른 관절염 개선 효과 (

)
본 발명의 Nutlin-3a 화합물의 처리에 따른 관절염의 치료 효과를 알아보기 위하여, 관절염 동물 모델을 제조에 Nutlin-3a 화합물을 주입하여 p53을 과발현 시킨 후 관절염 수치, 세포의 증식, 염증 수치 및 연골 손상 정도를 측정하고 면역조직화학염색법을 통한 관절파괴 정도 분석하였다.
실시예
<2-1> 관절염 동물 모델 제작
관절염 마우스 모델 (collagen-induced arthritis: 이하 간단하게 ‘CIA'라 함)을 제조하기 위해 6주령의 DBA/1J 마우스에 제2형 콜라겐과 CFA(complete freud's adjuvant) 각각을 100μl 씩 혼합하여 피내주사로 1차 면역하였다. 1주일 후에 Nutlin-3a 화합물을 3일마다 200μg/mice, 복강내 주사하였다 (총 9회).
실시예
<2-2> 관절염 수치 측정
본 발명의 Nutlin-3a 화합물 처리에 따른 관절염의 치료 효과를 알아보기 위하여, 상기 실시예<2-1>에서 제조된 관절염 동물 모델에, Nutlin-3a 화합물을 주입하여 p53을 과발현 시킨 후 관절염증의 임상수치(clinical score)를 측정하였다.
상기 임상수치는 관절염증 질환의 진행 정도를 매일 마우스의 관절염 병변을 육안으로 관찰하여, 발이나 꼬리 부위의 종창 및 발적 여부 대한 각각의 점수를 매기는 임상점수체계를 사용하여 평가하였다.
그 결과 도 5에서 나타낸 바와 같이, 본 발명의 Nutlin-3a 화합물을 처리한 경우 CIA군에 비하여 관절염 수치가 유의적으로 낮아지는 것을 확인할 수 있었다.
실시예
<2-3> 세포의 증식 측정
분리한 비장세포 2×105 를 96 웰 플레이트에 분주하고, 제2형 콜라겐 100 μg으로 자극하여 3일간 배양하였다. 최종 배양 시간 16시간 전에 [3H]thymidine 1 μCi을 세포에 처리하고, 16시간 후에 (총 3일 배양) Betaplate scintillation counter 기계를 사용하여 세포의 증식을 측정하였다.
그 결과 도 6에서 나타낸 바와 같이, 본 발명의 Nutlin-3a 화합물을 처리한 경우 CIA군의 경우 세포 증직이 억제되는 것을 확인할 수 있었다.
실시예
<2-4> 면역조직화학염색법을 통한 관절파괴 정도 분석
면역조직화학염색법은 다음과 같은 과정으로 실시하였다. 상기에서 사용하는 마우스 군의 관절을 채취 후, 10% 중성완충포르말린에 고정하고 EDTA로 뼈를 탈회시킨 다음 파라핀에 포매한 후 관절 조직을 7um 두께의 절편을 만들어 슬라이드에 부착하였다. 기본 염색을 진행하기 전에 자일렌을 이용하여 탈 파라핀 과정을 통과 후, 에탄올을 고농도에서 저농도까지 함수시켰다. 염색과정은 헤마톡실린/에오신 (hematoxylin/eosin) 염색을 수행하였으며, 연골에 포함된 프로테오글리칸을 감지할 수 있는 Safranin O와 Toluidine blue 방법을 사용하였다. 마우스 관절에서 면역조직화학염색법을 시행하였으며, 광학현미경으로 분석하였다.
그 결과 도 7에서 나타낸 바와 같이, 관절염 마우스 동물 모델에 본 발명의 Nutlin-3a 화합물을 처리한 경우 CIA군에 비하여 마우스의 골수 부위에서 관절의 파괴가 완화되는 것을 확인할 수 있었다.
실시예
<2-5> 염증 수치 및 연골 손상 정도 분석
상기 실시예<2-4>의 조직염색 결과를 바탕으로 염증 수치 및 연골 손상 정도를 수치화하였다. 면역조직화학염색법 수행 후, 염증 수치는 H&E 염색 결과를, 연골 손상은 safranin O와 Toluidine blue 염색 결과를 수치화하였다. 수치화 기준은 아래와 같다.
0 = no destruction
1 = minimal erosion, limited to single spots
2 = slight to moderate erosion in a limited area
3 = more extensive erosion
4 = general destruction
그 결과 도 8에서 나타낸 바와 같이, 관절염 마우스 동물 모델에 본 발명의 Nutlin-3a 화합물을 처리한 경우 CIA군에 비하여 염증 수치 및 연골 손상이 확연이 줄어드는 것으로 나타났다.

본 발명의 Nutlin-3a 화합물이 관절염 증상 완화에 효과가 보이는 것을 상기 실시예 2를 통해 확인할 수 있었는데, 이러한 기작이 IL-17 발현 억제를 통해 이루어진다는 것을 확인하기 위하여, 관절염 동물 모델에 본 발명의 Nutlin-3a 화합물을 처리한 후 IL-17 발현여부를 측정하였다.
IL-17 발현여부는 CIA군(관절염 동물 모델 군) 및 Nutlin-3a 주입군의 비장에서 세포를 분리하여 PMA (25 ng/ml)와 ionomycin (250 ng/ml)에 노출시켜 4시간 자극한 후, FACS stain 분석을 통하여 IL-17 발현을 조사하였다.
그 결과 도 9에서 나타낸 바와 같이, Nutlin-3a 주입군에서 CIA에 비하여 IL-17의 발현이 억제됨을 관찰할 수 있었다.
결론적으로 관절염 동물 모델에서 IL-17의 발현이 억제됨에 따라 관절염증의 개선효과가 나타나며, 이러한 IL-17의 발현 억제는 Nutlin-3a에 의해 조절될 수 있다는 사실을 알 수 있었다.
이제까지 본 발명에 대하여 그 바람직한 실시예들을 중심으로 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 특허청구범위에 나타나 있으며, 그와 동등한 범위 내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.
