Description
The present invention obtains a liquid component, a solid component, and a lipid extract from an velvet antler or a hard antler by using an ultrahigh pressure, low temperature, enzyme-added manufacturing process for the preparation of health foods, functional drinks, and functional cosmetics. It relates to a method for developing as a component.
Deer antler, which has been used as an essential component of oriental traditional Chinese medicine for thousands of years, is covered with a lot of hair and is composed of soft bone tissue. It is known to improve body metabolism and to treat various diseases.
These antlers or rusts are harvested from red deer (Cervus elaphus) and deer (Cervus nippon Temminck), which begin to grow each spring and harvest in the fall.
Historically, Dongbobom (1613) reported that antler is a branched horn, composed of soft tissues, and rusted, composed of hard tissues. Deer antler or rust is characterized by warm, sweet, sour, non-toxic, flammable, limbs and lower back and pain, weak kidneys and cold, weak leg knees, oil well, dream snow, indigestion of woman, redness It's good, it's easy to wear, has been used as a correction tonic
In China, antler was used as a preventive medicine to treat erectile dysfunction, dysmenorrhea, uterine bleeding, dizziness and insomnia, amnesia, wounds, pain and arthritis in 206-220 BC. In the Roman Plinius Secundus era, antler or rust was used as a therapeutic drug to treat baldness and epilepsy. Topsell (1607) also powdered antler or rust, and was used to treat baldness, acne, toothache and even snake bites. Therefore, antler and rust shell have been used as drugs and therapeutic agents in the East as well as in the West.
In the Orient, known methods for producing products using antler or rust are known to extract active ingredients using hot air or freeze-drying. First it was dried for 40 days at 60 ℃ or more in hot air to store the antler or rust. Then the fire was scorched to remove the fur and then the scraped fur was scraped off. The hot water extraction method was used as a traditional method of extracting the active ingredient after thinly cutting the dried antler and rust shell. This method destroys the biological function of the active ingredient due to high production cost, time consuming and heat treatment. The disadvantage of this method is also that production quality control is very difficult because the components of the active ingredient are not constant.
On the other hand, in order to more easily produce a product containing such antler or rust rust, a method that can more effectively solubilize (sollubilization) or powder the antler has been continuously developed.
For example, Patent Literature 1 introduces a hot water extraction method of antler and green antler, and Patent Literature 2 (Method of manufacturing antler concentrate powder through antler enzyme treatment) and Patent Literature 3 (Method for producing hydrolyzate of antler) are proteins. A hydrolase extraction method is disclosed, and Patent Document 4 (a new antler decomposing bacterium and its separation method) discloses a decomposition method using a antler decomposition strain, and Patent Document 5 (method of preparing an antler extract) and Patent Document 6 ( A soft capsule containing deer antler extract and Patent Document 7 (a bone formation promoting substance extracted from deer antler and a method of extracting thereof) disclose an organic solvent extraction method, and Patent Document 8 (Manufacturing method of oxidized powder and health containing lyophilized lysis) Nutrition composition) and Patent Document 9 (Method for preparing concentrated antler extract and dry antler extract fine powder and health food using same) discloses rust blood powder method. The mechanical antler grinding method is disclosed in 10 (Method of manufacturing antler powder taken without decoction) and Patent Document 11 (determination of antler capsule), and Patent Document 12 (method of extracting an active ingredient from antler or rust) uses an electric method. Extraction methods are disclosed.
However, in the boiling water extraction method (heat treatment) of Patent Literature 1, the active ingredient of antler or green rust is destroyed by heat, and in the decomposition method using the antler decomposition strain of Patent Literature 4, it is easy to extract components of new metabolites, Due to the nature of the deer antler, the active ingredient is changed, the function may be lost, causing odor or difficult storage. In the organic solvent extraction method of Patent Documents 5, 6 and 7, it is difficult to extract other active ingredients because only the lipid component possessed by the antler is extracted, and the organic solvent is expensive.
In common, when using the above-described method, only a part of the active ingredient of antler or rust is extracted, and other active ingredients are lost. In addition, the development of new methods is urgently needed because of the time and cost required for extraction.
The heat treatment method (hot water extraction method) includes various heating steps and increases unnecessary costs, and since hydrolysis and decomposition methods using antler decomposition strains generate new metabolites, allergies and diabetes side effects may occur in some people. Heat-treated materials cause protein denaturation, which leads to loss of biological function and reduced digestibility and absorption. For example, collagen components present in bone tissue are denatured to gelatin when heat treated. Therefore, it is necessary to develop a technology for extracting in high yield without changing the active ingredient. In addition, special extraction processing methods are needed to improve digestibility and maintain the biological function of the active ingredient.
The present invention reduces the cost of production and maintain the functionality of the active ingredient by the ultra-high pressure and low temperature treatment without the use of heat and chemicals in order to extract the active ingredient of antler or rust shell It provides a way to completely extract the.
Therefore, liquid components, solid components, and lipid extracts are extracted from the soft internal tissues of antler and skin using ultra-high pressure, low temperature, enzymatic decomposition and processing methods. Carbohydrates, proteins and lipolytic enzymes are added at 30-40 ℃, the most active temperature, to extract solid solution and minerals. The extracted active ingredient is prepared in powder and liquid form and is used as a raw material for health foods, functional drinks, and functional cosmetics.
The inventors of the present invention use a method of adding an enzyme to extract solid solution and minerals by using low temperature to decompose solid tissues by using ultra-high pressure, which is a physical method, and to maintain the biological functionality of the active ingredient. As a result of intensive research to develop, when extracting the active ingredient of antler or green antler, not only the active ingredient of antler or antagonist can be purified, separated and extracted in high yield, but also as a raw material of health food, functional beverage and functional cosmetics. Can be used.

By improving the extraction efficiency of the production cost can be lowered, the active ingredient is extracted as a powder, liquid, lipid extract can be widely used in a variety of products, that is, health food, functional drinks, functional cosmetics.
Hereinafter, the present invention will be described in detail through specific examples.
The present invention is a method for extracting the active ingredient from the antler or green rust and using the extract ingredients as a raw material for health foods, functional drinks, functional cosmetics. Deer antler or antler uses antler and antler harvested from elk deer, red deer, chica deer and similar animals.
The health food, functional beverage, and functional cosmetics of the present invention are for improving human health and well-being, and the health food, functional beverage, and functional cosmetics contain a functional ingredient that is functional for disease prevention and health promotion.
In general, antler or rust shell consists of the outer skin layer and internal tissues.
Extracting the active ingredient, liquid component, solid component, and lipid extract of the antler or green rust of the present invention, and extracting the active ingredient of the skin layer, all start with fresh deer antlers, which are harvested directly from the farm Freeze at -20 ° C for long-term storage and transportation over long distances. The surface of this frozen deer antlers is first removed with 70% alcohol to remove any foreign or foreign matter adhering to the surface and washed with cold water.
The method of extracting the active ingredient from the antler or rust is cut 1-5 cm, which is a convenient length for the entire antler or rust, including the deer's horn skin tissue layer, hair, and internal tissue. Purified water (purified water) in a ratio of 50 wt% of the cut antler or rust was put in a closed plastic bag, and also amylase, protease, trypsin, lipase, papain. Proteolytic enzymes, including papain, pineapple, or kiwi juice, which are papain, pepsin, or natural enzymes containing natural enzymes, are added within the range of 0.001 to 1 wt.
Subsequently, the sealed plastic bag was immersed in a chamber of a high pressure machine filled with water, and the pressure was set at 500 atm to 2000 atm, the temperature was set at 4 to 50 ° C, and the ultrahigh pressure time was set at 2 to 24 hours. Pressurize for 2-30 minutes at 5000 atmospheres to inactivate the added enzymes and sterilize unwanted microorganisms, including bacteria, fungi, parasites and viruses.
The sterilized extract is ground in a blender (Waring) at high speed for about 5 minutes and then filtered to remove undesirable hairs contained in the ground extract. The sterilized and pulverized extract is lyophilized and then pulverized in a blender (trade name: Waring) to be powdered.
The method of separating and extracting the liquid and solid components from the antler or rust is carried out in the same process as the method of extracting the active ingredient from the antler or rust, and the sterilized and pulverized extract is put into a centrifuge and the centrifuge is operated at 2000 rpm. After 30 minutes of operation, the liquid component and the solid component are separated, and then the separated liquid component (supernatant) and the solid component (precipitate) are lyophilized, and then powdered with a blender (trade name: Waring). The liquid component is mainly composed of water-soluble protein, but also contains carbohydrate components, and the solid component is a mineral component.
Lipid extraction from the antler is performed by cutting 5-10 cm of the antler end or tip section (top section of the antler) to remove the skin layer, and then removing the inner layer of the antler from which the skin layer has been removed. After ultra high pressure treatment in the air, the microorganisms are sterilized, and then the sterilized extract is pulverized in a blender at a high speed for about 5 minutes to dissolve only the lipid component, and the lipid is separated by adding an organic solvent to the sterilized extract. Thereafter, the organic solvent is removed to extract the lipid extract and packaged in a sterile container.
Since rust is a hard horn, there is almost no lipid, and since the lipid exists only at the end of the antler, often called a bone bone, the present invention extracts a lipid extract from the antler.
The method of extracting the active ingredient from the skin layer of the antler is to cut 30-50 cm of the upper part of the antler or the upper part (upper section) and peel off the skin layer, and then after the skin layer of the antler is subjected to ultra high pressure in a high pressure machine. After sterilizing the unwanted microorganisms, the sterilized extract is pulverized in a blender for about 5 minutes at high speed to separate the liquid and solid components from the ground extract, and the separated liquid component is lyophilized and then powdered with a blender.
Since the rust shell is almost horned and the skin layer is also ossified, only the skin of the antler part can be used, so the active ingredient is extracted from the skin layer of the antler.
The method of extracting the active ingredient from the antler or the rust shell using the ultra-high pressure low temperature extraction method of the present invention as described above will be described in more detail with reference to the following Examples.
1]Example 1
Extraction of active ingredients from deer antler or rust
(1) Start with fresh deer horns. Deer horns are harvested directly from the farm and frozen and used at -20 ° C for long distance storage and transportation.
(2) The surface of frozen deer antlers is first removed from the outside or foreign matter on the surface with 70% alcohol, followed by rinsing with cold water.
(3) Cut the entire antler or green rust, including deer's horn skin tissue, hair, and internal tissue, into 1-5 cm, which is a convenient length.
(4) Purified water (purified water) in a ratio of 50 wt% of the cut antler or rust was put in a closed plastic bag, and also amylase, protease, trypsin, lipase. Proteolytic enzymes, including papain, pineapple, or kiwi juice, papain, pepsin, or natural enzymes containing natural enzymes, are added within the range of 0.001 to 1 wt.
(5) After the sealed plastic bag is immersed in the chamber of the high pressure filled with water, the pressure is set to 500 atm ~ 2000 atm, the temperature is set at 4 ~ 50 ℃, and the ultra high pressure is set at 2 ~ 24 hours. Pressurize for 2-30 minutes at atmospheric pressure to 5000 atmospheres to inactivate the added enzymes and sterilize unwanted microorganisms, including bacteria, fungi, parasites and viruses.
By using the sealed plastic bag as described above, air in the water is released due to the ultra-high pressure, thereby inhibiting the oxidative action generated during the hydrolysis due to the addition of the enzyme, thereby extracting the safe and unoxidized active ingredient.
(6) The sterilized extract is ground at high speed for about 5 minutes in a blender (trade name: Waring).
(7) Filter the ground extract to remove foreign matter such as hair.
(8) The sterilized and pulverized extract is lyophilized and then pulverized in a blender (trade name: Waring) to powder.
As described above, the active ingredient extracted and produced by the active ingredient extraction method from the fresh antler or rust is about 40% of the antler or rust used in the extraction method.
