Abstract
Description
도 2a는 SV40 프로모터 및 하이그로마이신 저항 마커를 포함하는 SEAP 유전자 발현 플라스미드 구성을 나타낸 것이다.
도 2b는 제조된 플라스미드가 HCT-8 세포에 도입되어 안정적으로 SEAP 를 발현하고 있는지를 RT-PCR 로 확인한 결과를 나타낸 것이다. 도 2c는 소포체 스트레스 유발 물질로 알려진 thapsigargin을 처리하여 SEAP 분비 발현 결과를 확인한 것이다.
도 3은 in vitro 에서 8-케토 트리코테센의 SEAP mRNA 및 효소적 활성에 미치는 영향을 나타낸 것이다. 1은 vehicle처리, 2는 8-케토 트리코테센 DON 처리, 3에는 15-아세틸 DON 처리, 4에는 3-아세틸 DON 처리, 5에는 NIV 처리를, 6에는 4-아세틸 NIV처리를 24시간동안 하였다. 그 중 A는 RNA를 RT-PCR 로 측정한 결과이고, B는 재조합 SEAP를 ID50 또는 2xID50의 양으로 8-케트 트리코테센을 처리하여 얻은 SEAP의 활성도 결과이다.
도 4는 8-케토 트리코테센 중 DON, 15-아세틸 DON, 3-아세틸 DON, NIV 및 4-아세틸 NIV를 각각 SEAP 발현 세포에 처리하여 SEAP의 활성을 나타낸 것이다. 상대적인 SEAP 활성은 전체 단백질에 대해서 배지 안에서의 SEAP 활성정도를 %로 계산하였으며, 모든 실험은 3번씩 반복하였다.
도 5는 SEAP 기반 DON 모니터링법에서 곡물 매트릭스 효과를 나타낸 것이다. DON 을 함유하는 옥수수 (A) 또는 벼 (B)와 SEAP 발현 세포주를 반응시켜, 실제 DON 양과 SEAP 기반 바이오 모니터링법에서 측정된 DON 양의 상관관계를 확인하였다.

(b) 상기 (a) 단계의 인간 상피 세포주에서 분비형 알칼리 포스파타아제 (SEAP)의 분비가 억제되는지 여부를 관찰하는 단계;
를 포함하는 8-케토 트리코테센의 검출 방법. - 제 10항에 있어서, (c) 상기 (b) 단계에서 SEAP의 분비가 억제되면 시료가 8-케토 트리코테센에 오염되었다고 판단하는 단계;가 더 포함되는 것을 특징으로 하는, 8-케토 트리코테센의 검출 방법.
- 제 10항에 있어서, 상기 8-케토 트리코테센은 데옥시니발레놀 (Deoxynivalenol, DON), 3-아세틸 DON, 15-아세틸 DON, 니발레놀 및 4-아세틸 니발레놀으로 이루어진 군에서 선택된 1종 이상인 것을 특징으로 하는, 8-케토 트리코테센의 검출 방법.
