Abstract
본 발명은 틸라피아 성장 호르몬에 비해 증가된 효과를 보이는 폴리펩티드에 관한 것이다. 이러한 폴리펩티드는 어류 및 갑각류 유생의 성장, 생존율 및 질을 자극할 수 있다. 또한, 기생충에 대한 저항을 증가시키고 수생 생물의 면역계에 관한 파라미터를 자극한다. 유전자를 코딩하는 폴리펩티드를 피치아 파스토리스 발현 벡터 내에 클로닝하였다. 이러한 벡터는 목표 단백질의 세포외 발현을 가능하게 한다. 목표 폴리펩티드를 함유하는 배양 상청액을 액침 배스 또는 먹이 첨가물로서 수생 생물에 투여하였다.

본 발명은 수생 생명공학 분야, 특히 틸라피아 성장 호르몬 보다 높은 활성을 갖는 폴리펩티드 및 액침 또는 사료 첨가물로서 처리 유생의 생존율 및 질을 향상기키기 위한 폴리펩티드의 용도에 관한 것이다.
수생 생물의 성장을 촉진하는 약제의 조사는 여러 조사 연구소에서 관심을 기울여 왔다. 성장 호르몬(Growth hormone; GH)는 이러한 조사의 주요 과제이다.
포유동물, 조류, 파충류, 양서류 및 어류에서의 GH의 존재는 보고되었다. 촉진된 성장, 생존율 증가 및 유생 질적 발달은 수산양식에서 주요 문제 중 하나이다.

미국 특허 제6,239,100호는 어류 유생의 성장을 촉진하고 사망률을 감속시키기 위해 어류 GH와 유사한 활성을 갖는 폴리펩티드의 사용에 대해 기재하고 있다.이 폴리펩티드에 대한 서열은 틸라피아 GH와 73%의 상동성을 가지며 N-말단의 51개 누클레오티드가 결여되어 있다. 본 발명의 폴리펩티드는 틸라피아 성장 호르몬으로부터 수득한 것이다. 폴리펩티드 SEQ ID NO. 7은 C-말단에 상응하는 46개의 아미노산의 단편이 결여되어 있다. 폴리펩티드 SEQ ID NO. 8은 C-말단에서 동일한 단편이 결여되어 있으며 N-말단에서 17개의 아미노산이 결여되어 있다.
GH와 유사한 활성을 갖는 미국 특허 제6,239,100호의 폴리펩티드와는 다르게, 본 발명의 폴리펩티드는 GH보다 높은 활성을 갖는다. 게다가, C-말단에서 단편이 결여된 폴리펩티드 SEQ ID NO. 7은 폴리펩티드 SEQ ID NO. 8에 비해 우수한 성장 촉진 활성을 갖는다.
본 발명의 구체화에서, 아미노산 서열 SEQ ID NO. 7 및 8에서 정의된 폴리펩티드의 어떤 부분을 포함하는 조성물은 어류 및 갑각류의 성장을 자극한다.
본 발명은 피치아 피스토리스 발현 벡터 pPS10으로의 상기 언급한 폴리펩티드의 코딩 서열의 클로닝을 포함한다.
주형으로서 pR17에 미리 클로닝한 tiGH의 cDNA를 사용하여 폴리머라제 사슬 반응에 의해 이러한 폴리펩티드에 대한 코딩 서열을 수득한다(Guillen I. - 폴리펩티드를 포함하는 재조합 파스토리스 MP36 균주의 배양 상청액
- 틸라피아 GH를 포함하는 재조합 파스토리스 MP36 균주의 배양 상청액
- 비-형질전환 파스토리스 MP36의 배양 상청액을 포함하는 음성 대조
- 비처리군.
이러한 결과는 폴리펩티드를 처리한 군은 tiGH, 음성 대조 및 비처리군에 비해 중량이 증가함을 보여준다.
면역계에 대한 효과를 분석하기 위해, 20마리 동물을 균질화하였다. 라이소좀 및 렉틴과 같은 다른 비-특이 체액성 인자의 존재 및 산화 스트레스의 하기 파라미터를 측정하였다.
- 감소된 글루타티온: GSH는 효소 글루타티온 페록시다제의 보조인자이다. 이는 산화 스트레스에 대한 방어 메카니즘 중 하나이다. 그럼에도 불구하고, 폴리펩티드 1 및 2로 처리한 군에서 증가하는 경향이 있다.

과산화 잠재성은 생물에서 산화 스트레스 지표이기 때문에, 비처리군에 비해 일부 산화 스트레스를 생성하는 폴리펩티드를 검정하는 것을 추론할 수 있다. 산화 스트레스는 중요한 조절 사건이다. 실험을 시작한 후 45일에, 지질 산화에 대한 효과를 갖는 산화 스트레스가 있을 때조차도, 비처리군엥 비해 처리군 사이의 카탈라제 수준이 상당히 감소하였다. 이러한 사건은 유리 라디칼 수준 및 산화 스트레스의 감소를 나타낸다.
지질은 에너지를 저장하는 다른 생분자에 비해 두드러진 비율 에너지/질량을 제공한다. GH가 지방생성을 저해하고 지방분해를 활성화하며 생성된 자방산이 이상적인 산화성 기질임이 증명되었다. 결과로서, 아미노산은 산화 과정으로부터 성장으로 이동한다. 본 출원인의 실험에서 발견된 지질 산화는 이러한 과정에 관련될 수 있다.
면역계에 관련된 다른 파라미터는 리소자임과 렉틴이다.
리소자임은 바이러스 및 박테리아 감염엥 대한 비-특이 면역 기능의 구성요소 중 하나이다. 어류에서의 리소자임 수준 및 GH 사이의 상관관계가 관찰되었다(Yada T., 등의 (2002) Immunomodulatory effects of prolactin and growth hormone in the tilapia, Oreochromis mossambicus. J Endocrinol . 173: 483-492).
렉틴은 병원체의 다양한 배열에 반응하며 생물에 고유의 면역을 제공한다(Alexander J.B. and Ingram G.A., (1992) Noncellular nonspecific defence mechanismof fish. Annu . Rev . Fish . Dis . 2: 249-279).
비처리군에 비해 폴리펩티드 1 및 2 처리군에서 두 파라미터의 통계적 유의차를 발견하였다.
이러한 폴리펩티드들의 효과는 또한 관상어 유생에서 시험되었다. 이들은 대조에 비해 폴리펩티드-처리군에서 성장 및 생존을 촉진시킬 수 있다. 또한 어류 색소침착을 향상시킨다. 이 파라미터는 관상어 상업화에 결정적이다. 이러한 폴리펩티드에 의해 생성된 효과는 처리를 종료한 후 15일에 보여진다.

상술된 폴리펩티드에 대한 코딩 서열을 pR17 벡터 내로 미리 클로닝된 주형으로서 틸라피아 cDNA를 사용하는 특이 올리고누클레오티드와 함께 PCR하여 수득하였다. 폴리펩티드 SEQ ID No. 7에 대한 코딩 서열을 증폭하기 위해, 서열 SEQ ID No. 1 및 SEQ ID No. 2에 상응하는 올리고누클레오티드를 사용하였다. 폴리펩티드 SEQ ID No. 8에 대한 코딩 서열을 증폭하기 위해, 서열 SEQ ID No. 3 및 SEQ ID No. 4에 상응하는 올리고누클레오티드를 사용하였다.
tiGH에 대한 코딩 서열을 서열 SEQ ID No. 5 및 SEQ ID No. 6에 상응하는 올 리고누클레오티드를 사용하여 벡터 pR17로부터 PCR하여 증폭시켰다. PCR 산물을 폴리누클레오티드 키나제 효소로 처리하여 유전자 말단을 인산화시키고 발현 벡터로의 클로닝을 촉진하였다. 피치아 파스토리스 발현 벡터 pPS10을 제한 엔도누클레아제 Nae I로 효소 절단시키고 알칼리성 포스파타제로 처리하여 말단을 탈인산화시켰다. 대상 폴리펩티드 및 tiGH에 대해 코딩하는 유전자를 T4 DNA 리가제를 사용하여 발현 벡터내로 결합시켰다(도 1).
형질전환 전에, 플라스미드를 효소 SphI로 선형화시켰다. 피치아 파스토리스 MP36 균주를 재조합 발현 벡터와 함께 전기천공법을 사용하여 형질전환시켰다. 이 균주는 형질전환 후 His+ 표현형을 취득하게 되는 영양요구성 돌연변이체이다.
도트 블롯(Dot Blot)에 의해 확인된, 형질전환체를 또한 재조합 플라스미드의 발현 카세트(cassette)에 대한 피치아 파스토리스의 유전자 AOX1의 치환에 의해 삽입이 이루어지는 것을 증명하기 위해 서던 블롯(Southern blot)으로 분석하였다. 이러한 삽입 이벤트는 MutS(메탄올 이용의 저수준) 및 His+ 표현형을 생성한다. 대상 폴리펩티드 및 틸라피아 성장 호르몬을 코딩하는 유전자는 메탄올에 의해 유도될 수 있는 AOX1 프로모터의 조절하에 있으며 이들은 시그널 펩티드를 갖는 다. 피치아 파스토리스는 낮은 수준으로 자신의 단백질을 분비하며 이의 배양 배지는 보충물로서 단백질을 필요로 하지 않는다. 그러므로, 분비된 이종의 단백질은 배지에서 총 단백질 중 높은 비율을 차지할 것이다(80% 이상)(Tschopp y col.; Bio/Technology 1987, 5: 1305-1308; Barr 등의; Pharm. Eng. 1992, 12: 48-51). 본 발명에서 설명된 재조합 단백질의 생성은 배지에 메탄올을 첨가한 5L의 생물반응기에서 이루어진다. 도 2에 나타난 바와 같이, 틸라피아 GH에 특이적인 모노클로날 항체를 사용한 웨스턴 블롯을 배양 상청액에서 단백질의 발현을 확인하기 위해 사용하였다.
실시예 2: 대상 폴리펩티드를 함유하는 피치아 파스토리스 배양 상청액으로 액침함에 의한 틸라피아에서의 성장 실험. 면역계에서의 효과.
실험은 일주일에 3회 90분간 액침하여 이루어졌다. 투여량은 물 ℓ당 0.1㎎의 목표 단백질이다. 실험군은 다음과 같다:
1. 폴리펩티드 1을 함유하는 피치아 파스토리스 배양 상청액으로 처리한 군(처리 1).
2. 폴리펩티드 2를 함유하는 피치아 파스토리스 배양 상청액으로 처리한 군(처리 2).
3. 틸리피아 GH를 함유하는 피치아 파스토리스 배양 상청액으로 처리한 군(처리 3).
4. 비-형질전환된 피치아 파스토리스 배양 상청액으로 처리한 음성 대조(음성 대조).
5. 비-처리군.
실험 시작 후 45일에, 폴리펩티드 SEQ ID No.7을 처리한 유생은 비처리군, 음성 대조 및 tiGH 처리군에 비해 각각 3.6, 2.1 및 1.6배 높은 체중 증가를 나타낸다. 반면, 폴리펩티드 SEQ ID No.8을 처리한 유생은 비처리군에 비해 3.2배, 음성 대조에 비해 1.9배 및 tiGH 처리군에 비해 1.4배 높은 체중 증가를 나타낸다(표 1, 표1.1, 도3).

이는 경제적으로 중요한 어류의 초기 발생 단계에서 성장 촉진이 배양 생산성을 증가시키기 위해 결정적인 생존율 증가를 이끌어내므로 매우 중요한 결과이다.
면역계에 대한 효과를 분석하기 위해, 군 당 20마리 동물을 실험 시작 후 21일 및 45일에 포획하였다. 유생을 균질화시키고 산화 스트레스의 다른 파라미터를 측정하였다: GSH, SOD, 총 기관 과산화물, 카탈라제, 과산화 잠재성 및 단백질 산화.
실험 시작 후 21일에 음성 대조에 비해 처리군간의 과산화 잠재성에서 차이가 발견되었다. 실험 시작 후 45일에 유생 추출물에서 리소자임 활성을 측정하였다. 시료(유생 균질액)의 리소자임 활성을 박테리아 마이크로콕커스 리소데이크티쿠스(Micrococcus lysodeikticus)를 용해시키기 위해 리소자임의 활성에 기초한 방법을 사용하여 측정하였다. 96-웰 마이크로플레이트에, 인산염 완충용액에 42배 계열희석한 시료 100μL를 마이크로콕커스 리소데이크티쿠스(시그마)의 3㎎/㎖ 현탁액 100μL와 혼합하였다. 마이크로플레이트를 22℃에서 항온하고 450nm에서 O.D.를 0, 15, 30 및 60분에 판독하였다. 양성 대조에 대해 유생 균질액을 닭의 달걀 화이트 리소자임(hen egg white lysozyme)(8㎍/㎖에서 시작하는 계열 희석)으로 대체하고, 음성 대조에 대해 완충용액을 유생 균질액으로 대체하였다. 리소자임 활성의 유닛은 0.001분-1의 O.D. 판독에서 감소를 유발하는 유생 균질액의 양으로서 규정하였다. 비처리군에 비해 폴리펩티드 SEQ ID No. 7 및 SEQ ID No. 8을 갖는 처리군간의 통계학적 유의차가 발견되었다(표 2).
유생 균질액에서 렉틴을 측정하기 위해 혈구응집 검정을 또한 실시하였다. 유생 추출물의 2배 계열 희석을 새로 제조한 2% 적혈구 현탁액(PBS 내 토끼)의 동일 부피를 첨가한 U자형 바닥 마이크로플레이트(96 웰, 그레이너, 아이그로론)에 pH 7.2인 PBS를 사용하여 실행하였다. 웰을 1시간 동안 실온에서 항온하고 역가를 시각적으로 판독하고 응집을 보이기 위해 마지막 웰에서의 희석을 동일하게 하였다(모든 웰 바닥에서의 세포의 고르게 분배된 층에 의해 명백해진 바와 같다). 시료의 혈구응집 활성을 시험하고 각 역가 수치를 수득하였다. 활성을 역가로서, 즉 완전한 응집을 나타내는 가장 높은 희석의 역수로서 표현하였다.
비처리군에 비해 폴리펩티드 SEQ ID No. 7 및 SEQ ID No.8을 갖는 처리군간에 통계학적 유의차를 관찰하였다(표 3).

실시예 3: 성장 자극 실험 및 대상 폴리펩티드를 함유하는 피치아 파스토리스 배양 상청액을 갖는 액침 배스에 의해 금붕어에서의 유생의 질 개선.
4개의 실험군을 계획하였다. 일주일에 3회 90분간 액침 배스에 의해 처리하였다. 투여량은 물 ℓ당 0.1㎎의 목표 단백질이다. 실험군은 다음과 같다:
1. 폴리펩티드 1을 함유하는 피치아 파스토리스 배양 상청액으로 처리한 군(처리 1).
2. 폴리펩티드 2를 함유하는 피치아 파스토리스 배양 상청액으로 처리한 군(처리 2).
3. 틸리피아 GH를 함유하는 피치아 파스토리스 배양 상청액으로 처리한 군(처리 3).
4. 비-형질전환된 피치아 파스토리스 배양 상청액으로 처리한 음성 대조(음성 대조).
실험 시작 후 45일에, 처리 1 및 2는 처리 3 및 4에 비해 상당한 무게 증가를 보여준다.
75일에, 폴리펩티드 SEQ ID No.7 및 SEQ ID No.8로 처리한 유생은 음성 대조 2배, tiGH 처리군에 비해 1.5배 높은 무게 증가를 나타낸다(표 4; 표 4.1; 도 4). 게다가, 처리 1, 2 및 3은 음성 대조에 비해 높은 생존율을 나타낸다(도 5).
관상어의 색은 상업화의 매우 중요한 파라미터이다. 이 실험에서, 처리 1 및 2에서의 유생이 음성 대조에 비해 이른 발달 단계에서 색을 나타냄을 발견하였다. 실험 시작 후 75일에 이들은 좀더 선명한 색을 갖는다(도 6).

실시예
4: 다른 투여량의 폴리펩티드 1을 갖는
액침
배스에 의한 잉어의 성장 실험.
잉어 유생의 성장에 대한 다른 투여량의 폴리펩티드 1의 효과를 실험하였다. 4개의 실험군을 다음과 같이 계획하였다:
1. 물 리터당 0.02mg의 폴리펩티드 투여량에서 폴리펩티드 SEQ ID 7을 포함하는 피치아 파스토리스 배양 상청액을 처리한 군(처리 1).
2. 물 리터당 0.1mg의 폴리펩티드 투여량에서 폴리펩티드 SEQ ID 7을 포함하는 피치아 파스토리스 배양 상청액을 처리한 군(처리 2).
3. 물 리터당 0.5mg의 폴리펩티드 투여량에서 폴리펩티드 SEQ ID 7을 포함하는 피치아 파스토리스 배양 상청액을 처리한 군(처리 3).
4. 비처리군.
실험 시작 후 30일에 물 리터당 0.5mg의 폴리펩티드 SEQ ID 7의 투여량을 처리한 유생은 비처리군에 비해 2.75배 많은 무게를 갖는다. 반면에 물 리터당 0.1mg의 폴리펩티드 SEQ ID 7의 투여량을 처리한 유생은 비처리군에 비해 1.8배 많은 무게를 갖는다. 비처리군 및 최소 투여량 사이에 통계학적 차이는 없었다(표 5; 표 5.1; 도 7).

3개의 실험군을 계획하였다. 일주일에 3회 1시간 30분 동안 액침 배스에 의해 처리하였다. 투여량은 물 리터당 0.1mg의 대상 단백질이다:
1. 폴리펩티드 SEQ ID 7을 포함하는 피치아 파스토리스 배양 상청액을 처리한 군(처리 1).
2. 폴리펩티드 SEQ ID 8을 포함하는 피치아 파스토리스 배양 상청액을 처리한 군(처리 2).
3. 비-형질전환된 피치아 파스토리스 배양 상청액을 처리한 음성 대조군(음성 대조).
실험 결과로서, 대상 폴리펩티드를 처리한 새우에서 유생의 질이 개선되었다. 폴리펩티드 SEQ ID No. 7 및 SEQ ID No.8을 처리한 유생은 음성 대조에 비해 2.3배 높은 체중 증가율을 나타내며 좀더 많은 분지 형성 및 부리 변이를 나타낸다. 상응하는 통계 테스트를 각 파라미터에 대해 실시하고 통계학적 유의차를 도든 경우에서 발견하였다.

72.99±3.25g의 초기 무게를 갖는 미숙한 틸라피아의 성장에 대한 두 투여량의 폴리펩티드 SEQ ID No. 7의 효과를 측정하였다. 시험된 투여량은 먹이 Kg 당 4.8mg의 폴리펩티드 SEQ ID No. 7 및 먹이 Kg 당 38.4mgmg의 폴리펩티드 SEQ ID No. 7이다. 경구 제형은 기초 사료 내에 대상 폴리펩티드를 함유하는 피치아 파스토리스 배양 상청액을 첨가하여 제조된다. 음성 대조는 비-형질전환된 피치아 파스토리스 배양 상청액이 풍부한 기초 사료를 갖는 틸라피아 먹이이다. 실험 기간 동안 동물들에게 6주간 체중의 5%에 상응하는 먹이를 하루에 2회 공급하였다.
먹이 Kg 당 4.8mg의 폴리펩티드 SEQ ID No.7의 투여량으로 사료에 포함되는 폴리펩티드 SEQ ID No. 7은 대조군에 비해 성장률을 22% 증가시키며 높은 통계학적 유의차를 갖는다(표 6; 표 6.1; 도 8).

오늘날에는, 수생 생물은 단백질의 중요한 원천이나, 이의 자연환경에서의 포획은 완전히 개발되었다. 이러한 이유에 있어서, 생산을 증가시키기 위해 이러한 수생종의 포획이 필수적이다. 대부분의 생물에ㅎ서 호르몬 투여, 유전자 변이 등에 의해 성장 조절을 가능하게 하는 광범위한 범위가 존재한다(Pullin y col.; Conference Proceeding 7, 432 p. 본원에 기재된 폴리펩티드는 종-특이적이지 않으므로 증가된 성장, 생존율 및 유생의 질과 같은 여러 기능에 대해 다른 수생 생물에 적용될 수 있으며 면역 반응에 관해 다른 파라미터를 증강시키는 장점을 갖는다. 게다가,성장 호르몬에 비해 상당히 높은 성장 효과를 갖는다. 다른 한편으로, 이러한 폴리펩티드들은 인간에서 생물학적 활성을 갖지 않아 인간이 소비하는 동물에 적용할 수 있어 불편함을 줄인다. 게다가, 관상어에서의 이러한 폴리펩티드의 사용은 성장 촉진의 결과로서 양식 시간을 감소시킬 뿐만 아니라 물고기의 색에 대한 효과도 가지므로 매우 효과적이다.

- 서열 SEQ ID No.7 및 SEQ ID No.8 중 하나에 포함되는 아미노산 서열인 폴리펩티드
- 제1항에 있어서, 상기 폴리펩티드는 피치아 파스토리스(Pichia pastoris)에서 수득된 것임을 특징으로 하는 폴리펩티드.
- 어류 및 갑각류의 성장을 촉진시키기 위한, 제1항의 폴리펩티드를 포함하는 조성물.
- 먹이 Kg 당 4.8mg의 폴리펩티드의 효과적인 투여량으로 어류 및 갑각류의 성장을 촉진하고 질병을 저해하기 위한 먹이 제형에서의 제1항의 폴리펩티드를 함유한 피치아 파스토리스 배양 상청액을 포함한 조성물.
- 물 리터당 0.05 및 0.5mg 사이의 폴리펩티드의 농도로 어류 및 갑각류의 성장, 생존율 및 유생의 질을 촉진시키기 위한 제1항의 폴리펩티드를 함유한 액침 처리용 피치아 파스토리스 배양 상청액을 포함한 조성물
