Abstract
본 발명은 왕겨초액을 유효성분으로 함유하는 염증성 질환의 예방 또는 치료용 약학 조성물에 관한 것이다. 본 발명에 따른 왕겨초액은 비만세포주 및 호염구 세포주인 RBL-2H3 세포에서 β-헥소스아미니다제의 분비를 현저히 억제하고, 대식세포주인 RAW 264.7 세포에서 산화질소(NO)의 생성과 ROS의 생성을 현저히 억제하여 대식세포의 활성화를 강하게 억제하며, TPA로 유도된 마우스의 귀 부종을 억제시키고, 염증 관련 유전자인 iNOS, 5-LOX, COX-2를 mRNA 수준 및 단백질 수준에서 모두 억제시키고, 부종이 일어난 마우스의 귀 조직 내에서 염증 관련 사이토카인 (TNF-α, IL-1β, IL-6)과 에이코사이드(PGE2, LTB4)의 양을 현저히 감소시키고, 마우스 귀 부종 모델의 귀 조직 내에서 비만세포의 활성화 마커인 트립타제를 발현시키지 않아 비만세포 활성화를 억제하여 염증 억제 효과가 우수하므로, 염증성 질환의 예방 또는 치료에 유용하게 사용될 수 있다.
Description
본 발명은 왕겨초액을 유효성분으로 함유하는 염증성 질환의 예방 또는 치료용 약학 조성물에 관한 것이다.
염증성 질환은 세균의 침입에 의하여 형성되는 농양의 병리적 상태를 의미한다. 염증은 병원체, 이물질 및 조직 손상에 대한 중요한 생체의 방어 반응으로, 열, 쇠약감, 식욕감퇴 및 오한 등의 전신적 증상, 또는 발적, 부종, 동통 및 기능장애 등의 국소적 증상을 동반한다.
염증은 지속 기간에 따라 급성 염증, 아급성 염증 및 만성 염증으로 분류된다. 급성 염증은 혈관을 기준으로 반응이 일어나며, 호중구(neutrophil)가 주로 관여한다. 특히, 화농성 염증의 경우에는 호중구의 증가가 현저히 나타난다. 만성 염증은 수주 또는 수개월 동안 지속되는 염증으로서, 조직의 손상 및 치유가 동시에 일어나며, 지연된 과민감성 반응을 일으키는 계속적인 감염(예를 들어, 결핵, 매독, 진균 감염), 지속적인 내독소(예를 들어, 증가된 혈장 지질) 또는 외독소(예를 들어, 실리카, 석면, 타르, 수술 봉합사)에 대한 노출, 또는 자가 조직에 대한 자가면역 반응(예를 들어, 류마티스성 관절염, 전신성 홍반성 낭창, 다발성 경화증, 건선)의 결과로서, 시간이 경과됨에 따라 진행되는 잠행성 과정을 나타낸다. 아급성 염증은 급성과 만성의 중간 단계의 염증을 말한다.
일반적으로, 염증성 질환은 염증성 사이토카인의 불균형과 효과세포 (effector cell)의 상호작용에 의해 야기되며, 주요 염증성 질환으로는 감염성 비염, 알레르기성 비염, 만성 비염, 급성 부비동염 및 만성 부비동염 등과 같은 비염 및 부비동염; 급성화농성 중이염 및 만성화농성 중이염 등과 같은 중이염; 세균성 폐렴, 기관지 폐렴, 대엽성 폐렴, 레지오렐라 폐렴 및 바이러스성 폐렴 등과 같은 폐렴; 급성 또는 만성 위염; 감염성 소장결장염, 크론씨 병(Crohn's disease), 특발성 궤양성 대장염 및 위막성 대장염 등과 같은 장염; 및 화농성 관절염, 결핵성 관절염, 퇴행성 관절염 및 류마티스 관절염 등과 같은 관절염 등이 있다. 그 외에 초기에 극심한 전신적 염증반응이 동반되는 패혈증(septicemia)이 있는데, 패혈증은 숙주가 그람-음성균의 내독소인 LPS(lipopolysaccharide) 등에 대해 과도하게 반응함으로써 발생한다.
염증의 발생에는 다양한 생화학적 물질 및 현상이 관여하고 있으며, 특히 산화질소 합성효소(nitric oxide synthase; NOS) 및 시클로옥시게나제 (cyclooxygenase; COX)는 염증반응을 매개하는데 있어 중요한 역할을 하는 것으로 알려져 있다.
상기 NOS는 L-아르기닌으로부터 NO(nitric oxide)를 생성시키는 효소로, 뇌 에 존재하는 bNOS(brain NOS), 신경계에 존재하는 nNOS(neuronal NOS) 및 혈관계에 존재하는 eNOS(endothelial NOS)는 체내에서 항상 일정수준으로 발현되고 있으며, 이들에 의해 소량 생성되는 NO는 신경전달이나 혈관확장을 유도하는 등 정상적인 신체의 항상성 유지에 중요한 역할을 한다. 하지만, iNOS(induced NOS)는 각종 사이토카인이나 외부 자극 물질에 의해 유도되어 과량의 NO를 급격히 발생시켜 세포독성이나 각종 염증반응을 일으키는 것으로 알려져 있다. 특히, 만성 염증은 iNOS 활성의 증가와 밀접한 관련이 있다고 알려져 있다. 일반적으로 NO는 대기 상태에서 가스의 형태로 존재하며, 자유 라디칼 구조이기 때문에 매우 유독할 뿐만 아니라 반감기가 6~10초로 매우 불안정하여 안정화된 최종 산물인 NO3(nitrate)와 NO2 (nitrite)로 신속히 변환되는 것으로 알려져 있다.
또한, 상기 COX는 아라키돈산으로부터 프로스타글란딘류를 합성하는데 관여하는 효소로, COX-1 및 COX-2의 2 종류가 있다. COX-1은 세포 내에 항상 존재하여 세포보호작용에 필요한 프로스타글란딘을 합성하는 작용을 나타내는 것에 반하여, COX-2는 염증 반응시 세포 내에서 급격히 증가되어 염증반응을 일으키는데 있어서 중요한 역할을 수행하는 것으로 알려져 있다.
따라서, iNOS 또는 COX-2의 발현을 억제하면 염증성 질환을 치료할 수 있을 것이며, iNOS 및 COX-2를 동시에 억제할 수 있는 물질은 바람직한 염증성 질환 치료제로 사용될 수 있을 것으로 생각된다.
현재까지 NOS 저해제로는 NG-메틸-L-아르기닌(NG-methyl-L-arginine), NG-니 트로-L-아르기닌(NG-nitro-L-arginine), NG-니트로-L-아르기닌 메틸 에스터(NG-nitro-L-arginine methyl ester), 아미노구아니딘(aminoguanidine), L-티오시트룰린(L-thiocitrulline) 및 S-메틸-L-티오시트룰린(S-methyl-L-thiocitrulline) 등이 알려져 있으나, 이들 대부분은 NOS의 기질인 L-아르기닌과 경쟁적으로 작용하여 NOS를 저해하는 것으로, iNOS에 선택적이지 못한 단점이 있다. 또한, 최근에 합성방법으로 제조된 COX-2에 대한 저해제로는 NS-398, CGP-29238, SC-58125, L-745337 및 메록시캄 등이 보고되고 있으나 아직 널리 이용되지는 않고 있으며, 그들의 부작용에 대해서도 확실히 검증되지 않은 상태이다.
한편, 왕겨초액은 왕겨를 태우는 과정에서 발생한 연기를 증류 및 냉각하여 얻은 액체를 6개월 이상 숙성 및 정치하여 얻은 적갈색의 투명한 액체를 일컫는다. 왕겨초액은 초산을 주성분으로 하는 pH 3 전?후의 산성 액체로서, 200여 종의 각종 유기산으로 혼합되어 있다. 왕겨초액은 10~20%의 유기 화합물과 80~90%의 수분을 포함하고 있으며, 유기 화합물 중에는 200여 종류의 화합물을 함유하고 있다. 왕겨초액은 과일과 채소의 당도를 높이고, 식물의 생장 촉진, 해충 방제, 강한 살균력, 토양 산성화 방지 등에 탁월한 효과가 있다고 알려져 있다. 따라서, 왕겨초액은 토양 살균, 축산 분뇨화, 탈취, 작물의 해충 기피, 퇴비 발효 촉진, 식물 생장 및 뿌리생육 촉진 효과 등 환경 정화 분야에 활용되고 있으며, 일부 식품첨가제로도 시판되고 있다. 최근 들어서는 친환경 유기농법에 이용되고 있으며, 생분해성이 제고되는 팽연화 및 미세 분쇄기술이 개발되어 활용도가 높아지고 있다. 또한, 왕겨초액의 항산화력, 항균력, 항당뇨성 등을 이용한 가공제품 또는 식용유지의 저 장성을 향상시킬 수 있는 보존제로의 가능성도 연구되고 있다.
상기한 바와 같이, 왕겨초액을 이용한 용도가 다양하게 개발되고 있지만, 왕겨초액을 염증 질환 치료에 이용한 연구는 거의 전무한 상태이다.
따라서, 왕겨초액을 이용하여 염증성 질환의 예방 또는 치료에 효과적인 새로운 의약품 개발에 대한 연구의 필요성이 요구되고 있다.
본 발명자들은 왕겨초액을 이용하여 염증성 질환의 예방 또는 치료에 효과적인 새로운 의약품에 대해 연구하던 중, 왕겨초액이 β-헥소스아미니다제의 분비를 현저히 억제하고, 산화질소(NO)의 생성과 ROS의 생성을 현저히 억제하며, TPA로 유도된 마우스의 귀 부종을 억제시키고, 염증 관련 유전자인 iNOS, 5-LOX, COX-2를 mRNA 수준 및 단백질 수준에서 모두 억제시키고, 부종이 일어난 마우스의 귀 조직 내에서 염증 관련 사이토카인(TNF-α, IL-1β, IL-6)과 에이코사이드(PGE2, LTB4)의 양을 현저히 감소시키고, 마우스 귀 부종 모델의 귀 조직 내에서 비만세포 활성화를 억제하여 염증 억제 효과가 우수함을 확인하고, 본 발명을 완성하였다.
본 발명은 왕겨초액을 유효성분으로 함유하는 염증성 질환의 예방 또는 치료용 약학 조성물을 제공하고자 한다.
본 발명은 왕겨초액을 유효성분으로 함유하는 염증성 질환의 예방 또는 치료용 약학 조성물을 제공한다.
이하, 본 발명에 대해 상세히 설명한다.
본 발명에 따른 왕겨초액은 비만세포주 및 호염구 세포주인 RBL-2H3 세포에서 β-헥소스아미니다제의 분비를 현저히 억제하고, 대식세포주인 RAW 264.7 세포 에서 산화질소(NO)의 생성과 ROS의 생성을 현저히 억제하여 대식세포의 활성화를 강하게 억제한다. 또한, 왕겨초액은 TPA로 유도된 마우스의 귀 부종을 억제시키고, 염증 관련 유전자인 iNOS, 5-LOX, COX-2를 mRNA 수준 및 단백질 수준에서 모두 억제시키고, 부종이 일어난 마우스의 귀 조직 내에서 염증 관련 사이토카인(TNF-α, IL-1β, IL-6)과 에이코사이드(PGE2, LTB4)의 양을 현저히 감소시킨다. 또한, 왕겨초액은 마우스 귀 부종 모델의 귀 조직 내에서 비만세포의 활성화 마커인 트립타제를 발현시키지 않아 비만세포 활성화를 억제하여 염증 억제 효과가 우수하다.
따라서, 본 발명의 왕겨초액을 유효성분으로 함유하는 조성물은 염증성 질환의 예방 또는 치료에 유용하게 사용될 수 있다.
상기 염증성 질환으로는 감염성 비염, 알레르기성 비염, 만성 비염, 급성 부비동염 및 만성 부비동염 등과 같은 비염 및 부비동염; 급성화농성 중이염 및 만성화농성 중이염 등과 같은 중이염; 세균성 폐렴, 기관지 폐렴, 대엽성 폐렴, 레지오렐라 폐렴 및 바이러스성 폐렴 등과 같은 폐렴; 급성 또는 만성 위염; 감염성 소장결장염, 크론씨 병(Crohn's disease), 특발성 궤양성 대장염 및 위막성 대장염 등과 같은 장염; 화농성 관절염, 결핵성 관절염, 퇴행성 관절염 및 류마티스 관절염 등과 같은 관절염; 및 패혈증 등을 포함하나, 이에 한정되지 않는다.
본 발명의 조성물은 왕겨초액과 함께 염증성 질환의 예방 또는 치료 효과를 갖는 공지의 유효성분을 1종 이상 함유할 수 있다.
본 발명의 조성물은, 투여를 위해서 상기 기재한 유효성분 이외에 추가로 약 학적으로 허용가능한 담체를 1종 이상 포함하여 제조할 수 있다. 약학적으로 허용 가능한 담체는 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로오스 용액, 말토덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 이상을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다. 또한 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립 또는 정제로 제제화할 수 있다. 더 나아가 당분야의 적정한 방법으로 또는 Remington's Pharmaceutical Science(최근판), Mack Publishing Company, Easton PA에 개시되어 있는 방법을 이용하여 각 질환에 따라 또는 성분에 따라 바람직하게 제제화할 수 있다.
본 발명의 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구 투여(예를 들어, 정맥 내, 피하, 복강 내 또는 국소에 적용)할 수 있으며, 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설율 및 질환의 중증도 등에 따라 그 범위가 다양하다. 상기 왕겨초액은 희석하여 사용하는 것이 바람직하며, 1%로 희석한 왕겨초액의 일일 투여량은 약 500㎕~1㎖/㎏, 바람직하게는 약 400~800㎕/㎏이며, 하루 일회 내지 수회에 나누어 투여하는 것이 더욱 바람직하다.
본 발명의 조성물은 염증성 질환의 예방 또는 치료를 위하여 단독으로, 또는 수술, 호르몬 치료, 약물 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
실시예 1
: DPPH 라디칼에 대한 왕겨초액의 전자공여능 측정
화학적 라디칼인 DPPH(1,1-diphenyl-2-picrylhydrazyl) 라디칼에 대한 왕겨초액의 전자공여능을 측정하기 위하여, 하기와 같은 실험을 수행하였다.
실험에 사용한 왕겨초액은 (주)대원 GSI로부터 구입하여 사용하였다. 에탄올 1㎖, 100mM 아세트산나트륨 완충용액(pH 5.5) 990㎕, 농도별(0.5%, 1%) 왕겨초액을 각각 분주한 시험관에 에탄올에 용해시킨 0.5mM DPPH 용액 0.5㎖를 첨가한 후, 실온에서 30분 동안 반응시켰다. 그 다음 DPPH 라디칼의 잔존 수준을 UV/VIS 분광광도계(Beckman DU-65, USA)를 이용하여 517㎚에서 흡광도를 측정하였다. 전자공여능은 하기 수학식 1로 계산하였다. 양성대조군으로는 합성 항산화제인 BHT(butylated hydroxytoluene)을 사용하였다.
결과는 표 1에 나타내었다.

표 1에 나타난 바와 같이, 본 발명의 왕겨초액의 전자공여능은 양성대조군인 BHT와 비슷한 수준의 전자공여능을 나타내었다.
실시예 2
: 왕겨초액이 세포 생존율에 미치는 영향
왕겨초액이 세포 생존율에 미치는 영향을 확인하기 위하여, 미토콘드리아 탈수소효소 활성(mitochondrial dehydrogenase activity)의 지표를 나타내는 MTT colorimetric reduction assay를 수행하였다.
실험에 사용한 백서 유래의 비만세포주 및 호염구 세포주인 RBL-2H3 세포는 Health Source Research Bank(Osaka, Japan)로부터 구입하였으며, 마우스 유래 대식세포주인 RAW 264.7 세포는 ATCC(Manassas, VA, USA)로부터 구입하였다.
RBL-2H3 세포와 RAW 264.7 세포는 각각 10% FBS, 100 units/㎖의 페니실린, 스트렙토마이신이 포함된 DMEM 배지와 37℃, 5% CO2를 포함하는 포화습도 공기 조건에서 배양하였다. RBL-2H3 세포와 RAW 264.7 세포의 세포수를 각각 1×106 cells/㎖ 및 5×105 cells/㎖로 조절한 후, 96 웰 플레이트에 200㎕씩 분주하고 37℃, 5% CO2 배양기에서 24시간 동안 배양하였다. 그 다음 왕겨초액 시료를 농도별(0.1%, 0.5%, 1%)로 각각 처리하였다. 처리 후 48시간 동안 배양한 다음 배양액을 제거하고 0.5㎎/㎖의 MTT를 포함한 배지를 200㎕씩 처리하였다. 다시 3시간 동안 배양한 후 상징액을 제거하고 200㎕의 DMSO를 처리하여 세포 내의 포마잔 결정을 용출시켰다. ELISA reader(Model-550, Bio-Rad, USA)를 사용하여 570㎚ 및 655㎚에서 흡광도를 측정하였다. 측정 결과는 세포독성의 지표로 사용하였다.

표 2에 나타난 바와 같이, 지시세포로 사용한 RBL-2H3 세포와 RAW 264.7 세포에 왕겨초액 원액을 처리한 경우 대식세포주인 RAW 264.7 세포에서 세포독성이 나타남을 확인하였다. 상기와 같이, 강한 세포독성으로 인하여 세포 수준에서 실험이 불가능하여, NaOH를 사용하여 강산성인 왕겨초액을 pH 5 수준으로 중화시킨 다음 RBL-2H3 세포와 RAW 264.7 세포에 처리한 결과 대부분의 세포독성이 사람짐을 확인하였다. 따라서, 이후의 실험에서는 pH 5 수준의 왕겨초액을 사용하였다.
실시예 3
: 왕겨초액의 β-헥소스아미니다제(β-hexosaminidase) 분비 억제 활성 측정
왕겨초액이 비만세포주 및 호염구 세포주가 분비하는 염증매개 물질인 β-헥소스아미니다제의 분비를 억제하는지 확인하기 위하여, 백서 유래의 비만세포주 및 호염구 세포주인 RBL-2H3 세포를 지시세포로 하여 하기와 같은 실험을 수행하였다.
RBL-2H3 세포를 10% FBS, 100 units/㎖의 페니실린, 스트렙토마이신이 포함된 DMEM 배지와 37℃, 5% CO2를 포함하는 포화습도 공기 조건에서 배양하였다. 세포수를 1×106 cells/㎖로 조절한 후, 96 웰 플레이트에 200㎕씩 분주하고 37℃, 5% CO2 배양기에서 24시간 동안 배양하였다. 배양 후 배지를 제거하고 티로이드 완충용액(tyroid buffer) (137mM NaCl, 2.7mM KCl, 1.8mM CaCl2, 1.1mM MgCl2, 11.9mM NaHCO3, 0.4mM NaH2PO4, 5.6mM glucose, pH 7.2)으로 3회 세척한 후, 왕겨초액 시료를 농도별(0.1%, 0.5%, 1%)로 티로이드 완충용액에 첨가하여 세포에 처리하였다. 37℃ 배양기에서 15분 동안 처리한 후 시료를 제거하고 티로이드 완충용액으로 3회 세척하였다. 여기에 칼슘 이오노포(calcium ionophore)인 A23187을 최종농도 10μM이 되도록 티로이드 완충용액에 희석하여 처리한 후 37℃ 배양기에서 다시 20분 동안 배양하였다. 배양 후 상징액 50㎕를 취하고, 여기에 동량의 1mM p-니트로페닐-β-아세틸-글루코사미드를 넣은 다음 37℃ 진탕배양기에서 100rpm으로 1시간 동안 반응시켰다. 반응 후, 200㎕의 중탄산나트륨(sodium bicarbonate, pH 10.2)을 첨가하여 발색시킨 후 ELISA reader를 이용하여 405㎚에서 흡광도를 측정하였다. β-헥소스아미니다제 분비지표(release index)는 하기 수학식 2로 계산하였다.
결과는 표 3에 나타내었다.

실시예 4
: 왕겨초액의 산화질소(nitric oxide) 생성 억제 활성 측정
왕겨초액이 대식세포 활성화의 주요 지표 중 하나인 산화질소(NO)의 생성을 억제하는지 확인하기 위하여, 마우스 유래 대식세포주인 RAW 264.7 세포를 지시세포로 하여 무라카미(Murakami) 등의 방법에 따라 측정하였다.
구체적으로는, RAW 264.7 세포를 10% FBS, 100 units/㎖의 페니실린, 스트렙토마이신이 포함된 DMEM 배지와 37℃, 5% CO2를 포함하는 포화습도 공기 조건에서 배양하였다. 세포수를 5×105 cells/㎖로 조절한 후, 96 웰 플레이트에 200㎕씩 분주하고 37℃, 5% CO2 배양기에서 24시간 동안 배양하였다. 배양 후 배지를 제거하고 새로운 DMEM 배지를 이용하여 농도별(0.1%, 0.5%, 1%)의 왕겨초액과 100ng/㎖의 LPS(lipopolysaccharide)를 함께 처리한 후 37℃, 5% CO2 배양기에서 48시간 동안 배양하였다. 배양 후 상징액 100㎕를 회수한 후, 여기에 동량의 그리스 용액 (griess solution)을 첨가하여 상온에서 30분 동안 방치하였다. 그 다음 상징액의 발색도를 ELISA reader를 이용하여 570㎚ 및 655㎚에서 흡광도를 측정하였다.
결과는 표 4에 나타내었다.

표 4에 나타난 바와 같이, 왕겨초액은 대식세포주인 RAW 264.7 세포에서 산화질소(NO)의 생성을 현저히 억제하였다. 따라서, 왕겨초액은 대식세포의 활성화를 강하게 억제함을 알 수 있다.
실시예 5
: 왕겨초액에 의한 ROS(reactive oxygen species) 생성 억제 활성 측정
왕겨초액이 대식세포 활성화의 주요 지표 중 하나인 ROS의 생성을 억제하는지 확인하기 위하여, 하기와 같은 실험을 수행하였다.
마우스 유래 대식세포주인 RAW 264.7 세포를 96 웰 플레이트에 1×105 cells/well로 접종한 후, 24시간 동안 배양하였다. 배양 후, DCFH-DA(2',7'-dichlorofluorescein diacetate)를 20μM이 되도록 HBSS(Hank's balanced salt solution)에 희석하여 암실 조건에서 20분 동안 처리하여 세포를 표지하였다. 표지된 세포에 0.5% 왕겨초액과 LPS를 함께 처리하여 1시간 동안 배양한 다음 여기파장 485㎚, 방출파장 528㎚에서 ROS의 방출양을 측정하였다.
결과는 도 1에 나타내었다.
도 1에 나타난 바와 같이, 왕겨초액은 대식세포주인 RAW 264.7 세포에서 ROS의 생성을 현저히 억제하여 ROS가 거의 발생되지 않음을 확인하였다.
실시예 6
: 왕겨초액의 염증반응(부종) 억제 효과
왕겨초액의 염증반응(부종) 억제 효과를 확인하기 위하여, 하기와 같은 실험을 수행하였다. 실험에 사용한 마우스는 6주령의 암컷 CD-1 마우스를 샘타코(Osan, Kyunggido, Korea)에서 구입하였으며, 실험에 사용하기 전까지 실내온도를 22±2℃로 유지하면서 충분한 물과 사료를 공급하여 사육하였다.
6주령의 암컷 CD-1 마우스의 한쪽 귀에 1% 왕겨초액 20㎕를 2시간 동안 전처리하였다. 그 다음 PKC(protein kinase C)의 활성제인 TPA(12-O-tetradecanoyl phorbol 13-acetate) 20㎕를 처리하여 염증반응(부종)을 유도하였다. 24시간 후, 염증반응(부종)이 유도된 마우스의 귀 두께를 측정하였다.
측정 결과는 도 2에 나타내었다.
도 2에 나타난 바와 같이, TPA로 유도된 마우스의 귀 부종에 왕겨초액을 처리한 경우 부종이 억제됨을 확인하였다.
실시예 7
: 염증 관련 유전자의 발현량 측정
왕겨초액에 의한 염증 관련 유전자(iNOS, 5-LOX, COX-2)의 발현 정도를 확인하기 위하여, 하기와 같은 실험을 수행하였다.
1. RT-PCR
TPA를 이용하여 염증반응(부종)을 유도한 마우스를 희생한 후, 부종이 발생한 귀를 적출하고 액체질소를 이용하여 분쇄하였다. 그 다음 산 페놀 - 구아니딘 티오시아네이트 - 클로로포름 추출 방법으로 세포 내 총 RNA를 추출하였다. 추출 후 1㎍의 RNA를 주형으로 AMV 역전사효소와 올리고(dT)를 사용하여 cDNA를 합성하였다. 그 다음 thermocycler(PTC-200, MJ-research, USA)를 이용하여 PCR을 수행하였다. 증폭된 DNA는 1% 아가로오스 겔에서 전기영동하였다.
결과는 도 3에 나타내었다.
도 3에 나타난 바와 같이, 왕겨초액은 염증 관련 유전자인 iNOS, 5-LOX, COX-2을 mRNA 수준에서 모두 억제하였다.
2. 웨스턴 블롯
TPA를 이용하여 염증반응(부종)을 유도한 마우스를 희생한 후, 부종이 발생한 귀를 적출하고 액체질소를 이용하여 분쇄하였다. 그 다음 1㎖의 RIPA 완충용액 (50mM Tris-Cl, 150mM NaCl, 1% NP-40, 0.5% sodium deoxycholate, 0.1% SDS, 1mM EDTA, pH 7.4)을 이용하여 세포를 용해시켰다. 분리된 단백질은 브래드포드 방법 (bradford method)을 이용하여 정량하였고, 30~50㎍의 단백질을 이용하여 10% 폴리아크릴아미드 겔에서 SDS-PAGE를 수행하였다. 전기영동 후, 니트로셀룰로오스 막으로 옮기고, 5% 탈지우유와 0.1% 트윈-20이 포함된 PBS를 사용하여 상온에서 1시간동안 블록킹(blocking)하였다. 블록킹 후 각각의 항체를 5% 탈지우유, 0.1% 트윈-20이 포함된 PBS에 희석하여 처리하고 상온에서 3시간 동안 반응시켰다. 반응 후 PBS-T를 이용하여 막을 5분씩 3회 세척한 후, HRP(horseradish peroxidase)가 결합된 2차 항체를 넣고 상온에서 1시간 동안 반응시켰다. 반응이 끝난 후 PBS-T로 5분씩 3회 세척하였고, 웨스턴 블롯 루미놀 시약(western blotting luminol reagent)을 넣고 1분 동안 반응시킨 후, Las-1000 luminescent image analyzer(Fuji Photo Film, Tokyo, Japan)를 이용하여 단백질 밴드를 확인하였다.
결과는 도 4에 나타내었다.
도 4에 나타난 바와 같이, 왕겨초액은 염증 관련 유전자인 iNOS, 5-LOX, COX-2을 단백질 수준에서 모두 억제하였다.
실시예 8
: 왕겨초액이 염증 관련 사이토카인의 생성에 미치는 영향
왕겨초액이 염증 관련 사이토카인(TNF-α, IL-1β, IL-6)의 생성에 미치는 영향을 확인하기 위하여, 부종이 일어난 마우스의 귀 조직 내의 염증 관련 사이토카인(TNF-α, IL-1β, IL-6)의 양을 Biosource사의 사이토카인 ELISA 키트를 이용하여 제조사의 지시에 따라 측정하였다. 구체적으로는, 추출한 단백질 50㎕를 항체가 코팅된 96 웰 플레이트에 넣고 실온에서 1시간 동안 반응시킨 후 세척완충용액으로 3회 세척하였다. 여기에, 다시 항체를 넣고 2시간 동안 반응시켜 결합시킨 후 세척하고, 비오틴(biotin)이 결합된 2차 항체를 넣어 반응시켰다. 다시 아비딘 (avidin)이 결합된 HRP를 넣어 반응시킨 후 기질을 첨가하여 30분 더 반응시켰다. 반응이 끝난 후 ELISA reader를 이용하여 420㎚에서 흡광도를 측정하여 각 사이토카인의 양을 계산하였다.
결과는 표 5에 나타내었다.

표 5에 나타난 바와 같이, 왕겨초액은 부종이 일어난 마우스의 귀 조직 내에서 염증 관련 사이토카인(TNF-α, IL-1β, IL-6)의 양을 현저히 감소시켰다.
실시예 9
: 왕겨초액이 에이코사이드의 생성에 미치는 영향
왕겨초액이 에이코사이드의 생성에 미치는 영향을 확인하기 위하여, 부종이 일어난 마우스의 귀 조직 내의 COX-2의 대사산물인 PGE2(prostaglandin E2)와 5-LOX의 대사산물인 LTB4(leukotriene B4)의 양을 Biosource사의 ELISA 키트를 이용하여 제조사의 지시에 따라 측정하였다.
측정 결과는 표 6에 나타내었다.

표 6에 나타난 바와 같이, 왕겨초액은 부종이 일어난 마우스의 귀 조직 내에서 에이코사이드(PGE2, LTB4)의 양을 현저히 감소시켰다.
실시예 10
: 왕겨초액이 마우스 귀 조직 내 비만세포 활성화에 미치는 영향
왕겨초액이 마우스 귀 조직 내 비만세포 활성화에 미치는 영향을 확인하기 위하여, 비만세포 활성화 마커인 트립타제의 발현을 면역조직화학적검사 (immunohistochemistry)를 이용하여 확인하였다.
구체적으로는, 마우스의 적출한 귀를 4% 파라포름알데히드에 넣고 4℃에서 12시간 동안 처리하여 고정하였다. 고정된 조직을 조직처리기(Leica, Germany)와 파라핀 포매(Leica, Germany)를 이용하여 파라핀 블록을 제작하였다. 파라핀 블록을 초박편절편기(microtome) (Leica, Germany)를 이용하여 4㎛의 두께로 자른 후 코팅된 슬라이드 글라스(Fisher scientific, USA)에 부착시켰다. 자일렌을 이용하여 파라핀을 제거하였고, 수분을 충분히 공급한 후, 각각의 일차 항체를 넣고 37℃에서 1시간 동안 반응시켰다. 여기에 HRP가 결합된 이차 항체를 넣고 37℃에서 1시간 동안 반응시킨 후, DAB(diaminobenzidine)를 처리하여 발색시키고 광학현미경으로 관찰하였다.
결과는 도 5에 나타내었다.
도 5에 나타난 바와 같이, TPA만 처리한 마우스에서는 트립타제가 발현되어 염색된데 반하여, 왕겨초액을 처리한 마우스에서는 트립타제가 발현되지 않음을 확인하였다. 따라서, 왕겨초액은 마우스 귀 부종 모델의 귀 조직 내 비만세포 활성화를 억제하여 염증 억제 효과가 우수함을 알 수 있다.
하기에 본 발명의 조성물을 위한 제제예를 예시한다.
제제예 1
: 산제의 제조
왕겨초액 0.1 g
유당 1.5 g
탈크 0.5 g
상기의 성분들을 혼합하고 기밀포에 충진하여 산제를 제조하였다.
제제예 2
: 정제의 제조
왕겨초액 0.1 g
락토오스 7.9 g
결정성 셀룰로오스 1.5 g
마그네슘 스테아레이트 0.5 g
상기의 성분들을 혼합한 후 직타법(direct tableting method)으로 정제를 제조하였다.
제제예 3
: 캡슐제의 제조
왕겨초액 0.1 g
옥수수전분 5 g
카르복시 셀룰로오스 4.9 g
상기의 성분들을 혼합하여 분말을 제조한 후, 상기 분말을 통상의 캡슐제의 제조방법에 따라 경질 캡슐에 충전하여 캡슐제를 제조하였다.
제제예 4
: 주사제의 제조
왕겨초액 0.02~0.2 g
주사용 멸균 증류수 적량
pH 조절제 적량
안정화제 적량
통상의 주사제의 제조방법에 따라 1 앰플 당(2㎖) 상기의 성분 함량으로 제조하였다.
제제예 5
: 액제의 제조
왕겨초액 0.1 g
이성화당 10 g
만니톨 5 g
정제수 적량
통상의 액제의 제조방법에 따라 정제수에 각각의 성분을 가하여 용해시키고, 레몬향을 적량 가한 다음 상기의 성분을 혼합하였다. 그 다음 정제수를 가하여 전체 100㎖로 조절한 후 갈색병에 충전하고 멸균시켜 액제를 제조하였다.
본 발명에 따른 왕겨초액은 비만세포주 및 호염구 세포주인 RBL-2H3 세포에서 β-헥소스아미니다제의 분비를 현저히 억제하고, 대식세포주인 RAW 264.7 세포에서 산화질소(NO)의 생성과 ROS의 생성을 현저히 억제하여 대식세포의 활성화를 강하게 억제하며, TPA로 유도된 마우스의 귀 부종을 억제시키고, 염증 관련 유전자인 iNOS, 5-LOX, COX-2를 mRNA 수준 및 단백질 수준에서 모두 억제시키고, 부종이 일어난 마우스의 귀 조직 내에서 염증 관련 사이토카인(TNF-α, IL-1β, IL-6)과 에이코사이드(PGE2, LTB4)의 양을 현저히 감소시키고, 마우스 귀 부종 모델의 귀 조직 내에서 비만세포의 활성화 마커인 트립타제를 발현시키지 않아 비만세포 활성화를 억제하여 염증 억제 효과가 우수하다. 따라서, 본 발명에 따른 왕겨초액은 염증성 질환의 예방 또는 치료에 유용하게 사용될 수 있다.
도 1은 대식세포주인 RAW 264.7 세포에서 왕겨초액에 의한 ROS의 생성 억제 활성을 나타낸 도이다.
도 2는 TPA로 유도된 마우스의 귀 부종에서 왕겨초액에 의한 부종 억제 효과를 나타낸 도이다.
도 3은 왕겨초액에 의한 염증 관련 유전자(iNOS, 5-LOX, COX-2)의 발현 정도를 RT-PCR로 관찰한 도이다.
도 4는 왕겨초액에 의한 염증 관련 유전자(iNOS, 5-LOX, COX-2)의 발현 정도를 웨스턴 블롯으로 관찰한 도이다.
도 5는 왕겨초액이 마우스 귀 조직 내 비만세포 활성화에 미치는 영향을 면역조직화학적검사를 이용하여 비만세포 활성화 마커인 트립타제의 발현으로 관찰한 도이다.
Claims (3)
- 왕겨초액을 유효성분으로 함유하는 환경적 요인에 의한 과민성 염증반응으로 인한 염증성 질환의 예방 또는 치료용 약학 조성물.
- 제 1항에 있어서, 상기 염증성 질환은 감염성 비염, 알레르기성 비염, 만성 비염, 급성 부비동염, 만성 부비동염, 급성 또는 만성 위염, 크론씨 병(Crohn's disease), 특발성 궤양성 대장염, 퇴행성 관절염, 류마티스 관절염 및 패혈증으로 이루어진 군으로부터 선택된 1종 이상인 것을 특징으로 하는, 염증성 질환의 예방 또는 치료용 약학 조성물.
- 제 1항에 있어서, 상기 조성물은 정제, 산제, 캡슐제, 과립, 액제, 현탁액, 유탁액, 또는 주사제로 제형화되는 것을 특징으로 하는, 염증성 질환의 예방 또는 치료용 약학 조성물.
