Figure 2 is a graph and table showing the nematicidal effect of the conventional basic control drug substance.

The present invention relates to nematicidal compositions containing anisomycin, and more particularly to nematicidal compositions containing anisomycin having nematicidal effects on pretty little nematodes, diploscaptors and pine re-nematodes.

The nematode Diploscapter sp.strain LKC25 is known to mediate a variety of pathogenic microorganisms such as Listeria monocytogenes and Salmonella entertica, including Enterocoliform Escherichia coli O157: H7. Due to the lack of awareness of nematodes, research on the development of nematicides for the nematodes has not been conducted.

Meanwhile, pine worm nematodes (Bursaphelenchus. Xylophilus) are plant parasitic nematodes, which are transmitted by the mediator of the horned beetle. It is a plant parasite. In 1971 in Japan and in Korea in 1988, it was first reported at Geumjeongsan, Busan.

For the past 30 years, many researches have been carried out on the pine bark, a medium-borne insect, for the control of pine re-nematode diseases. However, the development of inhibitors directly against pine re-nematodes using biological approaches is very poor. The only way is to inject water into the live pine tree.

Morantel tartrate is currently used as an agent for injecting pine re-nematodes. Morantel tartrate was originally developed as a repellent for livestock animals and was marketed as a preventive drug in 1985. Morantel tartrate (Morantel tartrate) is a drug developed in Japan is effective, but expensive, there is a problem that is difficult to apply in Korea at present. In addition, as a preventive agent, pine trees with pine nematode disease cannot be cured, and there is a problem in that they must be unconditionally cut and treated with fumigation and crushing.

On the other hand, anisomycin (Anisomysin) is a compound (C14H19NO4; molecular weight 265.30) of 2- (para-methoxybenzyl) -3, 4-pyrrolidinediol 3-acetate of the following formula 1, Flazecidin (Flagecidin) It is known under the trade name of and is a mechanism that inhibits the synthesis of proteins and deoxyribonucleic acid (DNA) made from microorganism Streptomyces griseolus or Streptomyces roseochromogenes. It is an antibiotic.

Anisomycin is known to specifically inhibit Mitogen-activated protein (MAP) kinases, known as stress-activated protein kinases, which are activated by intracellular stimulation and stress. It has been shown to inhibit protein synthesis by inhibiting thidyl transferase or the ribosomal system of 80S, suggesting that it may be useful as a clinical and medical antibiotic.

However, the insecticidal effect by anisomycin is not known yet, and in the present invention, the composition containing anisomycin has a particularly strong nematicidal effect against pretty little nematode, diploxcaptor and pine re-nematode. It is expected to be developed as a high efficiency nematicide.

Therefore, it is an object of the present invention to develop an nematode composition containing anisomycin as an effective nematicide against difloscaptor and pine re-nematode.

In order to achieve the above object, the present invention provides a nematicidal composition containing anisomycin of the following structural formula 1.

1Structural Formula 1

Figure 112007044622212-pat00003
Figure 112007044622212-pat00003

The anisomycin nematode composition of the present invention has an nematode effect against a diploscaptor or pine re-nematode.

The nematicidal composition of the present invention may use anisomycin in an amount of 0.0001-2 mM.

In the present invention, in order to determine the insecticidal effect on the diploxcapter and pine re-nematode of the anisomycin substance of the above formula 1,

1. The screening was performed using a high speed screening method,

2. A 50% mortality of anisomycin (IC 50) was compared with the known pine re-nematode drug to verify the nematicide effect of anisomycin.

(Effects of Nematodes on Diplospcaper and Pine Re-nematode by Fast Screening of Anisomycin Substance)

High-speed screening was carried out with anisomycin to investigate the nematode effects on dicapscaper and pine re-nematode.

Anisomycin was dissolved in dimethyl sulfoxide (DMSO) and used in less than 1% of the total amount to exclude the effect on dimethyl sulfoxide (DMSO) when screening. Anisomycin was dissolved in dimethyl sulfoxide (DMSO) to make a stock solution at 100-200 mM, and diluted to 200, 100, 10, 1, 0.1 mM.

In the case of DiploCaptor, nematodes are collected in 50 ml tubes using sterile distilled water when nematodes are collected from the food-rich Enmuem medium. ENGM medium is a medium for growing E. coli, which is a food for Diploscaptor, and is a general medium for growing nematodes for feeding E. coli. Pine re-nematodes were isolated using the Berman funnel separation method and diluted to a concentration of 20.000 birds / ml. Diluted nematodes were dispensed into Eppendorf-tubes or 96 well plates. The total amount was 200 μl, and the isolated nematodes were divided by 100 μl, and the diluted anisomycin was divided into 2 μl of 1% of the total amount at each concentration, and the remainder was quantified with sterile distilled water. Final concentrations of anisomycin were 2, 1, 0.1, 0.01, and 0.001 mM, and 6 hours after the dispensing, the dicloscaper was incubated at 20 ° C. and the pine re-nematode at 25 ° C.

After 6 hours, 20 μl was taken from the sample and the total number of nematodes, live and dead were counted, respectively. And 50% lethality (IC50) was measured. Dead nematodes were considered dead by anisomycin if they did not respond to physical impulses using the platinum wire.

(Compared 50% mortality (IC50) between anisomycin and control)

As a result of the screening of Example 1, it was confirmed that anisomycin was effective, and this experiment was conducted using Morantel tartrate, which is an existing pine nematode disaster prevention drug, as a control.

Experimental methods were compared in the same manner as in Example 1 to obtain a 50% lethality (IC50) value.

The results are shown in FIGS. 1 and 2.

1 is a high-speed screening method of anisomycin was observed 1, 6 hours after the drug mixture nematode effect on the dicapscaper and pine re-nematode. After 6 hours of drug mixing, the pharmacological action was fast enough to derive 50% lethal concentration.

Figure 2 is to examine the nematicidal effect of high-speed screening method Morantel tartrate, a control known as pine re-nematode control drug. Morantel tartrate, which is widely used as a control agent for pine re-nematodes in Japan, has a 50% lethal concentration of 25 mM or more, indicating that the nematicidal effect on pine re-nematodes is not greater than that of anisomycin.

As shown in the experimental results of Figure 1, it was confirmed that the anisomycin has a nematicidal effect on the diplosecaper and pine re-nematode. In addition, as shown in the graph of Figure 2 it can be seen that the anisomycin composition of the present invention shows a very fast medicinal effect such that 50% lethality (IC50) is measured in 6 hours, showing the potential as an nematicide.

In addition, it was found that the anisomycin composition of the present invention exhibits a higher nematicidal effect than Morantel tartrate, which is developed in Japan and widely used as a pine re-nematode control drug, as shown in the experimental results of FIG. 2.

According to the present invention, the nematicidal drug is confirmed by confirming that anisomycin, which is known as a substance that does not adversely affect various ecosystems including the human body, has an nematicidal effect against various harmful nematodes such as difloscaper and pine re-nematode. It can help you develop. In addition, in the case of pine re-nematodes, the basic control agent is effective only as a preventive agent, whereas the anisomycin composition of the present invention has been confirmed to kill pine re-nematodes in a short time, so it is developed not only as a preventive agent but also as a therapeutic agent for pine re- nematode diseases.
