Claims (8)
- - 기판(2)과 기판 상에 고정된 탐침 핵산(1), 상기 탐침 핵산과 혼성화된 타겟 핵산(3) 및 상기 탐침 핵산과 타겟 핵산이 혼성화된 이중나선 핵산(4)에 선택적으로 결합하고 페로센이 결합되어 있는 페로센-결합 인터컬레이터(5)를 포함하는 핵산 칩;- 작업전극(working electrode, 9), 기준전극(reference electrode, 10) 및 대전극(counter electrode, 8)을 포함하는 전기화학 장치;- 상기 전기화학 장치에 인적전압을 가하는 포탠시오스탯(15); 및- 얻어지는 순환 전압-전류 곡선 결과를 제어, 측정하는 컴퓨터(16)를 포함하는, 전기화학적 핵산 혼성화 검출기.
- 제 1 항에 있어서, 상기 인터컬레이터에가 DAPI(4',6-diamidino-2-phenylindole), 노갈라마이신(nogalamycin), hoechest 33258, 디스타마이신 A(distamycin A), EtBr, 독소루비신(doxorubicin) 및 다우노루비신(daunorubicin)으로 구성되는 군 중에서 선택되는 핵산 혼성화 검출기.
- 제 1 항에 있어서, 상기 핵산을 올린 기판과 별도의 작업전극을 사용하며, 기판과 작업전극 사이의 간격이 0.4 내지 2.0 mm로 유지되도록 구성된 핵산 혼성화 검출기.
- 제 3 항에 있어서, 상기 기판이 유리, 실리콘, PCB(printed circuit board) 또는 플라스틱류에 금판을 얇게 코팅한 것인 핵산 혼성화 검출기.
- 제 1 항에 있어서, 상기 핵산을 올린 기판을 작업전극으로 사용하여 전압을 가하는 핵산 혼성화 검출기.
- 제 5 항에 있어서, 상기 기판으로서 금판을 사용하는 핵산 혼성화 검출기.
- - 기판 위에 탐침 핵산을 고정시키고, 여기에 타겟 핵산을 공급하여 혼성화시킨 후, 이중나선 핵산에만 선택적으로 결합하고 페로센과 결합된 페로신 결합 인터컬레이터를 공급하여 상기 혼성화된 이중나선에 선택적으로 결합시키는 단계를 포함하는 핵산 칩 제작 단계:- 작업전극이 상기 인터컬레이터가 결합된 이중나선 핵산을 포함하는 핵산 칩과 일정한 간격을 두고 위치하도록 하여 전기화학 장치를 위치시키고, 상기 전기화학장치를 포탠시오스탯에 연결시키는 단계; 및- 포탠시오스탯을 이용하여 인가전압을 가하고 산화-환원 반응을 유도하여 얻어지는 순환 전압-전류 곡선을 측정하는 단계로 이루어지고, 제 1 항 내지 제 4 항 중 어느 한 항에 따른 핵산 혼성화 장치를 이용하는 핵산 혼성화 검출방법.
- - 기판 위에 탐침 핵산을 고정시키고, 여기에 타겟 핵산을 공급하여 혼성화시킨 후, 이중나선 핵산에만 선택적으로 결합하고 페로센과 결합된 페로신 결합 인터컬레이터를 공급하여 상기 혼성화된 이중나선에 선택적으로 결합시키는 단계를 포함하는 핵산 칩 제작 단계:- 상기 핵산이 고정된 기판을 작업전극으로 하고 기준전극과 대전극을 위치시켜 전기화학 장치를 구성하고, 상기 전기화학장치를 포탠시오스탯에 연결시키는 단계; 및- 포탠시오스탯을 이용하여 인가전압을 가하고 산화-환원 반응을 유도하여 얻어지는 순환 전압-전류 곡선을 측정하는 단계로 이루어지고, 제 5 항 또는 제 6 항에 따른 핵산 혼성화 장치를 이용하는 핵산 혼성화 검출방법.
