前記リポソームは、膜構成成分としてカチオン性脂質を含み、
前記リポソームの表面が温度応答性ポリマーで修飾された、核酸送達用キャリア。
前記核酸送達用キャリアと核酸との複合体をヒトを除く動物に投与する工程を有する、細胞内への核酸送達方法。
本発明の核酸送達用キャリアは、膜融合性能を有するリポソームからなる、細胞内への核酸送達用キャリアであって、リポソームは、膜構成成分としてカチオン性脂質を含み、リポソームの表面が温度応答性ポリマーで修飾されたものである。本発明の核酸送達用キャリアは、かかる構成により、核酸送達後の細胞の死滅の抑制効果に優れ、かつ、核酸の細胞内への送達効率に優れる。
本発明の核酸送達用キットは、上述の核酸送達用キャリアを含むものである。
本発明の核酸送達方法は、上述の核酸送達用キャリアと核酸との複合体と、単離された細胞とを接触させる工程、又は、核酸送達用キャリアと核酸との複合体をヒトを除く動物に投与する工程を有する、細胞内への核酸送達方法である。

式(2)で表されるNIPAAm(N−イソプロピルアクリルアミド)と式(3)で表されるDMAPAAm(メチルアミノプロピルアクリルアミド)とのモル比率が95:5の割合になるようにDMF(ジチルホルムアミド)に溶解させた。なお、NIPAAmとDMAPAAmとの総量は10gとした。溶解後の溶媒に、MPA(3−メルカプトプロピオン酸)をNIPAAmの0.028mol倍となるように加え、更にAIBN(アゾビスイソブチロニトリル)を58mg加えた。その後、溶媒に対して窒素置換と超音波による脱気を行った後、70℃で5時間ラジカル重合を行った。反応後の液体の温度を室温に戻してから、あらかじめ冷やしておいたジエチルエーテルに滴下し、再沈精製を行った。その後、沈殿物を濾過で回収した。この再沈精製を更に2回行い、式(1)で表される温度応答性ポリマーを回収した。この式(1)で表される温度応答性ポリマーについて水により透析を行い、その後、凍結乾燥によって式(1)で表される温度応答性ポリマーを更に精製した。なお、式(1)で表される温度応答性ポリマーの分子量は5500であり、水に対する下限臨界溶解温度は40℃である。この反応スキームを以下に示す。
式(1)で表される温度応答性ポリマーを活性エステル化させるために、モル比が、式(1)で表される温度応答性ポリマー:DCC(N,N’−ジシクロヘキシルカルボジイミド):NHS(N−ヒドロキシスクシンイミド)=1:2.5:2.5の割合となるように塩化メチルに溶解させてから、室温で24時間反応させた。反応後、吸引濾過により副生成物のジシクロヘキシル尿素を取り除き、ジエチルエーテルによる再沈精製を行い、沈殿物として、式(4)で表されるスクシニルポリマーを回収した。式(4)で表されるスクシニルポリマーと膜融合性脂質であるDOPE(1,2−ジオレオイル−sn−グリセロ−3−ホスホエタノールアミン)がモル比1:1でとなるようにジオキサンに溶解させ、室温で24時間反応させた。反応後の溶液をエバポレーターでバキュームし、溶媒を飛ばすことによって、式(5)で表される脂質修飾温度応答性ポリマーを調製した。この反応スキームを以下に示す。
脂質総量が20mgとなり、モル比率が、カチオン性脂質であるDOTAP(1,2−ジオレオイル−3−トリメチルアンモニウムプロパン):DOPE(式(5)で表される脂質修飾温度応答性ポリマーを含む)=3:7の割合となるように、それぞれの脂質をクロロホルムに溶解した。なお、クロロホルムに溶解したDOPEのうち、DOPEと式(5)で表される脂質修飾温度応答性ポリマーとのモル比は、6.5:0.5とした。その溶液を、ナスフラスコに移動させ、エバポレーターで溶媒を飛ばし、リピッドフィルムを作製した。このリピッドフィルムに精製水を1ml加え、リピッドフィルムを水和させた後、ボルテックスで分散させた。その後、超音波槽で30分ソニケーションを行い、リポソームのサイズを小さくしてから、エクストルーダーを用いて、100nmのフィルターを通して、リポソームのサイズを整えた。その後、ゲル濾過によりリポソームを分離させて精製し、目的の、表面が式(1)で表される温度応答性ポリマーにより修飾された、実施例1に係るリポソームを作製した。
インビトロジェン社製のリポフェクタミン(Lipofectamine(登録商標)RNAiMAX)を準備し、これを比較例1に係るリポソームとした。
リポソームを作製する際に、DOPEに式(5)で表される脂質修飾温度応答性ポリマーを含まずに行わなかった点以外は、実施例1と同様の手順によって、比較例2に係るリポソームを作製した。比較例2に係るリポソームは、表面が式(1)で表される温度応答性ポリマーにより修飾されてない点以外は、実施例1に係るリポソームと同様である。
実施例1において、式(1)で表される温度応答性ポリマーの代わりに、PEG(ポリエチレングリコール、分子量:2000)を用いた点以外は、実施例1と同様の手順によって、比較例3に係るリポソームを作製した。
実施例1、比較例1〜3に係るリポソームについて、それぞれ、核酸との複合体の分散液を作製し、この分散液を用いて、核酸のHeLa細胞への送達を行った。核酸としては、ルシフェラーゼ活性を抑制するsiRNA(シグマジェノシス社製のsiRNA duplex)を用いた。以下に、実施例1、比較例1〜3についての、siRNAと複合体との分散液中における、それぞれのリポソームとsiRNAの量を示す。なお、siRNAと複合体との分散液中のsiRNA濃度は、全て終濃度を50nMとした。
実施例1:リポソーム−siRNA複合体/siRNA(N/P(窒素とリンとのモル比))=5:1
比較例1:siRNA:リポソーム=0.1nmol:10μl(インビトロジェン社製のリポフェクタミン(液体に分散した状態で)10μl)
比較例2:リポソーム−siRNA複合体/siRNA(N/P)=5:1
比較例3:リポソーム−siRNA複合体/siRNA(N/P)=5:1
まず、6wellプレートを用いて、それぞれのwellで最終濃度50nMとなるようにsiRNAを(non FBS(非ウシ胎児血清)(血清MEM(最少必須培地)))に混合して、合計125μlとして、5分間静置した。他方、実施例1、比較例1〜3に係るそれぞれのリポソームをnon FBS培地(血清MEM(最少必須培地))125μlに分散させ、5分間静置した。これら2つのnon FBS培地を混合することで、リポソーム−siRNA複合体の分散液を作製し、30分間静置した。その後、分散液をFBS培地で1mlにメスアップした。メスアップの後に、前日に5.0×104cells/mlの濃度で6wellプレートにまいておいたHeLa細胞(子宮頸癌細胞)の培地をリポソーム−siRNA複合体の分散液に変えて、37℃で48時間インキュベートした。インキュベート後、それぞれのwellから培地を取り除き、PBSで5倍希釈した細胞溶解剤(ピッカジーン細胞溶解剤Lcβ(東洋インキ))を250μl/well加えた。この状態で30分インキュベートした後、ピペッティングにより細胞をはがしてから、得られた溶液をマイクロチューブに移した。その後、マイクロチューブを−80℃で凍結させ、細胞を崩壊させた。細胞を崩壊させた後、37℃の浴槽で溶液を融解させ、12000rpm、4℃、5分の条件で遠心を行い、夾雑物を沈殿させた。得られた上澄みのうち90μlを他のマイクロチューブに移し、そこから10μlを96wellプレートに加え、更に50μlピッカジーン溶液を加えて発光測定を行い、HeLa細胞内へのsiRNA送達後のルシフェラーゼ活性を測定した。ルシフェラーゼ活性は、検出された発光量をC(Count per Second)とし、これをタンパク量P(Protein Abundant)で除したものを単位発光量(C/P)として、それぞれの単位発光量について評価を行った。なお、上記の他にsiRNAを細胞内に送達していないHeLa細胞の培地10μlを96wellプレートに加え、BCA(ビシンコニン酸)アッセイキットを使用して、BCA標準品を0〜1mg/mlの範囲の濃度として検量線を作成してからタンパク定量を行い、発光量C及びタンパク量Pから求められた単位発光量(C/P)を100%(control)とし、上記の実施例1、比較例1〜3に係るルシフェラーゼ活性を相対的に評価した。
実施例1に係るリポソームについて、siRNAとの複合体を形成せず、リポソーム単独で細胞内へ送達した点以外は、全て上述と同様の条件で、ルシフェラーゼ活性を測定した。

96wellプレートを用いて、それぞれのwellで最終濃度100nMとなるようにsiRNAを(non FBS(非ウシ胎児血清)(血清MEM(最少必須培地)))に混合して、合計125μlとして、5分間静置した。他方、実施例1、比較例1に係るそれぞれのリポソームをnon FBS培地(血清MEM(最少必須培地))125μlに分散させ、5分間静置した。これら2つのnon FBS培地を混合することで、リポソーム−siRNA複合体の分散液を作製し、30分間静置した。その後、分散液をFBS培地で1mlにメスアップした。メスアップの後に、前日に5.0×104cells/mlの濃度で6wellプレートにまいておいたHeLa細胞(子宮頸癌細胞)の培地をリポソーム−siRNA複合体の分散液に変え、37℃で48時間インキュベートした。インキュベートした後に、それぞれのwellにWST−8溶液(同仁化学製品 Cell Counting Kit−8 − 同仁化学研究所社製)を10μl加えて、更に37℃で30分インキュベートしてから、450nmの吸光度を測定し、細胞生存率を測定した。また、未処理のHeLa細胞について、450nmを測定し、この数値を細胞生存率100%(control)として、上記の実施例1、比較例1に係る細胞生存率を相対的に評価した。
実施例1に係るリポソームについて、siRNAとの複合体を形成せず、リポソーム単独で細胞内へ送達した点以外は、全て上述と同様の条件で、細胞生存率を測定した。

図1に、実施例1、比較例1、対照例1、2についてのルシフェラーゼ活性の測定結果を示す。図2に、実施例1、比較例1〜3、対照例2についてのルシフェラーゼ活性の測定結果を示す。図3に、実施例1、比較例1、対照例3、4についての細胞生存率の測定結果を示す。
実施例1、比較例1、比較例3に係るリポソームについて、それぞれ、核酸との複合体の分散液を作製し、この分散液を用いて、異なる温度条件下で核酸のHeLa細胞への送達を行った。核酸としては、ルシフェラーゼ活性を抑制するsiRNA(シグマジェノシス社製のsiRNA duplex)を用いた。以下に、実施例1、比較例1についての、siRNAと複合体との分散液中における、それぞれのリポソームとsiRNAの量を示す。なお、siRNAと複合体との分散液中のsiRNA濃度は、全て終濃度を50nMとした。
実施例1:リポソーム−siRNA複合体/siRNA(N/P(窒素とリンとのモル比))=5:1

(実施例2)
[温度応答性ポリマーの合成]
式(2)で表されるNIPAAm(N−イソプロピルアクリルアミド)と式(7)で表されるDMAAm(ジメチルアミノアクリルアミド)とのモル比率が69.5:30.5の割合になるようにDMF(ジチルホルムアミド)に溶解させた。なお、NIPAAmとDMAAmとの総量は10gとした。溶解後の溶媒に、MPA(3−メルカプトプロピオン酸)をNIPAAmの0.028mol倍となるように加え、更にAIBN(アゾビスイソブチロニトリル)を58mg加えた。その後、溶媒に対して窒素置換と超音波による脱気を行った後、70℃で5時間ラジカル重合を行った。反応後の液体の温度を室温に戻してから、あらかじめ冷やしておいたジエチルエーテルに滴下し、再沈精製を行った。その後、沈殿物を濾過で回収した。この再沈精製を更に2回行い、式(6)で表される温度応答性ポリマーを回収した。この式(6)で表される温度応答性ポリマーについて水により透析を行い、その後、凍結乾燥によって式(6)で表される温度応答性ポリマーを更に精製した。なお、式(6)で表される温度応答性ポリマーの分子量は5000であり、水に対する下限臨界溶解温度は41.8℃である。この反応スキームを以下に示す。
式(6)で表される温度応答性ポリマーを活性エステル化させるために、モル比が、式(6)で表される温度応答性ポリマー:DCC(N,N’−ジシクロヘキシルカルボジイミド):NHS(N−ヒドロキシスクシンイミド)=1:2.5:2.5の割合となり、かつ、ポリマーと膜融合性脂質であるDOPE(1,2−ジオレオイル−sn−グリセロ−3−ホスホエタノールアミン)とのモル比が1:1となるようにクロロホルムに溶解させ、室温で24時間反応させた。反応後の溶液をメタノール溶液により透析することにより生成物を精製した。さらにエバポレーターでバキュームして溶媒を飛ばすことによって、式(8)で表される脂質修飾温度応答性ポリマーを調製した。この反応スキームを以下に示す。
脂質総量が20mgとなり、モル比率が、カチオン性脂質であるDOTAP(1,2−ジオレオイル−3−トリメチルアンモニウムプロパン):DOPE(式(8)で表される脂質修飾温度応答性ポリマーを含む)=3:7の割合となるように、それぞれの脂質をクロロホルムに溶解させた。なお、クロロホルムに溶解したDOPEのうち、DOPEと式(8)で表される脂質修飾温度応答性ポリマーとのモル比は、6.5:0.5とした。その溶液を、ナスフラスコに移動させ、エバポレーターで溶媒を飛ばし、リピッドフィルムを作製した。このリピッドフィルムに精製水を1ml加え、リピッドフィルムを水和させた後、ボルテックスで分散させた。その後、超音波槽で30分ソニケーションを行い、リポソームのサイズを小さくしてから、エクストルーダーを用いて、100nmのフィルターを通して、リポソームのサイズを整えた。その後、ゲル濾過によりリポソームを分離させて精製し、目的の、表面が式(6)で表される温度応答性ポリマーにより修飾された、実施例2に係るリポソームを作製した。
実施例2に係るリポソームについて、核酸との複合体の分散液を作製し、この分散液を用いて、異なる温度条件下の核酸のHeLa細胞への送達を行った。核酸としては、ルシフェラーゼ活性を抑制するsiRNA(シグマジェノシス社製のsiRNA duplex)を用いた。以下に、実施例2についての、siRNAと複合体との分散液中における、それぞれのリポソームとsiRNAの量を示す。なお、siRNAと複合体との分散液中のsiRNA濃度は、全て終濃度を50nMとした。
実施例2:リポソーム−siRNA複合体/siRNA(N/P(窒素とリンとのモル比))=5:1

リポソームは実施例1に係るリポソームを準備し、リピッドフィルムの水和には精製水の代わりにカルボキシフルオレセイン(CF)2mMを含有したPBSを1ml加えた。準備したリポソームをPBSで20倍希釈し、蛍光光度測定器にてその蛍光強度を測定し、CF封入時蛍光強度とした。また、Triton10Xを1%加え、リポソームを完全崩壊させたのちに測定した蛍光強度を完全放出時の蛍光強度とした。そしてTritonを加えたときの蛍光強度の値から封入時の蛍光強度の値を引くことで、その差を封入量として算出した。Triton処理前と処理後の蛍光強度を図6に示す。図6に示すように、Triton処理後に蛍光強度が増加したことから、カルボキシフルオレセインを封入できたことが確認された。
上記のカルボキシフルオレセインを封入した実施例1に係るリポソーム、及び比較例1に係るリポソーム(リポフェクタミン)に対して、ルシフェラーゼ活性を測定し、siRNAトランスフェクション能を有するかを確認した。その結果を図7に示す。図7に示すように、比較例1と同等にルシフェラーゼ活性が低下しており、同等の効率のトランスフェクション能を有することが確認できた。すなわち、実施例1に係るリポソームは、リポソーム内に低分子物質であるカルボキシフルオレセインを封入することができ、かつそれを利用してsiRNAと複合体を作製することにより、リポフェクタミンと同等な効率のトランスフェクション能を示した。
上記の比較例1に係るリポソーム(インビトロジェン社製のリポフェクタミン(Lipofectamine2000))、比較例3に係るリポソーム(温度応答性ポリマーの代わりにPEGにより修飾)を準備した。また、上記の実施例1に係るリポソームを準備した。実施例1、比較例1及び比較例2に係るリポソームについて、核酸(プラスミド)との複合体の分散液をそれぞれ作製し、核酸(プラスミド)のHeLa細胞内への送達を行った。具体的には、以下のような方法で、核酸との複合体の分散液の作製と、核酸の細胞内への送達を行った。

- 膜融合性能を有するリポソームからなる、細胞内への核酸送達用キャリアであって、
前記リポソームは、膜構成成分としてカチオン性脂質を含み、
前記リポソームの表面が温度応答性ポリマーで修飾された、核酸送達用キャリア。 - 前記温度応答性ポリマーの水に対する下限臨界溶解温度が35.0〜45.0℃である、請求項1に記載の核酸送達用キャリア。
- 前記リポソームは、膜構成成分としてリン脂質を含む、請求項1又は2に記載の核酸送達用キャリア。
- 子宮頸癌細胞内への核酸の送達に用いられる、請求項1から3のいずれかに記載の核酸送達用キャリア。
- 請求項1から4のいずれかに記載の核酸送達用キャリアを含む、細胞内への核酸送達用キット。
- 請求項1から4のいずれかに記載の核酸送達用キャリアと核酸との複合体と、単離された細胞とを接触させる工程、又は、
前記核酸送達用キャリアと核酸との複合体をヒトを除く動物に投与する工程を有する、細胞内への核酸送達方法。
