Abstract
Description
骨髄性腫瘍も、無秩序な異形成血球産生や急性骨髄性白血病への移行を特徴とし、骨髄異形成症候群の関連障害である。
このアミノ酸の置換の検知には、アミノ酸置換に対応する遺伝子の変異を検出してもよいし、それから翻訳されるタンパク質やポリペプチドでの変異を検出してもよい。
なお、アミノ酸の略号は、SはSer(セリン)、FはPhe(フェニルアラニン)、YはTyr(チロシン)、QはGln(グルタミン)、RはArg(アルギニン)、PはPro(プロリン)、EはGlu(グルタミン酸)、XはXaa(未知またはその他のアミノ酸)、HはHis(ヒスチジン)、LはLeu(ロイシン)、RはArg(アルギニン)、KはLys(リシン)、DはAsp(アスパラギン酸)、NはAsn(アスパラギン)、TはThr(トレオニン)を示す。
これは、RNAスプライシング異常が発病の原因となることを、初めて証明できた例でもある。
詳しくは、U2AF35遺伝子から翻訳されるタンパク質の第34番目のアミノ酸におけるSからFまたはYへの置換、U2AF35遺伝子から翻訳されるタンパク質の第157番目のアミノ酸におけるQからRまたはPへの置換、ZRSR2遺伝子から翻訳されるタンパク質のすべての不活化変異、SFRS2遺伝子から翻訳されるタンパク質の第95番目のアミノ酸におけるPからHまたはLまたはRへの置換、SF3B1遺伝子から翻訳されるタンパク質の第700番目のアミノ酸におけるKからEへの置換、第622番目のアミノ酸におけるEからDへの置換、第662番目のアミノ酸におけるHからQまたはDへの置換、第666番目のアミノ酸におけるKからNまたはTまたはEまたはRへの置換である。なお、ZRSR2遺伝子の変異に関しては、実施例では第362番目のアミノ酸におけるEからXへの置換など挙げたが、翻訳されるタンパク質のすべての不活化変異が該当する。
なお、ポリペプチドとは、2以上のアミノ酸がペプチド結合で連結されたものであり、比較的短鎖のペプチドまたはオリゴペプチドと呼ばれるものからタンパク質と呼ばれる長鎖のものまでを含む。ポリペプチドを検出するために抗体を作製する場合に、ポリペプチドには、遺伝的にコードされている20種類のアミノ酸以外のアミノ酸や、修飾されたアミノ酸を含んでもよい。その修飾には、ペプチド結合の主鎖、アミノ酸側鎖、アミノ末端、カルボキシル末端において、アセチル化、アシル化、ADPリボシル化、アミド化、ビオチン化、脂質や脂質誘導体との共有結合、架橋結合の生成、ジスルフィド結合、糖鎖の付加、GPIアンカーの付加、リン酸化及びプレニル化などが挙げられる。
このようなポリペプチドは、化学合成によって直接ペプチドを調製する方法の他に、これらをコードするポリヌクレオチドを部位特異的突然変異誘発法などにより作製し、適当な系で発現させることによって調製することも可能である。
抗体は、ポリペプチドまたはそれと適当なアジュバントとの複合体を免疫原として用いて作製できる。例えばポリクローナル抗体の場合には、免疫原で動物を免役した後、血清から得ることができる。
Q−TOF/MS法などの質量分析によって検出することができる。
顕著なことに、単一の症例で突然変異していた3つの遺伝子(SF3A1、SF3B1、PRPF40B)をさらに含めると、スプライシング機構の6つの構成要素が32例中15例(47%)で相互排他的に突然変異していることが分かった。
RNAスプライシングの初期の段階では、U1 snRNPが、転写されている最中のPre−mRNAの5’SSにリクルートされると同時に、SF1及びU2補助因子(U2AF)の大きい方のサブユニットのU2AF2が、分岐点配列(BPS)及びその下流のポリピリミジン領域に結合する。U2AFの小さい方のサブユニット(U2AF35)は、3’SSのAGジヌクレオチドに結合し、それぞれ、そのUHDドメインとSRドメインを介してU2AF2とSFタンパク質(例えば、SFRS2(sc35))の両方と相互作用して、最初期のスプライシング複合体(E複合体)を構成する。ZRSR2、すなわち、UrpもU2AF及びSRタンパク質と相互作用して、RNAスプライシングに不可欠な役割を果たす。これらの因子によって3’SSが認識された後に、SF3A1とSF3B1を含む多成分タンパク質/snRNA複合体のU2 snRNPが3’SSにリクルートされて、スプライシング複合体Aを生成する。
骨髄異形成では、これらのスプライシング複合体の複数の構成要素が遺伝子突然変異によって影響を受けていて、ヒト癌における経路突然変異の新規かつ顕著な例を示している。
その結果、プールDNAスクリーニングにおける313の暫定的陽性事象を確認することにより、骨髄系腫瘍標本582例中209例で計219個の突然変異が同定された。4つの遺伝子、U2AF35(N=37)、SRSF2(N=56)、ZRSR2(N=23)及びSF3B1(N=79)の突然変異が突然変異の大部分を説明し、SF3A1(N=8)、PRPF40B(N=7)、U2AF65(N=4)及びSF1(N=5)の突然変異率は遙かに小さいものであった。
U2AF35及びSFRS2(場合によってはSF3B1も)の突然変異は、異なるホットスポットに関与しているが、ZRSR2の突然変異はその全長に沿って広範に分布していて、その大部分は、タンパク質の早期切断を引き起こすナンセンス突然変異またはスプライス部位突然変異、すなわち、インデルである。
スプライシング機構の突然変異は、環状鉄芽球の増加を伴うMDS(84.9%)または環状鉄芽球の増加を伴わないMDS(43.9%)、慢性骨髄単球性白血病(CMML)(54.5%)、及び治療関連AMLまたは骨髄異形成関連の変化を伴うAML(25.8%)など、骨髄異形成の特徴を示す疾患に極めて特異的であったが、初発AML(6.6%)及び骨髄増殖性腫瘍(MPN)(9.4%)では稀であった。なお、鉄芽球性不応性貧血(RARS)及び多血球異形成を伴う鉄芽球性不応性貧血(RCMD−RS)は、驚くことにSF3B1の変異が多く認められ、それぞれ82.6%、76%であった。
これらのスプライシング経路の遺伝子における突然変異の相互排他的なパターンが、この多数の症例シリーズで確認され、スプライシング調節における異なる遺伝子の突然変異が共通の結果をもたらすことが示唆された。
そのため、異なる突然変異は、E/Aスプライシング複合体に共通の影響を及ぼすと同時に、細胞機能に異なる影響も及ぼし、個別の疾患表現型の決定に寄与する可能性がある。例えば、SRSF2がDNA安定性の調節にも関与することや、SRSF2の減少がDNA高突然変異性をもたらし得ることが示されている。この関連で興味深いのは、疾患サブタイプを問わず、SRSF2突然変異を有するサンプルが、U2AF35突然変異と比べて、他の遺伝子の突然変異を有意により多く有することが示されたことである(p=0.001、重回帰分析)。
これらのスプライソソーム遺伝子に見られる頻出アミノ酸位置は、これらの突然変異の機能獲得型の性質を強く示唆するが、これは、コドン12、13及び61に生じるRAS突然変異のみならず、V617F JAK2突然変異、V600E BRAF突然変異、そして、ごく最近のY641 EZH2突然変異をはじめとする、他のオンコジーン突然変異でも十分に立証されているシナリオである。
しかしながら、dbSNPデータベース(ビルド131及び132)にも、1000人のゲノムデータベース(2011年5月時点のsnpコール)にも、これらのミスセンスヌクレオチドが含まれていなかったので、全てではないにせよ、これらのミスセンスSNVの多くが、特に男性に見られる機能的な体細胞変異を表す可能性が高いことが示唆される。U2AF35及びSRSF2におけるこれらの突然変異ホットスポットを調べたが、ALL(N=24)または非ホジキンリンパ腫(N=87)などのリンパ系腫瘍では突然変異が見られなかった。
特にRARSではSF3B1遺伝子の変異率が高く、実質的にこの病気を決定づける遺伝子変異になっている。SRSF2遺伝子の変異がある群は、他の遺伝子の変異を伴うことが多いことが統計学的に示された。SRSF2遺伝子変異はまたCMMLに多いことも特徴的である。
現在WHO分類でRCMDと分類される疾患群は、SF3B1、SRSF2遺伝子変異の有無により2つに分けられ、これらの変異のある群の予後は、変異のない群よりも良い可能性があるので、予後の予測に有用である。
これを理解するために、そしてまた、これらのスプライシング突然変異が与える生物学的/生化学的な影響に関する洞察を得るために、EGFPをマーカーとしたレトロウイルスによる遺伝子導入を用いて、野生型U2AF35及び突然変異体(S34F)U2AF35をHeLa細胞で発現させ。
図4(a)は、ウェスタンブロット解析写真である。遺伝子発現解析に用いられたHeLa細胞と、TF−1細胞における導入された野生型U2AF35または突然変異体(S34F)U2AF35の発現を示している。
次の段階で、核からのRNA輸送遺伝子セットからNMDに関与する遺伝子を選択して、さらなるNMD関連遺伝子を構築し、より完全なNMD経路遺伝子セットを構築した。突然変異体U2AF35導入HeLa細胞におけるNMD遺伝子セットのより有意なエンリッチメントを示している。遺伝子セットの有意性は、1,000遺伝子セットの並べ替えにより実験的に決定された。
図4(c)は、qPCRの結果である。NMD遺伝子セットのコアエンリッチメントに寄与する9つの遺伝子の発現のマイクロアレイ結果が確認されている。野生型U2AF35導入HeLa細胞の平均発現(+S.E.)で正規化した後に、模擬導入HeLa細胞、野生型U2AF35導入HeLa細胞及び突然変異体U2AF35導入HeLa細胞におけるNMD遺伝子の平均発現(+S.E.)をプロットした。マン−ホイットニーのU検定でP値を決定した。
突然変異体U2AF35導入TF−1細胞のGSEAの結果によると、U2AF35導入細胞におけるNMD経路遺伝子セットのエンリッチメントを示された。
さらに、有意に示差発現したコアセットプローブは、模擬導入細胞と比べて、野生型U2AF35導入細胞及び突然変異体U2AF35導入細胞において、それぞれ、上方調節及び下方調節される傾向にあり、また逆に、示差発現した非コアセットプローブの場合も同じであった。
すなわち、これらのエキソンアレイの結果から、野生型U2AF35が本物のRNAスプライシングを正しく促進するのに対し、突然変異体U2AF35はこのプロセスを阻害して非コアを生じさせ、そのために、イントロン配列がスプライシングされないまま残る可能性が高いことが示唆された。
突然変異体U2AF35導入細胞における異常なスプライシングは、様々な画分、すなわち、エキソン、イントロン、及び遺伝子間領域、並びにエキソン/イントロン接合部での読取りカウントを評価することによって、より直接的に証明された。まず、マッピングされた読取りの総数で調整した後、野生型U2AF35導入細胞は、突然変異体U2AF35導入細胞と比べて、エキソン画分での読取りの増加を示したが、他の画分でのカウントの低下を示した。突然変異体導入細胞からの読取りは、野生型U2AF35導入細胞からの読取りと比べて、より広範囲のゲノム領域にマッピングされたが、これは、非エキソン読取りによってほぼ説明されるものであった。
野生の遺伝子を導入した場合には、エクソンが多く、イントロンがほぼ無く、変異遺伝子を導入した細胞では、この逆で、イントロンが多く、エクソンがほぼ無いことがわかった。
図5(a)は、ウェスタンブロット解析写真である。誘導72時間後に行なわれたHeLa細胞及びTF−1細胞において、突然変異体(S34F)及び野生型U2AF35のドキシサイクリン誘導性の発現を示している。
突然変異体U2AF35の機能解析を調べたところ、HeLa細胞及びTF−1細胞の細胞増殖アッセイにおいて、U2AF35の発現を誘導した後、突然変異体U2AF35導入細胞では成長が有意に抑制されるが、野生型U2AF35導入細胞では抑制されなかった。
次に、競合的骨髄再構築アッセイで、これらの遺伝子導入細胞の骨髄再構築能を検討した。B6−Ly5.1/5.2 F1マウス由来の全骨髄細胞と混合した後、遺伝子導入細胞を致死量放射線照射B6−Ly5.2レシピエントに移植し、その6週間後に、フローサイトメトリーでGFP陽性細胞由来の末梢血キメリズムを評価した。
エクスビボ追跡調査で遺伝子導入細胞におけるEGFP%と全体的な増殖を評価することにより、各レシピエントマウスが、様々なレトロウイルス群間で同程度の数のGFP陽性細胞を受容したことを確認した。
野生型U2AF35導入細胞は、模擬導入細胞よりもわずかに高い骨髄再構築能を示した。他方、U2AF35突然変異体のどれかを導入した細胞のレシピエントは、模擬導入細胞または野生型U2AF35導入細胞よりも有意に低いGFP+細胞キメリズムを示し、U2AF35突然変異体を発現する造血幹/前駆細胞の骨髄再構築能が障害されていることを示した。野生型U2AF35コンストラクトと突然変異体U2AF35コンストラクトの挙動が反対であることから、これらの突然変異体が、おそらくは野生型タンパク質に対するドミナントネガティブな作用を介して、U2AF35の機能喪失を引き起こすことも示唆された。
RNAスプライシングシステムは後生動物種に固有の特徴である。このシステムは、コード配列を、ゲノムDNA上で分かれて存在する複数の断片として用意し、転写後に、介在する配列を欠失させることにより、これらを再結合させて、機能的なmRNAコピーを生成させる(非特許文献1)。回りくどいまたは無駄の多いやり方のように思われるが、これによって主なタンパク質源が提供され、そのために、遺伝子数が限られているにもかかわらず、選択的スプライシングによって機能的多様性が与えられる(非特許文献5)。
他方、こうした複雑な作業をスプライソソーム機構を用いて精巧に調節することによって、トランスクリプトーム全体の完全性を保証するならば、そのこと自体に当然の代償が必ず伴うことになる。RNAスプライシングの異常、すなわち、癌特異的選択的スプライシングは、その正確なメカニズムは解明されていないものの、骨髄異形成症候群や他の造血系腫瘍を含むヒト癌の発症への関与が示唆される。
骨髄異形成症候群の最も一般的な臨床所見が、無制限の細胞増殖ではなく、アポトーシスの増加を伴う無効造血による複数の細胞系統における重度の血球減少であることは注目に値する。この点について、RPS14のハプロ不全によって、赤血球前駆体のアポトーシスは増加するが、骨髄増殖は増加しないという5q−症候群の発病に関する研究結果が示唆を与え得る。
すなわち、被験物質が、被験者のヒト遺伝子を含む試料を用い、U2AF35遺伝子、ZRSR2遺伝子、SFRS2遺伝子、SF3B1遺伝子の少なくともいずれかの遺伝子から翻訳されるタンパク質の発現を抑制し得るか否か、または、その活性を阻害し得るか否かを評価する工程と、その遺伝子から翻訳されるタンパク質の発現を抑制し得るか、または、その活性を阻害し得る被験物質を、骨髄異形成症候群または骨髄性腫瘍に起因する状態または疾患を予防または治療する有効物質として選択する工程と、を設ける。
すなわち、被験物質が、被験者のヒト遺伝子を含む試料を用い、U2AF35遺伝子、ZRSR2遺伝子、SFRS2遺伝子、SF3B1遺伝子の少なくともいずれかの遺伝子の変異遺伝子から翻訳されるタンパク質の発現を抑制し得るか否か、または、その活性を阻害し得るか否かを評価する工程と、その変異遺伝子から翻訳されるタンパク質の発現を抑制し得るか、または、その活性を阻害し得る被験物質を、骨髄異形成症候群または骨髄性腫瘍に起因する状態または疾患を予防または治療する有効物質として選択する工程と、を設ける。
発現量の測定は、例えば、非ヒト動物から骨髄や血液等の生体試料を採取し、その試料中の転写産物等を測定することにより行ってもよい。
上記遺伝子またはその変異遺伝子から翻訳されるタンパク質の発現レベルを直接的に評価可能な細胞は、その発現細胞であり、発現レベルを間接的に評価可能な細胞は、上記遺伝子またはその変異遺伝子の転写調節領域についてレポーターアッセイを可能とする細胞である。
上記遺伝子またはその変異遺伝子の転写調節領域についてレポーターアッセイを可能とする細胞は、上記遺伝子またはその変異遺伝子の転写調節領域、その領域に機能可能に連結されたレポーター遺伝子を含む細胞である。
Claims (9)
- 骨髄異形成症候群または骨髄性腫瘍を生じ得る素因を有するか否かを評価する方法であって、
被験者のヒト遺伝子を含む試料を用い、U2AF35遺伝子、ZRSR2遺伝子、SFRS2遺伝子、SF3B1遺伝子の少なくともいずれかの遺伝子の変異を検知する工程を有する
ことを特徴とする骨髄異形成症候群または骨髄性腫瘍の素因の評価方法。 - U2AF35遺伝子から翻訳されるタンパク質の第34番目のアミノ酸におけるSからFまたはYへの置換、
U2AF35遺伝子から翻訳されるタンパク質の第157番目のアミノ酸におけるQからRまたはPへの置換、
ZRSR2遺伝子から翻訳されるタンパク質のすべての不活化変異、
