Abstract
Description
(A)カロテノイド類
(B)中鎖飽和脂肪酸トリグリセライド
(C)およびトコトリエノール
を含有することを特徴とするカロテノイド含有油性組成物である。
フコキサンチン濃縮物の調製(1):
特開2004−35528号公報記載の方法に従って培養したオキナワモズク盤状体50kgに150Lの86%エタノール(含水エタノール)を添加し、6時間攪拌抽出を行った。遠心分離により抽出液の固液分離を行い清澄化された抽出液を得た。この抽出液を合成吸着樹脂ダイヤイオンHP−20(三菱化学社製)を充填したカラムにロードし、フコキサンチンを吸着させ、次いで70%エタノール45Lにて洗浄を行った後、90%エタノール120Lにてフコキサンチンを溶出し、フコキサンチン含有画分を得た。このフコキサンチン含有画分を減圧下にて濃縮を行い、フコキサンチン濃縮物を得た。この濃縮物中のフコキサンチン含量を下記条件のHPLCにより定量分析を行ったところ、20%であった。
高速液体クロマトグラフィーLC−20ABシステム(島津製作所社製)を使用した。カラムはCOSMOSIL 5C22−AR−II(ナカライテスク製、内径4.6x250mm)を用い、移動相はアセトニトリル/水の混合液(80:20)を用いた。フコキサンチンの検出はフォトダイオードアレイにより波長450nmの吸収で行い、流速は1.5ml/minで分析を行った。
フコキサンチン含有油性組成物の調製:
製造例1で得られたフコキサンチン濃縮物に、O.D.O(トリ(カプリル/カプリン酸)グリセリル;日清オイリオ社製)およびトコリット92(トコトリエノール70%、トコフェノール30%;エーザイフードケミカル社製)を、フコキサンチンの終濃度が1%、トコリット92の終濃度が2%になるようにそれぞれ添加して混合した。
油性基剤および抗酸化剤を下記表1のように代えた以外は実施例1と同様にして油性組成物を調製した。使用した油性基剤および抗酸化剤は以下のとおりである。
(油性基剤)
O.D.O(トリ(カプリル/カプリン酸)グリセリル;日清オイリオ社製)
キャノーラ油ヘルシーライト(長鎖脂肪酸トリグリセライド;日清オイリオ社製)
オリーブオイル(長鎖脂肪酸トリグリセライド;日清オイリオ社製)
サラダ油(長鎖脂肪酸トリグリセライド;日清オイリオ社製)
(抗酸化剤)
トコリット92(トコトリエノール70%、トコフェロール30%;エーザイフードケミカル社製)
RM−21E(RME;ローズマリー抽出物;三菱化学フーズ社製)
抽出ビタミンE乳液(VE;ミックストコフェロール;三菱化学フーズ社製)
フコキサンチン保存安定性試験:
実施例1および比較例1〜11で得られた油性組成物をそれぞれ25mlバイアルに入れ、40℃のオーブンに保管した。7日後の油性組成物中のフコキサンチン含量を上記条件によるHPLCで測定した。結果を表1に併せて示す。
油性基剤および抗酸化剤を下記表2のように代えた以外は、実施例1と同様にして油性組成物を調製した。油性組成物を25mlバイアルに入れ、50℃のオーブンおよび−80℃の冷凍庫に保管した。0、1、5、11、15および26日後の油性組成物中のフコキサンチン含量を上記条件によるHPLCで測定した。フコキサンチン含量は、−80℃で同日保管したサンプルを100とした相対値として求めた。結果を表3に示す。
フコキサンチン含有油性組成物の調製およびフコキサンチン保存安定性試験:
製造例1で得られたフコキサンチン濃縮物に、スコレー64(トリ(カプリル/カプリン酸)グリセリル;日清オイリオ社製)、トコリット92(トコトリエノール70%、トコフェロール30%;エーザイフードケミカル社製)およびトコフェロール(日清オイリオ社製)を、フコキサンチンの終濃度が1%、トコトリエノールとトコフェロールの合計の終濃度が2%になるようにそれぞれ添加して混合した。トコトリエノールとトコフェロールの配合質量比は、1:0.43〜4.7の範囲で変化させた。
得られた油性組成物を25mlバイアルに入れ、40℃のオーブンおよび−80℃の冷凍庫に保管した。15日後の油性組成物中のフコキサンチン含量を上記条件によるHPLCで測定した。フコキサンチン含量は、−80℃で同日保管したサンプルを100とした相対値として求めた。なお、抗酸化剤を添加しないものをコントロールとした。結果を表4に示す。
フコキサンチン含有油性組成物の調製およびフコキサンチン保存安定性試験:
製造例1で得られたフコキサンチン濃縮物に、スコレー64(トリ(カプリル/カプリン酸)グリセリル;日清オイリオ社製)、トコリット92(トコトリエノール70%、トコフェロール30%;エーザイフードケミカル社製)、トコフェロール(日清オイリオ社製)およびビタミンCパルミテート(三菱フードケミカル社製)を、フコキサンチンの終濃度が1%、トコトリエノール、トコフェロールおよびビタミンCパルミテートの合計の終濃度が1〜8%になるようにそれぞれ添加して混合した。トコトリエノール、トコフェロールおよびビタミンCパルミテートの配合質量比は、1:0.43:0.48〜4.29の範囲で変化させた。
得られた油性組成物を25mlバイアルに入れ、40℃のオーブンおよび−80℃の冷凍庫に保管した。15日後の油性組成物中のフコキサンチン含量を上記条件によるHPLCで測定した。フコキサンチン含量は、−80℃で同日保管したサンプルを100とした相対値として求めた。なお、抗酸化剤を添加しないものをコントロールとした。結果を表5に示す。
フコキサンチン濃縮物の調製(2):
(精製1)特開2004−35528号公報記載の方法に従って培養したオキナワモズク盤状体50kgに150Lの86%エタノール(含水エタノール)を添加し、6時間攪拌抽出を行った。遠心分離により抽出液の固液分離を行い清澄化された抽出液を得た。この抽出液を合成吸着樹脂ダイヤイオンHP−20(三菱化学社製)を充填したカラムにロードし、フコキサンチンを吸着させ、次いで70%エタノール45Lにて洗浄を行った後、90%エタノール120Lにてフコキサンチンを溶出し、フコキサンチン含有画分(1)を得た。このフコキサンチン含有画分(1)を減圧下にて濃縮を行い、フコキサンチン濃縮物(1)を得た。この濃縮物について、フコキサンチン含量を製造例1と同様にしてHPLCにより分析した。また、下記条件によるHPLCにより、クロロフィルa、c1、c2の定量分析を行った。結果を表7に示す。
高速液体クロマトグラフィーLC−20ABシステム(島津製作所社製)を使用した。カラムはIntersil ODS−P 5μm(GLサイエンス製、内径4.6x150mm)を用いた。流速は0.8ml/min.で、分析はグラジエントにて行った。
溶媒A:メタノール(0.25M酢酸アンモニウム含有),アセトニトリル,水=4:3.5:2.5
溶媒B:アセトニトリル,酢酸エチル=3:7
グラジエント分析のタイムプログラムは下記表6の通りである。
フコキサンチン含有油性組成物の調製およびフコキサンチン保存安定性試験(耐光性):
製造例2で得られたフコキサンチン濃縮物(1)および(2)に、スコレー64(トリ(カプリル/カプリン酸)グリセリル;日清オイリオ社製)、トコリット92(トコトリエノール70%、トコフェロール30%;エーザイフードケミカル社製)、を、フコキサンチンの終濃度が1%、トコトリエノールの終濃度が2%になるようにそれぞれ添加して混合した(実施例12および13)。また、フコキサンチン濃縮物に代えて、試薬(和光純薬工業社製、純度94.0%以上)を用いて同様の組成で油性組成物を調製した(実施例14)。
得られた油性組成物を25mlバイアルに入れ、5000Lux、40℃の条件下で保存し、0、4、5、6、7日後における油性組成物中のフコキサンチン含量を上記製造例1の条件によるHPLCで測定した。フコキサンチン含量は、−80℃で同日保管したサンプルを100とした相対値として求めた。結果を表8に示す。
フコキサンチンソフトカプセルの調製:
下記処方のソフトカプセルを調製した。
(処方) (1カプセル400mgあたり)
(1)ゼラチン(皮膜) 150mg
(2)実施例1の油性組成物 250mg
合計 400mg
このソフトカプセルは保存安定性に優れるものである。
フコキサンチン含有飲料の調製:
下記処方の飲料を調製した。
(処方) (100mlあたり)
(1)実施例1の油性組成物 0.5g
(2)レシチン(大豆由来) 0.5g
(3)緑茶抽出物(三共ライフテック) 0.85g
(4)果糖 4g
(5)L-アスコルビン酸 0.05g
(6)香料 0.1g
(7)精製水
合計 100g(精製水で100gとする)
この飲料は保存安定性に優れるものである。
Claims (17)
- 次の成分(A)ないし(C);
(A)カロテノイド類
(B)中鎖飽和脂肪酸トリグリセライド
(C)トコトリエノール
を含有することを特徴とする油性組成物。 - 成分(A)カロテノイド類が、フコキサンチンまたはアスタキサンチンである請求項1記載の油性組成物。
- 成分(A)カロテノイド類が、フコキサンチンである請求項2記載の油性組成物。
- フコキサンチンが褐藻類抽出物に含まれるものである請求項3記載の油性組成物。
- 褐藻類抽出物が、褐藻類の含水エタノール抽出液を合成吸着樹脂で処理して得られる吸着画分である請求項4記載の油性組成物。
- 成分(B)中鎖飽和脂肪酸トリグリセライドが、炭素数8〜10の飽和脂肪酸から構成されるものである請求項1ないし5のいずれかの項記載の油性組成物。
- 成分(B)中鎖飽和脂肪酸トリグリセライドが、トリ(カプリル/カプリン酸)グリセリルである請求項1ないし6のいずれかの項記載の油性組成物。
- 成分(C)トコトリエノールを0.1〜5.0質量%含有する請求項1ないし7のいずれかの項記載の油性組成物。
- さらに次の成分(D);
(D)トコフェロール(D1)および油溶性ビタミンC誘導体(D2)よりなる群から選ばれる1種または2種以上の油溶性抗酸化剤
を含有する請求項1ないし8のいずれかの項記載の油性組成物。 - 成分(C)と成分(D)の配合質量比が、1:50〜50:1である請求項9記載の油性組成物。
- 成分(D)がトコフェロール(D1)であり、成分(C)と成分(D1)の配合質量比が、1:0.2〜8である請求項9記載の油性組成物。
- 成分(D)がトコフェロール(D1)および油溶性ビタミンC誘導体(D2)であり、成分(C)、成分(D1)および成分(D2)の配合質量比が、1:0.2〜8:0.2〜8である請求項9記載の油性組成物。
- 成分(D2)がビタミンCパルミテートである請求項12記載の油性組成物。
- 請求項1ないし13いずれかの項記載の油性組成物を含有する飲食品。
- 請求項1ないし13のいずれかの項記載の油性組成物を含有する医薬品。
- 請求項1ないし13のいずれかの項記載の油性組成物を含有する化粧料。
- 中鎖飽和脂肪酸トリグリセライドを含有する油性基剤中にカロテノイド類およびトコトリエノールを溶解せしめることを特徴とするカロテノイド類の安定化方法。
