Abstract
Description
一方、チタンと生体骨とを結合しやすくする手段として、チタンをアルカリ処理した後、そのまま加熱し、あるいは脱アルカリし次いで加熱することにより、チタン表面にチタン酸ナトリウム層あるいはアナターゼ層を形成する技術が知られている(特許文献2、3)。また、必要により続いて擬似体液に浸けることによりその上にアパタイト層を形成する技術も知られている(特許文献2、3)。
それ故、この発明の課題は、外観的にも性質的にも生体骨に近似していて骨伝導能及び骨誘導能に優れた人工骨、並びにそのための人工骨前駆体を提供することにある。
なお、この明細書において、生体骨が存在する所に人工骨を埋め込んだ場合に人工骨の気孔内に生体骨が進入し人工骨と結合する現象を骨伝導能という。また、筋肉内に人工骨を埋め込んだ場合に人工骨の気孔内に生体骨が形成される現象を骨誘導能という。
先ず、デジタル化された生体骨の三次元画像データにおいて、図1(a)に示すように海綿骨及び/又は皮質骨に相当する部分(図中のハッチング部分)を抽出し、図1(b)に示すように中心線を設定する(抽出工程)。次いで図1(c)に示すように前記中心線に沿って一様な直径又は肉厚の梁又は壁を描くことにより、人工骨画像データを形成する(画像形成工程)。ここで描かれる梁は海綿骨に対応する形状であり、壁は皮質骨に対応する形状である。その後、前記人工骨画像データに基づいて、金属、樹脂及びセラミックのうちから選ばれる1種以上の粉末をレーザー焼結し、焼結層を積み重ねる(造形工程)。
この発明の人工骨の製造方法は、こうして得られた人工骨前駆体を、前記粉末が金属であるときは、アルカリ処理した後、加熱することを特徴とする。これにより表面にアパタイト形成能を有する金属酸塩が形成されるからである。表面にアナターゼ等の金属酸化物層を形成するときは、アルカリ処理した後、加熱前に脱アルカリするかあるいは脱アルカリと同時に加熱するとよい。また、前記粉末がアパタイト、β−TCP、酸化チタンなどのように骨と結合しうるものであるときは、人工骨前駆体がそのまま人工骨となるし、樹脂や他のセラミックであるときは、骨と結合しうる粉末を混ぜてレーザー焼結することにより、人工骨前駆体がそのまま人工骨となる。
ガスアトマイズ法により製造した酸素含有量0.12重量%、最大粒径45μmのチタン粉末を準備した。
並行して三次元マイクロコンピュータトモグラフィ装置を用いて健康なヒトの第4腰椎の断層写真を撮像した。CT装置の撮影条件は、管電圧50kV、管電流40μA、スライス厚83.5μmとした。得られた約500個のファイルからなるDICOM(Digital Imaging and Communications in Medicine)データを三次元画像処理プログラム(ラトック社製TRI/3D−BON)にてSTL(Stereo Lithography Triangulation Language)形式データに変換し、海綿骨及び皮質骨に相当する部分を抽出した。そして、中心線を設定し、その中心線に沿って解像度83.5μm/pixelで0.35mmの直径又は肉厚の梁及び壁を描くことにより、人工骨画像データを作成した。
輪郭部(Contour部):レーザースポット径100μm
積層面(Hatch部):レーザースポット径150μm
装置のプラットホームに粉末を1層分供給し、前記人工骨画像データに基づいてアルゴン雰囲気中でYbファイバーレーザー(λ=1060〜1100nm)を照射した。プラットホームを1層分の厚みだけ下降させ、次の層となる粉末を供給し、同様にレーザーを照射した。このように粉末供給とレーザー照射を必要数だけ繰り返すことにより、人工骨前駆体を製造した。得られた人工骨前駆体のSEM写真を図4に示す。
ヒトの体液とほぼ等しい無機イオン濃度を有する擬似体液(特許文献2、第6欄第43行〜49行)に人工骨を7日間浸けたところ、人工骨の表面にアパタイト相が形成されていた。
実施形態1におけるチタン粉末に代えてチタン合金(Ti6Al4V)粉末を用いた以外は、実施形態1と同一条件で人工骨前駆体を製造した。得られた人工骨前駆体の写真を図5に示す。

実施形態1におけるチタン粉末に代えてチタン合金(Ti15Mo5Zr3Al)粉末を用いた以外は、実施形態1と同一条件で人工骨前駆体を製造した。得られた人工骨前駆体の写真を図6に示す。
Claims (10)
- デジタル化された生体骨の三次元画像データにおいて海綿骨及び/又は皮質骨に相当する部分を抽出し、中心線を設定する抽出工程と、
前記中心線に沿って一様な直径又は肉厚の梁又は壁を描くことにより、人工骨画像データを形成する画像形成工程と、
前記人工骨画像データに基づいて、金属、樹脂及びセラミックのうちから選ばれる1種以上の粉末をレーザー焼結し、焼結層を積み重ねる造形工程と
を備えることを特徴とする人工骨前駆体の製造方法。 - 前記画像形成工程において、梁又は壁を描くとともに所定の中空部を埋めることにより、前記人工骨画像データを形成する請求項1に記載の人工骨前駆体の製造方法。
- 前記粉末が、金属の粉末である請求項1又は2に記載の人工骨前駆体の製造方法。
- 前記金属が、コバルト、タンタル、ジルコニウム、ニオブ及びチタン並びにそれらの合金のうちから選ばれる1種以上である請求項3に記載の人工骨前駆体の製造方法。
- 前記梁の交点の一部又は全部が、更に太く描かれている請求項1〜4のいずれかに記載の人工骨前駆体の製造方法。
- 前記三次元画像データが、生体骨のコンピュータ断層撮影データの群から変換されたSTL形式データである請求項1〜5のいずれかに記載の人工骨前駆体の製造方法。
- 前記造形工程の後、1000℃以上で焼成する焼成工程を更に備える請求項3又は4に記載の人工骨前駆体の製造方法。
- 請求項3又は4のいずれかの製造方法で得られた人工骨前駆体を、アルカリ処理した後、加熱することを特徴とする人工骨の製造方法。
- 前記アルカリ処理後、前記加熱と同時に又は前記加熱の前に、脱アルカリする請求項8に記載の人工骨の製造方法。
- 前記加熱後、擬似体液に浸ける請求項8又は9に記載の人工骨の製造方法。
