Abstract

PURPOSE:To enhance bacterium cell concentration of Escherichia coli and improve the productivity of useful protein, by cultivating a recombinant Escherichia coli in a culture medium containing an iron compound or manganese compound. CONSTITUTION:Escherichia coli retaining an useful protein expressing plasmid and transformed is cultivated in a culture medium obtained by adding 1muM-10mM, preferably 3-100muM bivalent iron compound such as iron (II) chloride or iron (II) sulfate and/or 1muM-10mM, preferably 5muM-100muM bivalent manganese compound such as manganese chloride or manganese sulfate to a culture medium for propagation such as LB culture medium containing carbon source, nitrogen source, inorganic salts, etc. and the culture medium is replaced by a producing culture medium limited in an addition amount of iron or manganese and added by 3XM-1mM, preferably 5-500muM zinc compound to provide also useful protein such as human beta type interferon.
