Abstract
PURPOSE:To enhance bacterium cell concentration of Escherichia coli and improve the productivity of useful protein, by cultivating a recombinant Escherichia coli in a culture medium containing an iron compound or manganese compound. CONSTITUTION:Escherichia coli retaining an useful protein expressing plasmid and transformed is cultivated in a culture medium obtained by adding 1muM-10mM, preferably 3-100muM bivalent iron compound such as iron (II) chloride or iron (II) sulfate and/or 1muM-10mM, preferably 5muM-100muM bivalent manganese compound such as manganese chloride or manganese sulfate to a culture medium for propagation such as LB culture medium containing carbon source, nitrogen source, inorganic salts, etc. and the culture medium is replaced by a producing culture medium limited in an addition amount of iron or manganese and added by 3XM-1mM, preferably 5-500muM zinc compound to provide also useful protein such as human beta type interferon. Cultivation by culture medium for propagation and cultivation for producing culture medium are preferably carried out at 5-8pH and 20-37 deg.C for 1-4 days.
