PURPOSE:To produce a stable aqueous solution of enzyme, by efficiently separating and concentrating alpha-amylase from a low-purity solution containing alpha-amylase or a diluted solution containing alpha-amylase. CONSTITUTION:A solution obtained by removing a solid substance from a liquid culture solution of an alpha-amylase producing microorganisms or a crude alpha- amylase-containing solution such as an extracted solution obtained by extracting a solid culture mixture or a culture cell and removing a solid substance is brought into contact with starch particles at a temperature not to exceed a freezing point or not to cause substantial liquefying reaction of starch, the alpha-amylase is adsorbed on the starch particles and the starch particles having adsorbed the alpha-amylase are separated. The starch particles are washed with water at a temperature not to cause substantial liquefying reaction of starch, an alpha-amylase-containing solution obtained from the separated starch particles is treated with an enzyme containing at least one of glucoamylase and beta-amylase and hydrolyzed to a saccharide having <= molecular weight of oligosaccharide to produce purified alpha-amylase.
