Abstract

PURPOSE: To produce an anthocyan based pigment used for coloring foods, by inducing and cultivating a plant body of Perilla frutescens Britton, var. acuta Kudo and cultivating the obtained calluses capable of producing a red pigment in a culture medium containing an auxinic action substance in a low concentration.
CONSTITUTION: Ripe leaves of Perilla frutescens Britton var. acute Kudo are sterilized and cultivated in a culture medium prepared by adding trace amounts of organic substances, such as vitamins or amino acids, carbon source, plant hormones, etc., to an inorganic synthetic agar culture medium, such as White's culture medium, at 20W30°C, preferably 24W26°C while being irradiated with rays at 600W900nm or 190W500nm wavelength for about 1 month to induce calluses capable of producing a red pigment. The resultant calluses are then cultivated and propagated in a culture medium containing 0.05W0.5ppm auxinic action substance, such as α-NAA, at 20W30°C, preferably 24W26°C for 3W5 weeks. After completing the cultivation, the pigment is extracted with a 0.1% hydrochloric acid-methanol solution and the solvent is removed to afford the aimed red pigment.
