Description
本発明は5−アセチル−2−アルキルベンゼン
スルホンアミドの製造法に関する。
さらに詳しくは4−アルキルアセトフエノンを
スルホン化し、N・N−ジアルキルアニリンと反
応させてアニリン塩となし、これを塩素化して得
られた5−アセチル−2−アルキルベンゼンスル
ホニルクロライドをアンモニアと反応させて5−
アセチル−2−アルキルベンゼンスルホンアミド
を製造する方法に関するものである。
5−アセチル−2−アルキルベンゼンスルホン
アミドは心臓薬として知られるフエニルエタノー
ルアミン誘導体製造の中間原料として有用な化合
物である。
従来5−アセチル−2−アルキルベンゼンスル
ホンアミドを製造する方法は種々知られている。
例えばIndian J.Chem 18B、277〜9(1979)
には4−メチルアセトフエノンをクロロホルム中
でクロルスルホン酸と加熱反応させて5−アセチ
ル−2−メチルベンゼンスルホニルクロライドと
なし、これをアンモニアと反応させて5−アセチ
ル−2−メチルベンゼンスルホンアミドを得る方
法が記載されているが、その収率は30%と低い。
またGer.Offen.2843016(1979)には3−アミノ
−4−メチルアセトフエノンを低温でジアゾ化し
た後、氷酢酸中で塩化銅の存在下に亜硫酸ガスと
反応させて5−アセチル−2−メチルベンゼンス
ルホニルクロライドとなし、これをアンモニアと
反応させて5−アセチル−2−メチルベンゼンス
ルホンアミドを製造する方法が記されているが、
原料が高価な上、ジアゾ化反応は低濃度で行なう
ため容積効率が悪く、かつ全体の工程が長いため
工業的に有利な反応とは云えない。
本発明者らは、このような状況に鑑み工業的に
入手しやすい4−アルキルアセトフエノンを原料
として5−アセチル−2−アルキルベンゼンスル
ホニルクロライドを経て5−アセチル−2−アル
キルベンゼンスルホンアミドを製造する方法につ
いて検討し、4−アルキルアセトフエノンを濃硫
酸中で無水硫酸または発煙硫酸と反応させ、さら
に塩化チオニルと反応させて得られた5−アセチ
ル−2−アルキルベンゼンスルホニルクロライド
をアンモニアと反応させれば工業的に収率よく5
−アセチル−2−アルキルベンゼンスルホンアミ
ドが得られることを知り、さきに「5−アセチル
−2−アルキルベンゼンスルホンアミドの製造法
として特許出願した。〔特願昭56−213889、(特開
昭58−118557)〕
さらに本発明者らは、この製造法について鋭意
検討を加えた結果、4−アルキルアセトフエノン
を濃硫酸中で無水硫酸または発煙硫酸と反応させ
て得た5−アセチル−2−アルキルベンゼンスル
ホン酸または、スルホン酸塩にN・N−ジアルキ
ルアニリンまたは、そのプロトン酸塩を反応さ
せ、5−アセチル−2−アルキルベンゼンスルホ
ン酸N・N−ジアルキルアニリン塩となし、この
塩に塩化チオニルを反応させて得られた5−アセ
チル−2−アルキルベンゼンスルホニルクロライ
ドとアンモニアを反応させると高収率で5−アセ
チル−2−アルキルベンゼンスルホンアミドが得
られることを知り、本発明に到つた。
即ち前記製造法には
Na2SO4が同時に析出するので濾過時の温度
コントロールが困難である。
スルホン酸ソーダが一水塩となるため、乾燥
温度が高く乾燥が因難であるばかりか、乾燥後
粉砕する必要がある。
などの問題がある。これに対し本発明の方法はジ
アルキルアニリン塩とすることにより抽出が可能
となり、前記の様な困難な工程が必要なくスムー
ズに次工程へ移ることが出来る。
本発明の目的は工業的に有利な5−アセチル−
2−アルキルベンゼンスルホンアミドの製造法を
提供するにあり、その要旨は4−アルキルアセト
フエノンを濃硫酸中で無水硫酸または発煙硫酸と
反応させてスルホン化し、得られたスルホン酸ま
たはスルホン酸塩にN・N−ジアルキルアニリン
または、そのプロトン酸塩を反応させ、5−アセ
チル−2−アルキルベンゼンスルホン酸のN・N
−ジアルキルアニリン塩となし、得られたアニリ
ン塩に塩化チオニルを反応させて、5−アセチル
−2−アルキルベンゼンスルホニルクロライドと
なし、これをアンモニアと反応させることを特徴
とする5−アセチル−2−アルキルベンゼンスル
ホンアミドの製造法である。
即ち本発明の特徴は上記反応式のごとく4−ア
ルキルアセトフエノンに硫酸を配位させて硫酸コ
ンプレツクスを生成せしめ、低温でSO3を作用さ
せ選択的に5−アセチル−2−アルキルベンゼン
スルホン酸を合成し、N・N−ジアルキルアニリ
ンを反応させ、5−アセチル−2−アルキルベン
ゼンスルホン酸N・N−ジアルキルアニリン塩と
なし、これに塩化チオニルを作用させて5−アセ
チル−2−アルキルベンゼンスルホニルクロライ
ドとなし、さらにアンモニアと反応させて5−ア
セチル−2−アルキルベンゼンスルホンアミドを
製造する方法である。その作用機構は充分詳かで
はないが、スルホン化反応において4−アルキル
アセトフエノンに硫酸を配位させることにより選
択的に5−アセチル−2−アルキルベンゼンスル
ホン酸を収率よく得ることができる。さらに生成
した5−アセチル−2−アルキルベンゼンスルホ
ン酸をアニリン塩となし、これを塩化チオニルと
反応させることにより高収率で5−アセチル−2
−アルキルベンゼンスルホニルクロライドを得る
ことができる画期的にすぐれた方法である。4−
アルキルアセトフエノンを濃硫酸を使用しないで
無水硫酸または発煙硫酸と反応させるとアセチル
基がスルホン化され5−アセチル−2−メチルベ
ンゼンスルホン酸はほとんど得られない。
本発明のスルホン化に用いる濃硫酸は98%程度
のものを用い、4−アルキルアセトフエノン1モ
ルに対し3〜15モル、望ましくは4〜8モルを混
合する。3モルより少ないと収率が低下し、15モ
ル以上を用いてもさしたる効果がないので不経済
である。
濃硫酸と混合した後に添加する無水硫酸もしく
は発煙硫酸中のSO3の量は4−アルキルアセトフ
エノン1モルに対し、2〜15モル、望ましくは4
〜10モルが適当である。2モル未満では収率が低
く、15モルを超えてもさしたる効果がない。この
際の反応は20℃以下の低温で行なうのがよく、就
中0〜10℃の範囲で行なうと好結果が得られる。
20℃を超えると収率が低下するので好ましくな
く、また0℃以下の低温は工業的実施に困難を伴
う。反応で得られたスルホン酸のままでは分離に
困難であるので、その塩の形で取出すのが望まし
い。経済的にはアルカリ特に必要量の水酸化ナト
リウムを加え、スルホン酸ナトリウムとして取出
すのがよいが、さらに本発明の特徴たるアニリン
塩として取出すのが収率もよく抽出も可能で経済
的にもすぐれている。
この際、アニリン塩を単離することなくそのま
ま塩素化反応に移ることもできるが、工程管理上
これを晶析単離した後塩化チオニルと反応させる
ことが望ましい場合もある。
次に塩素化反応には触媒として例えばジメチル
ホルムアミド,ピリジン,N−メチルピロリド
ン,ジメチルアセトアミド等第3級窒素を有する
塩基を存在させると好結果を得られる場合もある
が、無触媒でも充分反応する。特にN・N−ジメ
チルホルムアミドと塩化チオニルをほぼ等モル予
め混合したものを塩素化剤として使用するとさら
に好結果が得られる。反応は通常有機溶媒中で行
なう。溶媒としては活性水素を有しない有機溶媒
であれば如何なるものでも使用できるが、例えば
テトラヒドロフラン、ジオキサン、ジエチルエー
テル、アニソール、エチレングリコールジメチル
エーテル、エチレングリコールジエチルエーテ

反応に用いる塩化チオニルは5−アセチル−2
−アルキルベンゼンスルホン酸に対し等モル以上
望ましくは1.1〜1.5モルの範囲が好結果を与え
る。また反応温度は−10〜100℃、望ましくは0
〜40℃にて行なうのがよい。高すぎると副反応等
のために収率が低下する場合があるので好ましく
ない。
次にアミド化反応においては前記反応により生
成した5−アセチル−2−アルキルベンゼンスル
ホニルクロライドを単離することなく、そのまま
アンモニアを添加して反応させることができる。
アンモニアは通常アンモニア水の形で用いるのが
よいが、これに限定されない。経済性を考えると
28%安水を用いるのが好ましい。反応温度は一般
に低温がよく、前記スルホン化の場合と同様0〜
20℃の範囲で行なうのが望ましい。20℃を超える
と収率が低下するので好ましくなく、0℃以下に
保つには工業的実施に困難を伴う。
本発明の反応においてスルホン化に際し、一旦
硫酸を配位せしめることにより選択的に3の位置
をスルホン化すること、および生成した5−アセ
チル−2−アルキルベンゼンスルホン酸をアニリ
ン塩とすることに特徴を有し、また塩素化剤とし
て塩化チオニル,アミド化剤としてアンモニアを
用いることにより収率よく、5−アセチル−2−
アルキルベンゼンスルホンアミドを得ることがで
きるのは、これまでに見られないすぐれた方法で
ある。
なお、本発明の方法に適用されるアルキル基と
してはC1〜C5の低級アルキル基が一般に用いら
れるが、その他アルコキシ基、アリール基等の炭
化水素を含む基にも広く利用できる。
また本発明で用いるN・N−ジアルキルアニリ
ンとしては、N・N−ジメチルアニリン、N・N
−ジエチルアニリン、N・N−ジ−n−プロピル
アニリン等が挙げられるが、反応性,経済性等か
ら考えてN・N−ジエチルアニリンを用いると最
も好結果が得られる。工業的にはスルホン化の反
応液を中和してN・N−ジアルキルアニリンのプ
ロトン酸塩、例えばN・N−ジエチルアニリン塩
酸塩と反応させて、アニリン塩として分離するこ
とができる。
N・N−ジアルキルアニリンの使用量は、4−
アルキルアセトフエノンに対し等モル以上であり
さえすればよいが、1.0〜1.5モル倍用いると好結
果が得られる。
以下実施例を示して本発明をさらに詳細に説明
するが、これが本発明を限定するものではない。
実施例 1
98%硫酸250g(2.5モル)中に、4−メチルア
セトフエノン134g(1.0モル)を15℃に保持しな
がら滴下し、30分間同温度に保つて撹拌を続け
た。
次いで5〜10℃に冷却しながら、65%発煙硫酸
861g(SO3として7.0モル)を滴下した後、5℃
に保つて5時間撹拌を続けた。
水2に得られた反応液を添加して希釈した
後、45%水酸化ナトリウム水溶液2187g(24.6モ
ル)を加えて中和した。この中和した反応液に50
%N・N−ジエチルアニリン塩酸塩の水溶液444
g(1.196モル)を加え、さらにクロロホルム
1050gを加えて、40℃に昇温した後分液してクロ
ロホルム相を分離し、脱水した後N・N−ジメチ
ルホルムアミド93.9g(1.286モル)と塩化チオ
ニル153g(1.286モル)の混合物を2〜3℃で添
加し、同温度に保つて1時間撹拌した。次いで20
〜30℃に昇温して1時間撹拌を続けた。この反応
液に水1266gを添加し、分液してクロロホルム相
を取り、さらに水400gで2回水洗した。得られ
た反応液を高速液体クロマトグラフイーで分析し
たところ、5−アセチル−2−メチルベンゼンス
ルホニルクロライド203.1g(0.874モル)を得
た。
その収率は、4−メチルアセトフエノンに対し
ては87.4%であつた。
上記5−アセチル−2−メチルベンゼンスルホ
ニルクロライドの反応液に10℃以下で、アンモニ
ア69g(4.05モル)をガス状で吹き込み、1時間
撹拌した後、水544gを添加して5−アセチル−
2−メチルベンゼンスルホンアミドを晶析させ
た。
これを取、乾燥して5−アセチル−2−メチ
ルベンゼンスルホンアミド176.8g(0.830モル)
を得た。融点は、150.5〜151.5℃であり、収率は
5−アセチル−2−メチルベンゼンスルホニルク
ロライドに対して95%であり、4−メチルアセト
フエノンに対して83.0%であつた。
実施例 2
4−メチルアセトフエノンのかわりに、4−n
−ブチルアセトフエノンを使用した以外は、実施
例1と同様にして、5−アセチル−2−n−ブチ
ルベンゼンスルホニルクロライド230.7g(0.842
モル)を得た。収率は4−n−ブチルアセトフエ
ノンに対して84.2%であつた。
上記5−アセチル−2−n−ブチルベンゼンス
ルホニルクロライドの反応液にジオキサン230g、
28%アンモニア水350g(5.76モル)を添加し、
実施例1と同様にして5−アセチル−2−n−ブ
チルベンゼンスルホンアミド189.2g(0.742モ
ル)を得た。
収率は、5−アセチル−2−n−ブチルベンゼ
ンスルホニルクロライドに対して88.1%であり、
4−n−ブチルアセトフエノンに対して74.2%で
あつた。
実施例 3
N・N−ジエチルアニリンのかわりに、N・N
−ジメチルアニリンを使用した以外は、実施例1
と同様にして5−アセチル−2−メチルベンゼン
スルホンアミド167.3g(0.786モル)を得た。そ

塩化チオニルを反応させて5−アセチル−2−ア
ルキルベンゼンスルホニルクロライドとなし、こ
れをアンモニアと反応させることを特徴とする5
−アセチル−2−アルキルベンゼンスルホンアミ
