Description
本発明は鼓脹症予防剤、又は治療剤に関する。
更に詳しくは、少なくとも糖脂肪酸エステルを含
有して成る鼓脹症予防剤、又は治療剤に関するも
のである。
鼓脹症は、牛や羊の如き反芻動物の第1胃及び
第2胃が、内部に蓄積した発酵ガスによつて過度
に膨張する疾病である。鼓脹症にかかつた反芻動
物は、食欲不振に陥り、増体重や泌乳量の低下を
来し、又、甚だしい時は呼吸困難となつて窒息死
することがある為、肉牛、乳牛、羊等の飼養上、
最も恐ろしい病気の一つである。
鼓脹症の原因については従来から種々の説が唱
えられているが、現在では豆科牧草の多給や濃厚
飼料の多給等が定説となつている。即ち、豆科牧
草を多給した場合には豆科牧草中に含まれている
サポニンが植物蛋白等の起泡性物質の作用によつ
て第1胃内容物が泡沫化し易くなり、又、粗飼料
不足・濃厚飼料多給の場合には第1胃内における
異常発酵、内容液の粘稠性の増大等を起し易くな
る為、いずれの場合も〓気によるガスの排出が妨
害され、第1胃及び第2胃が過度の拡張の起すと
考えられている。
従来、鼓脹症を予防、又は治療する方法として
は、(1)牧草地に大量の油脂を散布する方法、(2)濃
厚飼料に対する粗飼料の併用量を適正に保つ方
法、(3)飲水やミネラルブロツク中にシリコーン、
ポリプロピレングリコール(別名ポリオキシプロ
ピレングリコール)、ポリオキシプロピレンポリ
オキシエチレンブロツクコポリマー等の消泡剤を
常時添加する方法、(4)胃カテーテルや套管針を用
いてガスを排出する治療方法、(5)多量のシリコー
ン、ポリプロピレングリコール、ポリオキシプロ
ピレンポリオキシエチレンブロツクコポリマー、
鉱物油、植物油等の消泡剤を投与する治療方法等
が知られているが、夫々次の様な欠点を有してい
る。即ち、(1)は広大な牧草地に油脂を散布する必
要がある為、手間がかかる上、経済的ではなく、
又、(2)は我国の様な濃厚飼料多給による肉牛の早
期肥育方式等と矛循する一面を有すると共に、完
壁な効果を期待するのは困難であり、又、(4)は専
門家を必要とし、しかも鼓脹症の原因が除去され
ない限り再発を繰り返す可能性が高い。又、(3)や
(5)は比較的良好な効果を発揮することができる
が、現在知られている大部分の消泡剤は長期連続
投与や一時の多量投与によつて消化障害を引き起
すので問題がある。従つて、消泡効果その他の点
でより優れた性能と高い安全性とを兼ね備えた薬
剤の出現が待ち望まれているのが現状である。
本発明者らはかかる現状に鑑み、鋭意検討を重
ねた結果、糖脂肪酸エステルが優れた消泡効果、
第1胃液の粘度低下効果等の諸特性を有し、しか
も高安全性である為、鼓脹症の予防剤、治療剤と
して好適であることを見い出し、本発明を完成さ
せるに至つたものである。
次に本発明を詳細に説明する。
本発明の鼓脹症予防剤、又は治療剤が対象とす
る動物は、肉用牛、乳用牛、子牛、羊、山羊等の
反芻動物である。
又、本発明に使用される糖脂肪酸エステルと
は、アラビノース、キシロース、リボース、リキ
ソース、リブロース、キシルロース、グルコース
(ブドウ糖)、ガラクトース、タロース、マンノー
ス、フルクトース(果糖)、ソルボース、タガト
ース、プシコース、マルトース(麦芽糖)、イソ
マルトース、セロビオース、ゲンチオビオース、
トレハロース、ラクトース(乳糖)、シユクロー
ス(シヨ糖)、マルトトリオース、ゲンチアノー
ス、ラフイノース、スタキオース等によつて代表
される糖類の脂肪酸エステルを指し、1分子中に
エステル結合を1個有するモノエステル、又は2
個以上のエステル結合を有するジエステル、トリ
エステル、テトラエステル等のポリエステル、又
はこれらのモノエステルやポリエステルの混合物
のいずれでも良い。
上記した糖脂肪酸エステルのうち、特にシヨ糖
脂肪酸エステルは工業的に大量に生産されている
為、入手が容易であり、かつ、比較的安価である
ので、本発明に最も有利に使用し得る糖脂肪酸エ
ステルの一つである。
糖脂肪酸エステルを構成する脂肪酸成分として
は、通常、炭素数6〜24程度の脂肪酸が適当であ
る。脂肪酸は飽和、又は不飽和の脂肪酸のいずれ
でも良い。又、脂肪酸の炭素鎖は直鎖型に限定さ
れず、分岐型のものでも良く、更に、水酸基等の
置換基を有する脂肪酸でも差支えない。尚、脂肪
酸は必ずしも一塩基酸に限定されず、二塩基酸等
も用いることができる。
尚、脂肪酸は油脂等の天然物由来の脂肪酸に限
定されず、パラフインの液相接触酸化、α−オレ
フインのカルボニル化(オキソ法)や分岐オレフ
インのカルボキシル化(Koch法)等によつて製
造される合成脂肪酸も用いることができる。
これらの脂肪酸の代表的なものとしてカプロン
酸、エナント酸、カプリル酸、ペラルゴン酸、カ
プリン酸、ラウリン酸、トリデカン酸、2−メチ
ルテトラデカン酸、5−メチルテトラデカン酸、
2,2−ジメチルテトラデカン酸、ミリスチン
酸、パルミチン酸、マルガリン酸、ステアリン
酸、アラキン酸、ベヘン酸、リグノセリン酸、オ
レイン酸、エライジン酸、リノール酸、リノレン
酸、リシノール酸、アラキドン酸、エイコサペン
タエン酸、エルカ酸、アゼライン酸、セバシン
酸、1,20−エイコサメチレンジカルボン酸等を
例示することができる。
尚、上記した糖脂肪酸エステルは必ずしも単品
である必要はなく、糖脂肪酸エステルを構成する
糖成分や脂肪酸成分の異なるもの、エステル結合
数の異なるもの等を二種以上任意の割合で使用す
ることができる。
これらの糖脂肪酸エステルは、糖と脂肪酸メチ
ルエステル、脂肪酸エチルエステルの如き脂肪酸
低級アルキルエステルとを水、又はプロピレング
リコールの如き高安全性の溶媒の存在下にアルコ
ーリシスする方法、糖と油脂、即ち、脂肪酸トリ
グリセリドとを溶媒の不存在下にアルコーリシス
する方法、糖と脂肪酸塩化物、又は脂肪酸無水物
とを反応させる方法等の公知の方法に従つて容易
に製造することが可能であり、いずれの方法によ
つて製造したものでも本発明に使用することがで
きる。
尚、糖脂肪酸エステルは必ずしも高度に精製さ
れたものである必要はなく、例えば、糖脂肪酸エ
ステルを製造する際に粗反応生成物中に一部未反
応のまま残存、又は残存する可能性のある糖、脂
肪酸抵級アルキルエステル、脂肪酸グリセリド
等、反応で副生するアルコール、グリセリン、脂
肪酸塩類等、精製の過程で副生する脂肪酸等の如
く、高安全性のものであれば、含有されていても
特に支障はない。
本発明の鼓脹症予防剤、又は治療剤は粉末状、
粒状、ペレツト状、クランブル状、キユーブ状、
タブレツト状、半湿状、ペースト状、水溶液状、
水性懸濁液状等の任意の形態に調製して用いるこ
とができる。
尚、上記の各種形態に調製する際、水の他、小
麦粉、デンプン、デキストリン等や飼料原料とし
て広く使用されているトウモロコシ、マイロ(コ
ーリヤン)等の穀類、米ぬか、脱脂米ぬか、ふす
ま等の糟糠類、大豆油粕、菜種油粕、綿実油粕、
亜麻仁油粕等の油粕類、牛脂、大豆油、パーム
油、ココナツ油、魚油等の油脂類等を希釈剤とし
て用いても良い。
本発明の鼓脹症の予防剤、又は治療剤を動物に
投与する場合には、上記の各種形態に調製したも
のを飲水や飼料とは別個に給与するか、又は強制
的に第1胃内に注入しても良いが、通常、飲水に
添加するか、又は飼料に添加、配合して給与する
方法が最も便利である。尚、牧草主体の飼養方式
の場合には、本剤を牧草に噴霧する方法を採用す
ることも可能である。
本発明の鼓脹症予防剤、又は治療剤の使用量
は、対象とする動物の種類、日令、体重等や予防
剤として用いる場合と治療剤として用いる場合の
区分、投与方法、鼓脹症の程度、給与飼料の種類
等によつて異なり、一律に規定することはできな
いが、例えば飲水や飼料に添加して投与する場合
には、最終的に動物に給与する飲水や飼料に対し
て糖脂肪酸エステルの純分換算で通常、0.01〜10
重量%、好ましくは0.05〜5重量%、最も好まし
くは0.3〜3重量%程度が適当である。
糖脂肪酸エステルの使用量が、上記した範囲の
下限値未満の場合には本発明の効果を充分に発揮
しにくくなり、又、上限値よりも多量に用いても
特別の効果は発揮されず、寧ろ経済的でなくなる
為、いずれも好ましくない。
尚、予防剤として使用する場合には比較的低濃
度でも連続投与すれば充分に効果を発揮すること
が可能であるが、治療剤として使用する場合には
予防剤の場合よりも高濃度で用いる方が短期間で
本発明の効果を発揮することができる。
尚、本発明の鼓脹症予防剤、又は治療剤は、通
常、単独で使用されるが、無論、既存のシリコー
ン、ポリプロピレングリコール、ポリオキシプロ
ピレンオキシエチレンブロツクコポリマー、油脂
等の消泡剤やその他の薬剤と併用することもでき
る。
本発明の鼓脹症予防剤、又は治療剤は従来から
使用されている薬剤よりも優れた消泡効果を有し
ている為、反芻動物の第1胃液の泡沫化を未然に
防止したり、又、泡沫化した第1胃液を正常な状
態に戻すことが可能である。
又、本発明の鼓脹症予防剤、又は治療剤は第1
胃液の粘度を低下させる作用を有する為、第1胃
内で発生するガスが口から速やかに体外に排出さ
れるのを容易にする効果を発揮する。
しかも、本発明の鼓脹症予防剤、又は治療剤に
使用される糖脂肪酸エステルは、極めて安全性が
高く、かつ、生体内で代謝され易い為、動物に長
期間連続投与したり、又、一時に多量投与しても
安全性の点で全く問題がない。
以上に詳述した如く、本発明の薬剤は優れた消
泡効果、反芻動物の第1胃液の粘度低下効果等の
諸特性と高い安全性とを兼備している為、鼓脹症

特に消泡効果をモデル的に検討する為、0.25重量
%濃度のサポニン水溶液に対して“少なくとも糖
脂肪酸エステルを含有する表−1に記載した各種
の本発明の鼓脹症予防剤、又は治療剤”を所定濃
度(表−1に記載)になる様添加した際の該水溶
液の起泡力を測定した。結果を表−1に示した。
尚起泡力の測定は下記の通りである。
起泡力の測定方法
0.25重量%濃度のサニポン水溶液に本発明の鼓
脹症予防剤、又は治療剤を所定量添加して混合
後、25℃、又は40℃の温度でRoss&Miles法(日
本工業規格JIS K 3362)により起泡後並びに5
分後の泡高(mm)を測定する。
比較例 1〜2
本発明の鼓脹症予防剤、又は治療剤を全く添加
せず、0.25重量%濃度のサポニン水溶液のみの場
合の起泡力を実施例1〜6と同様の方法で測定し
た(測定温度は比較例1が25℃、比較例2は40
℃)。結果を表−1に示した。

体重450Kgのホルスタイン雄去勢牛10頭を用い、
各群5頭づつの2群(試験群、対照群)に分け、
各群に表−2に記載した組成の濃厚飼料を不断給
餌し、4週間の試験を実施した。
その効果、対照群の牛のうち2頭に腹部の過度
の膨張が断続的に認められ、軽度の鼓脹症が発生
したが、試験群の牛には鼓脹症の発生が認められ
なかつた。
尚、試験期間中、2週間及び4週目に全ての牛
の第1胃液を採取し、粘度を測定した効果、試験
群は9.4センチポイズ/25℃、対照群は15.1/25
℃(いずれも2週目と4週目の測定値の平均値)
