に分離されていたものである。
前記式()で示されるML−236B誘導体もし
くはこの閉環ラクトン体はいずれもコレステロー
ル合成阻害作用を有するが、特にM−4のコレス
テロール合成阻害作用は顕著であり、ML−236B
ラクトンの10倍以上の阻害作用を示す。しかしな
がらM−4はML−236Bラクトンを投与した動物
の代謝産物として得られていたものであり、量産
性に乏しく、従つて本物質の経済的な生産方法を
検討していたところ、微生物の作用により、ML
−236B化合物を式()を有するML−236B誘
導体もしくはこの閉環ラクトン体に変換せしめ得
ることを見出し、本発明を完成した。
ML−236BラクトンおよびML−236Bカルボン
酸自体は既知物質であり、青カビの一種ペニシリ
ウム・チトリヌムの代謝産物より分離、精製され
た物質で、次式に示される化学構造を有してお
り、実験動物から分離した酵素系や培養細胞系に
おいてコレステロールの生合成をその律速酵素の
3−ヒドロキシ−3−メチルグルタリル・コエン
ザイムAリダクターゼと競合することにより阻害
し、動物の個体レベルにおいても強力な血清コレ
ステロールの低下作用を示すことが知られている
(特開昭50−155690号、ジヤーナル・オブ・アン

これらの微生物の中、特にムコール・ヒイマリ
ス・ホルマ・ヒイマリス(Mucor hiemalis f.
hiemalis)はML−236B化合物を90%以上の変換
率で式()の化合物に変換する能力を有する。
本発明において好適に用いられる微生物はいず
れも微工研に寄託されているか、もしくは公的な
保存機関(IFO、CBS、IAM、ATCCまたは
NRRL)より入手可能である。
ML−236B化合物を前記式()で示される
ML−236B誘導体もしくは閉環ラクトン体に変換
せしめるには、ML−236B化合物を含む培地で微
生物菌(生菌または休止菌体系)を培養するか、
場合によつてはこれらの微生物の酵素抽出液(無
細胞抽出液)をML−236B化合物と接触せしめる
ことによつても達成される。
この場合、微生物(生菌)培養では培養条件に
よつてカルボン酸型、ラクトン型、アルカリ金属
塩または低級アルキルエステル型として生成す
る。また休止菌体系および酵素抽出液(無細胞抽
出液)ではアルカリ金属塩として得られる。
あるいは、ML−236B化合物から微生物変換に
よつて得られるカルボン酸型をそのまま、または
ラクトン型に変換した後、所望により化学的常
法、例えばジアゾアルカンまたは塩を形成する物
質と処理することにより、これらを低級アルキル
エステル型もしくはアルカリ金属塩として得るこ
ともできる。また、同様に例えばアルカリ金属塩
を所望により化学的常法によつて処理することに
より、低級アルキルエステル型に変換することも
できる。
上記菌株の培養菌体をML−236B化合物に作用
させて、生成するM−4の定量法を以下に示す。
M−4ラクトンの定量法
M−4ラクトンの定量法は、高速液体クロマト
グラフイにより実施した。すなわち、担体とし
て、マイクロボンダ・パツクC18(ウオーターズ
製)を用い、溶媒として62%メタノール水溶液、
液量1ml/min、検出方法としては、紫外部吸収
237nmで検出すると、M−4ラクトンは保持時
間10分にピークが見られ、これにより定量した。
次に実施例を示す。
実施例 1
下記組成の培地100mlを含有する500ml容三角フ
ラスコ20本にムコール・ヒイマリス・ホルマ・ヒ
イマリス IFO 5834を植菌し、26℃、220rpmで
振盪培養し、4日後、ML−236Bラクトンを最終
濃度で0.05%になるように添加して更に6日間26
℃、220rpmで培養する。
培地組成

培養終了後、変換反応液を過し、液をトリ
フルオロ酢酸でPH3に調整した。次いで、1の
酢酸エチルで3回抽出するとM−4を含む区分が
得られる。M−4は薄層クロマトグラフイー
(TLC)(プレート;メルク社製シリカゲル
Art5715溶媒;ベンゼン:アセトン:酢酸=50:
50:3)によりRf値0.45を示す。変換率90%。上
記抽出液を飽和食塩溶液で洗浄し、ジアゾメタン
のエーテル溶液を加え、30分放置後、減圧乾固し
た。残渣をローバー・カラム(メルク社製Si60、
サイズA)にかけ、ベンゼン:酢酸エチル=1:
1の系で精製するとM−4メチルエステルが約
600mg得られた。なお本操作におけるジアゾメタ
ンに代えて、適当なジアゾアルカンを使用すると
該当するM−4のアルキルエステルが得られる。
M−4メチルエステルは次の特性を有する。

IFO 5834を用いて実施例1と同様に操作して、
ML−236Bラクトンの変換反応液1.9を得た
(変換率90%)。変換反応液を過し、液をトリ
フルオロ酢酸でPH3に調整した。次いで、1の
酢酸エチルで3回抽出するとM−4を含む区分が
得られる。上記抽出液を飽和食塩水で洗浄し、次
いで硫酸ナトリウムで脱水後、触媒量のトリフル
オロ酢酸を添加してラクトン化した。次いで上記
抽出液を5%炭酸水素ナトリウム水溶液で洗浄
し、硫酸ナトリウムで脱水後、濃縮乾固してラク
トンを得た。これを酢酸エチルから結晶化して約
560mgのM−4ラクトンを得た。収率56%。
M−4ラクトンは次の物性値を有する。
(1) NMRスペクトル
重クロロホルム中、内部標準にTMSを使用
して、60MHzで測定したNMRスペクトルを第
1図に示す。

酢酸エチルで3回抽出するとM−4を含む区分が
得られる。ただちに、5%NaHCO3水に転溶す
ることによりM−4 Na塩区分を得る。次に2N
−HClでPH7.0に調整し、M−4 Na塩区分を
HP−20樹脂(三菱化成工業(株)社製)に吸着さ
せ、水洗後、50%アセトンでM−4 Na塩を含
有する区分を溶出し、凍結乾燥品570mgを得た。
収率52%。

媒;ベンゼン:アセトン:酢酸=50:50:3)に
より、Rf値0.45を示す。上記抽出液を飽和食塩水
で洗浄し、次いで硫酸ナトリウムで脱水後、触媒
量のトリフルオロ酢酸を添加してラクトン化し
た。次いで上記抽出液を5%炭酸水素ナトリウム
水溶液で洗浄し、硫酸ナトリウムで脱水後、濃縮
乾固してラクトンを得た。残渣をローバー・カラ
ム(メルク社製Si60サイズA)、酢酸エチルの系
で精製し、イソM−4′ラクトン12mgが得られた。
イソM−4′ラクトンは次の特性を有する。

変換反応液を過し液をトリフルオロ酢酸でPH
3に調整した。次いで、10の酢酸エチルで3回
抽出すると、M−4、M−3を含む区分が得られ
る。この中、M−3は薄層クロマトグラフイー
(TLC)(プレート;メルク社製シリカゲル
Art5715、溶媒;ベンゼン:アセトン:酢酸=
50:50:3)によりRf値0.2を示す。
上記抽出液を飽和食塩水で洗浄し、硫酸ナトリ
ウムで脱水後、触媒量のトリフルオロ酢酸を添加
してラクトン化した。次いで上記抽出液を5%炭
酸水素ナトリウム水溶液で洗浄し、硫酸ナトリウ
ムで脱水後、減圧乾固した。残渣をローバー・カ
ラム(メルク社製、RP−8サイズA)を用い、
25%アセトニトリルで溶出し、M−3のラクトン
体として5mgを得た。
M−3ラクトンの特性値
(1) 融点:63〜67℃
(2) 質量分析値(M+):424(C23H36O7)

実施例7に示したのと同様な方法で表3の微生

コレステロール合成阻害作用
前記式()で示される化合物はコレステロー
ル合成経路上の律速酵素として知られる3−ヒド
ロキシ−3−メチルグルタリル・コエンザイムA
リダクターゼ(3−hydroxy−3−methyl−
glutaryl−Co A reductase)を特異的に阻害す
ることが分つた。これら化合物のコレステロール
合成阻害作用〔ジヤーナル・オブ・バイオロジカ
ル・ケミストリー(J.Biol.Chem.)234巻2835頁

酸、その低級アルキルエステルまたはそのアルカ

ン体に変換しうるムコール属、リゾープス属、チ
ゴリンクス属、シルシネラ属、アクチノムコール
属、ゴングロネラ属、フイコマイセス属、モルチ
エレラ属、ピクノポラス属またはリゾクトニア属
に属する微生物を、ML−236B化合物を含有する
培地で培養するか、あるいはこれらの微生物の酵
素抽出液とML−236B化合物とを接触せしめて、
ML−236B化合物を前記式()を有するML−
236B誘導体またはこの閉環ラクトン体に変換せ
しめ変換反応物を含む系より前記式()を有す
るML−236B誘導体またはこの閉環ラクトン体を
採取することを特徴とする前記式()を有する
ML−236B誘導体またはこの閉環ラクトン体の製
