Description
本発明は新規なマイトマイシン類およびその製
造法に関する。
マイトマイシン類は一般的に抗腫瘍活性を有す
る物質として知られ、その代表的な化合物として
マイトマイシンA、B、C、ポルフイロマイシン
(The Merck Index 9th Ed.)などが知られてい
る。
また、本出願人によつてマイトマイシンD(特
願昭53−29238)、マイトマイシンE(特願昭53−
29238)、マイトマイシンF(特願昭53−117836)
に関する発明について出願がなされている。
これらのマイトマイシン類は以下の構造を有す
る。

またこれらの化合物の種々の誘導体が知られて
いる。例えば1a−N−アセチルマイトマイシン
C、1a−N−ブチリルマイトマイシンC(以上、
特公昭40〜18117、米国特許第3514452号)、7−
エチルアミノ−7−デメトキシマイトマイシンA
(特公昭40−20718、米国特許第3514452号)、10−
デカルバモイルマイトマイシンA、BおよびC
(特公昭46−31229、米国特許第3738998号)、1a−
N−アセチル−10−デカルバモイルマイトマイシ
ンC(特公昭49−17279)、1a−N−アセチル−10
−デカルバモイル−10−パラトルエンスルホニル
マイトマイシンC、10−デカルバモイル−10−パ
ラトルエンスルホニルポルフイロマイシン(以
上、特公昭47−37639)などがある。
最近、9、10位に2重結合を有し、すぐれた抗
菌活性を有する、次式に示す一連のマイトマイシ
ン類が見い出された(特願昭53−86748)。
(式中、XBはNH2またはアルコキシを表わし、Y
AはHまたはアルキル基を表わす)。
優れた性質を有する化合物は常に求められてお
り、本発明者らは新規マイトマイシン類の合成に
ついて検討した結果、下記一般式〔〕で表わさ
れるマイトマイシン類〔以下化合物〔〕という
場合がある。他の式番号の化合物についても以下
同様とする。〕が抗菌性を有する新規な化合物で
あることを見い出し、本発明を完成した。
〔式中、Xはアミノ、置換アミノ、ヒドロキシま
たはアルコキシ、Yは水素、−COR1(式中、R1
は水素、アルキル、置換アルキル、アリールまた
は置換アリールを表わす)または−CH2CH2Z
(式中、Zはアルキルオキシカルボニル、アシル
またはシアノを表わす)、RA、RBはRAが水素で
RBが−CH2OHもしくは−CH2OSO2R2(式中、
R2はアルキル、置換アルキルまたはアリールを
表わす)か、RA、RBが一体となつて=CH2を表
わす。ただし、RAが水素でRBが−CH2OHの場
合はYは−CH2CH2Zであるとし、またRAが水素
でRBが−CH2OSO2R2の場合はYは水素または−
CH2CH2Zであるとする。〕。
本発明の目的化合物(化合物〔〕)は広範囲
な抗菌活性を有する。したがつて病院における器
具等の殺菌のために、あるいは洗剤と共に洗浄剤
として、また医薬として有用である。また化合物
〔〕は新たな有用な化合物を作る中間体として
も有用である。
次に本発明をさらに詳しく説明する。
化合物〔〕の基Xにおいて、アルコキシとし
ては炭素数1〜4のアルコキシ、例えばメトキ
シ、エトキシ、i−プロポキシ、n−ブトキシ等
があげられる。基R1においてアルキルとしては
炭素数1〜3のアルキル、例えばメチル、エチ
ル、i−プロピル等、置換アルキルとしては例え
ばクロロメチル、アリールとしては例えばフエニ
ル、置換アリールとしては例えばp−メトキシフ
エニルなどがあげられる。
基Zにおいて、アルキルオキシカルボニルのア
ルキルとしては炭素数1〜4のアルキル、例えば
メチル、エチル、t−ブチル等、アシルとしては
−COR4〔式中、R4は水素または炭素数1〜3の
アルキル、例えばメチル、エチル、イソプロピル
等、を表わす〕があげられる。
基R2としては例えばメチル、トリフルオロメ
チル、p−メチルフエニルなどが適当である。

次に化合物〔〕の製造法について説明する。
化合物〔〕の製造工程図の1例が次に示され
る。
上記工程図において、Xaはアミノ、置換アミ
ノまたはアルコキシを表わす(Xにおいてヒドロ
キシの場合を除いた基である)。Xaaはアミノ、
置換アミノを表わす(aにおいてアルコキシの場
合を除いた基である)。Xabはアルコキシを表わ
す(Xaにおいてアミノ、置換アミノの場合を除
いた基である)。
Z、R1、R2は前記と同義である。
化合物〔−1〕〜〔−7〕および〔−2
−1〕、〔−2−2〕はいずれも化合物〔〕に
包含される。
次に上記各工程にいて説明する。
化合物〔〕→〔〕の工程
化合物〔〕でXa=NH2の場合はマイトマイ
シンC、Xa=OCH3の場合はマイトマイシンAで
ある。化合物〔〕でXaがアルコキシ(OCH3の
場合を除く)の場合の化合物〔〕およびその製
法は特公昭42−22380、米国特許第3558651号、特
公昭54−28250に、Xaが置換アミノの場合の化合
物〔〕およびその製法はJ.Med.Chem.、14、
103(1971)に記載がある。
化合物〔〕も既知化合物があり化合物〔〕
を出発物質として、リチウムアルミニウムハイド
ライド法(特公昭46−31229、米国特許第3738998
号)、アルカリ加水分解法(特公昭47−24558、英
国特許第1250063号)、ナトリウムアルコラート法
〔特公昭47−24560、英国特許第1250063号、J.
Med.Chem.、64109(1971)〕等の方法によつて
得ることができる。
後述の参考例1で示すナトリウムアルコラート
法につい以下に簡単に説明する。
化合物〔〕をメタノール、エタノール、t−
ブチルアルコール等のアルコラートで加水分解す
ることにより化合物〔〕を製造する。溶媒とし
てはメタノール、エタノール、イソプロパノー
ル、テトラヒドロフラン、ジオキサン、ジメチル
ホルムアミド、ベンゼン等を使用する。
化合物〔〕に対するアルコラートの使用量は
5〜7倍molが適当である。
反応は一般に常温で進行し、通常数時間から数
日で終了する。
化合物〔〕→化合物〔〕の工程
化合物〔〕は化合物〔〕と
R1COOH
(式中、R1は前記と同義である)
で表わされるカルボン酸の反応性誘導体、例えば
酸ハロゲン化物、酸無水物等(以下、アシル化剤
という)とを塩基の存在下、不活性溶媒中で反応
させることによつて得られる。この方法はすでに
報告がなされているが(特公昭49−17279号)、以
下簡単に説明する。
アシル化剤としてはアセチルクロライド、プロ
ピオニルクロライド、無水酢酸等が用いられる。
塩基としては炭酸ナトリウム、水素化ナトリウ
ム、トリエチルアミン、ピリジン等が用いられ
る。溶媒としてはテトラヒドロフラン、ジオキサ
ン、クロロホルム等が用いられるが塩基が溶媒を
かねる場合がある。
アシル化剤は化合物〔〕に対し1〜2倍モ
ル、塩基は化合物〔〕に対し1〜100倍モル用
いるのが適当である。
反応温度は−78℃〜30℃が適当である。
化合物〔〕→化合物〔〕の工程
化合物〔〕は化合物〔〕と一般式
R2SO2OH
(式中、R2は前記と同義である)
で表わされる化合物の反応性誘導体(以下、スル
ホニル化剤という)とを、塩基の存在下不活性溶
媒中で反応させることによつて得られる。
スルホニル化剤としては、上記一般式の化合物
のハライド、酸無水物等、例えばP−トルエンス
ルホン酸クロリド、メタンスルホン酸クロリド、
トリフルオロメタンスルホン酸無水物等が用いら
れる。
塩基としては、無機塩基(例えば炭酸ナトリウ
ム、ナトリウムハイドライド等)、有機塩基(例
えば、トリエチルアミン、ピリジン等)が用いら
れる。これらの有機塩基は溶媒をかねることもあ
る。溶媒としてはテトラヒドロフラン、ジオキサ
ン、クロロホルム等が用いられる。
スルホニル化剤は化合物〔〕に対し、1〜2
倍モル、塩基は化合物〔〕に対し1〜100倍モ
ル程度用いる。
反応は−78〜30℃で行うのが適当である。
化合物〔〕→化合物〔−1〕の工程
化合物〔−1〕は化合物〔〕を塩基の存在
下、反応に不活性な溶媒中、脱アルキル(もしく
はアリール)スルホン酸化することにより得られ
る。
塩基としては無機塩基、例えば水酸化ナトリウ
ム、ナトリウムハイドランド等、および有機塩
基、例えばカリウム−t−ブトキシド、1・5−
ジアザビシクロ〔5・4・0〕ウンデセン−5、
トリエチルアミン、ナトリウムメトキシド、特に
ナトリウム−t−ブトキシド、1・5−ジアザビ
シクロ〔5・4・0〕ウンデセン−5等が用いら
れる。
溶媒としてはエーテル類(例えばエチルエーテ
ル、テトラヒドロフラン、ジオキサン、エチレン
グリコールジメチルエーテル等)、炭化水素類
(例えばn−ヘキサン、石油エーテル、ベンゼン
等)、アミド類(N・N−ジメチルホルムアミド
等)、エステル類(酢酸エチル等)、ケトン類(ア
セトン等)、ハロゲン化炭化水素類(塩化メチレ
ン等)、ジメチルスルホキシド等が用いられる。
塩基は化合物〔〕に対して1−20倍モル程度
用いられる。
反応は0〜80℃で行なわれ、用いられる塩基の
種類、反応温度等により異なるが、通常1時間か
ら1週間で終了する。
化合物〔−1〕→化合物〔−2〕の工程
化合物〔−2〕は化合物〔−1〕を塩基の
存在下、水および不活性溶媒中で加水分解するこ
とにより得られる。
塩基としては炭酸水素ナトリウム、アンモニ
ア、ジエチルアミン、ヒドラジン等が用いられ
る。不活性溶媒としては、メタノール、エタノー
ル、テトラヒドロフラン、ジメチルホルムアミ
ド、酢酸エチル、アセトン、クロロホルムなどが
用いられる。
塩基は化合物〔−1〕に対し1〜20倍モル用
いられる。
反応は0〜60℃で行なわれ、通常1〜10時間で
終了する。
化合物〔〕→化合物〔−3〕の工程
化合物〔−3〕は化合物〔〕と一般式
CH2=CHZ
(式中、Zは前記と同義である)
で表わされる化合物(アルキル化剤)とを不活性
溶媒中反応させて、化合物〔〕の1a位をアルキ
ル化することによつて得られる。
反応に活性なアルキル化剤(例えばアクロレイ
ン)を用いる場合は化合物〔〕と混合するだけ
で反応が進行するが、必要に応じて有機塩基(例
えば、トリエチルアミン、ナトリウムメトキサイ
ド、カリウム−t−ブトキサイド等)を用いると
反応が速やかに進行する。
溶媒としてはエーテル類(エチルエーテル、テ
トラヒドロフラン、ジオキサン、エチレングリコ
ールジメチルエーテル等)、炭化水素類(n−ヘ
キサン、石油エーテル、ベンゼン等)、アミド類
(N・N−ジメチルホルムアミド等)、エステル類
(酢酸エチル等)、ケトン類(アセトン等)、ハロ
ゲン化炭化水素類(塩化メチレン、クロロホルム
等)ジメチルスルホキシドなどの有機溶媒が用い
られる。
アルキル化剤は化合物〔〕に対して1〜30倍
モル用いられる。
反応は好ましくは20〜80℃で行なわれ、通常1
時間から1週間で完結する。
化合物〔−3〕→化合物〔−4〕の工程
化合物〔〕→化合物〔〕の工程と同様にし
て目的物を得ることができる。
化合物〔−4〕→化合物〔−5〕の工程
化合物〔〕→化合物〔−1〕の工程と同様
にして目的物を得ることができる。
化合物〔−5〕→化合物〔−2〕の工程
化合物〔−2〕は化合物〔−5〕を酸また
は塩基の存在下、反応に不活性な溶媒中、レトロ
マイケル反応に服せしめることにより得られる。
酸としては無機酸(例えば過塩素酸)、塩基と
しては三級アミン(例えばN・N−ジメチルアニ
リン)が用いられる。
溶媒としてはエーテル類(例えばエチルエーテ
ル、テトラヒドロフラン、エチレングリコール、
ジメチルエーテル)、炭化水素類(n−ヘキサ
ン、石油エーテル、ベンゼン等)、アミド類(NN
−ジメチルホルムアミド等)、エステル類(酢酸
エチル等)、ケトン類(アセトン等)、ハロゲン化
炭化水素類(塩化メチレン、クロロホルム等)、
ジメチルスルホキシドなどが用いられる。
酸または塩基は化合物〔−5〕に対し、1〜
100倍モル程度用いる。
反応は0〜80℃で行うのが好適であり、通常10
分〜数時間で終了する。
化合物〔−4〕→化合物〔−6〕の工程
化合物〔−5〕→化合物〔−2〕の工程と
同様にして目的物を得ることができる。
化合物〔−6〕→化合物〔−2〕の工程
化合物〔〕→化合物〔−1〕の工程と同様
にして目的物を得ることができる。
化合物〔−2−1〕→化合物〔−7〕の工程
化合物〔−7〕は化合物〔−2−1〕を塩
基の水溶液中、加水分解することにより得られ
る。
塩基としては無機塩基(カセイソーダ、炭酸ソ
ーダ等)、有機塩基(トリエチルアミン等)が用
いられる。
塩基の濃度は用いられる塩基によるが、通常
0.01〜1Nで行うのが適当である。
反応は通常室温で行われ、30分〜数時間で完了
する。
化合物〔−7〕→化合物〔−2−2〕の工程
化合物〔−2−2〕は化合物〔−7〕とジ
アゾアルカン(ジアゾメタン等)、アルキルハラ
イド(塩化メチル等)、硫酸ジアルキル(硫酸ジ
メチル等)とを溶媒(酢酸エチル、エチルエーテ
ル等)中で反応させることにより得られる。
反応時に酸が生成する場合、酸結合剤として炭
酸カリウム、トリエチルアミン等を共存させる。
化合物〔−2−2〕→〔−2−1〕の工程
化合物〔−2−1〕は化合物〔−2−2〕
とアンモニアまたはアミンとを不活性溶媒(例え
ばメタノール)中で反応させることにより得られ
る。
アンモニアまたはアミンは化合物〔−2−
2〕に対して1〜100倍モル用いられる。
反応は通常20〜80℃で行われ、1〜数時間で終
了する。
なお、上記各工程において、反応終了後、反応
液から目的物の回収は常法により、例えば、対応
する各実施例および参考例に示した方法により行
われる。
次に実施例および参考例を示す。
実施例 1
(化合物〔〕−〔−1〕)
1a−N−アセチル−10−デカルバモイル−10−
メタンスルホニルマイトマイシンC38mgをエチレ
ングリコールジメチルエーテル8mlに溶かし、
1・5−ジアザビシクロ〔5・4・0〕ウンデセ
ン−5 150mgを加え、窒素気流下2時間加熱還
流する。反応混合物を飽和炭酸水素ナトリウム水
溶液に注ぎ、酢酸エチルで抽出する。抽出液を水
洗後、無水硫酸ナトリウムで乾燥し、溶媒を減圧
留去する。残渣をクロロホルム:アセトン=6:
4(容量比。以下のクロマトグラフイーでも同
様)を展開溶媒に用いるシリカゲルカラムクロマ
トグラフイーにより精製する。濃緑色結晶24mgを
得る。
この結晶物性値は以下に示される。
マイトマイシン:分子イオンピークをm/e315に
与える。1
HNMRスペクトル(pyridine−d5中、δ
ppm):2.03(s、3H)、2.05(s、3H)、3.17

1656(m)(C=C stretch.)を示す。
上記により、この物質は、1a−N−アセチル−
10−デカルバモイルオキシ−9−デヒドロマイト
マイシンCと特定される。収率82%。
実施例 2
(化合物〔−1〕→〔−2〕)
1a−N−アセチル−10−デカルババモイルオキ
シ−9−デヒドロマイトマイシンC57mgをメタノ
ール1mlに溶かし、10%炭酸水素ナトリウムを1
ml加え、室温で6時間撹拌する。反応終了後、反
応液を酢酸エチルで抽出、無水硫酸ナトリウムで
乾燥し、溶媒を減圧留去する。残渣を、クロロホ
ルム:アセトン=1:1を展開溶媒として用いる
シリカゲルカラムクロマトグラフイーにより精製
する。濃緑色の結晶36mgを得る。
この結晶の物性値は以下に示される。
マススペクトル:分子イオンピークをm/e273に
与える。
高分解能マススペクトルの結果は実測値
2731130を示す(計算値2731113)1
HNMRスペクトル(pyridine−d5中、δ
