本発明は制癌効果増強剤に関する。
癌化学療法に関する研究は従来より広範囲な分
野において行なわれているが、核酸代謝制御をね
らいとする癌の化学療法は1940年代後半より始ま
り、核酸代謝抑制剤として先ず6―メルカプトプ
リンが合成され、次いで5―フルオロウラシルが
見い出された。
5―フルオロウラシルは1957年ダシンスキーに
よつて合成され、ハイデルベルガーらによりその
制癌活性が見い出されて以来、広い制癌スペクト
ルをもち、特に腺癌に優れた効果をもつことから
現在臨床で最も広く用いられている制癌剤の1つ
である。5―フルオロウラシルは核酸関連代謝拮
抗物質の代表的な化合物であることから、これを
基本骨格とする化合物の研究開発は現在もなお活
発に行なわれており、最近では2、3の優れた化
合物の報告がなされている。例えば、ソ連で開発
された1―(2―テトラヒドロフリル)―5―フ
ルオロウラシルは、いわゆる5―フルオロウラシ
ルのマスクドタイプ(masked type)であり、
生体内で徐々に5―フルオロウラシルに変換する
ため、5―フルオロウラシルを投与した場合に見
られるような直接的な毒性がほとんどなく、経口
投与が可能な制癌剤として、我国においてその治
療方法が確立された。しかしながら、その反面、
効果は5―フルオロウラシルよりも若干劣るとい
われており、制癌効果と副作用のバランスの上
で、さらに優れた5―フルオロウラシル誘導体の
開発が望まれている。
1―(2―テトラヒドロフリル)―5―フルオ
ロウラシルは前記のように直接的副作用、例えば
経口投与した場合の消化器系への影響が軽減され
た点でその存在意義は大であるが、残された問題
としてはいかにして制癌効果を高めるかにあろう
と思われる。一般に5―フルオロウラシルを基本
骨格とする誘導体は、生体内で5―フルオロウラ
シルに変換することによりその制癌効果が発現さ
れるといわれているが、これが充分な効果を発揮
し得ない現由の1つは変換された5―フルオロウ
ラシルが、さらに分解、不活性化されてしまうた
めにあると考えられる。ちなみに5―フルオロウ
ラシルを静脈内投与した場合の血中半減期は約15
〜20分といわれている。一方下山らによれば5―
フルオロウラシルのような代謝拮抗物質の効果発
現の作用機序である殺細胞様式は、時間依存性で
あり、癌組織の一定レベルの濃度を長時間持続す
るのが望ましいとされている。このような点か
ら、5―フルオロウラシル類の制癌効果を高める
には生体内で変換された5―フルオロウラシルの
分解、不活性化を減ずる必要があり、望ましくは
この現象が正常組織よりも癌組織においてより特
異的なものであれば好ましいと考えられる。以上
の観点より、本発明者は鋭意研究を重ねた結果、
5―フルオロウラシル類にウラシルを配合するこ
とにより所期の目的が達成され得ることを見い出
し先に特許出願した。
本発明者らは前記発明に引続き、5―フルオロ
ウラシル類の制癌効果を高めた制癌剤を得るべく
更に研究を重ねた結果、5―フルオロウラシル類
に対してチミジンが制癌効果増強作用を有するこ
とを見い出し、本発明を完成するに至つた。
即ち本発明は、チミジンを有効成分として含有
し、5―フルオロウラシル類の有する制癌効果を
増強させることを特徴とする制癌効果増強剤に係
る。
本発明で5―フルオロウラシル類としては、一
般式
〔式中R1及びR2は同一又は相異なつて水素原
子、アルコキシメチル基、テトラヒドロフリル基
又はアルキルカルバモイル基を示す〕
で表わされる5―フルオロウラシル類を挙げるこ
とができる。
本発明に依れば、チミジン自体には制癌効果は
ほとんど認められないが、これと5―フルオロウ
ラシル類とを併用することによつて制癌効果が著
しく増大し、治療係数も著しく増大する。
本発明で使用される5―フルオロウラシル類は
上記一般式(1)で表わされる化合物であり、これら
の式で表わされる化合物を広く使用し得る。ここ
で一般式(1)のR1及びR2において、アルコキシメ
チル基のアルコキシ基としては炭素数1〜6のも
のを、またアルキルカルバモイル基のアルキル基
としては炭素数1〜8のものを使用できる。具体
的には、エトキシメチル、ブトキシメチル、ヘキ
シルオキシメチル、エチルカルバモイル、ブチル
カルバモイル、n―オクチルカルバモイル等の基
を挙げることができる。
一般式()で表わされる化合物のうち代表的
なものを以下に掲げる

またこれら一般式で表わされる化合物以外の化
合物でも5―フルオロウラシル誘導体であれば生
体内で5―フルオロウラシルに変換され、チミジ
ンと併用することにより制癌効果が増強されるも
のと考えられる。
一般式()で表わされる化合物はいずれも公
知の化合物であり、これらは公知の方法に従つて
あるいは公知の方法に準じて製造される。例えば
化合物1は特公昭36―3873号に、化合物2は特公
昭49―10510号に、化合物3は特開昭50―50384号
に、化合物4は特開昭50―14836号に、化合物5
は特開昭50―37787号に、化合物6は特開昭52―
51373号に記載されている。
本発明の制癌効果増強剤において5―フルオロ
ウラシル類とチミジンとの使用割合は通常前者の
1モルに対して後者を0.5モル以上使用するのが
良い。該使用割合は5―フルオロウラシル類の種
類に応じて異なり一概には言えないが、一般には
前者1モルに対して後者を100モルまで使用する
こともできる。また前者1モルに対して後者を50
モル以下、更には20モル以下使用すれば良い場合
もある。本発明では5―フルオロウラシル類とチ
ミジンとをそれぞれ別個に投与することもできる
が、両者を予め配合しておきこれらを配合剤とし
て投与するのが好ましい。本発明に係る制癌効果
増強剤又はその配合剤である制癌済の投与単位形
態としては各種の形態を治療目的に応じて選択で
き、例えば錠剤、カプセル剤、顆粒剤等の経口
剤、注射剤、坐剤等の非経口用剤等を挙げること
ができる。斯かる投与単位形態に成形するに際し
ては、担体としてこの分野で従来公知のものが使
用され、この分野で慣用されている手段に従つて
製造される。斯かる種々の投与単位形態中に配合
されるべき5―フルオロウラシル類の量は5―フ
ルオロウラシル類の種類及びチミジンの使用割合
に応じて異なり特に限定することはできないが、
一般に経口剤では10〜200mg、注射剤では50〜
1000mg、坐剤では250〜1000mgが望ましい。また
1日当りの投与量も5―フルオロウラシル類の種
類及びチミジンの使用割合に応じて異なり特に限
定することはできないが、実際の臨床用量及び基
礎的効力実験から推定される臨床用量は1日当
り、5―フルオロウラシル類として一般的には経
口剤では20〜1200mg、注射剤では50〜2000mg、坐
剤では250〜2000mgであるのが望ましい。
次に本発明制癌効果増強剤の配合剤である制癌
剤の代表的な処方例を掲げる。

次に本発明制癌効果増強剤の配合剤である制癌
剤のマウスにおける急性毒性、制癌効果及び治療
係数を示す。
(1) 急性毒性試験
体重22±1gのICR系雄性マウスを1群5匹と
して実験に使用した。使用薬剤は下記第1表に示
す割合で5―フルオロウラシル類とチミジンとを
配合し、5%アラビアゴム溶液にて懸濁し、経口
ゾンデにて強制的に投与した。投与量は1ml/
100gとなるように調製した。試験開始より3週
間にわたつて一般中毒症状、体重および死亡の有
無を連日観察した。LD50値の算出は3週間後に
Up and Down法により行なつた。その結果は第
1表の通りであり、その値は5―フルオロウラシ
ル類の量(mg/Kg)で表わした。
(2) 制癌試験
ザルコーマ180癌細胞2×106個をICR系雄性マ
ウス(1群6匹)の背部皮下に接種した。癌細胞
接種24時間後から下記第1表に示す割合で5―フ
ルオロウラシル類とチミジンとを配合したものを
5%アラビアゴム溶液に懸濁し、1日1回連続7
日間経口投与した。投与量は各薬剤とも4doseを
設け、1ml/100gとなるように調製した。癌細
胞接種後10日目に癌を摘出し、その重量を測定し
て薬剤投与群(T)と対照群(C)との平均癌重量比
(T/C)を求め、投与量と効果(T/C)の用
量―反応曲線から50%癌抑制を示す用量
(ED50)を求めた。その結果は第1表の通りであ
り、その値は5―フルオロウラシル類の量(mg/
Kg)で表わした。
(3) 治療係数
前記試験で得られた薬剤処置群のLD50値と
ED50値の比(LD50/ED50)をもつて治療係数とし
た。結果を第1表に示す。
