Abstract

PURPOSE:To attain to shorten a migration time, by superposing two glass plates having different length dimensions each having a horizontal upper edge having no protruded part so as to interpose spacers to both left and right end parts therebetween to form a superposed glass plate pair and closing both end parts at the length difference part by packings. CONSTITUTION:A glass plate pair 1 for a gel slab is constituted by superposing two rectangular glass plates 2, 3 with different length dimensions having horizontal upper edges 21, 31 each having no protruded part in such a state that a length different part 4 is formed to the upper parts of said glass plates and strip like spacers 5, 6 defining a gel thickness are interposed to both left and right end parts. An electrophoretic gel, for example, a polyacrylamide or agarose gel is grasped in the gel grasping space of the glass plate pair 1 and the both side parts of the length different part 4 are closed by packings 7 and thus formed structure is mounted to an electrophoretic apparatus to make it possible to shorten an electrophoretic time.

[Detailed Description of the Invention] [Industrial Application Field] The present invention uses a vertical slab as a gel holding tool for an electrophoresis device, more specifically, in a vertical electrophoresis device having a buffer tank at the upper and lower portions. The present invention relates to a gel clamping tool for clamping a flat gel.

In general, vertical slab electrophoresis is superior to disk electrophoresis in terms of operability and resolution, and because it allows secondary development, it is suitable for the separation and analysis of biological components such as proteins and nucleic acids. This method has been frequently used recently as a suitable method.

As shown in FIG. 6, the conventional gel holding plate used in such a vertical slab type electrophoresis method consists of two glass plates 2a * 3a 'x spacer 5a that defines the gel thickness.
* 6a were stacked on top of each other with the glass plate 2a interposed therebetween, but both had a so-called concave upper edge 21at in which the entire upper center part of the glass plate 2a on the buffer tank side was cut out and both ends were left to protrude.
It was forming.

However, such conventional gel clamping plates have difficulties in workability due to their special shape of concave upper edges, and
In particular, in order to maintain the strength of the remaining protruding ends 22a, it was necessary to use a thick glass plate, and the glass plate thickness had to be at least 3 mm or more.

Therefore, in the past, it was difficult to dissipate heat even though it is important to suppress or dissipate Joule heat during electrophoresis. Because of the difficulty, it had to be operated at low voltage and low current, and it usually took a long time, 2 to 3 hours when using a gel with a size of 10 cm x 8.6 z x 0.1 m. There were disadvantages such as distortion caused by heat generation and the glass plate being easily damaged during electrophoresis.

Therefore, the inventors of the present invention have made various studies in order to eliminate such conventional drawbacks, and as a result, they have discovered the gel clamping tool of the present invention which can use a glass plate having a thickness of 1 inch or less.

That is, the present invention provides a pair of glass plates in which two glass plates of different lengths each having a horizontal upper edge without a protruding part are stacked with a protruding part formed on the upper part thereof and spacers are entirely interposed on both the left and right sides. This is a gel clamping tool for a vertical slab type electrophoresis device, which comprises: and a backing that closes on both sides of the glass plate-to-length difference portion.

Reference numeral 1 designates a pair of glass plates for gel slabs, each of which has two rectangular glass plates 2 and 3t-1 of different lengths, each having horizontal upper edges 21 and 31 without a protrusion. , are constructed by overlapping each other with band-shaped spacers 5 and 6 interposed on both left and right sides to define the gel thickness. The glass plate pair 1 is preferably held in such a way that it can be disassembled as appropriate using an adhesive tape or the like. 7 is a backing which closes both sides of the glass plate pair protrusion 4. The specific shape of the backing 7 may be any shape as long as it can close both sides of the protrusion 4 and prevent the buffer solution from leaking from both sides, but it may be a short glass plate. Thick protruding backing parts 72 and 73t that close both sides of the protruding part 4 are integrally formed on both ends of the thin band-shaped backing part 71 that comes into contact with the upper part of the protruding part 2. This is advantageous since it is possible to seal the short glass plate 2 and the buffer tank described later. The material for the backing 7 is preferably one that has chemical resistance, heat resistance, and external force resistance, such as rubber,
Examples include plastics and urethanes.

Reference numeral 8 denotes a vertical slab type electrophoresis apparatus to which the gel clamping device of the present invention is applied, and the upper buffer tank 12 and the lower buffer tank 13 are disposed on a support stand 9 via a standing plate 10 on all the stairs. It is structured as follows.

Incidentally, a backing mounting groove 11 is formed in the upper part of the vertical plate 10 along the frontage of the upper buffer solution tank 12, and terminals 14 and 15 are formed in the upper and lower buffer solution tanks 12 and 13, respectively. And platinum wires 16 and 17 connected thereto are attached.

Since the present invention is constructed as described above, an electrophoresis gel such as polyacrylamide, 5DS-polyacrylamide, agarose, etc. can be prepared and held in the gel holding space of the pair of glass plates 1 according to a conventional method. At the same time, by closing both sides of the protrusion 4 with a backing 7 and mounting it on the electrophoresis apparatus 8 as described above, electrophoresis can be performed and analyzed using a conventional method.

In particular, since the present invention is constructed of a glass plate that does not have both ends of the remaining protrusion as in the prior art, even a thin glass plate has a sufficient strength-retaining effect, and the protruding portion 4 The presence of the backing 7 that closes both sides of the buffer completely prevents the buffer solution from leaking.

When a 5O8-polyacrylamide gel of 1 x 0.1 tan is used, the electrophoresis time can be shortened to 30 to 40 minutes, and the resolution can also be improved.
