Description

【発明の詳細な説明】 (発明の技術分野) 本発明は、エンドトキシンの毒性を減刑させる中和剤に
関する。
(従来技術とその問題点) エンドトキシンは、@乳動物の血中に入ると発熱を起す
物質であり、その量が多いときには致死作用すら示ず。
このエンドトキシンが血中に入る機会は、肝W:4瘍、
胆のう炎などの重度の細菌感染症、胃穿孔や外科手術後
の感染症のときであるが。
そのほか最近では人工透析や輸液の際にも、器具等のエ
ンドトキシン汚染によって起りうる。ポリミキシンはエ
ンドトキシンの中和作用を有する抗生物質であるが、血
中に入ると腎毒性が強いのでその使用は経口投与あるい
は局所投与に限られ。
その使用mにも制限がある。
〈発明の目的) ポリミキシンのエンドトキシン中和作用を生かしつつ、
副作用を減刑すること。
(本発明の構成) 本発明は、ポリミキシンを繊維状担体に固定化してなる
エンドトキシン中和剤を12供づるものである。
本発明でいうポリミキシンとは、 3 aci l l
uspolymaxにJ:り産生される抗性物質であり
、ポリミキシンA、ポリミキシンB1.ポリミキシンB
2.ポリミキシンD1.ポリミキシン「1およびポリミ
キシン上2笠の、グラム陰性菌に対づる抗菌作用を有づ
−るものを意味づる。
本発明で君う繊維状担体とは、0.1〜100m2 /
(Iの表面積を有するIli維状成型物であり。
使用条件下において、化学的J3よび物理的に安定であ
るとともに、ポリミキシンを化学的に固定しうる官能基
を有するものを意味ブる。イの一例をあげると、α−ハ
ロゲン化アシル基、ハロゲン化ベンジル基またはイソシ
アン酸基を有するポリスチレン繊維または架橋ポリスチ
レン繊維、アクリル酸・アクリロニ1〜リル共重合体繊
維、カルボキシル阜をイボするポリビニルアルコール繊
維等がある。本発明中和剤に固定化されているポリミキ
シンの吊は、少なすぎるとエンドトキシンの中和ができ
な(なるので、10mg/(J担体以上、より好ましく
は50mg/q担体以上であることが必要である。
本発明中和剤の調製は、繊維状担体とポリミキシンを混
合することにJζす、あるいは、さらにベプヂド縮合剤
(ジシクロへキシルカルボジイミド。
1−エチル−3−(3−ジメチルアミノプロピル)−カ
ルボジイミドなど)を添加づ−ることにより容易に達成
できる。
本発明中和剤の用途は、輸液および透析液中のエンドト
キシン除去のほか、エンド1ヘキシン血症忠者の治療な
どがあり、その使用方法の代表例として、該中和剤を充
填したカラムに輸液、透析液または血液を循環させる方
法がある。
(発明の作用機構) 本発明の土ンドl〜キシン中和剤の作用賎(1へは明確
ではないが、エンド1−キシンの吸着作用よりも分解作
用に基づくと考えられる。
(発明の効果) 本発明のエンドトキシン中和剤は、ポリミキシンを高い
密度で固定化しているため、溶液中のLンドトキシンを
分解・無毒化りるとともに、ポリミキシンが溶液中に溶
出しないため、該中和剤で処理された溶液が毒17Iを
小さ41いことにfj徴を有する。
以下に実施例を示す。
実施例1゜ ポリプロピレン(三井゛ノーブレン“”J311G)5
0部を島成分とし、ポリスチレン(パスタイ[]ン”6
66)46部、ポリプ[1ピレン(イ]友“′ノーブレ
ン”WF−727−F)4部の)昆合物を尚成分とする
海鳥型複合繊維(島数16.中糸繊度2.6デニール、
引張強度2 、9 g/d 、 (11[印50%、フ
ィラメント数42)50gを、N−メチロール−α−ク
ロルアセトアミド50(]、ニトロベンゼン400(1
,98%硫酸400(IおよびパラホルムアルデヒドO
,,85gからなる温合溶液中に浸し、20℃で1時間
反応させた。繊維を反応液から取り出し、0℃の氷水5
u中に投じて反応停止させたのら、水で洗浄し1次に、
繊維にイ」着している二[・ロベンUンをメタノールで
抽出除去した。この繊維を50℃で真空乾燥して、クロ
ルアレ1−アミドメチル化繊維(繊組A>71gを得た
ポリミキシンBフフイザー(含糖ファイIf−a式会社
>4.8g (50万中位のもの80本)を4.00m
1の水にとかし、この中に8JJ ill△11gを加
え、5時間振どう後、酸化マグネシウム1.5gを添加
し、室温で60時間振とうした。
つぎに、この反応母液からとり出し、クロマトカラムに
つめて、0.1N−塩酸4800m1で洗浄した。この
洗浄液と反応母液を混合した溶液について、ミクロビユ
レット法で溶液中のポリミキシン量を算出した。一方2
反応した謀層1はさらに水25αおよび生理食塩水6α
て洗って、ポリミキシンB固定化繊維(繊維B)を15
1だ。固定化mは残存法から110mg/gであった。
実施例2 E 5cberichia Co11 O111: [
3/Iのリボ多糖体(ディフコ・ラボラ[−リーズ社製
、)]−ノール抽出法)の0.5 mg/mlの水溶液
20m1に繊維B2(+を加え、37℃で60分振と°
)後、上澄を1 ml リ”つ、BΔL、 +3 / 
Cマウスに尾静脈から投与して、48時間後の死亡率を
調べたところ。
13匹中4匹(31%)が死亡した。織組]3の代りに
繊維Aを用いて同様な実験を行ったところ。
マウス11匹中11匹(100%)が死亡1ノだ。
この実験からポリミニ1;シン固定化繊維がリボ多糖体
を111(毒化していることがわかる。
実施例3 E 5cl)ericia Co11 O26: 13
6 (デイフニ1φラボラトリーズ社製、フェノール抽
出法)の025 mg /1の水溶液に繊維B2(]を
加え、37°Cで60分間振どう後、上澄を1 111
1ずつ、’BALB/Cマウスに尾静脈から投与して、
48時間後の死亡率を調へたどころ、8匹中1四(12
,5%)が死亡した。対照として、 18雑Bで処理し
なかった水溶液について同様の投与を行ったところ。
13匹中12匹(92%)が死亡した。この実験からポ
リミギシン固定化繊維がリボ多糖体を無毒化しているこ
とがわかる。

繊IB2gを加え、37℃で60分間振とうしたのち、

ずつ投与して、48時間後の死亡率を調べたところ、0
.5mg/+++lでは15匹中15匹。
0、25 mg/mlでは13匹中1匹、0.125m
g/mlでは13匹中O匹の死亡率であった。
