PURPOSE: To increase the production of an objective substance remarkably in the cultivation of lymphoidoblast cells capable of continuously producing the objective substance, by extracting the filtrate of the cultivation medium during cultivation, and continuing the cultivation while replenishing the medium with fresh one.
CONSTITUTION: A cultivation tank (pref. spin filter cultivation tank) is charged with a medium for the cultivation of mammalian cells (e.g. Eagle's MEM medium containing about 0.1W15v/v% blood serum of bovine fetus). Cells (capable of continuously producing interferon, e.g. human lymphoidoblast cells transformed with Epstein-Barr Virus) are added to the medium in concentration of about 105W107 cells/ml, and subjected to the aerobic cultivation. The cultivation is continued by extracting a half to the whole quantity of the cultivation medium about every 0.5W2 days, and adding the same amount of fresh medium.
