Abstract

PURPOSE:To obtain a novel plasmid usable as a vector, by separating a plasmid from Streptococcus faecalis, introducing (transducing) the separated plasmid into Bacillus substilis, and multiplying the Bacillus subtilis. CONSTITUTION:Streptococcus faecalis (FERM-P No.5772) is cultivated by the conventional method, and the collected microbial cells are subjected to the lysis with a lysozyme and then separated by the density gradient centrifugation method of CsCl to give the aimed plasmid (PTP62) having a molecular weight of 9.1X10<6> daltons and properties shown in the table. The resultant plasmid is then introduced (transduced) into a microorganism, e.g. a strain of Bacillus subtilis 168, requiring adenine, and deficient in a restriction enzyme and modifying enzyme, and the Bacillus subtilis is then cultivated to separate the plasmid PTP62 from the multiplied microorganisms in the same way as in the separation from the Streptococcus faecalis and introduction (transduction) into the Bacillus subtilis 168.
