Abstract
PURPOSE: To obtain an immunoglobulin for intravenous injection useful for the infectious prevention and treatment of an antibody-deficient patient in high purity and yield, by treating a blood plasma or plasmatic fraction obtained by the Cohn method with an acid, and fractionating the resultant plasma or plasmatic fraction with an alkylene oxide polymer or copolymer.
CONSTITUTION: A blood plasma or fraction (I+II+III), fraction (II+III), fraction (II) or fraction (III) obtained by subjecting the blood plasma to the cold alcoholic fractionation by the Cohn method is treated with an acid, e.g. hydrochloric or acetic acid, under the conditions of pH 3.2W5.0 and temperature of 4W15°C for 30W180min to dissociate the polymeric immunoglobulin to a nonpolymeric type. The resultant treated blood plasma or fraction is then fractionated with an alkylene oxide polymer (1W4C alkylene oxide polymer) or a copolymer, e.g. Pluronic, having a molecular weight of 2,000W20,000 at a pH of 4W5.5 in a polymeric concentration of 3W 5W/V% to remove the polymeric immunoglobulin. A polymer is then added to the fraction free from the polymeric immunoglobulin at a pH of 6.0W 8.5 in a polymeric concentration of 6W15W/V% to give the aimed immunoglobulin containing no polymeric type.
