Abstract

PURPOSE: To purify acidic carboxypeptidase, in an industrial scale, by passing acidic carboxypeptidase prepared by the cultivation of microorganisms through a column packed with carboxy-sephadex.
CONSTITUTION: A bacterial strain capable of producing acidic carboxypeptidase and belonging to Penicillium genus, e.g. Penicillium caseicolum, is cultured, and the filtrate of the cultured product is passed through a column packed with carboxy-sephadex at an ionic strength of ≤0.2M and a pH of 3.0W4.0 to effect the adsorption of the acidic carboxypeptidase from the filtrate to the sephadex. The column is washed, and the adsorbed carboxypeptidase is eluted at an ionic strength of ≥0.5M and a pH of 3.0W4.0.
