Abstract

PURPOSE:To hemolyze many blood samples at the same time, by arranging a filter layer absorbed with hemolysis agent to a porous material, in adjacency to separation medium layer used for chromatography and so on. CONSTITUTION:A porous material made of vinyl acetate resin is dipped in 10mul/ ml aqueous surfactant solution, then, dried at 50 deg.C for 3hr in an air bath. This filter layer is pushed into a plastic column horizontally, which is filled with weak acidic ion-exchange resin as a separation medium for measurement of glycosylhemoglobin, and added 200mul buffer solution for elution of glycosylhemoglobin. When 10mul whole blood sample measured by a trippet is put on the filter layer, hemocyte is hemolyzed, and the hemolyzed sample is entered into the separation medium layer to measure the amount of glycosylhemoglobin, followed by taking off a stopper at the bottom of the column. Since the whole blood sample can be used as it is without adjusting the hemolyzed sample as mentioned above, this method is preferable to analyze many samples.
