Abstract

PURPOSE: An enzyme mix extracted from readily available human placentas is combined with coenzymes, enzyme activity stabilizer and a nonionic surfactant to form an enzyme solution, which is freeze dried to produce a human standard enzyme sample with high stability.
CONSTITUTION: The washings of human placentas with physiological salt solution is centrigfuged to collect erythrocytes. The resultant erthrocytes are combined with the same volume of distilled water to form an erythrocyte suspension. A 3:1 mixture of ethanol and chloroform is added to the suspension at a ratio of 0.44 part to 1 part of the erythrocyte suspension to extract almost all the enzymes included in etythrocytes such as catalase. Then, the extract is combined with a solution containing NADH2 (reduction type of β-nicotinic amide adenine-2-nucleotide), an enzyme activity stabilizer such as polyvinyl pyrrolidone and a nonionic surfactant such as Triton X-100, e.g., a phosphoric buffer solution and they are thoroughly mixed and freeze dried.
