Abstract

PURPOSE: To remove impurities having an anticomplementary action and obtain γ-globulin suitable for intravenous injection, by suspending a raw material in an aqueous solution of a saccharide or sugaralcohol, adjusting the pH, adding dextran to the suspension, removing the formed precipitate, and precipitating γ-globulin with ammonium sulfate.
CONSTITUTION: A monosaccharide, e.g. glucose, disaccharide, e.g. fructose, or sugaralcohol, e.g. mannitol, is suspended in a 3W15% aqueous solution, and the pH is adjusted to 7.0W9.0. Dextran having an average molecular weight of 10,000W 70,000 is then added to the suspension to give the dextran concentration of 2W 10w/v%. The precipitate thus formed is fractionated and removed, and ammonium sulfate is added to the suspension at 0W10°C to give the ammonium sulfate concentration of 15W35w/v%. Thus, the purified γ-globulin is precipitated. Preferably, the raw material for the γ-globulin is suspended in an aqueous solution in a volume of 25W60ml/g of the protein content.
