Abstract
PURPOSE: Substance intracellularly and extracellularly protecting cells from freeze damage are added to a hepatocyte suspension and the suspension is frozen by gradual cooling, whereby higher than 80% viability of hepatocytes are ensured.
CONSTITUTION: To a suspension of hepatocytes are added at least one of substances intracellularly protecing from freeze damage selected from glycerol and dimethyl sulfoxide and at least one of substances extracellularly protecting from freeze damage selected from dextrin and albumin wherein the concentration of the intracellularly protecting substance is adjusted to 20w/v% or lower as glycerol or to 15w/v% or lower as dimethyl sulfoxide and in the range from 2W20w/v% as the total, while the extracellularly protecting substance is adjusted to 20w/v% or lower and the cooling rate is controlled to 0.1W3°C/min.
