Abstract
PURPOSE:To enable sample slices to be cut with the temperature thereof maintained accurately, by a method wherein a metal foil plate supporting sample slices fitted in slits in instantaneously pressure-bonded by two metal blocks cooled to ultralow temperature to form a glasslike frozen layer having no formation of cryohydrate on the surface. CONSTITUTION:Living body slices 1 are fitted in a plurality of slits in a metal foil plate 3 of excellent heat conductivity. After metal blocks 7 and 8 have been cooled in a cooling chamber 4, an electromagnet 10 is operated to raise the metal block 8, and a supporting base 12 is fitted. The metal foil plate 3 is placed on a supporting member 20, which is attached to the lower end of a cylinder 6 by using a rubber band 21, and a lid-shaped body 5 is lowered to cover the cooling chamber 4. A rod-shaped weight 11 is inserted into the cylinder 6, and the metal block 8 and the metal foil plate 3 are made to drop. Consequently, the slices 1 are pressure-bonded from both sides by means of the cooled metal blocks 7 and 8 and instantaneously frozen to form a glasslike frozen layer 2 having no formation of cryohydrate. The slices 1 are made to bite into the metal foil plate 3 and firmly fixed, each being formed into an I-shape. Accordingly, controlling the temperature of the metal foil plate 3 permits that of the slices to be controlled.
