Abstract

PURPOSE: To enable the test over a wide range of pH by carrying out the red blood cell agglutination (HA) or anti-agglutination (HI) for the arbovirus by means of the fixed bird red blood cell.
CONSTITUTION: The bird red blood cell (the chicken of within 48 hours after hatching and with no food given is used preferably) is processed via the fixing agent such as formalin, glutamyl aldehyde or the like. The above mentioned fixed red blood cell and the arbovirus special HA antigen are diluted by the buffer solution of pH6W12 to secure the HA reaction. At the same time, the above mentioned fixed red blood cell, the arbovirus special HA antigen and the blood serum to be tested are diluted by the buffer solution of pH6.6W12 to secure the HI reaction. In this case, the phosphorus buffer brine, HEPES buffer solution and the like are used for the buffer solution, and about 0.1W1% of the blood serum albumin and about 0.05W0.00025% of gelatin are added preferably.
