Abstract

PURPOSE: To prepare a monoglyceride-lipase having strong decomposition activity against triglycerides and diglycerides as well as monoglycerides, by culturing a specific strain of bacteria belonging to Staphylococcus genus.
CONSTITUTION: The objective monoglyceride-lipase can be prepared by culturing Staphylococcus aureus 226 (FERM-P No.1212). The lipase has the following characteristics: (a) substrate specificity, effectively decomposes tributyrin, monocaprin and monoolein; (b) optimum pH, alkaline when an α-monoolein is used as the substrate; (c) optimum temperature, 50°C for α-monoolein; (d) thermal degeneration, rapidly deactivated abouve 60°C; (e) isoelectric point, pH 2.55; (f) molecular weight, about 174,000 (by gel filtration); and (g) elemental analysis, c about 41.3%, H about 5.7%, N about 8.6%, etc.
