Abstract

PURPOSE: To obtain the blood platelet counting suspension by a simple operation in a short time, by adding to blood a specific erythro-clastic liquid and a specific erythrocyte membrane-dissolving liquid.
CONSTITUTION: An erythro-clastic liquid consisting of alcohol, ethylenediamine tetraacetate and distilled water is added to blood to be examined, to destroy the erythrocytes and prepare a blood solution. An erythrocyte membrane-dissolving diluting liquid consisting of an aqueous solution which contains dodecyl sulfate and alkali chloride and which is isotonic with blood is added to the resulting blood solution to reduce the amount of residual membrane of the erythrocytes after they have been destroyed, and thereby form a suspension of blood platelets. The blood plate- lets are then counted with a particle counter by using the mentioned suspension as a blood platelet counting diluting liquid.
